Effect of Hot Water Soluble Extract from Angelicae Radix on the Lipid Metabolism and Antioxidant Defense System (당귀열수추출물이 종류가 다른 지방식이를 공급한 흰쥐의 지질대사와 항산화계에 미치는 영향)

  • The aim of this study is to investigate the effects of hot water soluble extract from Angelicae Radix on the components of serum and liver and the effects on the antioxidant system. For this purpose, five experimental groups were set up. And for fat source, perila oil enough with unsaturated fatty acid and beef tallow enough with saturated fatty acid were supplemented to the rats together with hot water soluble extract from Angelicae Radixs. Five experimental groups kept eight Sprague-Dawley rats respectively. They were CO group supplemented with basic diet of AIN-93, PO group supplemented with perila oil, POA group supplemented with perila oil and hot water soluble extract from Angelicae Radix, BT group supplemented with beef tallow, and BTA group supplemented with beef tallow and hot water soluble extract from Angelicae Radix. The results were; 1) Final weight, weight gain, fluid intake and FER were not different significantly among the experimental groups, 2) Significant difference of food intake was observed(p<0.05) in BTA group only, 3) No significant difference was observed in serum total lipid, serum triglyceride and HDL cholesterol among experimental groups. Serum total cholesterol and LDL cholesterol were significantly low(p<0.05) in the group supplemented with beef tallow which was with hot water soluble extract from Angelicae Radix (BTA group). 4)Liver total cholesterol in liver was low in groups supplemented with perila oil and hot water soluble extract from Angelicae Radix. In summary, hot water soluble extract from Angelicae Radix did not affect the weight gain, fluid intake and food efficiency ratio among the experimental groups, but had an effect of lowering food intake, serum total cholesterol and serum LDL cholesterol significantly in the groups which were supplemented with beef tallow and hot water soluble extract from Angelicae Radix. The effect of lowering liver total cholesterol with the supplementation of hot water soluble extract from AnRelicae Radix was observed in perila oil group only. The effect of lowering cholesterol with the supplementation of hot water soluble extract from Angelicae Radix was observed both in serum and in liver.

Blood lipid lowering effect of butanol extract from Galkun-Whanglyeon-Whanggum-Tang (갈근황련황금탕 부탄올 추출물의 혈중에서의 지질 개선효과)

  • The Galkun-Whanglyeon-Whanggum-Tang, an officially standardized mixture of traditional herbal medicines used in Korea and China, consists of Pueraria lobata, Scullellaria baicalensis, Coptis chinensis and Glycyrrhiza uralensis at a ratio of 6:9:3:2.4. In this study, we evaluated the effect of lowering lipid accumulation in blood by treatment of Galkun-Whanglyeon-Whanggum-Tang in Apo E(-/-) atherosclerotic animal model. ApoE/mice fed with 1.25% cholesterol, 7.5% cocoa butter and 0.5% sodium cholate diet were orally given vehicle or Galkun-Whanglyeon-Whanggum-Tang(10, 100 and 300 mg/kg/day) for 12 weeks. Serum levels of triglyceride(TG), total cholestrerol(TC), low-density lipoprotein(LDL) and high-density lipoprotein(HDL) were analyzed, and PPAR-${\alpha}$ and PPAR-${\gamma}$ were examined by Western blotting analysis. Galkun-Whanglyeon- Whanggum-Tang decreased serum levels of TG, TC and LDL, but not HDL in ApoE/mice. In parallel, Galkun-Whanglyeon-Whanggum-Tang treatment showed the increased activity of PPAR-${\alpha}$ and PPAR-${\gamma}$ in hepatocytes. In summary, Galkun-Whanglyeon-Whanggum-Tang can reduce lipid accumulation in blood, and this effect might be accompanied by the upregulation of PPAR-${\alpha}$ and PPAR-${\gamma}$ in Apo E(-/-) atherosclerotic mouse model.

Effects of Different Kinds of Korean Soybeans on Body Lipids and Lipid Peroxidation in Rats (다양한 콩종류가 흰쥐의 체내지질과 지질과산화에 미치는 영향)

  • This study was conducted to investigate the effect of different kinds of traditional Korean soybeans on blood and liver lipids and lipid peroxidation. SD male rats (n=48) were assigned to six different groups and provided experimental diets for 28 days. The protein source of the diet was casein, isolated soy protein (ISP), or casein plus traditional soy powders (yellow bean, huktae, jinuni or seorietae). The ISP, jinuni and seorietae diet decreased blood triglyceride compared to casein group. The ISP groups had lower hepatic total lipid and triglycerides than casein groups. The yellow bean and seorietae group had hepatic lipid lowering effects, but the effect was not found in huktae and jinuni group. The glutamate-oxaloacetate transaminase activity was significantly decreased in ISP and jinuni groups than in casein group. The glutamate-pyruvate transaminase activity was also decreased in ISP group. The results of this study suggest that jinuni, yellow bean and seorietae groups are more effective in lowering body lipids than huktae group.

Biological Effects of Allium monanthum Extracts on Lipid Metabolism, Anti-oxidation and the Production of Pro-inflammatory Cytokines in Rats Fed a High-Fat Diet

  • This study is a basic study on the development of functional substances involved in obesity prevention, lipid metabolism, and immune regulation. Male Sprague-Dawley rats were fed a high-fat diet for 10 weeks. Allium monanthum extracts (AME) were administered orally to obesity-induced rats, and their lipid-lowering, antioxidative and various types of biological effects related to the immune system were examined. Blood free fatty acid and triglyceride concentrations decreased as the dose of AME increased. Total cholesterol and LDL cholesterol concentrations in the blood decreased as the dose of AME increased. The total cholesterol concentrations in the liver of the AME-treated groups were lower than the control group. The thiobarbituric acid reactive concentrations were lower in the plasma and liver of all AME-treated groups than the control group. Plasma AST and ALT activities did not show any significant differences among the treatment groups. IL-$1{\beta}$ and IL-6 concentrations in the liver tended to decrease as the dose of AME increased. TNF-${\alpha}$ and IL-10 concentrations did now show any significant differences compared to the control group. Lower expression levels of TNF-${\alpha}$, Apo-B and Apo-E genes were found in the AME-treated groups. Taken together, these results indicate that AME may show positive effects in lipid lowering, antioxidation and anti-inflammation.

Antihyperglycemic and lipid lowering effect of Tectona grandis in alloxan induced diabetic rats

  • In India, Tectona grandis is traditionally used in the treatment of diabetes mellitus and lipid disorder. In the present study, the antihyperglycemic and lipid lowering effect of ethanolic extract of bark of Tectona grandis (TG) was evaluated using alloxan induced diabetes in rats. Alloxan was given at the dose of 140 mg/kg, i.p. After induction of diabetes, TG was administered for 42 days p. o. and simultaneously different biochemical parameters like plasma glucose, liver glycogen content, serum triglyceride, cholesterol, LDL-cholesterol and HDL-cholesterol were estimated. Diabetic control showed significant increase (P < 0.01) in plasma glucose, serum triglyceride, cholesterol, LDL-cholesterol and significant decrease (P < 0.01) in serum HDL-cholesterol and liver glycogen content. Treatment with TG showed significant reduction (P < 0.01) in plasma glucose when compared with diabetic control. The elevated levels of serum triglyceride and cholesterol levels were significantly reduced (P < 0.01) by TG. TG treatment for 42 days showed significant decrease in serum LDL-cholesterol (P < 0.01) and significant increase in serum HDL-cholesterol level (P < 0.01). Moreover, diabetic control there was significant decrease in liver glycogen content which was significantly increased (P < 0.05) by treatment with TG. Hence, from the result obtained in the present study it can be concluded that Tectona grandis has the potential to treat diabetes condition and associated lipid disorder.

The influence of obesity on the effects of spirulina supplementation in the human metabolic response of Korean elderly

  • BACKGROUND/OBJECTIVES: Spirulina, a blue-green alga, is widely produced and commercialized as a dietary supplement with bio- and immune-modulatory functions. We have previously shown that spirulina had favorable effects on lipid profiles, immune functions, and antioxidant capacity in healthy Korean elderly. Despite favorable effect of spirulina supplementation, some sub-populations have shown a poor response to supplementation. Obesity is a factor related to poor-response. Therefore, the purpose of this study was to determine the immuno-modulation, antioxidant capacity, and lipid-lowering effect of spirulina in obese and non-obese Korean elderly. SUBJECTS/METHODS: The subjects were 78 elderly aged 60-87 years. In a randomized double blind, placebo-controlled study, subjects were fed either placebo or spirulina daily, at 8 g for 12 weeks. Subjects were divided into the non-obese group and the obese group based on body mass index (BMI) criteria for Asians suggested by the International Obesity Task Force: $BMI<25kg/m^2$ (non-obese) and $BMI{\geq}25kg/m^2$ (obese). RESULTS: In the non-obese group, spirulina supplementation showed a significant lowering effect on plasma concentration of total cholesterol and LDL-cholesterol, a significant increase in interleukin (IL)-2 concentration (P < 0.01) and a significant increment (P < 0.05) in IL-2/IL-6 ratio, and a significant increase in total antioxidant status level and a significant decrease in thiobarbituric acid reactive substances level. However, these effects were not observed in the obese group. CONCLUSION: These results demonstrated that blood lipid lowering and immune and antioxidant improving response for spirulina supplement was affected by obesity in Korean elderly.

Biological Functions of Organosulfur Compounds in Allium Vegetables (Allium속 식물 유래 함유황 유기화합물의 생리적 유용성)

  • This review contains a discussion of the physiological activity of the components of Allium vegetables. Organosulfur compounds in Allium vegetables, such as ajoene, diallyl sulfides and S allylcysteine, have cancer preventive activity in chemically induced animal cancer models. They also have inhibitory effects on proliferation of cancer cells in vitro. Allium vegetables have lipid and cholesterol lowering effect, and platelet aggregation inhibitory activity that help the prevention of cardiovascular diseases. Sulfur con taining compounds, especially allicin and ajoene, have antimicrobial activities against gram negative, positive bacteria and fungi. Moreover, Allium organosulfur compounds such as S allylcysteine showed reducing effects on the senescence related symptoms including cognition. Allium organosulfur compounds have significant importance in food industry as both biologically active ingredients and savory.

Effect of Meristotheca papulosa on Lipid Concentration of Serum and Liver in Rats Fed High Fat Diet (해조류 갈래곰보가 고지방 식이를 급여한 흰쥐의 혈액 및 간장 지질 농도에 미치는 영향)

  • This study was carried out to investigate the effect of Meristotheca papulosa on lipid concentration of serum and liver in rats fed high fat diet for 4 weeks. Experimental groups ere divided into normal group (5% corn oil), high fat diet group (5% corn oil+20% lard), high fat diet and 5% or 10% Meristotheca papulosa treated group. The weights of kidney, heart and tests were not significantly different among the group, but weight of liver was significantly decreased by supplement of Meristotheca papulosa (10%). The contents of serum total lipid, trigly-ceride, total cholesterol, free cholesterol, cholesterol ester and atherogenic index were decreased, while HDL-cholesterol was increased by Meristotheca papulosa supplementation compared to high fat diet group. Meristotheca papulosa supplement has obvious effects on lowering hepatic total lipid, triglyceride and total cholesterol. Our findings suggest that Meristotheca papulosa supplement can improve lipid metabolism in high fat fed rats.

Effect of Polygonatum Odoratum on Lowering Lipid and Antioxidation (옥죽(玉竹)의 지질강하 및 항산화효과)

  • Objectives : This study was designed to examine the effects of Polygonatum odoratum EtOH ext. on lowering lipid and anti-oxidation using hyperlipidemic rat. Methods : Male rats weighting $195.21{\pm}4.93g$ were divided into 4 groups and fed high fat diet for 8 weeks. Each of 7 rats was divided into a control and sample group. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats basal diet and administered an extract of Polygonatum odoratum(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of anti-oxidative activity and tumor necrosis factor-$\alpha$($TNF-{\alpha}$). Results : 1. Concentration of plasma free fatty acid, low density lipoprotein-cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of plasma total cholesterol, triglyceride showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. However, concentration of plasma high density lipoprotein-cholesterol was not significantly different in all the treatment groups. 2. Concentration of liver total cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of liver triglyceride(TG) showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. 3. Concentration of plasma thiobarbituric acid reactive substance, and liver thiobarbituric acid reactive substance showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 4. The values of glutathione peroxidase activity showed a significant increment in all Polygonatum odoratum EtOH ext. groups than that of control group. The values of superoxide dismutase activity and catalase activity showed a significant increment in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 5. The values of plasma aspartate aminotransferase and alanine aminotransferase activities were not significantly different in all treatment groups. 6. Concentration of liver $TNF-{\alpha}$ showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Conclusions : Based on the results in this study, the Polygonatum odoratum EtOH ext. showed a positive effect in lowering lipid and anti-oxidation.

Antihyperlipidemic Activity of Several Traditional Herbal Medicines on Experimental Hyperlipidemia in Rats (고지혈증 랫트를 이용한 수종 전통 한약제의 항고지혈 효과)

  • Three herbal medicine recipes were tried on the animal model of diet induced hypercholesterolemia to screen for the lipid-lowering effect. The recipes adopted were based on the prescription for atherosclerosis-related symptoms by folk-medicine practioners. Hyperlipidemia was induced in rats by giving the high lipid/cholesterol diet for 7 days. Then, the recipes started with the normal diet. Blood was sampled at 1,2,3 and 5 week-points of the treatment, and levels of total cholesterol, triglyceride, high density lipoprotein and lower density lipoprotein were measured. Dae-shiho-tang decreased total cholesterol level significantly. Hwangryun-haedok-tang and Samhwang-sasim-tang slightly decreased total cholesterol level, although it was not statistically significant.
