Studies on the grassland Development in the Forest V. Effect of sharing degrees on the early growth characteristics and winter survival of main grasses (임간초지 개발에 관한 연구 V. 차광정도가 주요 초목의 초기생육과 월동에 미치는 영향)

  • This field experiment was conducted to determine the effects of shading degrees (0 : full sunlight, 25, 50 and 75%) on the early growth characteristics and winter survival of grasses. For the test different artificial shading houses were established and pasture species used were orchardgrass, timothy and perennial ryegrass. The experiment was performed at Livestock Experiment Station in Suweon, and sowing date was Aug. 25, 1984. The results are summarized as follows : 1. Generally, root length, new root length, number of new root and tillers of grasses were increased at 0% (full sunlight) of shading, followed by 25, 50 and 75% of shading degrees, in that order. Plant length and leaf elongation, however were increased at 50% of shading, followed by 75, 25 and 0%. 2. The percentage of winter killing of grasses was significantly (P<0.05) increased as the shading degrees increased. Also the percentages of winter killing were 10.1% in orchardgrass, 9.5% in timothy and 14.2% in perennial ryegrass, respectively. 3. A significant negative correlations were observed in the relationship between winter killing and tillers, new root length and number of new root of grasses. However, there were positive correlations between winter killing and plant length or leaf elongation under shading conditions. 4. As the shading degrees are increased by 0, 25, 50 and 75%, the NAR of grasses was tended to be decrease to 10.98, 11.52, 9.81 and 6.12 g/$m^2$/day, in that order. Also there was a negative correlation (r=-0.5943*) between winter killing and NAR of grasses, and NAR of orchardgrass, timothy and perennial ryegrass were 8.58, 8.53 and 11.71 g/$m^2$/day, respectively.

Some Characteristics of Interspecific and Intergeneric Hybrids in Herbage Grasses (禾本科 牧草의 種屬間雜種 Hybrid ryegrass와 Festulolium의 生育特性)

  • Even though grass species are one of major importance in livestock industry, the response of intergeneric hybridization of Lolium and Festuca and interspecific hybridization of Lolium m.X Lolium p. to Korean climatic condition has not been adequately documented. This reasearch was conducted to examine the growth characteristics and dry matter yield of Festulolium, cv. Felopa and C-Stamm N 66 and hybrid ryegrass, cv. Maja and Tetilelite. 1. The early growth of Italian ryegrass was better than that of hybrid ryegrass and Festulolium. The plant length and height of Italian ryegrass were longer/taller than those of hybrid ryegrass and Festulolium, but the number of tillers of Italian ryegrass was less than that of hybrid ryegrass and Festulolium. 2. Festuloliu and hybrid ryegrass were more tolerant than Italian ryegrass to winter killing, 70-80% of tillers was survived. 3. After wintering, early spring growth of hybrid ryegrass and Festulolium were better than that of Italian ryegrass. Dry matter yield of hybrid ryegrass and Festulolium were more than that of Italian ryegrass. 4. Spikes of hybrid ryegrass and Festulolium (C-Stamm N66) were more than those of Italian ryegrass. Seed yield per $m^2$ of hybrid ryegrass and Italian ryegrass were almost same, but that of Festuloliu was lower.

  • Purpose: The purpose of this study is to analyze the characteristics of animal carcasses treatment and quarantine management of infectious diseases of livestock, and to present the effectiveness of quarantine and follow-up management of livestock infectious diseases. Method: Actually, a livestock epidemic occurred and the burial site and burial management facility that treated livestock carcasses were visited on-site to analyze the management status and problems. Result: The burial method of livestock outbreaks of livestock infectious diseases and the long-term follow-up management accordingly requires a large amount of manpower and budget to be spent. For example, it can be seen that it is an inefficient quarantine system. Conclusion: It is necessary to review plans for the establishment of integrated livestock infectious animal carcasses treatment facilities at the level of metropolitan cities, districts, and municipalities with the government, where there is no fear of secondary infectious disease transmission and no follow-up management is required.

Validation of method for chemical treatment of pig carcasses for the control of livestock infectious diseases (가축 전염병 관리를 위한 돼지 사체의 화학적 처리 방법)

  • This is a study on the improvement of the chemical treatment method of the livestock carcass treatment newly introduced in the livestock infectious disease prevention method in order to improve the problems of the existing burial-centered carcass treatment method when a livestock infectious disease occurs. It was conducted to establish detailed treatment standards for the chemical treatment method of pig carcasses based on the results of proof of the absence of infectious diseases in pigs. After inoculating pig carcasses with 10 pathogens (6 viruses [FMDV, ASFV, CSFV, PCV2, PRRSV, PEDV] and 4 bacteria [Lawsonia intracellularis, Clostridium perfringens type C, E. coli, Salmonella Typhimurium]) It was treated at 90℃ for 5 hours in a potassium hydroxide (KOH) liquid solution corresponding to 15% of the body weight. This method liquefies all cadaveric components and inactivates all inoculated pathogens. Based on these results, it was possible to prove that chemical treatment of pig carcasses is effective in killing pathogens and is a safe method without the risk of disease transmission. Although there are problems to be solved in the processing and operation of the chemical treatment products of livestock carcasses, the chemical treatment method of livestock carcasses can be suggested as an alternative to the current domestic burial-centered livestock carcass treatment method, preventing environmental pollution, and contributing to public health.

Determination of Heat Killing Temperature of Birdsfoot trefoil and Italian ryegrass (버즈풋 트레포일 및 이탈리안 라이그라스의 치사온도 결정)

  • To determine lethal temperature of birdsfoot trefoil(BFT) and italian ryegrass(IRG) at heat-stressed conditions, seedlings grown in a small pots fur 4 weeks were subjected to different temperature regimes of heat treatment. No apparent damage was observed BFT and IRG were treated at 45, 50 or $60^{\circ}C$ for 1 h. And also heat treatments at 60, 65 and $70^{\circ}C$ for 1 h, both of them were withered and showed damage symptom on their leaves but it was not lethal conditions for the whole plants. By contrast, most of plants were prominently withered within one day after heat treatment at $80^{\circ}C/60min$. When BFT was exposed to $80^{\circ}C/60$ min, they were died within 6 days but there was found that new shoots were regenerated from the plants that had been treated at $80^{\circ}C$ within 55 min. IRC was also died within 2 days that exposed to $80^{\circ}C/20$ min but there was found that new shoots were regenerated from the plants that had been treated at $80^{\circ}C$ within 15 min. These results indicate that heat killing temperatures of BFT and IRG plants are $80^{\circ}C/60$ min an $80^{\circ}C/20$ min respectively. Simple viability assay system established in this study will be useful for selection and characterization of heat-tolerant transgenic BFT and IRG plants.

Determination of Heat Killing Temperature of Orchardgrass(Dactylis glomerata L.) (오차드그라스 (Dactylis glomerata L.)의 치사온도 결정)

  • To determine lethal temperature of orchardgrass (Dactylis glomerata L. cv. Janbeol 102) developed in Korea at heat-stressed conditions, seedlings grown in a amall pots for 4 weeks were treated at $45^{\circ}C$, $50^{\circ}C$ or $55^{\circ}C$ for 1 h. Heat treatments at $60^{\circ}C$ and $65^{\circ}C$ for 1 h, several plants were withered and showed damage symptom on their leaves. When the plants were exposed to $70^{\circ}C$ for 1 h, most of leaves were severely withered, but it was not lethal conditions for the whole plants. By contrast, most of plants were died within one day after heat treatment at $80^{\circ}C$ for 1h. Furthermore, plants exposed to $80^{\circ}C$ for 55 min were also died within 7 days. It was found that new shoots were regenerated from the plants that had been treated at $80^{\circ}C$ within 50 min. These results indicate that heat treatment at $80^{\circ}C$ for 55 min is an optimum condition to distinguish the lethality of orchardgrass plants. Simple viability assay system established in this study will be useful for selection and characterization of heat-tolerant transgenic orchardgrass plants.

Application of chemical treatment for cattle and chicken carcasses for the control of livestock infectious diseases (가축 전염병 발생에 따른 소와 닭 사체의 화학적 처리 방법의 적용)

  • In the event of an outbreak of a livestock epidemic, it has been considered that the existing burial-centered carcass disposal method should be improved ecofriendly for prevention of leachate and odors from burial basically in regard of pathogen inactivation. Therefore, the aim of this study is whether it was possible to treat the carcass of cattle and chickens using the chemical carcass treatment method. It was conducted to establish detailed treatment standards for the chemical treatment method of cattle and chicken carcasses based on the results of the proof of the absence of infectious diseases in cattle chickens. After inoculating cattle carcass with 10 pathogens (foot and mouth disease virus, bovine viral diarrhea virus, Mycobacterium bovis, Mycobacterium avium subsp. Paratuberculosis, Brucella abortus, Bacillus anthracis, Clostridium chauvoei, Clostridium perfringens, Escherichia coli, and Salmonella Typhimurium) and chicken carcasses with low pathogenic avian influenza virus, Clostridium perfringens type C, E. coli and Salmonella Typhimurium, these were treated at 90℃ for 5 hours in a potassium hydroxide liquid solution corresponding to 15% of the body weight. This method liquefies all cadaveric components and inactivates all inoculated pathogens by PCR and culture. Based on these results, it was possible to prove that chemical treatment of cattle and chicken carcasses is effective in killing pathogens and is a safe method without the risk of disease transmission. The chemical treatment method of livestock carcasses can be suggested as an alternative to the current domestic burial-centered livestock carcass treatment method, preventing environmental pollution, and contributing to public health.

Comparative Genomics Approaches to Understanding Virulence and Antimicrobial Resistance of Salmonella Typhimurium ST1539 Isolated from a Poultry Slaughterhouse in Korea

  • Non-typhoidal Salmonella (NTS) is one of the most frequent causes of bacterial foodborne illnesses. Considering that the main reservoir of NTS is the intestinal tract of livestock, foods of animal origin are regarded as the main vehicles of Salmonella infection. In particular, poultry colonized with Salmonella Typhimurium (S. Typhimurium), a dominant serotype responsible for human infections, do not exhibit overt signs and symptoms, thereby posing a potential health risk to humans. In this study, comparative genomics approaches were applied to two S. Typhimurium strains, ST1539 and ST1120, isolated from a duck slaughterhouse and a pig farm, respectively, to characterize their virulence and antimicrobial resistance-associated genomic determinants. ST1539 containing a chromosome (4,905,039 bp; 4,403 CDSs) and a plasmid (93,876 bp; 96 CDSs) was phylogenetically distinct from other S. Typhimurium strains such as ST1120 and LT2. Compared to the ST1120 genome (previously deposited in GenBank; CP021909.1 and CP021910.1), ST1539 possesses more virulence determinants, including ST64B prophage, plasmid spv operon encoding virulence factors, genes encoding SseJ effector, Rck invasin, and biofilm-forming factors (bcf operon and pefAB). In accordance with the in silico prediction, ST1539 exhibited higher cytotoxicity against epithelial cells, better survival inside macrophage cells, and faster mice-killing activity than ST1120. However, ST1539 showed less resistance against antibiotics than ST1120, which may be attributed to the multiple resistanceassociated genes in the ST1120 chromosome. The accumulation of comparative genomics data on S. Typhimurium isolates from livestock would enrich our understanding of strategies Salmonella employs to adapt to diverse host animals.

Seed Production Studies in Italian Ryegrass ( Lolium multiflorum Lam. Itailcum ) I. Effect of seeding time and seed rates of Italian ryegrass, cv. Tetrone on seed production (이탈리안 라이그라스의 종자생산에 관한 연구. 제1보. 파종기와 파종량이 종자에 미치는 영향)

  • In order ti find out the optimum seeding time(OST) and optimum seed rate(OSR) of Italian ryegrass on seed production, this studies with tetraploid cv. Tetrone were carried out on the experimental field of Livestock Experiment Station. Treatments included seed rates of 1, 2, 3 and 4 kg per 10a and combined with seeding time on 20, 30 Aug, 9, 19 and 29, September 1983. Seeds were sown in rows 50 cm apart and were spaced in a continuous line with width of 15 cm within the rows. The results are summarized as follows: 1. Autumn tillers could be classified into three groups from winter-killing point of view, namely winter-killing completely, damaged growing point only and living tillers. 2. The young inflorescence-bearing stem in Italian ryegrass which were sown earlier than 9. September were more susceptible to winter killing. Tiller buds in those stems which originated from an axillary buds at the stem base within senescent leaf-sheaths emerged lately in spring, and consequently heading was delayed, culm length shortened and seed yield reduced. 3. Tiller buds which originated from damaged growing point only and living tillers in moderate shoots emerged early in spring and those tillers became mainly spike-bearing culm. 4. The emergence-time of tillers influenced on culm-, spike- length and ripenness more than seeding time and seed rate. 5. Seed yield was mostly affected by the number of spikes per unit area. 6. For the safety of over-wintering and enough spikes on seed production, OST and OSR at Suweon were the last part of September and 2-3 kg per 10a, respectively. Especially OSR was 2 kg per 10a for early and 3 kg per 10a for late OST.
