Effect of Environmental Conditions on Growth and Quality of Log-cultured Oak Mushroom(Lentinus edodes(Berk) Sing) under Protected Cultivation (시설내 환경조건이 원목재배 표고버섯의 생육과 품질에 미치는 영향)

  • Since the growth and quality of oak mushrooms are sensitively affected by environmental conditions, an adequate environmental control is very essential to improve the yield and quality under protected cultivation. In this study, the diameter, thickness, length and hardness of cap and stipe, the dry weight, fresh weight and moisture content of the mushrooms were analyzed under the environmental conditions such as night air temperatures (control, $5{\~}11^{\circ}C$ and $12{\~}18^{\circ}C$), relative humidities in the daytime (control, humidifying for 6 h and 12 h), and wind velocities (control, fan-operation for 6 h and 12 h). At a lower night temperature, the quality of the mushrooms was improved, and considered to be optimum at $3{\~}8^{\circ}C$ in terms of fresh weight and quality. At $70{\~}80{\%}$ of relative humidity in the daytime, the fresh weight was increased, but the quality decreased. Considering the quality, the relative humidity in the daytime is adequate at around $50{\%}$. At 6 h of maintaining the wind velocity at $1{\~}2\;my{\cdot}s^{-1}$, the growth and quality were better, but the lightness became higher at 12 h although the fresh weight was reduced by evaporation. It is concluded that even though both of the yield and quality are not easily improved at a same time, optimum approaches should be required by using the relationship among environmental factors and the growth and quality.

The Effect of UV-B Irradiation and Hot-Air Drying on the Vitamin D$_2$Content of Shiitake Mushroom (Lentinus edodes) (자외선 B파 조사와 열풍건조가 표고버섯의 비타민 D$_2$함량에 미치는 영향)

  • Fresh Shiitake mushrooms were irradiated by UV-B or dried by using hot air to increase the vitamin D$_2$content. Vitamin D$_2$content was determined by high-performance liquid chromatography Irradiating the inside of the mushroom cap was more effective in increasing vitamin D$_2$content than irradiating the outside of the mushroom cap. Irradiation of UV-B at 0, 2.5, 5.0, 7.5 and 10 J/㎠ increased the content of vitamin D$_2$in the mushroom to 21.51, 120.78, 144.38, 168.08, and 187.69 $\mu\textrm{g}$/g, respectively. Vitamin D$_2$content of Shiitake mushrooms with irradiation of 10 J/㎠ decreased from 187.69 to 98.78 $\mu\textrm{g}$/g by 6 hours of hot-air drying. When three kinds of drying method (UV-B irradiation followed by hot-air drying, hot-air drying, and sunlight drying) were compared, the combination of UV-B irradiation and hot-air drying was the most effective in terms of increasine the vitamin D$_2$content in the mushroom.

Studies on the Extracts Preparation of Korean Shiitake Mushroom(Lentinus edodes) (한국산 표고버섯 Extract 제조에 대한 연구)

  • Pileus and stipe extracts of shiitake mushroom were prepared with various ethanol concentration by different extraction time at $60^{\circ}C$. Yields, total reducing sugars, free amino acids and nucleotides in ultrafiltrated extracts were analyzed. Yields were higher in hot water extracts but there was no difference depending on changes of extraction time. Total reducing sugar contents got higher by hot water extraction than by solvent extraction. In hot water extracts of the pileus and stipe, reducing sugar content were in the range of $416.18{\sim}488.18\;mg%\;and\;435.37{\sim}452.12\;mg%$, respectively. Threonine+serine, glutamic acid, lysine and arginine were dominent in the free amino acids pool of raw material. The contents of free amino acids in hot water extracts of pileus and stipe were about 528.46mg% in 2 hr and 221.01 mg% in 3 hr. The proportion of bitter amino acids in extracts to total free amino acid contents was in the range of $16{\sim}29%,\;35{\sim}37%$ in pileus and stipe extracts, respectively. Nucleotides contents were higher in pileus than in the stipe. When the 25% ethanol solution was used for extraction solvent, nucleotides contents in pileus and stipe extracts was high.

Drying and Antioxidant Characteristics of the Shiitake (Lentinus edodes) Mushroom in a Conveyer Type Far-Infrared Dryer (컨베이어 원적외선 건조기를 이용한 표고버섯의 건조 및 항산화 특성)

  • In an attempt to find ways of improving the quality of dried Shiitake mushroom, this study compared a conveyer-type far-infrared drying method with a traditional heated air drying method. The conveyer-type far-infrared dryer was performed at air velocity of 0.6 and 0.8 m/s under drying air temperature of 60, 70 and $80^{\circ}C$, respectively. Drying characteristics, antioxidant activities and the antioxidant compounds of Shiitake mushroom dried by far-infrared dryer were investigated. Generally, drying rate with the conveyer-type far-infrared drying method was faster than that with the traditional heated air drying method. In the conveyer type far infrared drying method, drying rates were increased with increasing temperature and air velocity. The loss of antioxidant activities during the conveyer-type far-infrared drying method at 60-0.6, 60-0.8, and $70^{\circ}C-0.6m/s$ was less than the traditional drying method. However, the loss of antioxidant activities at 70-0.8, 80-0.6, and $80^{\circ}C-0.8m/s$ was higher than that of the traditional drying method. Therefore, the conveyer type far infrared drying conditions with below $70^{\circ}C$ and 0.6 m/s air velocity may produce dried Shiitake mushroom with relatively higher antioxidant activities and antioxidant compounds.

Antioxidant activity and quality characteristics on the maturation period of the soy sauce with Gastrodia elata and oak mushroom (Lentinus edodes) (천마 및 표고버섯 첨가 전통간장의 숙성기간별 품질특성 및 항산화 활성)

  • This study was performed to investigate the changes in antioxidant activity and quality characteristics of soy sauce with Gastrodia elata (GK) and oak mushroom (MK) in maturation period to develop a new Kanjang product with improved functional and sensory characteristics. The pH was decreased, and the total acidity was higher in GK at 25-day maturation period. Although the total sugar content increased at 15 days with GK, it was reduced after 20 days. The reducing sugar, however, showed no notable differences after 20 days. The protease activities of GK was rapidly increased after 20 days. The total polyphenol and flavonoid contents tended to be similar and dramatically increased after 15 days. On the other hand, the DPPH radical scavenging activity of MK was higher than that of GK. These results showed that the 20-day maturation period of GK greatly enhanced its antioxidant activity and quality characteristics. It is thus expected to be used as functional materials.

  • This study demonstrated that the physiochemical properties and anti-oxidants activities of soy sauce with Oak mushroom (MK) and Astragalus memyranaceus (AK) improved in sensory acceptability and functionality during the ripening period of 30 days. The pH content was reduced and the total acidity content was increased with time. The total sugar of AK and MK was higher than traditional soy sauce (TK), and the changes in amino nitrogen of the AK increased dramatically from 0.50 to 0.98%. During the antioxidant experiments, the total polyphenol and flavonoid contents increased at a steady rate and studies showed that the DPPH free radical scavenging activities were higher in all treatments except in the traditional soy sauce. According to the results of this study, the antioxidant activities of the AK and MK were higher than the TK, while the preference for the AK and MK was higher than the TK. In conclusion, the AK could be used as a functional soy sauce.

Drying characteristics and physical properties of medicinal and edible mushrooms (약용버섯과 식용버섯의 건조방법에 따른 품질특성)

  • In this study, the drying characteristics of medicinal (Ganoderma lucidum, Phellinus linteus) and edible (Pleurotus eryngii, Lentinus edodes) mushrooms were investigated. Their shrinkage ratios, Hunter's color values, and rehydration ratios were evaluated. Mushrooms were cut with rectangular (1 cm) and square ($1{\times}1cm$) shapes, and dried in the room temperature and at $50^{\circ}C$ and $70^{\circ}C$ using hot-air dryer. Initial moisture contents of edible mushrooms were higher than those of medicinal mushrooms, whereas final moisture contents were vice versa. Drying rate was the highest when drying at $70^{\circ}C$ (p<0.05). Moreover the drying rate of square slices was higher than that of rectangular slices (p<0.05). The shrinkage ratios of both medicinal mushrooms and edible mushrooms were the least when drying at $50^{\circ}C$ and $70^{\circ}C$, respectively (p<0.05). The changes of color values were less in the medicinal mushrooms dried in the room temperature than in the hot-air dried medicinal mushrooms (p<0.05). However, in case of edible mushrooms, the changes of color values were less in the hot-air dried mushrooms (p<0.05). Rehydration ratios of medicinal mushrooms dried at $50^{\circ}C$ was the highest (p<0.05). In contrast, rehydration ratios of edible mushrooms was the highest when drying in the room temperature (p<0.05). Thus, the hot-air drying at $50^{\circ}C$ would be suggested as the efficient drying method for both medicinal mushrooms and edible mushrooms.

Study of Lipoprotein Lipase Inhibitory Activity of Anti-obesity Herb Extracts (항비만소재의 lipoprotein lipase 억제 작용 연구)

  • In this study, we evaluated the lipoprotein lipase (LPL) inhibitory activity of 11 water extracts derived from Cinnamomum cassia Blume, Sarcodon aspratus, Cordyceps militaris, Crataegus pinnatifida Bunge, Corni fructus, Allium cepa, Coix lacryma-jobi, Plantago asiatica L., Lentinus edodes, Rosa rugosa, and Foeniculum fructus. The results of the LPL secretion and activity assay showed Sarcodon aspratus (NE) extract have an LPL secretion inhibitory acitivity. The cause of reduction in LPL secretion after NE treatment was investigated using molecular biology methods. NE treatment affected the LPL content in cells, but did not affect LPL mRNA expression. It also increased the mRNA expression level of sortilin-related receptor LDLR class A (SorLA), a receptor that induces endocytosis and intracellular trafficking of LPL. Finally, cell fractionation revealed that NE treatment induced the expression of CCAAT-enhancer-binding protein beta ($C/EBP{\beta}$), a SorLA transcription factor, in the nuclei of 3T3-L1 adipocytes. These results show that NE's anti-obesity effect involves inhibition of LPL secretion through $C/EBP{\beta}$-mediated induction of SorLA expression.

Inhibitory Effect of Shiitake Mushroom Extracts Cultured in Aloe-Supplement on Invasion/Migration of TNF-α-Induced MDA-MB-231 Breast Cancer Cells (알로에 배지에서 재배한 표고버섯 추출물의 종양괴사인자로 유도된 유방암세포 MDA-MB231에 대한 전이 저해 활성)

  • To investigate the anti-cancer effect of shiitake mushrooms (Lentinus edodes) cultured in aloe-supplement, we treated extract of shiitake mushroom cultured in aloe-supplement (ESA) to MDA-MB-231 human breast cancer cells. ESA-treated MDA-MB-231 cells showed decreased growth rate in XTT assay. In addition migration/invasion was noticeably inhibited by ESA in TNF-${\alpha}$-treated MDA-MB-231 cells. Western blot analysis showed that the molecular mechanism of cell migration/invasion was mediated by reduced intercellular adhesion molecule-1 expression via p-ERK signal transduction pathways. We found ESA had inhibition activity against cellular growth and migration/invasion. Taken together, ESA has putative anti-cancer activity against human breast cancer.

Physiological Activities of Extract from Edible Mushrooms (주요 식용버섯 추출물의 생리활성 효과)

  • This study was conducted to investigate the physiological activity of extracts of fresh mushrooms. The components were extracted by hot water; subsequently, the hot-water extract was subjected to 60% ethanol precipitation to yield high-molecular-weight (HMW) and low-molecular-weight (LMW) fractions. Total polyphenol contents, $\beta$-glucan contents, electron-donating ability (EDA), superoxide dismutase (SOD)-like activity, nitrite-scavenging activity, fibrinolytic activity, nitric oxide (NO) production, and inhibition of NO production of the mushroom extracts were measured using lipopolysaccharide (LPS)-stimulated murine macrophages, RAW 264.7 cells. The extracts of Lentinus edodes (Berk.) Singer and Pleurotus ostreatus (Fr.) Kummer contained the highest levels of $\beta$-glucan (33.5% and 25.57%, respectively). Further, the LMW fractions of the Phellinus linteus contained the highest levels of polyphenols (233.23 mg/g). The EDA of LMW fractions (10 mg/mL) of the Phellinus linteus and Agaricus bisporus were 80.74% and 51.35%, respectively. Further, SOD-like activities of the LMW fractions were high as compared to those of the HMW fractions. Nitrite-scavenging activities of the LMW fractions (pH 1.2; concentration, 10 mg/mL) of the Phellinus linteus and Pleurotus ostreatus (Fr.) Kummer were 75.95% and 41.05%, respectively. The fibrinolytic activity of the LMW fractions of all mushrooms showed no enzyme activity by fibrin plate assay. The fibrinolytic activity of the extracts of Tricholoma matsutake was the greatest inhibitory activity at 60.4%. Further study revealed that the mushroom extracts exhibited anti-inflammatory effects on RAW 264.7 cells. The LMW fraction ($500\;{\mu}g/mL$) of the Phellinus linteus considerably inhibited NO production (100%).
