Characteristics of Mutant Lines through M6 to M7 Generation Induced by Radiation in Rapeseed (Brassica napus L.) (방사선 조사에 의한 유채 M6~M7세대 변이계통의 특성)

  • To develop new varieties of rapeseed (Brassica napus L.), the seeds of three varieties, 'Naehan', 'Tammi', and 'Halla' were irradiated with proton ion beams and gamma rays with 0 to 2,000 Gy. We had selected 9 lines in $M_5$ generation, and their useful characteristics were investigated by progressing from $M_6$ to $M_7$ generation for checking uniformity and stability. The 9 lines selected in $M_5$ generation were maintained their characteristics in terms of flowering date, maturing date, and plant height through $M_6$ to $M_7$ generations. Especially, 2 lines of NP600-1-1-198-2-1 and NP1000-13-2-362-4-1 selected in $M_5$ generation derived from 'Naehan' had characteristics of early maturity and shorter stem than original variety, and they also were maintained characteristic of early maturity such as 10~11 days earlier flowering date and 6~9 days earlier maturing date through $M_6$ to $M_7$ generations. For stem length, they showed characteristics of shorter stem in 2 lines of NP600-1-1-198-2-1 line and NP1000-13-2-362-4-1 line about 16%, 25% shorter stem than original variety respectively through $M_6$ to $M_7$ generations. Furthermore, some characteristics of 2 lines compared to the original variety were similar or higher in weight of 1,000 seeds, number of branches per plant, number of siliqua per panicle, number of seeds per silique, oil contents, and oleic acid contents. The line with large and plump flowers selected in $M_5$ generation also showed large and dark yellow flowers through $M_6$ to $M_7$ generations. The lines with High oleic acid and low saturated fatty acid contents selected in $M_5$ generation were maintained characteristics through $M_6$ to $M_7$ generation and these useful characteristics were expected for developing a new variety for bio diesel uses.

Histamine Release by Hydrochloric Acid is Mediated via Reactive Oxygen Species Generation and Phospholipase D in RBL-2H3 Mast Cells

  • In order to investigate the underlying mechanism of HCI in oesophagitis, the inflammatory response to HCI was observed in RBL-2H3 mast cells. Rat basophilic leukemia (RBL-2H3) cells were used to measure histamine release, arachidonic acid (AA) release, reactive oxygen species (ROS) and peroxynitrite generation induced by HCI. Exogenous HCl increased the level of histamine release and ROS generation in a dose dependent manner, whereas it decreased the spontaneous release of [$^3$H] M and the spontaneous production of peroxynitrite. Mepacrine (10 $\mu$M), oleyloxyethyl phosphorylcholine (10 $\mu$M) and bromoenol lactone (10 $\mu$M) did not affect both the level of histamine release and ROS generation induced by HCI. U73122 (1 $\mu$M), a specific phospholipase C (PLC) inhibitor did not have any influence on level of histamine release and ROS generation. Propranolol (200 $\mu$M), a phospholipase D (PLD) inhibitor, and neomycin (1 mM), a nonspecific PLC and PLD inhibitor, significantly inhibited both histamine release and ROS generation. Diphenyleneiodonium (10 $\mu$M), a NADPH oxidase inhibitor, and tiron (5 mM), an intracellular ROS scavenger significantly inhibited the HCI-induced histamine release and ROS generation. These findings suggest that the inflammatory responses to HCI is related to histamine release and ROS generation, and that the ROS generation by HCI may be involved in histamine release via the PLD pathway in RBL-2H3 cells.

Variation of Agronomic Traits of Rice Mutant Lines Induced by Sodium Azide

  • To investigate the mutagenecity of sodium azide (SA) and to select mutants having various agronomic characteristics in rice (Oryza sativa L. ssp. japonica), dry seeds of rice variety 'Suweon 472' were treated with 0, 0.5, 1.0, 2.0, and 4.0 mM SA solutions prepared in 0.1 M phosphate buffer (pH 3.0). Germination rate, seedling height and sterility were investigated in $M_1$ generation and chlorophyll mutations were observed in $M_3$ generation. Germination rate and seedling height decreased as the increase of SA concentration in $M_1$ generation, the maximum seed sterility (40.8%) was found at 4.0 mM SA concentration. Chlorophyll mutants were occurred in $M_3$ generation and the frequency calculated on a line basis was 13.5% at the same treatment. Many kinds of mutations for morphological and agronomic characters were observed and mutations with short culm and glabrous leaf were frequently found in $M_3$ generation. Interestingly, five mutant lines resistant to blast or bacterial blight (BB) were selected and evaluated with several isolates in $M_3$ generation although Suweon 472 has been known to be susceptible to blast and BB. These mutants showed all resistance to seven isolates of blast and a total of 76 lines among 2,567 lines evaluated showed resistant to race K1 of bacterial blight. Two mutant lines (440172 and 41272) showed different reaction to BB isolates from the other resistant mutants. A few kinds of endosperm mutants were also identified and most of them were waxy mutants.

Enhancing Effect of Organic Substances on Hydroxyl Radical Generation During Ozonation of Water: Stopped-Flow ESR Technique

  • Generation of hydroxyl radical, one of their major active species in ozonation of water was directly observed with spin-trapping/electron spin resonance (ESR) technique using 5,5-dimethyl-pyrrolidine-1-oxyl (DMPO) as a spin-trapping reagent. Hydroxyl radical was trapped with DMPO as a stable radical, DMPO-OH. 80 mM of ozone produced $1.08{\times}10^{-6}$M of DMPO-OH, indicating that 1.4% of ${\cdot}$OH is trapped with DMPO if ${\cdot}$OH is produced stoichiometrically from ozone. Humic acid suppressed DMPO-OH generation in a dose-dependent manner. Generation rate of DMPO-OH was determined with ESR/stopped-flow measurement. Phenol derivatives increased the amount and generation rate of DMPO-OH, indicating that phenol derivatives enhance·OH generation during ozonation of water.

Effects of Light and Photosynthetic Electron Transport System on the Generation of Singlet Oxygen ($^1$O$_2$) in Ginseng Thylakoid Membrane (인삼 틸라코이드에서 Singlet Oxygen($^1$O$_2$) 생성에 미치는 전자전달계의 영향)

  • In order to Investigate the mechanism of the leaf-burning disease of ginseng (Panax ginseng C.A. Meyer), studies on the generation of singlet oxygen (1O2) and the photooxidation of the pigments were carried out in comparison with the ones of soybean (G1ycine max L). The studies were mainly focalized on the effects of light intensity, light intensity, inhibitor and electron donor/acceptor of the Photosynthetic electron transport system. When we measured the amounts of 1O2 generated in the thylakoids of ginseng and soybean by the irradiation of light (300 w/m2) as a function its time. It was identified that a higher amount of 1O2 was formed in the ginseng thylakoid than the case of soybean. A generation ratio of lO2 between ginseng and soybean sltbstantially identical in the range of light intensities 50∼150w/m2 However much higher amount of 1O2 was generated in ginseng by irradiation of strong intensity of light (200 500w/m2). Wave length dependency on the generation of 1O2 and the pigment photooxidation was observed on ginseng thylakoids; red light (600-700 nm) gave a maximum effect in the contrast with blur green light (400-60 nm). When the ginseng thylalioid was treated with the electron donor (Mn2+) and acceptors (DCPIP, FeCy) of the photosynthetic electron transport system. a drastic inhibition of 1O2 generation was observed. However, treatment with its inhibitors (DCMU, KCW) activated 1O2 generation. An interesting fact that an electron donor or acceptor of the photosystem II(P680) Inhibited 1O2 generation, suggests an intimate relationship between 1O2 generation and photosystem II.

  • This study was conducted to establish an effective radiation treatment and selection method for induced mutants in M_1 population of soybean treated with gamma-ray. About 64% of total M_1 plants was reduced in plant height up to 50 - 60% and among which 60 - 70% of the plants were contained mutations in M_2 generation. About 60% of the MI plants have born 6 - 15 seeds per plant and 50 - 60% of their progenies produced mutants in M_2 generation. Positive correlation between plant height and number of seeds per plant in M_1 population was found. Higher visible macro-mutation rate in M_2 was observed in the groups of reduced plant height and seed number in the M_1 generation, whereas the frequency of chlorophyll mutation was increased in the group of less damaged plants. The size of mutation sector was increased with reduction in number of seeds per M_1 plant and the mutants were occurred at random in all the parts of M_1 plants. For the effective selection of mutants in soybean mutation breeding, the M_1 seeds should be harvested from the radiation damaged M_1 plants with the application of higher doses of mutagens, and handling M_2 generation by bulk population method is recommendable.

An Experimental Study on a Windheat Generation System with a Savonius Wind Turbine

  • A windheat generation system with a Savonius windturbine was developed and the performance was evaluated through field tests. The system consisted of a heat generation drum, heat exchanger, water storage tank, and two circulation pumps. Frictional heat is created by rotation of a rotor inside the drum containing thermo oil, and was used to heat water. In order to estimate the capacity of this windheat generation system, weather data was collected for one year at the site near the windheat generation system. Wind Power from the savonius wind turbine mill was transmitted to the heat generation system with an one-to-three gear system. Starting force to rotate the savonius wind turbine and the whole system including the windheat generation system were 1.0 and 2.5 kg, respectively. Under the outdoor wind condition, maximum speed of the rotor in the drum was 75rpm at wind speed 6.5 m/sec, which was not fast enough to produce heat for greenhouse heating. Annual cumulative hours for wind speeds greater than 5 m/sec at height of 10, 20, 30 m were 190, 300 and 1020 hrs, respectively. A $5^{\circ}C$ increase in water temperature was achieved by the windheat generation system under the tested wind environment.

CFD study of an iterative focused wave generation method

  • An iterative focused wave generation method is developed and implemented in a local analytic based Navier-Stokes solver. This wave generation method is designed to reproduce the target focused wave by matching the target amplitude spectrum and phase angle. A 4-waves decomposition scheme is utilized to obtain the linearised component of the output wave. A model test studying the interaction between different focused waves and a fixed cylinder is selected as the target for the wave generation approach. The numerical wave elevations and dynamic pressure on the cylinder are compared with the experimental measurement and other state-of-the-art numerical methods' results. The overall results prove that the iterative adjustment method is able to optimize the focused wave generated by a CFD approach.

Selection of Salt Tolerant Rapeseed (Brassica napus L.) Mutant Lines Induced by Irradiation of Proton Ion Beams and Gamma Ray (양성자 및 감마선처리에 의한 유채 내염성 변이계통 선발)

  • This study was carried out to develop salt tolerant varities of rapeseed (Brassica napus L.) which can be grown in the high salty reclaimed land. The seeds of three varieties 'Naehan', 'Tammi', and 'Halla' were treated by proton ion beams and gamma rays with 0 to 2,000 Gy. For the selection of salt tolerant lines, emergence and survival rate, and growth characteristics of $M_2$ to $M_4$ generations were investigated in the Saemangeum reclaimed fields with the different salt concentrations. The lines with potential salt tolerance were selected in the $M_4$ generation and tested indoor for their growth characteristics. There was no significant changes in the soil pH for $M_4$ generation during growth period. However, soil EC was higher in early spring than sowing period (mid October). In $M_4$ generation test, the seeds of original and selected line showed high rates of emergence and survival, as determined one month after sowing. After wintering, however, the original varieties showed the significant reduction in the survival rate, while the selected lines showed a higher survival rate and good growth, leading to the completion of their life cycle. Consequently we selected 9 lines from $M_4$ generation with better performance in growth and yield. Soil EC was $2.8{\sim}4.3dS{\cdot}m^{-1}$ during $M_4$ generation growth period. The laboratory test of the lines selected from $M_4$ generation was made for their salt tolerance potential. The selected lines showed higher chlorophyll and proline contents than the original varieties. There was also no significant difference in the emergence rate of seed between the original and selected varieties. In 200 mM natural sea salt, the $N{\gamma}600-21-1-641$ line derived from 'Naehan' was the highest in growth rate, leaf chlorophyll and proline contents. $T{\gamma}800-20-2-461$ line derived from 'Tammi' didn't show significant difference in growth rate compared to original variety in 200 mM and withered in 250 mM like other lines as time passed. $H{\gamma}200-7-1-740$ line showed similar growth and chlorophyll content compared to its original variety.

Study on Production of Cloned Animals by Recycling Nuclear Transplantation II. Improved Second Generation Cloning of Rabbit Embryos Using Donor Nuclei with Synchronized Cell Cycles (반복핵이식에 의한 복제동물 생산에 관한 연구 II. 토끼에서 공핵배의 세포주기 조절에 의한 제2세대 복제배의 생산효율 개선)

  • large scale production of cloned embryos requires the technology of multiple generation nuclear transplantation(NT) using NT embryos as the subsequent donor nuclei. The purposes of this study were producing the second generation cloned rabbit embryos, and also to determine the electrofusion rate and in vitro developmental potential comparatively in the cloned embryos of the first and second NT generation. The embryos of 16-cell stage were collected from the mated does by flushing oviducts with Dulbecco's phosphate buffered saline(D-PBS) containing 10% fetal calf serum(FCS) at 47 hours after hCG injection In the first generation NT, the nuclear donor embryos were synchronized in the phase of Gi /S transition of 32-cell stage. The first generation NT embryos which were developed to 8-cell were synchronized in Gi /S transition phase of the following 16-cell stage and used as donor nuclei for second generation Synchronization of the cell cycle of blastomeres was induced, first, using an inhibitor of microtuble polymerization, colcemid for 10 hours to arrest blastomeres in M phase, and secondly, using a DNA synthesis inhibitor, aphidicolin for 1.5 to 2 hours to arrest them in Gi /S transition boundary. The recipient cytoplasms were obtained by removing the nucleus and the first polar body from the oocytes collected at 14 hours after hCG injection. The separated donor blastomeres were injected into the enucleated recipient oocytes by micromanipulation and were electrofused by electrical stimulation of three pulses for 60 $\mu$sec at 1.25 kV /cm in 0.28 M rnannitol solution The fused oocytes were co-cultured with a monolayer of rabbit oviductal epithelial cells in M-199 solution containing 10% FCS for 120 hours at 39$^{\circ}C$ in a 5% $CO_2$ incubator. Following in vitro culture of the first and second generation cloned embryos to blastocyst stage, they were stained with Hoechst 33342 dye for counting the number of blastomeres by fluorescence microscopy. The results obtained were summarized as follows: 1. The electrofusion rate was found to be similar as 79.4 and 91.5% in the first and second generation NT rabbit embryos, respectively. 2. The in vitro developmental potential to blastocyst stage of the second generation NT embryos (23.3%) was found significantly(p<0.05) lower, compared with that of the first generation NT embryos (56.8%). 3. The mean blastomeres counts of embryos developed to blastosyst stage following in vitro culture for 120 hours and also their daily cell cycles during the culture period were decreased significantly (p<0.05) to 104.3 cells and 1.33 cylces in the second NT generation, compoared with 210.4 cells and 1.54 cycles in the first NT generation, respectively.
