The Experimental Study on Anti-bacterial Potency of Sean-tang on Three species of causative bacteria of Keratitis (세안탕(洗眼湯)이 다종(多種)의 각막염(角膜炎) 유발균(誘發菌)에 미치는 효과(效果)에 관한 실험적 연구)

  • Objective : This experimental study was performed to investigate the effect of herbal eye drops, Sean-tang and Coptidis rhizoma on Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa keratitis. Methods : After administering herbal eye drops (Sean-tang, Coptidis rhizoma) on Staphylococcus ausreus, Staphylococcus. epidermidis and Pseudomonas aeruginosa, I measured MIC and the size of inhibition zone. MIC was measured by dropping to $50{\mu}{\ell}$ according to density(l00%, 10%, 1%, 0.1%). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Result : On Staphylococcus aureus, Staphylococcus epidermidis, Coptidis rhizoma showed the highest anti-bacterial potency and on Pseudomonas aeruginosa, Sean-tang, Coptidis rhizoma was nor anti-bacterial potency. Conclusions : The present author think that the herbal eye drops, Sean-tang and Coptidis rhizoma can be used to cure Staphylococcus aureus, Staphyloccus epidermidis keratitis and if further study is performed, the use of the herbal eye drops will be valuable and beneficial in the clinical medicines.

THE INHIBITING EFFECT OF TISSUE CONDITIONER COMBINED WITH ANTIFUNGAL AGENTS ON THE GROWTH OF CANDIDA ALBICANS (항진균제를 첨가한 수종의 점막 조정제가 Candida albicans 성장 억제에 미치는 효과)

  • Tissue conditioners have been used for treatment of denture stomatitis caused by wearing of dentures. Early studies pointed out Candide albicans (C. albicans) as main etiologic factor, and antifugal agents were added for control of the species. But there is a little information about broad comparison on the effect of tissue conditioners and antifungal agents added. The purpose of the present study was to compare the inhibiting effect of four tissue conditioners and one temporary soft liner on the growth of C. albicans for treatment of denture stomatitis using gel diffusion method by measuring diameter of the zone of growth inhibition. Three antifungal agents were added to each material for evaluation of the effect of added agents. Finally, observation was made to evaluate the effect of the loss of antifungal elements by aging of the specimen. The results of this study were obtained as follows : 1. Tempo had remarkable antifungal effect showing the zone of growth inhibition as 2.35 mm at 1st day, and was most effective on End: 4th and 7th day from incubation (p<0.05). But Coecomfort, Dura conditioner, Visco-gel, Coe-soft had little antifungal effect from the 1st day of incubation. 2. Nystatin was most effective showing 9.60-12.04 mm of zone of inhibition at the 1st day from incubation. The antifungal properties were reduced to amphotericin B, chlorhexidine and materials without agent (p<0.05), and the effect was diminished by time. 3. As pretreatment with amphotericin B, nystatin, chlorhexidine, Tempo was very effective at the 1st day from incubation showing zone of inhibition as 3.65, 12.04, 4.78 mm with addition of each agent. Dura conditioner had strongest antifungal effect at the next day as 2.86, 5.33, 1.29 mm of zone of inhibition, and yielding results of Coe-comfort, Tempo, Coe-soft was shown at 4th and 7th day from incubation (p<0.05). Taken all together, tissue conditioners have little antifungal effect except Tempo. Formation of the zone of growth inhibition was due to agents amphotericin B, nystatin, chlorhexidine and nystatin was most effective. Conclusively, it is advisable to select material which is effective on the growth of C. albican and consider addition of antifungal agents for treatment of denture stomatitis.

  • This study was divided into two parts. In the first experiment, the in vitro antimicrobial effect was tested in order to evaluate the effect of vapors, and the effectiveness of the nonspecific endodontic medicaments (formocresol, camphorated parachlorophenol and eugenol). In the second experiment, the intracanal effect was tested in vitro under simulated clinical condition. The actual bactericidal effect of the nonspecific endodontic medicaments (formocresol, camphorated parachlorophenol and eugenol) was quantitated. The results were obtained as follows: 1. The zone of inhibition was appeared on the vapors of formocresol only, however there were no zone of inhibition appeared on the vapors of camphorated parachlorophenol and eugenol. 2. Formocresol produced the widest zone of inhibition and eugenol, the next and camphorated parachlorophenol, the narrowest. 3. All of the tested medicaments were vaporized in the root canal. They proved to be the effective antimicrobial activity in the root canal. 4. All of the tested medicaments were showed more bactericidal effect at 72 hours than 48 hours. 5. In comparing with the bactericidal effect of the tested medicaments in the root canal, formocresol was showed the most bactericidal medicament, camphorated parachlorophenol was showed the least. 6. Complete sterilization of the root canal was not achieved in any medicaments applied in this study.

Antibiotic, Antioxidant and Whitening effects of Jwa Kum-Whan and Soo Ryeon-Whan (좌금환과 수련환의 항균, 항산화 및 미백 효과)

  • Objectives : The purpose of this study was to research the antibiotic, antioxidant and whitening effects of Jwa Kum-Whan(JKW) and Soo Ryeon-Whan(SRW). Both Jwa Kum-Whan and Soo Ryeon-Whan are composed of Coptidis rhizoma and Evodiae fructus, but the ratios of the two species are different. JKW is composed of Coptidis rhizoma and Evodiae fructus by ratio of 6:1 and SRW's ratio is 1:1. Methods : Antibiotic activities of JKW and SRW's water extracts were studied by paper disc diffusion method. Four kinds of bacteria were applied in paper disc, and each extract was dropped, individually. The diameter of inhibition zone was measured. DPPH assay was used for studying free radical scavenging activity. DPPH is quantitatively decolorized from purple color by antioxidant material. Change of color was measured by spectrophotometer. Whitening activity was analyzed by tyrosinase inhibition assay. Tyrosinase is major enzyme for control process of making eumelanin. Optical Density was measured by spectrophotometer. Results : Candida albicans and Staphylococcus aureus's inhibition zone were made large available by Both prescriptions. Inhibition zone's diameter of JKW was preferable to SRW's. Radical scavenging activity was better at SRW, but JKW's activity was available, too. Tyrosinase inhibition activity was found out available only for JKW, not for SRW. Conclusions : JKW had antibiotic effects for Candida albicans, Staphylococcus aureus. And had antioxidant, whitening effects. SRW had antibiotic, antioxidant activities but not whitening effect.

The Experimental Study on Antifungal effects of mixture of Sophorae Subprostratae Radix, Acoti Radix, and Hibisci Syriaci Cortex by the Three Types of Extraction on three species of the dermatophytes. (다양한 山豆根·川烏·木槿皮 혼합물 용액이 足部白癬菌에 미치는 영향)

  • This experimental study was performed to investigate the antifungal effects of mixture of Sophorac Subprostratae Radix, Aconti Radix, and Hibisci Syriaci Cortex by the three types of extraction in the dermatophytes such as Trichophyton rubrum, Trichophyton mentagrophytes, and Epidermophyton floccosum. The following results were obtained by using Minimum Inhibition Concentration(MIC) and Inhibition Zone. 1. MIC on Trichophyton mentagrophytes in Sample S-2, Sample S-3, and Sample-Y were 10${\mu}l$, respectively, showing the highest antifungal effect. 2. MIC on Epidermophyton floccosum in Sample S-2, Sample S-3, and Sample-Y were 10㎕ respectively, showing the highest antifungal effect. 3. MIC on Trichophyton rubrum in Sample S-3, Sample-Y were 10${\mu}l$ respectively, showing the highest antifungal effect. 4. The size of inhibition zone on Trichophyton mentagrophytes for Sample S-3 were 22.3mm in 10${\mu}l$, showing the highest antifungal effect. 5. The size of inhibition zone on Epidermophyton floccosum for Sample S-3 was 34.3mm in 10${\mu}l$, showing the highest antifungal effect. 6. The size of inhibition zone on Trichophyton rubrum for Sample-Y was 26.5mm in 10${\mu}l$, showing the highest antifungal effect. Whereas. in extractions, that of Sample S-3 was 12.5mm in 10${\mu}l$, showing the highest antifungal effect. 7. From the above results, Sample S-3 on Trichophyton mentagrophytes, Epidermophyton floccosum and Sample-Y on Trichophyton rubrum showed the highest antifungal effects. This results suggests that vinegar extraction excels other extraction method using water or ethanol to measure the antifungal effect on dermatophytes. In addition, the result shows that the extract of herbal medicines can be used to tinea pedis and if further study is performed, the use of the extract of herbal medicines will be valuable and beneficial in the clinical medicines.   • Objectives : This experimental study was performed to investigate antibiosis of decoctions made by Cassiae Semen(CaS), Celosiae Semen(CeS) and Buddlejae Flos(BF). Methods : Decoctions made by CaS. CeS and BF were prepared. After administering decoctions made by CaS. CeS and BF on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Condida albicans) which cause Keratitis, the size of inhibition zone and MIC(Minimum Inhibition Concentration) were measured Results : The inhibition zone on bacterial species didn't appear, after administering decoctions made by CaS, CeS and BF. Conclusions : This experimental study is showed that decoctions made by CaS, CeS and BF don't have antibiosis on bacterial species which cause Keratitis.

Melanin Biosynthesis Inhibitory Activities of Coumarins Isolated from Angelica polymorpha MAXIM (궁궁이(Angelica polymorpha MAXIM)로부터 분리한 Coumarin 계열 화합물의 Melanin 생합성 억제 활성)

  • During the screening for inhibitors of melanin biosynthesis from plant extract, Angelica polymorpha MAXIM which showed a high level of inhibition was selected. The inhibiting substances were purified form methanol extract of Angelica polymorpha MAXIM followed by silica gel column chromatography and HPLC. The inhibitors were identified as heraclenin, isosaxalin and heraclenol 3'-Me ether, by spectrescopic methods of ESI-MS, H-NMR, C-NMR, DEPT, HMQC and HMBC. These compounds did not have mushroom tyrosinase inhibitory activity, but showed a highly potent melanin biosynthesis inhibition zone in the plate culture of Streptomyces bikiniensis, a bacterium used as an indicator organism in this work. These compounds did not show any growth inhibition against S. bikiniensis at the same concentration of melanin biosynthesis test.

Effectiveness of Vicryl $plus^{(R)}$ (Ethicon, USA) in Nosocomial Bacteria (실제 병원성 균주에서 Vicryl $plus^{(R)}$ (Ethicon, USA)의 효용성)

  • Purpose: Surgical site infections (SSIs) are the third most frequently reported nosocomial infection. Of these SSIs, mostly were confined to the incision associated with underlying disease as diabetes, cigarette smoking, systemic steroid use, obesity, operating room environment, suture and surgical technique. This study has been planned to reduce the SSIs by using Vicryl $plus^{(R)}$ (Ethicon, USA) which contains triclosan, a broad-spectrum antibacterial agent, into the infected wound to evaluate whether or not Vicryl $plus^{(R)}$ (Ethicon, USA) is effective to nosocomial bacteria using a zone of inhibition assay. Methods: We did a comparison of Vicryl $plus^{(R)}$ suture (with triclosan) size 2-0, 5-0 with $Vicryl^{(R)}$ suture (without triclosan) size 4-0 each as treatment and control group, applied in Mueller-Hinton agar infected by following mircroorganisms: Methicillin-sensitive $Staphylococcus$ $aureus$ (MSSA), Methicillin-resistant $Staphylococcus$ $aureus$ (MRSA), Acinetobacter baumanii, $Escherichia$ $coli$, Enterobacter faecalis, Pseudomonas aeruginosa, Candida albicans. Cultures were made of the selected mircroorganisms, seeding the study strain in agar plates for 24 and 48-hour period in an oven at $37^{\circ}C$ followed by zone of inhibition assay. Results: Vicryl $plus^{(R)}$ group has demonstrated to create a zone of inhibition against MRSA, MSSA and $A.$ $baumanii$, but no effect on $E.$ $faecalis$, $P.$ $aeruginosa$, $C.$ $albicans$. Vicryl $plus^{(R)}$ suture size 2-0 also had antibactericidal effect while Vicryl $plus^{(R)}$ suture size 5-0 did not. $Vicryl^{(R)}$ group had no zones of inhibition showing colonization at all mircroorganisms. Conclusion: Our results seem to warrant the use of Vicryl $plus^{(R)}$ as absorbable buried suture when concerning SSIs as a prophylaxis against surgical nosocomial infection.

In vitro Study on the Antimicrobial Activity of Human Tears with Respect to Age

  • Lysozyme is present in tears and has the ability to inhibit bacterial growth. In addition, it acts as a physiological scavenger for harmful substances. In the present study, sixteen tear samples from people of different ages were evaluated for their antibacterial spectrum against selected bacterial strains (Escherichia coli, Shigella sonnei, Staphylococcus aureus, Salmonella enterica Typhi). A radial diffusion assay was used to evaluate the antibacterial potential of tear samples. To correlate the antibacterial activities of these tear samples, the concentration of lysozyme in the tear samples was also determined. Ampicillin was used as a standard drug. The zone of inhibition (mm) was used to measure the antibacterial property of the tears. All samples showed good antibacterial activities. The tear samples of children showed antibacterial activities in the range of 4.40~5.00 mm inhibition zones against the selected bacterial strains. The tear samples from the young and adults showed good antibacterial potential with a zone of inhibition in the range of 3.20~4.00 and 4.00~5.50 mm, respectively. The tear samples from the old age group showed inhibition zones from 1.50~5 mm. The adult tear samples showed the maximum inhibition against the selected bacterial strains among all groups. The lysozyme concentration was 1.7 mg/mL, 1.95 mg/mL, 2.13 mg/mL, and 1.76 mg/mL for children, young, adults, and elderly, respectively. In conclusion, the tears from adults have the high inhibition potential. In addition, this data also showed that the lysozyme contents in the tear sample increased with age until 40~42 years.

Haldane Inhibition at CAH DNAPL Source Zone in Soil and Groundwater

  • Two biokinetic models (\circled1 Mrichaelis-Menten kinetics with competitive inhibition \circled2 with both competitive inhibition and Haldane inhibition) for reductive dechlorination were developed and compared with results from batch kinetic tests conducted over a wide range of PCE and TCE concentrations with two different dechlorinating cultures. At PCE concentrations lower than 300 $\mu$M, both model simulated the experimental results well. However, The kinetic model that incorporated both competitive and Haldane inhibitions much better simulated experimental data for PCE concentrations greater than 300-400 $\mu$M, and TCE concentrations at half its solubility limit (4000 $\mu$M). The PM culture showed Haldane inhibition constants of 900, 6000, 7000 $\mu$M for TCE, c-DCE and VC, indicating very weak Haldane inhibition for c-DCE and VC, while the EV culture had lower Haldane inhibition constants for TCE, c-DCE, and VC of 900, 750, and 750 $\mu$M, respectively. The BM culture had better transformation abilities than the individual cultures over a wide range of PCE and TCE concentrations. Modeling results indicated that a combination of competitive and Haldane inhibition kinetics is required to simulate dechlorination over a broad range of concentrations up to the solubility limits of PCE and TCE.
