This study was aimed to investigate the difference for carbohydrate accumulation in both the red skinned tuber and white skinned tuber of Jerusalem artichoke (Helianthus tuberosus L.), and to evaluate their tuber yield of seven lines collected from Korea. Jerusalem artichoke tubers were divided into two groups regarding to their skinned colors. Red skinned tuber collected from Euisung region showed the lowest tuber yield as 3,100 kg per 10a, otherwise white skinned tuber collected from Imdong region resulted in the highest tuber production as 6,300 kg per 10a among the six kinds of white skinned tubers. Yield of white skinned tuber was higher than that of red skinned tuber. It was inferred from the result that carbohydrate accumulation in white skinned tuber was highly increased compared to red skinned tuber since after early tuber enlargement.
This study investigated in vitro antioxidant, anti-inflammatory and cytotoxicity on human lung epithelial A549 cells of different solvent extracts from Jerusalem artichoke (Helianthus tuberosus L.) tuber. The EtOH extract contained amounts of phenolics (22.20 tannic acid equivalent ㎎/ɡ) and exhibited the highest antioxidant activity and anti-inflammatory activity. Several methods were employed for measure the antioxidant activity: 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity (IC50 = 206.79 ㎍/㎖), reducing power activity (21.26 ascorbic acid equivalent ㎎/ɡ) and total antioxidant activity (19.05 ascorbic acid equivalent ㎎/ɡ). Meantime, the EtOH extract inhibited the NO production completely with a concentration of 800 ㎍/㎖. Besides, the H2O extract exhibited more potent effect on human lung epithelial A549 cells. This study suggested that Jerusalem artichoke tuber had antioxidant, anti-inflammatory and cytotoxicity on human lung epithelial A549 cells.

In order to utilize the tuber of Jerusalem artichoke (Jerusalem tuberosus L.) which has been considered one of the most economical resources for bioenergy, the tubers of the plant were processed into silage in 11 lab-silo with three levels of mixture of concentrates i.e. 10, 20, 30%. The silage was stored for 20 to 40 days before analyses. The quality of silage was compared with those of the tube-only silage without concentrates. Aproximate composition, pH, ammonium nitrogen, total nitrogen, and organic acids were analyzed. The results obtained are summarized ; 1. With the mixture of concentrates, the water content of the silage was able to be adjusted to 70 percent and NDF significantly increased (P<0.05) while ADF significantly decreased. 2. The content of water-soluble carbohydrate (WSC) was significantly decreased in silages of the mixture of concentrate and the tuber (P<0.05), while total nitrogen and ammonium nitrogen increased. 3. Tube-only silage gave satisfaction in fermentation, particularly organic-acid fermentation. As the storage period extended, Flieg's score of the silage was improved. Conclusively, the tuber of Jerusalem artichoke found to satisfy the requirements of good silage and to be recommended as a good resource for all-inane silage provided the tuber was ensilaged with 20 to 30 percent concentrate.

Nineteen clones of Jerusalem Artichoke (JA) from several countries were collected through the series of experiments about JA started in 1979. Collected clones were screened for adaptibility in Korea and showed introduction path way. The results about an ecological response of collected clones including flowering, tuberization, biomass production, sugar contents and grouping of collected clones for use of genetic material were as follows; 1. Nineteen clones collected were ffom Korea(2), Japan(l), USA(Z), Canada(2), France(4), Germany(7), and USSR(1). 2. Through the characteristics of top collected clones were divided to the types of branch-non branch, short and long plant height, and early and late maturity. Tuber characteristics were mainly grouped to the types of white skin color-violet skin color, clusters-single unit, round-elongate, and knotty-smooth. 3. Total sugar yields 6-om top at flowering time were 490 - 630kgl10a and 6-om the tuber were 420 -490 kg/ IOa through the high yielding clones. The top-high yielding clones were Mammoth French White, Fuseau 60, Nahodka, and JA3. The higher tuber yields were got from the clones of D- 19, Colombia, Bianka and Mammoth French White. 4. Collected clones were grouped to three and first group was characterized to early maturity and short plant height and second group to medium and finally, third group to late maturity and high plant height. 5. High yielding of top was 6-om the I group of early maturity and short plant height and high yielding of tuber h m III group of late maturity and high plant height.
To produce inulase from Kluyveromyces fragilis No.351 using Jerusalem artichoke tuber extract, optimization of the condition was conducted. As results, the optimal concentration of artichoke tuber extract was 3.5 % and bactocasitone showed better production than yeast extract. The optimal temperature and pH were $30^{\circ}C$ and 5.5, respectively. The addition of $NH_4H_2PO_4$ increased the enzyme production. Inulase synthesis was growth-associated and the enzyme production increased as concentration of dissolved oxygen increased. A higher quantity of industrial grade inulase was prepared by the combination of ultrafiltration and ethanol precipitation with 72% recovery.
In order to examine the possibility of seed propagation of jerusalem artichoke (Helianthus tuberosus L.) which has been propagated vegatatively so far, it was investigated the growth characteristics of seedlings and the biomass productivity by seed propagation in jerusalem artichoke. The results indicated that the seedlings from seeds exhibited the physiological dwarfing as a rosette habit. The physiological dwarfing as a rosette habit in jerusalem artichoke seedlings from seeds was a kind of appearance affected by photoperiod. This abnormal habit in seedlings from JA6 seeds during the growing period was persisted in short photoperiod of 10 hrs but was disappeared in long photoperiod of 14 hrs. In the condition of long photoperiod, seedlings from seeds grew normally and the average tuber yields obtained from seed propagation was finally 3.51 ton/10a, which can be the satisfying level to the use of seed propagation despite its complexity such as the needs of specific seedling time because it gives a high biomass productivity. This potential utilization of seed propagation in jerusalem artichoke can be basically applied to the collection or preservation of germ plasm and the improvement of jerusalem artichoke.
Polyphenol oxidase from Jerusalem artichoke(Helianthus tuberosus L.) tubers was partially purified by precipitation with ammonium sulfate, followed by gel filtration on Sephadex G-100. The enzyme showed maximal activity at pH 6.5 and $4^{\circ}C$. Kinetic studies indicated $K_{m}$ value of 3 mM for catechol and activation energy of 72.6 kcal/mole. As for substrate specificity of polyphenol oxidase the enzyme showed high affinity towards diphenol compounds, but not towards monophenols. The enzamatic browning was completely inhibited at 1 mM concentration of L-ascorbic acid, sodium hydrosulfite and L-cystein(HCl). The activity of polyphenol oxidase in 0.1 M potassium phosphate buffer(pH 6.5) was fairly stable for a week at $4^{\circ}C$, while it decreased remarkably at $25^{\circ}C$.

The inulase(EC 3.2.1.7) was isolated from the tuber of Jerusalem artichoke by conventional purification methods including ammonium sulfate fractionation, Sephadex G-100 filtration, and DEAE-cellulose column chromatography. The enzyme was purified 6,470 fold with 42% recovery, The enzyme was consisted of a polypeptide of Mw 57,000. The optimum temperature and the optimum pH for the enzyme action was $33^{\circ}C$ and pH 5.0, respectively. The enzyme was highly specific for inulin as a substrate. The km for inulin was 20mM. The inulase was not a metalloenzyme and was inhibited completely by 10mM $Mg^{2+},Ca^{2+},\;or\;Hg^{2+}$.
Drought is more concerned to be a huge problem for agriculture as it affects plant growth and yield. Endophytic bacteria act as plant growth promoting bacteria that have roles for improving plant growth under stress conditions. The properties of four strains of endophytic bacteria were determined under water deficit medium with 20% polyethylene glycol. Bacillus aquimaris strain 3.13 showed high 1-aminocyclopropane-1-carboxylate (ACC) deaminase production; Micrococcus luteus strain 4.43 produced indole acetic acid (IAA). Exopolysaccharide production was high in Bacillus methylotrophicus strain 5.18 while Bacillus sp. strain 5.2 did not show major properties for drought response. Inoculation of endophytic bacteria into plants, strain 3.13 and 4.43 increased height, shoot and root weight, root length, root diameter, root volume, root area and root surface of Jerusalem artichoke grown under water limitation, clearly shown in water supply at 1/3 of available water. These increases were caused by bacteria ACC deaminase and IAA production; moreover, strain 4.43 boosted leaf area and chlorophyll levels, leading to increased photosynthesis under drought at 60 days of planting. The harvest index was high in the treatment with strain 4.43 and 3.13 under 1/3 of available water, promoting tuber numbers and tuber weight. Inulin content was unchanged in the control between well-watered and drought conditions. In comparison, inulin levels were higher in the endophytic bacteria treatment under both conditions, although yields dipped under drought. Thus, the endophytic bacteria promoted in plant growth and yield under drought; they had outstanding function in the enhancement of inulin content under well-watered condition.

In order to know the effect of planting date and cutting management on Jerusalem Artichoke (Helianthus truberosus L.) as a new forage or energy crop, JA tubes were planted from April 7 to June 27 with 20 day interval. Growth and yield could stay high as normal planting if tuber planting had been done until late May. The delayed planting after June caused deterioration of seed tuber, produced poor emergence and retarded growth. Previous harvested field did not need additional planting every spring because there were enough leftover tubers usually small size, less than 3gr., to provide a dense seedling establishment. This plot had too dense in population to adjust its spacing by thinning some of seedlings. One or two times cutting for forage or silage production reduced tuber yield very significantly. Therefore, it was not a good management to harvest for both forage and tuber production. Topping at 20cm from growing point in early July prevented from lodging and assured tuber yield.
