This study explores the interaction between the production of indole-3-acetic acid (IAA), a typical phytohormone auxin and the role of IAA biosynthetic pathways in each IAA producing rhizobacterial strain. The bacterial strains were isolated from rhizosphere of wild plants and identified as Acinetobacter guillouiae SW5, Bacillus thuringiensis SW17, Rhodococcus equi SW9, and Lysinibacillus fusiformis SW13. A. guillouiae SW5 exhibited the highest production of IAA using tryptophan-dependent pathways among the 4 strains. When indole-3-acetamide (IAM) was added, Rhodococcus equi SW9 showed the highest IAA production of $3824{\mu}g/mg$ protein using amidase activity. A. guillouiae SW5 also showed the highest production of IAA using two pathways with indole-3-acetonitrile (IAN), and its nitrile hydratase activity might be higher than nitrilase. B. thuringiensis SW17 showed the lowest IAA production, and most of IAA might be produced by the amidase activity, although the nitrilase activity was the highest among 4 strains. The roles of nitrile converting enzymes were relatively similar in IAA synthesis by Lysinibacillus fusiformis SW13. Tryptophan-independent pathway of IAA production was utilized by only A. guillouiae SW5.
Effect of quercetin, a kind of natural plant flavonoids, on auxin-induced ethylene production in barley coleoptiles was studied. Auxin-induced ethylene production was apparently stimulated by quercetin. This stimulatory effect of quercetin appeared after 4 h of incubation period. Ethylene production was stimulated 200% over the control after 8 h of incubation by $3{\times}10^{-5}\;M$ quercetin. The quercetin effect was most prominent at $10^{-4}\;M$ of IAA. Ethylene production induced by the synthetic auxin, 2,4-D and NAA, was not significantly affected by quercetin. Also ACC-based ethylene production was unaffected by the flavonoid. In an effort to elucidate mechanisms of quercetin action on auxin-induced ethylene production, the effect of quercetin on 1M metabolism was studied. Data obtained from these experiments indicate that quercetin treatment resulted in about 90% inhibition of IAA oxidase activity. IAA ($3{\times}10^{-5}\;M$) conjugation was found to be not affected by quercetin. This results suggest that the stimulatory effect of quercetin on auxin-induced ethylene production may be due to the fact that quercetin inhibits 1M oxidase activity, thus increasing the free IAA level.
This study was conducted to examine the effect of cytokinin ($N^6$-benzyladenine; BA) and/or an IAA on ethylene production of maize (Zea mays) primary roots. When the two hormones were applied exogenously, both hormones synergistically increased ethylene production, which was greater than the sum of the level of each hormone's effect. For example, the ethylene production was stimulated between about 87% and 170% of the control by $10^{-4}\;M$ BA with $10^{-4}\;M$ IAA for 8 hours respectively, whereas the ethylene production was increased by about 480% of the control when the two hormones were treated simultaneously. Such a synergistic effect was also found in changes in the activity and gene expression level of ACC synthase. However, in the case of ACC oxidase did not show any observable effects. Based on our results, it is possible to conclude that BA and IAA stimulated the ethylene production synergistically by affecting the ACC synthase in maize roots.
This study was conducted to evaluate the indole acetic acid (IAA) formation in soils as a biological indicator for the health of rice paddy soils with control, nitrogen sole, chemical fertilizer (NPK), and chemical fertilizer plus compost (CNPK) plots. There was a positive relationship between colorimetric method and high performance liquid chromatography for IAA in soils determined, and the values were similar between two methods, as $0.83{\sim}1.23{\mu}g\;IAA\;g^{-1}h^{-1}$ in colorimetric method, $0.80{\sim}1.29{\mu}g\;IAA\;g^{-1}h^{-1}$ in HPLC method. Numbers of dehydrogenase-producing bacteria and the IAA production in soils were high in NPK and CNPK plots comparing with control and nitrogen sole plots. Also there was high correlation between numbers of dehydrogenase-producing bacteria and IAA production in soils.
A total of 35 phosphate solubilizing bacterial strains were isolated from waste mushroom bed of Agaricus bisporus in Buyeo-Gun, Chungnam and screened for the production of indole acetic acid (IAA). The best IAA producing strain was identified as Pantoea rodasii using 16S rRNA analysis. In addition to the IAA production, this strain could act as an efficient phosphate solubilizer (1100 ${\mu}g$$ml^{-1}$ after 5 days of incubation) also. The selected strain was cultured under different conditions in order to assess the optimum conditions for maximum IAA production. The nutrient broth (NB) medium was recorded as the best medium, where the maximum IAA production (229 ${\mu}g$$ml^{-1}$) was recorded at the start of stationary phase (12 hours after inoculation) of the bacteria growth. The performance of the strain was found to be maximum at the temperature of $30^{\circ}C$ followed by $25^{\circ}C$. IAA production was found to be increased with increasing tryptophan concentration (from 0.1 to 0.6%), however beyond this limit, a slight reduction in IAA production was observed. The strains' ability to produce IAA was further confirmed by extraction of crude IAA and subsequent TLC analysis. A specific spot from the extracted IAA preparation was found corresponding with the standard spot of IAA with same $R_f$ value. The results of HPLC analysis conducted in identifying and quantifying the IAA production more precisely, are in agreement with the results of the assessment done with colorimetric method. As revealed by the results of the pot experiment, the isolated strain could significantly enhance the growth (as measured by shoot and root growth) of mung bean plants compared to that of non-inoculated plants. Therefore it can be concluded that the present strain, Pantoea rodasii has great potential to be used as bio-inoculants.

In this study, we optimized the composition of the indole-3-acetic acid (IAA) production medium using response surface methodology on Pantoea agglomerans SRCM 119864 isolated from soil. IAA-producing P. aglomerans SRCM 119864 was identified by 16S rRNA gene sequencing. There are 11 intermediate components known to affect IAA production, hence the effect of each component on IAA production was investigated using a Plackett-Burman design (PBD). Based on the PBD, sucrose, tryptone, and sodium chloride were selected as the main factors that enhanced the IAA production at optimal L-tryptophan concentration. The predicted maximum IAA production (64.34 mg/l) was obtained for a concentration of sucrose of 13.38 g/l, of tryptone of 18.34 g/l, of sodium chloride of 9.71 g/l, and of L-tryptophan of 6.25 g/l using a the hybrid design experimental model. In the experiment, the nutrient broth medium supplemented with 0.1% L-tryptophan as the basal medium produced 45.24 mg/l of IAA, whereas the optimized medium produced 65.40 mg/l of IAA, resulting in a 44.56% increase in efficiency. It was confirmed that the IAA production of the designed optimal composition medium was very similar to the predicted IAA production. The statistical significance and suitability of the experimental model were verified through analysis of variance (ANOVA). Therefore, in this study, we determined the optimal growth medium concentration for the maximum production of IAA, which can contribute to sustainable agriculture and increase crop yield.
Indole-3-acetic acid (IAA) biosynthesis by bacteria occurs widely in rhizospheres. Bacterial species able to synthesize IAAmay be exploited for beneficial interactions in crop management systems. The objective of this study was to determine the response of ivyleaf morningglory (Ipomoea hederacea) seedlings to IAA and to an IAA-producing rhizobacterum, Bradyrhizobium japonicum isolate GD3. IAA solution and isolate GD3 suppression of seedling growth measured as radicle length and biomass depended on IAA concentration. Seedling radicle length was significantly reduced by ca. 29% with more than $1.0{\mu}M$ of IAA solution, compared to the control, 48 h after application. The cell concentration at 50% growth reduction ($GR_{50}$) of the seedling radicle was IAA production by isolate GD3 at $10^{4.82}\;cfu$, the cell concentration for 50% growth reduction ($GR_{50}$) of seedling radicle was 0.24 iM, which was much lower than the IAA solution concentration ($117.48{\mu}M$) required for $GR_{50}$. Therefore, excess IAA production by isolate GD3 may be more detrimental to morningglory radicle growth than standard IAA solution. Results confirmed involvement of IAA in suppressive effects of isolate GD3 on morning-glory seedlings grown in a hydroponic system.
An auxin-producing bacteria (strain 5-1) was isolated from button mushroom compost in Boryeong-Si, Chungcheongnam-Do. The 5-1 strain was classified as a novel strain of Enterobacter aerogenes based on chemotaxonomic and phylogenetic analyses. The isolated E. aerogenes 5-1 was confirmed to produce indole-3-acetic acid (IAA), one of the auxin hormones, using TLC and HPLC analyses. When the concentration of IAA was assessed by performing HPLC quantitative analysis, a maximum concentration of IAA of $109.9mgL^{-1}$ was detected in the culture broth incubated in R2A medium containing 0.1% L-tryptophan for 24 h at $35^{\circ}C$. Acidification of the culture was deemed caused by an increase of IAA because a negative relationship between IAA production and pH was observed. Supplementation with a known precursor of IAA production, L-tryptophan, appeared to induce maximal production at 0.1% concentration, but it reduced production at concentrations above 0.2%. To investigate the growth-promoting effects to crops, the culture broth of E. aerogenes 5-1 was used to inoculate water cultures and seed pots of mung bean and lettuce. In consequence, adventitious root induction and root growth of mung bean and lettuce were two times higher than those of the control.
One of the important phytohormones produced by plant growth promoting bacteria is the auxin; indole-3-acetic acid (IAA), with L-tryptophan as the precursor. In this study, we focused on the investigation of optimal conditions for the production of IAA by Bacillus megaterium BM5. We investigated culturing conditions, such as incubation temperature, pH of the culture medium and incubation period, with varying media components such as inoculation volume, tryptophan concentration and carbon and nitrogen source. Besides, optimization study intended for high IAA production was carried out with fermentation parameters such as rpm and aeration. The initial yield of $42{\mu}g\;IAA\;ml^{-1}$ after 24 hr increased to $85{\mu}g\;ml^{-1}$ when 5% (v/v) of L-tryptophan was used in the culture broth. The maximum yield of $320{\mu}g\;IAA\;ml^{-1}$ was observed in trypticase soy broth (TSB) supplemented with starch and soybean meal as C and N sources with a C/N ratio of 3:1 (v/v) at $30^{\circ}C$, pH 8.0 for 48 hrs with 1.0 vvm and 250 rpm in 5 L working volume using 10 L scale fermenter. The bacterial auxin extracted from the culture broth was confirmed by thin layer chromatography and high-performance liquid chromatography and effect on plant growth was confirmed by root elongation test.
An auxin-producing bacterium Yangsong-1 was isolated from a button mushroom bed in Chung cheongnam-do. The strain Yangsong-1 was classified as a novel strain of Arthrobacter enclensis based on a chemotaxonomic and phylogenetic analysis. The isolated A. enclensis Yangsong-1 was confirmed to produce indole-3-acetic acid (IAA), which is one of the auxin hormones. When the concentration of IAA was assessed by HPLC quantity analysis, the maximum concentration of IAA, $152.903mg\;L^{-1}$, was detected from the culture broth incubated in R2A medium containing 0.2% L-tryptophan for 48 h at $35^{\circ}C$. A negative relationship between IAA production and pH was estimated to show that the increase in IAA caused pH acidification of the culture. The effect of the supplement on L-tryptophan, a known precursor of IAA production, appeared to be at maximal production at 0.2% concentration and was rather reduced at concentration above 0.4%. To investigate the growth-promoting effects on the crops, the culture broth of A. enclensis Yangsong-1 was placed in water cultures and seed pots of mung beans and lettuce. In consequence, the adventitious root induction and root growth of mung beans and lettuce were 1.5 and 1.9 times higher, respectively, than those of the control.
