Characterization of the Transglycosylation Reaction of 4-α-Glucanotransferase (MalQ) and Its Role in Glycogen Breakdown in Escherichia coli

  • We first confirmed the involvement of MalQ (4-${\alpha}$-glucanotransferase) in Escherichia coli glycogen breakdown by both in vitro and in vivo assays. In vivo tests of the knock-out mutant, ${\Delta}malQ$, showed that glycogen slowly decreased after the stationary phase compared to the wild-type strain, indicating the involvement of MalQ in glycogen degradation. In vitro assays incubated glycogen-mimic substrate, branched cyclodextrin (maltotetraosyl-${\beta}$-CD: G4-${\beta}$-CD) and glycogen phosphorylase (GlgP)-limit dextrin with a set of variable combinations of E. coli enzymes, including GlgX (debranching enzyme), MalP (maltodextrin phosphorylase), GlgP and MalQ. In the absence of GlgP, the reaction of MalP, GlgX and MalQ on substrates produced glucose-1-P (glc-1-P) 3-fold faster than without MalQ. The results revealed that MalQ led to disproportionate G4 released from GlgP-limit dextrin to another acceptor, G4, which is phosphorylated by MalP. In contrast, in the absence of MalP, the reaction of GlgX, GlgP and MalQ resulted in a 1.6-fold increased production of glc-1-P than without MalQ. The result indicated that the G4-branch chains of GlgP-limit dextrin are released by GlgX hydrolysis, and then MalQ transfers the resultant G4 either to another branch chain or another G4 that can immediately be phosphorylated into glc-1-P by GlgP. Thus, we propose a model of two possible MalQ-involved pathways in glycogen degradation. The operon structure of MalP-defecting enterobacteria strongly supports the involvement of MalQ and GlgP as alternative pathways in glycogen degradation.

Ban-mal and Hierarchical Structures : Professor Chung′s Theory Critically Examined (반말과 계층 구조 -정대현 교수의 반말 이론에 대한 비판적 검토-)

  • Professor Daihyun Chung has recently presented an intriguing view of Ban-mal(non-honorific expressions) in Korean. He criticizes uses of Ban-mal, on the grounds that they presuppose the existence of hierarchical structures in human beings. Professor Chung also advocates ″a relational theory of Ban-mal″ according to which ascriptions of 'n-mal' should be relativized to the speaker-hearer relationship. He employs this view in showing that uses of Ban-mal indeed presuppose the existence of hierarchical structures in human beings I claim that Professor Chung has not shown the credibility of his relational theory of Ban-mal, and that this very view leads to an unintended and undesirable consequence. Moreover, 1 show that even if we assume that this view is true for the sake of an argument, he is not able to show that uses of Ban-mal indeed presuppose the existence of hierarchical structures. All he can show is rather that uses of 'Ban-mal'(a meta-linguistic expression) presuppose the existence of hierarchical structures. Finally, 1 conclude that what really presupposes the existence of hierarchical structures is the asymmetrical uses of Ban-mal and honorific expressions.

Study on Manufacturing Actual Mal-gun from Joseon Dynasty -Based on the Excavated Mal-gun from Seok-nam-dong, In-chceon- (조선시대[朝鮮時代] 말군의 실물 제작법에 관한 연구)

  • In 2004, Mal-gun(抹裙) was excavated from a plastered tomb in Seok-nam-dong, In-cheon. This tomb is assumed to be from an upper-class woman. The excavated Mal-gun was used to fill in the empty spaces in coffin, and it was seriously damaged and foxed dark. The Mal-gun is made with thin, loose Sook-cho(숙초), and the width of the fabric is 70cm. Both sides of the crotches are overlapped in front, and the back is opened. The waist straps are detached, leaving a little part to show the width of the straps. The hems of the Mal-gun are sewed straight, without any pleats, except some spaces for the feet to go through. This study compared the Mal-gun of Joseon Dynasty from the documentary records and picture records with the excavated Mal-gun from Seok-nam-dong, In-chceon. Also, actual Mal-gun was manufactured according to the excavated Mal-gun to study the process of manufacturing and the formation. As a result, the excavated Mal-gun has the same form of that shown in Ak-hak-gwae-bum, a documentary record from the early period of Joseon Dynasty.

Identification and Characterization of the Vibrio vulnificus malPQ Operon

  • It is likely that maltose could provide a good substrate for the bacteria in the intestine, when the pathogenic bacteria invade and colonize in human gut. For better understanding of this organism's maltose metabolism, a mutant that was not able to grow with maltose as a sole carbon source was screened from a library of mutants constructed by a random transposon mutagenesis. By a transposon-tagging method, malPQ genes encoding a maltodextrin phosphorylase and a 4-${\alpha}$-glucanotransferase, were identified and cloned from Vibrio vulnificus. The deduced amino acid sequences of malPQ from V. vulnificus were 48 to $91\%$ similar to those of MalP and MalQ reported from other Enterobacteriaceae. Functions of malPQ genes were assessed by the construction of mutants whose malPQ genes were inactivated by allelic exchanges. When maltose was used as the sole carbon source, neither malP nor malQ mutant was able to grow to a substantial level, revealing that the MalP and MalQ are the only enzymes for metabolic utilization of maltose. The malQ mutant exhibited decreased adherence toward intestinal epithelial cells in vitro, but there was no difference in the $LD_{50}s$ of the wild-type and the malQ mutant in mice. Therefore, it appears that MalQ is less important in the pathogenesis of V. vulnificus than would have been predicted by considering maltose as a most common sugar in the intestine, but not completely dispensable for virulence in mice.

Effect of Mal-Cha (Powdered Green Tea) on the Quality of Jeung-Pyun (말차(抹茶) 첨가에 따른 증편의 품질 특성)

  • The purpose of this study is to find out the optimal mixing ratios of Mal-Cha for the preparation of Jeung-Pyun through sensory and mechanical tests. The proximate composition of Mal-Cha were $5.46\pm0.15\%$ of moisture, $4.43\pm0.11\%$ of total nitrogen, $7.52\pm0.21\%$ of crude lipid, $8.74\%$ of crude fiber, $8.51\pm0.09\%$ of ash. Overall quality of Jeung-Pyun with $1.5\%$ Mal-Cha was the worst compare with 0, 0.5 and $1.0\%$ ones(p<0.05). Especially, $1.0\%$ Mal-Cha Jeung-Pyun showed the best overall quality. But the sweetness, sourness, flavor, hardness, and moistnes were not significantly different among all the treatments. Acceptabilities of 0.5 and $1.0\%$ Mal-Cha Jeung-Pyuns were not significantly different from that of the control in their sensory and mechanical qualities. Total color difference increased with the amount of Mal-Cha significantly(p<0.001).

A Study on Influences of the Antibacterial Activity of Methampicillin Lysinate by the Several Medicinals (병용약물(倂用藥物)이 Methampicillin Lysinate의 항균력(抗菌力)에 미치는 영향(影響))

  • Drug interaction of a new antibiotic, methampicillin lysinate (MAL) with nine drugs were investigated using four species of gram positive and gram negative bacteria. The experimental results were as follows: 1. MIC of MAL were found to be decreased against E. coil when combined with mefenamic acid, probenecid, aluminium hydroxide gel or corticosteroids. The other drugs did not affect MIC of MAL against the same bacteria. 2. MIC of MAL were found to be increased against Staphylococcus aureus ATCC 6538-P, 9441 when combined with mefenamic acid, aluminum hydroxide gel or dexamethasone acetate. The other drugs did not affect MIC of MAL against the same bacteria. 3. MIC of MAL were found to be increased against Shigella dysenteriae when either of the nine drugs was combined. 4. MIC of MAL were found to be increased approximately 2.5 times when combined with Streptokinase-Streptodornase or hydrocortisone and to be decreased approximately 2 times when combined with probenecid or dexamethasone against Salmonella typhi(type 2). It seems the other drugs do not affect the MIC of MAL against the same bacteria.

Optimal Growth Conditions for the Two Euryhaline Cyanobacterial Clones, Anabaena sp. CB-MAL21 and CB-MAL22 Isolated from Mankyeong Estuary, Korea

  • As a result of the 2-year monthly monitoring of the phytoplankton community at 3 stations in Mankyeong Estuary, Korea, we learned that cyan bacterial species of the genus Anabaena occurred at most sampling points with huge salinity differences (0.1-32.5 psu). We isolated several clones of Anabaena spp. from the monitoring stations, and screen out two euryhaline and nitrogen-fixing Anabaena clones, CB-MAL21 and CB-MAL22. The two clones were grown under various environmental gradients such as temperature (20, 30, 35 and 40$^{\circ}C$), salinity (0, 2, 5, 15 and 30psu), and $PO_4^{3-}$-P concentration (0, 1.6, 8.0, 40 and 200 ${\mu}M$M). Growth of CB-MAL21 and CB-MAL22 was measured by daily monitoring of chlorophyll fluorescence from each experimental culture for more than three serial transfers. Both the two experimental clones did not grow at 0psu. Maximal growth rates of the two clones were markedly reduced at lower $PO_4^{3-}$-P concentrations showing negligible growth at 0 and 1.6 ${\mu}M$M. However, growth of CB-MAL21 was not affected by low $NO_3^--$ concentration in culture media, showing the nitrogen-fixing ability. Maximum biomass yields of the two clones decreased dramatically at 35 and 40$^{\circ}C$. Optimal growth conditions for the two experimental clones were determined to be 20-30$^{\circ}C$, 40 ${\mu}M$M $PO_4^{3-}$-P, and wide salinity range from 5.0 to over 30psu. Best growth of CB-MAL21 was shown at (20$^{\circ}C$-15psu), which is less saline and cooler condition than those (i.e., 30$^{\circ}C$-30psu) for the best growth of CB-MAL22. The euryhaline and nitrogen-fixing CB-MAL21 strain thus can be a candidate laboratory culture for the future cyan bacterial marine biotechnology in temperate coastal waters.

The comparative study on the anti-obesity effect of Arctium lappa L. roots with and without microwave processing (마이크로파 처리 유무에 따른 우엉뿌리의 항비만 효과 비교 연구)

  • Objective : The present study comparatively analyzed the anti-obesity effect of Arctium lappa L. roots with and without microwave processing. Methods : Four HFD groups except for the Normal group (n=8) were allocated: Control, microwave-processed dried Arctium lappa L. roots (MAL) extract 400 mg/kg/d (MAL400), MAL extract 800 mg/kg/d (MAL800), and the dried Arctium lappa L. roots (DAL) extract 800 mg/kg/d (DAL800). The efficacy of MAL and DAL was confirmed in terms of the serum biochemical index, protein expressions related to the synthesis of triglyceride (TG) and total cholesterol (TC) and 𝛽-oxidation, and histopathological staining. Results : Both MAL and DAL treatments significantly reduced final body weight and body weight gain. MAL800 treatment significantly reduced serum TG, TC, low-density lipoprotein cholesterol, and leptin levels, but the serum high-density lipoprotein cholesterol and adiponectin concentrations were dramatically increased. In particular, leptin in the MAL800 group was reduced by 14.1% compared with the DAL800 group. Moreover, the MAL800 treatment showed an effect superior to the DAL800 treatment in the reduction of serum TC, the sirtuin 1 (Sirt1) activation, and the inhibition of 3-hydroxy-3-methyl glutaryl coenzyme A reductase (HMGCR) gene expression. In particular, unlike the DAL800 treatment, MAL treatment significantly led to the activation of peroxisome proliferator-activated receptor alpha (PPAR𝛼). Subsequently, PPAR𝛼 meaningfully regulated downstream proteins associated with 𝛽-oxidation. Conclusion : These findings suggest that Arctium lappa L. roots with microwave processing effectively ameliorate obesity through the regulation of leptin and TC and the promotion of 𝛽-oxidation compared with Arctium lappa L. roots without microwave processing.

Translation and Content Validity of the Korean Version of the Motor Activity Log (한국어판 운동활동일지(Korean Version-Motor Activity Log) 번안과 내용타당도 연구)

  • Purpose: Translation and adaptation involve cross-cultural and conceptual aspects; they are not simply based on linguistic equivalence. This study aimed to produce a conceptually equivalent Korean version of the motor activity log (K-MAL) for the upper extremities that can be applied across the Korean population and its cultures. Methods: Following the procedures used in the translation of other cross-cultural evaluation tools, and the five steps of the translation process, the motor activity log (MAL) was translated into Korean (K-MAL). We then examined the content validity of the K-MAL. Twenty-two rehabilitation professionals (11 males and 11 females, mean length of clinical career = 101.54 months) assessed the content validity of the K-MAL. The content validity ratio and content validity index were used to verify the content validity. Results: There were inconsistencies found in three sub-items in the MAL during the forward translation process. These inconsistencies were corrected, and the complete K-MAL was produced. The exact critical values of the content validity ratio and the content validity index of the K-MAL were 0.45-0.95 and 0.77-1.00, respectively. Conclusion: The K-MAL was successfully developed using a systematic methodology, which included translation, adaptation, and evaluation of the content validity. We expect that stroke rehabilitation professionals working in both clinical and research settings will apply the K-MAL when evaluating the amount and quality of use of the upper extremities in post-stroke patients in Korea.

A Case Report of Lumbar Spinal Stenosis Improved with Diarrhea-Inducing Treatment by Gamsui-mal and Korean Medicine Treatment (감수말 공하법을 병행한 한방치료로 호전된 요추 척추관 협착증 환자의 증례보고)

  • The purpose of this study is to report the effectiveness of diarrhea-inducing treatment by Gamsui-mal for lumbar spinal stenosis. During hospitalization four patients with lumbar spinal stenosis were treated with diarrhea-inducing treatment by Gamsui-mal and acupuncture, herbal medicine, moxibustion, cupping treatment, Bee-venom pharmacopuncture. Before treatment, we explained in detail about diarrhea-inducing treatment by Gamsui-mal and patients agreed with treatment. The symptoms appeared in patients of diarrhea, abdominal pain, and vomiting in the treatment process. We evaluated the degree of improvement by visual analogue scale (VAS), oswestry disability index (ODI). After treatment, we found that pain was reduced and patient's symptoms were improved by visual analogue scale (VAS), oswestry disability index (ODI) score. The results show that diarrhea-inducing treatment by Gamsui-mal was rapidly improving symptoms. Further studies are needed to clarify the effect of diarrhea-inducing treatment by Gamsui-mal.
