A Study on Filed Application of Electro-Osmosis Soil Improvement Method with Nano-Coated Plastic Drain Baord (나노 코팅된 PDB를 이용한 동전기 지반개량 공법의 현장 적용성에 관한 연구)

  • The PBD (Plastic Board Drain) method is one of effective ground improvement methods on the soft dredging reclamation ground. This method has outstanding economic efficiency and constructability, and it is widely used for the soft ground improvement. However, the PBD method reduces permeability and drainage capacity of the ground due to the long construction period. Therefore, the nano coated Plastic drain board (PDB) was developed to solve problems. It is the non-metallic electrode and improves the weakness of the PBD method by using electric force of the electro-osmosis method. Various researches have been conducted to apply the nano coated PDB, but these researches were limited to model tests in laboratory. In this study, model and field tests were conducted to assess field applicability of the nano coated PDB. The result showed that the nano coated PDB had the better effect on the ground improvement compared to the normal PDB.

Compatibility for Proposed R.94 PDB Test (PDB 시험에 대한 충돌 상호 안전성)

  • Currently various safety tests are being performed in many countries with growing interest in vehicle safety. However the vehicles which have good safety performance in these tests could not secure the good performance in real car to car accident. So new test protocol using progressive deformable barrier (PDB) was proposed by EEVC in Europe, NHTSA in USA and some vehicle manufacturers, etc. The target of PDB test is to control partner protection in addition to self-protection on the same test. The proposal is to update current ECE R.94 frontal ODB test. So barrier, impact speed, overlap are changed to avoid bottoming-out in the test configuration. In this paper 3 different tests (R.94, EuroNCAP and PDB test) were carried out using current production vehicles with same structure. The results of these tests were compared to understand PDB test. As a result PDB test shows the highest vehicle deceleration and dummy injury because PDB offers a progressive increase in stiffness in depth and height. However vehicle intrusion was affected with rather test velocity than stiffness of deformable barrier. PDB deformation data is used for partner protection assessment using PDB software and it shows that the test vehicle is rather not aggressive.

Mycelial Production and Amylase Activity of Fungi for Brewing in Different Submerged Culture Conditions (액체배양에서의 양조용 곰팡이의 균체 생산 및 전분분해효소 활성)

  • In this study, twelve strains of brewing fungi were individually cultivated on wheat extract broth (WEB), potato dextrose broth (PDB) and malt extract broth (MEB) in order to determine the microorganism with good culture characteristics as well as with amylolytic activity. The strain cultured in PDB exhibited mycelia production from 12.6 g/L (Rhizopus oryzae KACC 45714) to a maximum of 48.0 g/L (Aspergillus oryzae KACC 46959), which was 2.3~9.2 times lower than that of the strain cultured in WEB and 1.7~14.6 times lower than that of the strain cultured in MEB. Accorfing the results, We found that the commercial strains of A. oryzae Suwon, CF1001 and CF1003 had a higher dry cell mass than the wild-type strains KACC 46421, 46423, 46424 and 46959. For Rhizopus sp., the acidity levels in WEB, PDB and MEB were 0.12~0.47%, 0.22~1.0% and 0.16~0.68% (equivalent lactic acid concentration) respectively. For A. oryzae, the acidity levels were 0.06~0.11%, 0.03~0.04% and 0.06~0.08% (equivalent lactic acid concentration), respectively. Amylase enzyme from Rhizopus delemar KACC 46419 exhibited an enzyme activity of 0.013 U and 0.019 U in WEB and MEB cultures, respectively. The enzyme activity of the amylase enzyme from A. oryzae was 0.019~0.037, 0.017~0.033 and 0.028~0.046 U in WEB, PDB and MEB cultures, respectively.

Effect of Cd treatment on the mycelial growth of Pleurotus ostreatus (Cd처리가 느타리버섯 균사체 생장에 미치는 영향)

  • In this study, we determined the effect of different concentrations of Cd on the growth of Pleurotus ostreatus mycelia, which was confirmed using PDA, PDB, and a column test. The efficacy of treatment was evaluated using Cd at two concentrations, 10 ppm and 100 ppm. The extent of mycelial growth on PDA and PDB was similar to that observed in the untreated groups and those treated with 10 ppm Cd. However, it was found that the mycelial growth was suppressed in a system treated with Cd at concentrations of 100 ppm. Moreover, the extent of mycelial growth observed upon conducting a column test was similar to that obtained using PDA and PDB. When the composition of free amino acids in PDB was determined, their levels in the group treated with 100 ppm Cd were found to be similar to those of the control. However, the amounts of 15 amino acids in the group treated with 10 ppm of Cd had increased compared to those detected in the control.

Effect of Protein Kinase C on Norepinephrine Release in the Rat Hippocampus (흰쥐 해마에서 Norepinephrine 유리에 미치는 Protein Kinase C 의 영향)

  • The effects and interactions of $4{\beta}-phorbol$ 12,13-dibutyrate(PDB) and polymyxin B(PMB) with adenosine on the electrically-evoked norepinephrine (NE) release were studied in the rat hippocampus. Slices from the rat hippocampus were equilibrated with $^3H-noradrenaline$ and the release of the labelled product, $^3H-NE$, which evoked by electrical stimulation$(3\;Hz,\;2\;ms,\;5\;VCm^{-1},\;rectangular\;pulses)$ was measured. PDB$(0.3{\sim}10\;{\mu}M)$, a selective protein kinase C(PKC) activator, increased the evoked NE release in a dose related fashion while increasing the basal rate of release. And the effects of $1\;{\mu}M$ PDB were significantly inhibited by $0.3\;{\mu}M$ tetrodotoxin(TTX) pretreatment or $Ca^{++}-free$ medium. $PMB(0.03{\sim}1\;mg)$, a specific PKC inhibitor, decreased the NE release in a dose dependent manner while increasing the basal rate of release. Adenosine $(1{\sim}10\;{\mu}M)$ decreased the NE release without changing the basal rate of release, and this effect was significantly inhibited by 8-cyclopentyl-1,3-dipropylxanthine$(2\;{\mu}M)$, a selective $A_1-receptor$ antagonist, treatment. Also, adenosine effects were significantly inhibited by PDB-and PMB-pretreatment. These results suggest that the PKC plays a role in the NE release in the rat hippocampus and might be participated in a post-receptor mechanism of the $A_1-adenosine$ receptor.

  • 현재의 단백질 구조비교 시스템들 사이의 호환성이나 상호작용성의 문제를 해결하고 단백질 구조를 비교하는 시스템을 신속히 개발하기 위해서 단백질 3차구조를 표현하기 위한 데이터를 추출하여 XML과 같은 표준 형식으로 기술된 데이터를 제공하는 것이 바람직하다. 이에 따라 단백질의 2차구조 구성요소와 그들 사이의 관계를 이용하여 단백질 구조를 기술하는 PSA가 제안되었으며, PSA를 기반으로 하여 단백질 데이터의 XML 표현기법인 PSAML이 제안되었다. 본 논문에서는 PSAML 데이터의 생성을 위하여 PDB에서 제공되는 데이터를 PSAML 형식으로 변환시키는 도구를 설계하고 구현하였다. 변환도구는 XML DOM과 Java를 이용하여 구현되었으며, 생성된 데이터는 단백질 구조 및 유사성을 비교하기 위한 단백질 구조비교 시스템에서 사용될 수 있다.

Soil Improvement using Vertical Natural Fiber Drains (연직천연섬유배수재를 이용한 연약지반 개량)

  • A pilot test using environmentally friendly drains, was carried out to evaluate their applicability potential in the field. The pilot test site was divided into 5 different areas, with several combinations of vertical and horizontal drains installed for evaluation. Conventional natural fiber drains (FDB), new developed straw drain board (SDB) and plastic drain board (PDB) were used as vertical drains, while sand and fiber mats were used as horizontal drains. Surface settlement rates and excess pore pressure generation/dissipation tendency of PDB and FDB are almost identical except those of SDB. Cone tip resistance obtained from cone penetration test measured at the end of 1st consolidation stage for upper soft layer definitely increased irrespective of types of vertical drains. The monitoring and site investigation test data obtained at the pilot test site prove the vertical natural fiber drains can be used as substitutes of conventional plastic and sand material.

Factors Affecting Pellet Formation of Phosphate-solubilizing Fungus, Aspergillus sp. PS-104 in Submerged Culture (인산가용화균 Aspergillus sp. PS-104의 액침배양중 Pellet 크기에 영향을 주는 요인)

  • In order to minimize the mycelial pellet size of a high phosphate-solubilizing fungus, Aspergillus sp. PS-104 in liquid media, one of the critical obstacles during the submerged culture of filamentous fungi, an investigation was focused on the culture conditions (media and inoculum size) and additives (different soils, surfactants and polyethylene glycol 200). When the fungus was cultured in PDB, SDB and YPD media. their pellet sizes decreased in the order of SDB=YPD>PDB. At the higher concentrations of initial inoculum ranging from $1{\times}10^3$ to $1{\times}10^7$ conidia/ml, the smaller size of pellet was formed in the PDB medium. In addition, the pellet size was effectively reduced by 1/6${\sim}$1/4 by the addition of 0.1% soil containing zeolite, diatomite, loess, kaoline and talc, excluding bentonite. The addition of 0.1% Tween 80, Triton X-100 and PEG 200 also decreased the pellet size, but SDS completely inhibited the fungal growth.

  • PDB에서 제공하는 단백질 3차원 고분자결정 구조에 대한 플랫파일은 인자들의 좌표, 서열정보, 실험정보 및 참조 정보가 포함된다. 이러한 정보를 포함하고 있는 플랫파일로부터 필수적인 구조정보 및 서열정보 등의 효율적인 검색을 위해서는 이러한 데이터를 추출하여 데이터베이스 구축이 요구되며 이 때 단백질 구조 및 서열 정보와 실험 및 탐조 정보의 관계에 대한 모델링이 중요하다. 따라서 이 논문에서는 PDB에서 제공하는 플랫파일들의 엔트리들을 분석하고 3차원 공간 객체의 기하적 특성을 갖는 단백질 3차 구조를 공간객체로 표현하고 공간객체 모델을 적용하여 모델링한다. 이렇게 함으로써 단백질 3차 구조 분자를 구성하는 인자 및 구조 정보 검색이 가능하며 위상 및 기하 연산자글 이용하여 단백질 구조 분석에 활용할 수 있다.
