Effects of Diet with Laminaria religiosa on Egg Quality (다시마 첨가 사료가 계란의 품질에 미치는 영향)

  • The effect of diet with Laminaria religiosa on egg quality were investigated. Isa Brown chicken (28 weeks old) were raised for 10 week. The addition level of Laminaria religiosa to each treatment was 0, 10 and 20%, respectively. No difference in egg yolk and white color was found among treatment. The contents of Ca, Fe, K and Mg were increased but of P decreased with the feeding of Laminaria religiosa. Total mineral content were increased from 860.9 mg% to 936.8 mg%. Tensile strength of egg shell was improved from $63g/cm^2$ to $77g/cm^2$. Total cholesterol content were reduced from 1,760 mg% to 1,113 mg%. Among the analyzed amino acids, the content of each amino acid was reduced except histidine increased from 2,024.0 mg% to 2,119.6 mg%. Essential amino acids in the total amino acids, 42.1% to 42.6%, were hardly changed among treatments.

Structural Characterization of Purified Fucoidan from Laminaria religiosa, Sporophylls of Undaria pinnatifida, Hizikia fusiforme and Sagassum fulvellum in Korea (국내산 다시마, 미역포자엽, 톳, 모자반 정제 fucoidan의 구성당 결합 특성)

  • Primary structure of purified fucoidans was analysed by methanolysis, methylation and HT-IR measurement. Basic linkage in the fucoidans of Laminaria religiosa, sporophylls of Undaria pinnatifida, Hizikia fusiforme and Sargassum fuivellum from Korea was $(1\rightarrow3)$ linkage of fucose, but a considerable amount of $(1\rightarrow3)$ linked galactose was also found in case of the purified fucoidan from sporophylls of U. pinnatifida. Ester sulfate was substituted mainly to C-4 position of fucose and galactose.

Host Specificity of Endophytic Kelp Gametophytes (내부공생 켈프 배우체의 숙주 선택)

  • Farmed kelp gametophytes were previously observed to be living endophytically in filamentous red algae. The interactions of two farmed kelp species and six red algae were examined in laboratory culture. Undaria pinnatifida (Harvey) Suringar and Laminaria religiosa Miyabe demonstrated the differing abilities of zoospores to become endophytic in four host red algae and neither kelp became endophytic in two non-filamentous red algae. There was a strong seasonal component regarding infectiousness that is associated with the changes in fron erosion in U. pinnatifida from April to June. At the same time, L. religiosa showed no significant changes in frond erosion, and there were no apparent changes in infection levels in the two species they were able infect. This study indicated clear differences between two keip species with regard to their symbiotic relationship to red algae in terms of host specificity and preference of kelp gametophytes.

Fucoidan Extract from Laminaria religiosa Suppresses Ischemia-induced Apoptosis and Cell Proliferation in the Hippocampus of Gerbils

  • Fucoidan has been shown to exhibit a host of biological activities, including anti-coagulant, anti-thrombotic, anti-tumourigenic, anti-inflammatory, anti-viral, anti-complementary and neuroprotective effects. In the present study, we attempted to determine the effects of Fucoidan on both apoptosis and cell proliferation in the hippocampal CA1 region and the dentate gyrus of gerbils after the induction of transient global ischemia. This experiment involved the use of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay as well as immunohistochemisty for caspase-3 and 5-bromo-2'-deoxyuridine (BrdU). The monosaccharide composition of the purified Fucoidan which had been extracted from Laminaria religiosa was utilized in this study. The present study clearly induces that apoptotic cell death and cell proliferation in the gerbil's hippocampal regions increased significantly following the induction of transient global ischemia and the results of this study also indicate that Fucoidan exerted a suppressive effect on this observed ischemia-induced increase in apoptosis within the CA1 and dentate gyrus, and also suppressed cell proliferation in the dentate gyrus.

Comparison of Alginic Acid Yields and Viscosity by Different Extraction Conditions from Various Seaweeds (Laminaria religiosa, Hizikia fusiforme, and Undaria pinnatifida) (추출조건에 따른 해조류의 알긴산 수율과 점도)

  • Alginates were extracted from the Laminaria religiosa, Undaria pinnatifida, and Hizikia fusiforme by using four different extraction methods and compared the yields of alginate. Acid-alkali soluble alginate (AASA) extraction method from Undaria pinnatifida resulted in the best yield of alginate among the seaweeds. The optimal condition for extracting alginate from Laminaria religiosa was 0.4 N H$_2$SO$_4$ and 3% NaCO$_3$ concentrations at the AASA extraction method. The alginate yields of hot water extractable material (HWEM) water soluble alginate (WSA), alkali soluble alginate (ASA) and AASA in Hizikia fusiforme were 18.6, 4.7, 22.5 and 26.5%, respectively. The alginates manufactured by the WSA extraction method showed more bright color than those of the ASA and AASA extraction methods. The alginate prepared by the ASA extraction method from Hizikia fusiforme showed the higher viscosity than that of the ASA extraction method. The molecular weight of the alginate from Hizikia fusiforme was 33.3 kDa to 121.6 kDa depending on the extraction method.

  • Rheological properties of partially purified fucoidans of Laminaria religiosa, sporophylls of Undaria pinnatifida, Hizikia fusiforme, and Sargassum fulvellum in Korea were investigated, The fucoidans produced aqueous solutions of low apparent viscosity with pseudoplastic flow behavior. The viscosity of $5\%$ solution of the fucoidan from sporophylls of U. pinnatifida was 2.2 mPa.s. The activaton energies of L. religiosa, sporophylls of U. pinnatifida, H. fusiforme and S. fulvellum were $8.269\times10^6,\;7.498\times10^6,\;7.141\times10^6,\;and\;6.677\times10^6\;J/Kg.mol$ at 1,500 1/s of shear rate, respectively. The viscosity of the fucoidan solution was increased by the addition of sugar, but that was not changed by addition of NaCl.

  • The possibility of utilizing Porphyra yezoensis, Undaria pinnatifida and Laminaria religionsa into algae-tea having biological functionality was investigated by analysis of functional component, experimentation for low viscosity and various pretreatments. In water soluble fraction of powdered algae extracted for 3 minutes with 30 times of hot wate, major functinal components were composed of 1.53% porphyran and 170.04mg% taurine in P. yezoensis, 1.09% fucoidan and 1.18% sodium sodium alginate in U. pinnatifida, and 1.28% fucoidan, 1.99% soidum laginate and 371.25mg% iodine in L.religiosa, on dry basis. For lowering viscosity and masking off-flavor in each water soluble fraction, it was desirable that P. yezoensis was washed for 12 hours in sea water and 30 mimutes in fresh water, dried at 3 to 5$^{\circ}C$, powdered to size of 30 mesh nd then roasted for 3 minutes at 12$0^{\circ}C$, and that both U. pinnatifida and L. religiosa were washed, semidried to 40~50% moisture content, heated for 40 minutes at 12$0^{\circ}C$ by autoclave, dried, powdered to size of 30 mesh and then roasted for 5 minutes at 11$0^{\circ}C$.

  • There have been many problems like plasmolysis in the ultrastructural studies with kelps. The present study was conducted with series of different fixatives and buffer solutions and several resins to solve the plasmolysis phenomena. The fixative of 1470 mosmol($3\%$ glutaraldehyde, $2.4\%$ paraformaldehyde in 0.05M cacodylate buffer with 0.2M sucrose and $0.5\%$ caffeine at pH of 7.2) showed the least frequency of plasmolysis. The relationship between the concentration of primary fixatives and the frequency of plasmolysis was not found. With above fixative the possibility of plasmolysis during infiltration period was not identified using ultra-low viscosity resin. Generally, the kelp(Laminaria religiosa) contains a lot of mucilage, which interupts the infiltration of fixative. The possible physiological mechanisms related with mucilage secretion may be unable to fix the region of plasmalemma properly.

Isolation and Purification of Fucoidans from Laminaria religiosa and Undaria pinnatifida in Korea (한국산 다시마 및 미역으로 부터 Fucoidan의 추출 및 정제)

  • The objective of this research was to characterize fucoidans isolated from Laminaria religiosa and Undaria pinnatifida in Korea to obtain basic data for Production of soluble dietary fiber materials with biological functionality. Fucoidans were successively extracted 3 times at $65\%$ for 1hr with arid solution of pH 2.0, and cetylpyridinium chloride was used for partial purification. The yields of partially purified fucoidans were $2.71\%$ for L. religiosa, $6.65\%$ for sporophylls of U. pinnatifida and $0.40\%$ for blade of U. pinnatifida. The yield from sporophylls of U. pinnatifida was highest among the sample tested, whereas the yield from blade of U. pinnatifida was lowest. It appeared that the fuconidans content in different parts of U. pinnatifida varied. Partially purified fucoidans were separated into 3 fractions by DEAE-Sephadex A-25 ion exchange column and the maior fractions were refractionated with tractional precipitation with ethanol. $60-70\%$ ethanol precipitated fractions of 1. religiosa and sporophylls of U. pinnatifida turned out to be homogeneous by cellulose acetate electrophoresis and gel filteration chromatography. The molar ratios of fucose, galactose, and sulfate in the purified fucoidans(ethanol precipitated fractions) were 1 : 0.31 : 2.43 for L. religiosa and 1 : 0.97 : 1.99 for sporophylls of U. pinnatifida. The averaged molecular weights of the purified fucoidans from L. religiosa and sporophylls of U. pinnatifida were 31,000 and 38,000, respectively.

Artificial Reestablishment of the Kelp and Red Algal Symbiosis

  • A type of symbiosis was previously described from nature in which the gametophytes of Laminariales were endophytic in filamentous red algae. Here we reconstruct this symbiosis for the first time in laboratory culture using zoospores of the kelp, Undaria pinnatifida, and the red alga, Aglaothamnion oosumiense. Zoospores of U. pinnatifida readily attached to A. oosumiense. In 48 h these spores germinated and the initial germ tube penetrated into the host cell wall leaving only an empty zoospore wall outside the host. Within ten days, four to five-celled endophytic gametophytes were present. Zoospores of Laminaria religiosa which were also inoculated into cultures of A. oosumiense rarely attached to the red alga and never became endophytic. Within ten days the free-living gametophytes of L. religiosa on cover slips became fertile and produced young sporophytes. These observations demonstrate the ability of U. pinnatifida to become endophytic, and show differences in host specificity among kelp species.
