Adsorption of CO2 on Amine-impregnated Mesorporous Silica (아민계 함침 메조포러스 실리카를 이용한 CO2 흡착)

  • Adsorption experiment of carbon dioxide was performed on MCM41 silica impregnated with two kinds of EDA(ethylenediamine) and MEA(monoethanolamine). The prepared adsorbents were characterized by BET surface area, X-ray diffraction and FT-IR. The $CO_2$ capture study was investigated in a U type packed column with GC/TCD. The results of XRD for MCM-41 and amine-impregnated MCM41 showed typical the hexagonal pore system. BET results showed the MCM 41 impregnated amine to have a surface area of 141 $m^2/g$ to 595 $m^2/g$ and FT-IR revealed a N-H functional group at about 1400$cm^{-1}$ to 1600$cm^{-1}$. The $CO_2$ adsorption capacity on EDA and MEA was as follow: MCM41-EDA30 > MCM41 -EDA40 >MCM41-EDA20 >MCM-EDA10 and MCM41-MEA40 >MCM41-MEA30 > MCM41-MEA20> MCM41-MEA10. The MCM41-EDA30 showed the highest adsorption capacity due to physical adsorption and chemical adsorption by amino-group content. The results suggest that mesoporous media with EDA is effective adsorbent for $CO_2$ capture from flue gases.

Identification and characterization of a rice MCM2 homologue required for DNA replycation

  • The pre-replication complex (pre-RC), including the core hexameric MCM2-7 complex, ensures that the eukaryotic genome is replicated only once per cell division cycle. In this study, we identified a rice $\underline{m}ini\underline{c}hromosome$ $\underline{m}aintenance$ (MCM) homologue (OsMCM2) that functionally complemented fission yeast MCM2 (CDC19) mutants. We found OsMCM2 transcript expression in roots, leaves, and seeds, although expression levels differed slightly among the organs. Likewise, the OsMCM2 protein was ubiquitously expressed, but it was downregulated when nutritients were limiting, indicating that MCM2 expression (and therefore cell cycle progression) requires adequate nutrition. Yeast two-hybrid and GST pull-down assays demonstrated that OsMCM2 interacted with the COP9 signalosome 5 (CSN5). Taken as a whole, our results indicated that OsMCM2 functions as a subunit of the rice MCM complex and interacts with CSN5 during developmental regulation.

Biochemical Properties of the Minichromosomal Maintenance Complex after the Phosphorylation by Cdc7 Kinase

  • Previous studies showed that Cdc7 kinase of Schizosaccharomyces pombe phosphorylated the minichromosome maintenance (Mcm) complex efficiently in the presence of spMcm10 protein. The biochemical properties of the phosphorylated Mcm complexes were examined to understand the activation mechanism of the Mcm complex by Cdc7 kinase. The phosphorylation of Mcm complex in the presence of spMcm10 by Cdc7 kinase did not affect the stability of the Mcm complex containing all six subunits, and the changes in the sedimentation properties were not observed after the phosphorylation. The reconstitution of the Mcm complex using the purified proteins showed that the phosphorylation of Mcm2 proteins did not affect the interactions between Mcm proteins. The phosphorylation of the Mcm2-7 complex at the same condition also did not activate the other biochemical activities such as DNA helicase and single stranded (ss) DNA binding activities. On the other hand, spMcm10 protein that was used for the stimulation of Mcm phosphorylation showed single stranded DNA binding activity, and inhibited the DNA helicase activity of the Mcm4/6/7 complex. These inhibitory effects were reduced by the addition of Cdc7 kinase, suggesting that the phosphorylation by Cdc7 kinase decreased the interactions between spMcm10 and the Mcm complex. Taken together, these results suggested that the phosphorylation by Cdc7 kinase alone is not sufficient for the remodeling and the activation of the Mcm complex, and the additional factors or the phosphorylations might be required for the activation of the Mcm complex.

Evaluation of electrical characterization and critical length of interconnect for high-speed MCM (고속 MCM 배선의 전기적 특성 및 임계길이 평가)

  • This paper examined the geometrical variables of microstrip to control the characteristic impedance of MCM interconnect and also with respect to the practical requirements, evaluated the critical lengths for attenuation, propagation delay, and crosstalk at 500 MHz frequency compared to at 50 MHz frequency. With the illustration of each MCM-L and MCM-D interconnect having 50 characteristic impedance, it was revealed that the most important geometrical variables to control the characteristic impedance of microstrip are eventually dielectric thickness and line width. In particular, the dielectric thickness of MCM-D interconnect must be controlled with tolerance below 2 m. It is clear that the attenuation does not give rise to signal distortion in the range of up to 500MHz frequency for both MCM-L and MCM-D interconnects. However, the propagation delay is so significant that both MCM-L and MCM-D interconnects should be matched with load at the 500 MHz frequency. For the MCM-D interconnect, the crosstalk voltage would not be high to generate the wrong signal on the neighboring line at 500 MHz frequency, but the MCM-L interconnect could not be used due to severe crosstalk. Eventually, it is clear that the transmission line behavior must be studied for the design of MCM substrate at the 500 MHz frequency.

The Replicon Initiation Burst Released by Reoxygenation of Hypoxic T24 Cells is Accompanied by Changes of MCM2 and Cdc7

  • Although MCM2 is obviously important for the initiation of eukaryotic DNA replication, its role in $O_2$ dependent regulation of replicon initiation is poorly understood. In this report, I analysed the changes of MCM2 during the transition from hypoxically suppressed replicon initiation to the burst of initiation triggered by reoxygenation in T24 cells. A high level of chromatin bound and nucleosolic MCM2 was found under the hypoxic replicon arrest. In contrast low cytosolic MCM2 was noticed. Recovery of $O_2$ induced phosphorylation and diminution of chromatin bound MCM2, whereas cytosolic MCM2 increased. The level of chromatin bound Cdc7 did not change significantly upon reoxygenation. However, after reoxygenation, significant phosphorylation of Cdc7 and an increase of coimmunoprecipitation with its substrate (MCM2) were observed. This provides a hint that reoxygenation may promote the kinase activity of Cdc7. These changes might be the critical factors in $O_2$ dependent regulation of replicon initiation. Moreover, phosphorylation of Cdc7 by Cdk2 can be observed in vitro, but seems to fail to regulate the level of chromatin bound Cdc7 as well as the changes of MCM2 in response to reoxygenation of hypoxically suppressed cells.

Effect of E-box and E2F Binding Site on Transcriptional Activity in MCM Promoter (MCM promoter에서 E-box와 E2F 결합부위가 전사활성에 미치는 영향)

  • MCM proteins are essential for eukaryotic DNA replication, playing roles in the initiation and elongation of DNA replication. MCM proteins expression is much higher in malignant tissues than normal tissues. Several reports have indicated the usefulness of MCM proteins as markers of cancer cells in histopathological diagnosis. However, the cause of enhanced expression of MCM proteins in cancer cells remain to be clarified. The purpose of this study is to examine the relative transcriptional activities of human mcm gene promoters in cancer and normal cells. The minimal promoter region required for transcription of a luciferase reporter gene was contained an E-box and one E2F site. In addition, luciferase activities from mcm7 and mcm2 promoter/luciferase gene reporter constructs were significantly increased in cancer cells at 8 times compared with normal cells. E-box and E2F binding site in the promoter of mcm genes are responsible for different mechanism of transcription regulation on the cellular environment.

Property and Catalytic Activity of Heteropoly Acid Supported on MCM-41, 48 Mesoporous Material and SiO2 (MCM-41, 48 메조포러스 물질 및 SiO2에 담지한 헤테로폴리산의 특성 및 촉매적 활성)

  • Heteropoly acid(HPW) catalysts supported on three different carriers, an amorphous silica, MCM-41 and MCM-48, with different loadings and calcination temperatures have been prepared and characterized by X-ray diffraction, nitrogen physorption, infrared spectroscopy, and $^{31}P$ magic angle spinning NMR. From the result of IR and NMR, it was shown that HPW retains the Keggin structure on the supported catalysts. No HPW crystal phase was developed even at HPW loadings as high as 35 wt % on the MCM-41 and 65 wt % MCM-48. Thus, HPW appeared to form finely dispersed species. In the hydrolysis reaction of di, bis, tri-pentaerythritol, HPW/MCM-41, 48 exhibited higher catalytic activity than $HPW/SiO_2$ or HPW.

Effect of Pt as a Promoter in Decomposition of CH4 to Hydrogen over Pt(1)-Fe(30)/MCM-41 Catalyst (Pt(1)-Fe(30)/MCM-41 촉매상에서 수소 제조를 위한 메탄의 분해 반응에서 조촉매 Pt의 효과)

  • The effect of Pt was investigated to the catalytic methane decomposition of CH4 to H2 over Pt(1)-Fe(30)/MCM-41 and Fe(30)/MCM-41 using a fixed bed flow reactor under atmosphere. The Fe2O3 and Pt crystal phase behavior of fresh Pt(1)-Fe(30)/MCM-41 were obtained via XRD analysis. SEM, EDS analysis, and mapping were performed to show the uniformed distribution of nano particles such as Fe, Pt, Si, O on the catalyst surface. XPS results showed O2-, O- species and metal ions such as Pt0, Pt2+, Pt4+, Ft0, Fe2+, Fe3+ etc. When 1 wt% of Pt was added to Fe(30)/MCM-41, automic percentage of Fe2p increased from 13.39% to 16.14%, and Pt4f was 1.51%. The yield of hydrogen over Pt(1)-Fe(30)/MCM-41 was 3.2 times higher than Fe(30)/MCM-41. The spillover effect of H2 from Pt to Fe increased the reduction of Fe particles and moderate interaction of Fe, Pt and MCM-41 increased the uniform dispersion of fine nanoparticles on the catalyst surface, and improved hydrogen yield.

Decomposition of 1,2-dichlorobenzene over Al-MCM-41 (Al-MCM-41을 이용한 1,2-dichlorobenzene의 분해 반응)

  • The aim of this study is to test the feasibility of Pt/Al-MCM-41 for the dioxin decomposition reaction. For model reaction, 1,2-dichlorobenzene was decomposed instead of dioxin. $Pt/{\gamma}-Al_2O_3$ was compared with Pt/Al-MCM-41. Al-MCM-41 catlaysts were prepared by post grafting method and the Pt/Al-MCM-41 catalysts with Si/Al = 15, 30 showed higher catalytic activity than $Pt/{\gamma}-Al_2O_3$. Their higher catalytic activities were related with acid amounts. Especially Al-MCM-41 with ion exchanged with $H^+$ enhanced catalytic activity.

Characteristics Analysis of MCM41 Impregnated with Ethylenediamine for CO2 Adsorption (CO2 흡착을 위한 Ethylenediamine 함침 MCM41의 특성 분석)

  • Adsorption experiment of carbon dioxide was performed on MCM41 silica with a 30 wt.% EDA(ethylenediamine) loading at different $CO_2$ inlet concentration and various adsorption temperature. The surface characteristics of $CO_2$ capturing agent were carried out using BET analysis, X-ray diffraction and FT-IR. The results of BET showed 781 $m^2/g$ for MCM41 and 464 $m^2/g$ for EDA/MCM41. X-ray diffraction results reveled typical hexagonal pore system. The higher sorption capacity of EDA/MCM41 was about 80 $mg_{CO2}/g_{sorbent}$ with 50% $CO_2$ inlet concentration and 303 K adsorption temperature. The isosteric heat of adsorption in 303-353 K ranged from -25.47 to -28.24 KJ/mole for EDA/MCM41, which indicates $CO_2$-EDA/MCM41 interaction with exothermic adsorption process. Finally, the performance of EDA/MCM41 in 10 consecutive sorption-desorption runs was a stable with only a minor drop in its sorption capacity.
