Klebsiella pneumonia로 부터 제한효소 Kpn I의 분리 (Isolation of kpn I restriction endonuclease from klebsiella pneumonia)

  • A restriction endonuclease, Kpn I has been isolated from Klebsiella pneumonia. Cells were broken by sonication. After ultracentrifugation the supernatant containing Kpn I activity was further purified by Sepharose-6B gel filtration, DEAD-Cellulose, Heparin-Agarose, and Aminohexyl-Agarose column chromatography. Final enzyme preparation was essentially free of contamination exonuclease and phosphatase, as judged by ligation-recut test. Total activity of the enzyme recovered from 10 grams of cells was $4.7\times 10^5$ units.

  • 광범위 폐괴저는 매우 빠른 폐실질의 파괴를 보이는 대엽성 폐렴의 치명적인 합병증으로서 적절한 치료를 하지 않으면 높은 사망률을 보인다. 주원인균으로는 Klebsiella pneumoniae, Pneumococcus, Aspergillus 등이 있다. 흉부X선 검사상 폐엽의 고형질화 및 폐용적 증가 \ulcorner의한 폐엽간구의 융기현상을 보이며, 컴퓨터 단층촬영에서 작은 공동을 포함한 광범위한 폐실질의 파괴를 나타낸다. 치료는 조기 수술적 절제와 함께 항생제 치료가 추천되며, 수술적 치료가 안되었을 경우 패혈증, 다발성 장기부전에 이어 사망에 이르는 것이 통상적인 진행으로 알려져 있다. 저자들은 Klebsiella 폐렴에 이어 빠르게 진행하는 폐괴저에 대한 2례의 수술을 시행하였기에 문헌 고찰과 함께 보고하는 바이다.

폐렴우와 Klebsiella pneumoniae 실험적 감염 랫드의 기관 및 기관지 점막층에서 Globule leukocyte의 출현양상 (Appearance of globule leukocytes in tracheal and broncheal mucosa in Korean native cattle with pneumonia and rats experimentally infected with Klebsiella pneumoniae)

  • The study was aimed at identifying the globule leukocytes (GL) of tracheal mucosa layer of Korean native cattle showing symptom of pneumonia which have died as enterotoxemia and normal Korean native cattle in Kyungpook local area. In another set of experiment, Isolated Klebsiella pneumoniae from suddenly died cattle specimens was subjected to rat for a determining globule leukocyte appearance by using histochemical and immunohistochemical method. In histochemical study, globule leukocytes generally was existed in all the case of postmortem of Korean native cattle and 3 heads of slaughtered cattle which showing symptoms of pneumonia and it showed significant increase in tracheal mucosa of rats experimentally infected with Klebsiella pneumoniae. These increased number of globule leukocytes was moderately remained on early infection stage and gradually decreased in timedependent manner after infection. The granule patterns were also determined as an acidmucopolysaccharide. In immunohistochemical study, serotonin intensity in the treacheal mucosaepithelial cells of rat experimentally infected observed a strong immunoreactivity during early infection and gradually decreased in dependent of infection stage while no IgE immunoreactivity observed. These data show that globule leukocytes were increased in a pneumonia, therefore it was considered as a valuable cell that was associated with early stage of inflammatory response.

  • Klebsiella pneumoniae의 감염은 대부분 원발성 간농양을 일으키며 전이성 병변으로 폐렴, 뇌수막염, 요로감염 등의 간 외 합병증을 수반한다. K. pneumoniae 감염의 간 외 합병증으로 화농성 슬관절염이 보고된 예는 드물며 원발성 간농양 없이 화농성 슬관절염이 보고된 예는 더욱 드물다. 이에 제2형 당뇨병을 가진 슬관절 골관절염 환자에서 원발성 간농양 없이 발생한 K. pneumoniae에 의한 화농성 슬관절염을 조기 발견하고, 관절경적 치료로 호전된 1예를 보고하고자 한다.

  • 암모니아는 현대 산업에서 빠질 수 없는 유용한 물질이다. 일반적으로 농업용 폐기물의 분해과정을 통해 배출되며 인체에 매우 해로운 독극물로 알려져 있다. 산업에서 흔하게 쓰이는 물질이기에 직접 누출이나 간접 누출로 인한 수질오염의 가능성이 있다. 이 경우 암모늄이온을 빠르게 제거하는 데는 제올라이트의 흡착능을 이용하는 것이 좋지만 흡착만으로는 충분히 제거할 수 없다. 본 논문에서는 상용 제올라이트의 흡착능을 통한 암모늄이온의 제거 효율과 미생물의 생물학적 메커니즘을 통한 제거효율을 비교하였다. 추가적으로 제올라이트에 미생물을 고정하여 화학적 흡착 및 생물학적 전환 기술을 병합하여 그 효율을 비교하였다. 그 결과 100 ppm 기준 상용 제올라이트의 경우 2-4 h 사이에 67-81%의 제거효율을 보이는데 반해 선정 미생물인 Klebsiella pneumoniae subsp. Pneumoniae를 이용한 경우 8 h 내에 최대 97%의 제거 효율을 나타냈었다. 그리고 미생물을 제올라이트에 고정시켰을 때 8 h 이내에 98.5%로 제거 효율 및 속도가 증가하는 것을 확인할 수 있었다.

  • This article presents the case of a bilateral chronic subdural hematoma which was contaminated with Klebsiella pneumoniae and resulted in a life-threatening central nervous system infection. After repeated of bilateral burr-hole drainage, the patient became hyperpyrexic and drowsy. Suppuration within the subdural space was suspected and then the patient underwent bilateral fronto-temporo-parietal craniotomies, and pus was evacuated. Its cultures revealed Klebsiella pneumoniae. Intravenous meropenem was given for 6 weeks. He recovered completely. Microorganisms like Klebsiella pneumoniae may directly infect the subdural space with iatrogenic contamination.

  • Inspiratory pneumoniais one of the common complications in stroke patients who have dysphagia. Pneumonia due to Klebsiella bacteria shows higher fatality than other bacteria. For this reason, K. pneumoniae can be fatal to immune deficient patients. The purpose of this paper is to report the adapting pattern identification according to principles of defense, Gi, nutrients and blood on K. pneumoniae treatment. In this case, improvement was seen in a stroke patient with dysphagia diagnosed with K. pneumoniae by X-ray, sputum culture, and by using Cheonghobyulgaptang as a way of nourishing Eum and clearing heat, because in this case K. pneumoniae was presumed to cause Eumdeficiency due to the chronic low fever, cough, sputum and weakness. This paper proposes that adapting pattern identification according to principles of defense. Gi, nutrients and blood can increase the efficiency of K. pneumoniae treatment.

  • Acute osteomyelitis caused by Klebsiella pneumoniae is rare in the oral and maxillofacial region. Klebsiella pneumoniae is a Gram-negative bacillus and the normal flora of the human body, but it can cause pneumonia, urinary tract infection, meningitis, and osteomyelitis in patient with compromised immune systems. These infections are mainly caused by nosocomial infection. Microbacterial osteomyelitis was developed by clinical cause such as tooth extraction, fracture, and surgical history, which requires long-term antibiotic administration and surgical treatment. This report describes that a 56-year-old male patient with acute osteomyelitis caused by Klebsiella pneumoniae infection after implant placement was treated with intravenous administration of ertapenem without open surgery treatment. Through this case, we report that antibiotic susceptibility test is essential for the treatment of acute osteomyelitis caused by a bacterial infection resistant to empirical antibiotics, and early administration of appropriate antibiotics can reduce the possibility of extensive bone destruction or additional open surgery.
