The Optimum Design of Reinforced Concrete Structures Based on the LRFD (하중일- 저항계수 설계법에 의한 철근콘크리트 구조물 최적설계)

  • In this study, an optimization design of reinforced concrete structures is performed by using the structural optimization techniques based on the LRFD criteria. The target reliability index is estimated by the optimal reliability index considering the expected cost which is taken as a sum of the structural cost and the expected costs due to failure of the structure. The load and resistance factors calculated by using level I reliability theory with the target reliability index are compared for each load combination (D+L, D+L+w). The results of this study show that the resistance factors are ${\phi}_{M}$=0.90, ${\phi}_{V}$==0.70, ${\phi}_{C}$==0.65 and the load factors are 1.20D + 1.70L, 1.07L + 0.07L + 1.10W. The optimization techinques used to this study are S.L.P. The optimization design based on the LRFD criteria is more economical and rational than other criteria.

Increased Production of Ginsenoside Compound K by Optimizing the Feeding of American Ginseng Extract during Fermentation by Aspergillus tubingensis

  • The ginsenoside compound K (C-K) is widely used in traditional medicines, nutritional supplements, and cosmetics owing to its diverse pharmacological activities. Although many studies on C-K production have been conducted, fermentation is reported to produce C-K with low concentration and productivity. In the present study, addition of an inducer and optimization of the carbon and nitrogen sources in the medium were performed using response surface methodology to increase the C-K production via fermentation by Aspergillus tubingensis, a generally recognized as safe fungus. The optimized inducer and carbon and nitrogen sources were 2 g/l rice straw, 10 g/l sucrose, and 10 g/l soy protein concentrate, respectively, and they resulted in a 3.1-fold increase in the concentration and productivity of C-K (0.22 g/l and 1.52 mg/l/h, respectively) compared to those used before optimization without inducer (0.071 g/l and 0.49 mg/l/h, respectively). The feeding methods of American ginseng extract (AGE), including feeding timing, feeding concentration, and feeding frequency, were also optimized. Under the optimized conditions, A. tubingensis produced 3.96 mM (2.47 g/l) C-K at 144 h by feeding two times with 8 g/l AGE at 48 and 60 h, with a productivity of 17.1 mg/l/h. The concentration and productivity of C-K after optimization of feeding methods were 11-fold higher than those before the optimization (0.22 g/l and 1.52 mg/l/h, respectively). Thus, the optimization for the feeding methods of ginseng extract is an efficient strategy to increase C-K production. To our knowledge, this is the highest reported C-K concentration and productivity via fermentation reported so far.

Optical Misalignment Cancellation via Online L1 Optimization (온라인 L1 최적화를 통한 탐색기 비정렬 효과 제거 기법)

  • This paper presents an L1 optimization based filtering technique which effectively eliminates the optical misalignment effects encountered in the squint guidance mode with strapdown seekers. We formulated a series of L1 optimization problems in order to separate the bias and the gradient components from the measured data, and solved them via the alternating direction method of multipliers (ADMM) and sparse matrix decomposition techniques. The proposed technique was able to rapidly detect arbitrary discontinuities and gradient changes from the measured signals, and was shown to effectively cancel the undesirable effects coming from the seeker misalignment angles. The technique was implemented on embedded flight computers and the real-time operational performance was verified via the hardware-in-the-loop simulation (HILS) tests in parallel with the automatic target recognition algorithms and the intra-red synthetic target images.

  • In this paper, a new smoothing approximation to the l1 exact penalty function for constrained optimization problems (COP) is presented. It is shown that an optimal solution to the smoothing penalty optimization problem is an approximate optimal solution to the original optimization problem. Based on the smoothing penalty function, an algorithm is presented to solve COP, with its convergence under some conditions proved. Numerical examples illustrate that this algorithm is efficient in solving COP.

L1-penalized AUC-optimization with a surrogate loss

  • The area under the ROC curve (AUC) is one of the most common criteria used to measure the overall performance of binary classifiers for a wide range of machine learning problems. In this article, we propose a L1-penalized AUC-optimization classifier that directly maximizes the AUC for high-dimensional data. Toward this, we employ the AUC-consistent surrogate loss function and combine the L1-norm penalty which enables us to estimate coefficients and select informative variables simultaneously. In addition, we develop an efficient optimization algorithm by adopting k-means clustering and proximal gradient descent which enjoys computational advantages to obtain solutions for the proposed method. Numerical simulation studies demonstrate that the proposed method shows promising performance in terms of prediction accuracy, variable selectivity, and computational costs.

High Density Cell Culture of Bifidobacterium by Optimization of Medium Composition and Culture Conditions. (배지조성 및 배양환경 최적화에 의한 Bifidobacterium의 고농도 배양)

  • Bifidobacterium strain was isolated from the feces of brast fed infants. The isolated strain was identified as Bifidobacterium longum by 16S rRNA sequence analysis and named as Bifidobacterium SH2. The MRS medium was modified to obtain high density cells of Bifidobacterium SH2. The optimal medium was determined to be 50 g/L lactose, 10 g/L beef extract, 10 g/L peptone, 5 g/L yeast extract, 7 g/L sodium acetate, 2 g/L ammonium citrate, 2 g/L disodium phosphate,1 g/L tween 80, 0.2 g/L MnSO$_4$ and 0.5 g/L L-cysteine. The pH and temperature were optimized as 5.0 and $37^{\circ}C$, respectively. Through out the optimization of medium composition and culture conditions, the dry cell weight and viable cell count were 2.5 times and 1.8 times higer than those in MRS medium, respectively.

  • Optimization of the medium and fermentation conditions for erythritol production has been studied. We have found that the optimal carbon source was glucose at the concentration of 400 g/L. The optimal temperature was 3$0^{\circ}C$ with excessive aeration. Improved erythritol productivity was achieved by reducing the yeast extract from 5 g/L to 3g/L while adding 2.7 g/L urea, 1.79g/L $K_2HPO_4, and 0.18g/L MgSO$_4$. 7$H_2O. The erythritol productivity increased from 0.747 g/L/h to 1.071 g/L/h and the yield increased from 31.4% to 45.2%. The byproduct glycerol was reduced from 96.6g/L to 45.7g/L as well. We have performed 5L fermentation with and without the pH control. The erythritol productivity with the pH control was about 30% lower than that without pH control. Excessive foaming of 5L fermentation has been observed during fermentation.

Improvement of Amidase Production by a Newly Isolated Delftia tsuruhatensis ZJB-05174 Through Optimization of Culture Medium

  • The R-amidase production by a newly isolated strain of Delftia tsuruhatensis ZJB-05174 was optimized in this paper. Effects of factors such as carbon sources, nitrogen sources, and inducers on amidase production were investigated. The medium composition was optimized using central composite designs and response surface analysis. The optimal medium components for enhanced amidase production were found to be as follows: glucose, 8.23 g/l; yeast extract, 11.59 g/l; 2,2-(R,S)-dimethylcyclopropane carboxamide, 1.76 g/l; NaCl, 1 g/l; ${KH_2}{PO_4}$ 1 g/l; and ${K_2}{HPO_4}$ 1 g/l. A maximum enzyme production of 528.21 U/l was obtained under the optimized conditions, which was 4.7 times higher than that obtained under initial conditions.

  • In this paper, a design method for minimizing the cogging torque of an Interior Permanent Magnet Motor (IPM) is proposed based on a hybrid algorithm. The suggested optimization algorithm is based on a combination of the Response Surface Method (RSM) and Simplex Method. The results show that the proposed method provides improved characteristics compared to the conventional methods, such as a shorter calculation time and the acquisition of a more correct solution.

Optimization of Growth Medium Composition for Overproduction of Bacillus licheniformis Amylase in Recombinant Escherichia coli (Bacillus licheniformis amylase(BLMA)의 생산성 향상을 취한 재조합 대장균의 배지 최적화)

  • The research is concerned with optimization of growth medium composition in an attempt to improve the product yield of Bacillus licheniformis amylase (BLMA) in recombinant E. coli containing the BLMA gene. BLMA has the catalytic activity of producing branched oligosaccharides from starch. The medium optimization was performed in flask cultures based on the Box and Wilson method. The optimized medium is composed of tryptone 18.0 g/l, yeast extract 22.4 g/l, NaCl 5.3 g/l and glucose 2.1 g/l. In a jar fermenter culture with the conventional LB medium, the recombinant E. coli yielded 1.39 g/l of final dry cell mass and 5.11 U/ml of enzyme activity. In the optimized medium, however, the final cell mass was increased to 6.01 g/l and the enzyme activity to 23.2 U/ml. Medium optimization improved cell mass by 4.3 times and enzyme activity by 4.5 times. Such an increase in enzyme activity is mainly due to an enhancement of cell mass.
