Application of multi dimensional NMR experiments to VBS RNAs of Yeast Saccaromyces cerevisiae virus

  • The structures of two VBS (viral binding site) RNAs, SL1 and SL2, of Yeast Saccharomyces cerevisiae vims have been studied by 2D and 3D NMR experiments. VBSs play a crucial role in viral particle binding to the plus strand and packaging of the RNA. The secondary structures of the two VBS RNAs share a common feature of the stem-internal loop-stem-hairpin loop structure although the size of the internal loops of SL1 and SL2 differs. 2D experiments were sufficient for fill assignments of SL1. However, isotope labeling of the sample and multidimensional experiments were required for 28-nucleotide-long SL2 due to the spectral overlap. Several 3D HCCH experiments have accomplished full assignment of SL2 RNA.

Implementation of the Feed and Swaddle Technique as a Non-Pharmacological Strategy to Conduct Brain Magnetic Resonance Imaging in Very Low Birth Weight Infants

  • Purpose: Magnetic resonance imaging (MRI) is a useful tool for evaluating brain injury and maturation in preterm infants and often requires sedation to acquire images of sufficient quality. Infant sedation is often associated with adverse events, despite extreme precautions. In this study, the swaddling technique was investigated as an alternative non-pharmacological strategy to obtain brain MRIs of sufficient quality. Methods: We applied the feed and swaddle technique during routine brain MRI as a quality improvement project and compared its morbidity with that of sedation in a historic age-matched group. Seventy-nine very low birth weight infants in the neonatal intensive care unit of Ajou University Hospital (Suwon, Korea) were enrolled. Thirty-two (40.5%) infants were in the feed and swaddling group, and 47 (59.5%) were in the sedation group. Results: The morbidity associated with the cardiopulmonary system (swaddling group vs. sedation group: 53.13% [n=17] vs. 63.83% [n=30], P=0.723) and central nervous system (40.63% [n=13] vs. 29.79% [n=14], P=0.217) were not significantly different between groups. The MRI failure rate was not significantly different (swaddling group vs. sedation group: 12.5% [n=4] vs. 4.3% [n=2], P=0.174). The MRI scanning time was longer in the swaddling group than in the sedation group (76.5±20.3 minutes vs. 61.5±13.6 minutes, P=0.001). Cardiopulmonary adverse events were significantly less common in the swaddling group than in the sedation group (3.13% [n=1] vs. 34.04% [n=16], P=0.002). Conclusion: The success rate of MRI was comparable between the swaddling technique and sedation. Furthermore, despite the drawback of prolonged scan time, cardiopulmonary adverse events are fewer with swaddling than with sedative agents. Therefore, swaddling can be an alternative to sedation or anesthesia when performing neonatal MRI scans.

Added Value of Diffusion Weighted Imaging for Detecting Pancreatic Abnormality in Patients with Clinically Suspected Acute Pancreatitis

  • Purpose: To evaluate the added value of diffusion weighted imaging (DWI) to computed tomography (CT) for detecting pancreatic abnormality in patients with clinically suspected acute pancreatitis (AP). Materials and Methods: 203 patients who underwent abdomen CT and subsequent DWI to do a workup for epigastric pain were analyzed. Two blinded radiologists independently performed an interval reading based on CT image sets first, then based on combined CT and DWI image sets. The diagnostic criterion on DWI was the increased signal intensity in the pancreas to that of the spleen. For quantitative analysis, the third radiologist measured ADC value of the pancreas in each patient. Results: For AP (n = 43), the sensitivity for detecting pancreatic abnormality increased, from 42% to 70% for reader 1 (P < 0.05) and from 44% to 72% for reader 2 (P < 0.05). For borderline pancreatitis (n = 42), the sensitivity also increased, from 10% to 26% for reader 1 (P < 0.05) and from 7% to 29% for reader 2 (P < 0.05). The mean ADC values (unit, ${\times}10^{-3}mm^2/s$) were significantly different among the three groups (for AP, $1.09{\pm}0.16$; for borderline pancreatitis, $1.28{\pm}0.2$; for control, $1.46{\pm}0.15$, P < 0.05). Conclusion: Sensitivity for detecting pancreatic abnormality increased significantly after adding DWI to CT in patients with clinically suspected AP.

Clinical Analysis Comparing Efficacy between a Distal Filter Protection Device and Proximal Balloon Occlusion Device during Carotid Artery Stenting

  • Objective : The main concern during transfemoral carotid artery stenting (CAS) is preventing cerebral embolus dislodgement. We compared clinical outcomes and intraprocedural embolization rates of CAS using a distal filter protection device or proximal balloon occlusion device. Methods : From January 2011 to March 2015, a series of 58 patients with symptomatic or asymptomatic internal carotid artery stenosis ${\geq}70%$ were treated with CAS with embolic protection device in single center. All patients underwent post-CAS diffusion-weighted magnetic resonance imaging (DW-MRI) to detect new ischemic lesions. We compared clinical outcomes and postprocedural embolization rates. Results : CAS was performed in all 61 patients. Distal filter protection success rate was 96.6% (28/29), whose mean age was 70.9 years, and mean stenosis was 81%. Their preprocedural infarction rate was 39% (11/28). Subsequent DW-MRI revealed 96 new ischemic lesions in 71% (20/28) patients. In contrast, the proximal balloon occlusion device success rate was 93.8% (30/32), whose mean age was 68.8 years and mean stenosis was 86%. Preprocedure infarction rate was 47% (14/30). DW-MRI revealed 45 new ischemic lesions in 57% (17/30) patients. Compared with distal filter protection device, proximal balloon occlusion device resulted in fewer ischemic lesions per patient (p=0.028). In each group, type of stent during CAS had no significant effect on number of periprocedural embolisms. Only 2 neurologic events occurred in the successfully treated patients (one from each group). Conclusion : Transfemoral CAS with proximal balloon occlusion device achieves good results. Compared with distal filter protection, proximal balloon occlusion might be more effective in reducing cerebral embolism during CAS.

CT and MRI image fusion reproducibility and dose assessment on Treatment planning system (치료계획시스템에서 전산화단층촬영과 자기공명영상의 영상융합 재현성 및 선량평가)

  • Objectives: The aim of this study is to evaluate the reproducibility and usefulness of the images through the fusion of CT(Computed tomography) and MRI(Magnetic resonance imaging) using a self-manufactured phantom. We will also compare and analyze the target dose from acquired images. Materials and Methods: Using a self-manufactured phantom, CT images and MRI images are acquired by 1.5T and 3.0T of different magnetic fields. The reproducibility of the size and volume of the small holes present in the phantom is compared through the image from CT and 1.5T and 3.0T MRI, and dose changes are compared and analyzed on any target. Results: 13 small hole diameters were a maximum 31 mm and a minimum 27.54 mm in the CT scan and the were measured within an average of 29.28 mm 1 % compared to actual size. 1.5 T MRI images showed a maximum 31.65 mm and a minimum 24.3 mm, the average is 28.8 mm, which is within 1 %. 3.0T MRI images showed a maximum 30.2 mm and a minimum 27.92 mm, the average is 29.41 mm, which is within 1.3 %. The dose changes in the target were 95.9-102.1 % in CT images, 93.1-101.4 % in CT-1.5T MRI fusion images, and 96-102 % in CT-3.0T MRI fusion images. Conclusion: CT and MRI are applied with different algorithms for image acquisition. Also, since the organs of the human body have different densities, image distortion may occur during image acquisition. Because these inaccurate images description affects the volume range and dose of the target, accurate volume and location of the target can prevent unnecessary doses from being exposed and errors in treatment planning. Therefore, it should be applied to the treatment plan by taking advantage of the image display algorithm possessed by CT and MRI.

Synthesis and Characterization of Fluorinated Polybenzimidazole Proton Exchange Membranes for Fuel Cell (연료전지용 불소화 폴리벤즈이미다졸 양성자 교환 멤브레인 합성 및 특성평가)

  • A fluorinated polybenzimidazole (FPBI) was synthesized from 3,3-diaminobenzidine (DAB) of tetraamine, 2,2-bis(4-carboxyphenyl)hexafluoropropane of aromatic biscarboxylic acid, and 4,4-sulfonyldibenzoic acid of aromatic biscarboxylic acid in polyphosphoric acid (PPA). A FPBI was easily cast and made into clear films. The structure of condensation polymers and corresponding membranes were analyzed using GPC (gel permeation chromatography), $^1H$-NMR ($^1H$ nuclear magnetic resonance) and FT-IR (fourier transform infrared). TGA (thermogravimetric analysis) analysis showed that the prepared membranes were thermally stable, so that elevated temperature fuel cell operation would be possible. The proton conductivity of the FPBI membranes increased with increasing temperatures in the polymer. A FPBI membrane has a maximum ion conductivity of 45 mS/cm at $90^{\circ}C$ and 100% relative humidity.

Microwave Absorbing Properties of M-type Barium Ferrites with BaTi0.5Co0.5Fe11O19 Composition in Ka-band Frequencies (BaTi0.5Co0.5Fe11O19 조성을 갖는 M형 바륨 페라이트의 Ka-밴드 전파흡수특성)

  • Magnetic and Ka-band absorbing properties have been investigated in Ti-Co substituted M-type barium hexaferrites with $BaTi_{0.5}Co_{0.5}Fe_{11}O_{19}$ composition. The ferrite powders were prepared by conventional ceramic processing technique and used as absorbent fillers in ferrite-rubber composites. The magnetic properties were measured by vibrating sample magnetometer. The complex permeability and dielectric constant were measured by using the WR-28 rectangular waveguide and network analyzer in the frequency range 26.5~40 GHz. For the Ti-Co substituted M-hexaferrites, the ferromagnetic resonance is observed at Ka-band (29.4 GHz). The matching frequency and matching thickness are determined by using the solution map of impedance matching. A wide band microwave absorbance is predicted with controlled ferrite volume fraction and absorber thickness.

Inhibitory Activity of Aralia elata Leaves on Protein Tyrosine Phosphatase 1B and α-Glucosidase (참두릅 잎의 Protein Tyrosine Phosphatase 1B와 α-Glucosidase 저해 활성)

  • Anti-diabetic potential of the leaves of A. elata through the inhibitory activity on PTP1B and ${\alpha}$-glucosidase has not been reported. In this study, the EtOAc fraction of methanolic extract from the leaves of A. elata showed potent inhibitory activity against the PTP1B and ${\alpha}$-glucosidase with $IC_{50}$ value of $96.29{\pm}0.3$ and $264.71{\pm}14.87{\mu}g/mL$, respectively. Three known triterpenoids, oleanolic acid, oleanolic acid-28-O-${\beta}$-D-glucopyranoside and oleanolic acid-3-O-${\beta}$-D-glucopyranoside were isolated from the most active EtOAc fraction. We determined the chemical structure of these triterpenoids through comparisons of published nuclear magnetic resonance (NMR) spectroscopic data. Furthermore, we screened these triterpenoids for their ability to inhibit PTP1B and ${\alpha}$-glucosidase over a range of concentrations ($12.5-50{\mu}M$). All three terpenoids significantly inhibited PTP1B in a concentration dependent manner and oleanolic acid effectively inhibited ${\alpha}$-glucosidase. In addition, these compounds revealed potent inhibitory activity with negative binding energies toward PTP1B, showing high affinity and tight binding capacity in the molecular docking studies. Therefore, the results of the present study clearly demonstrate that A. elata leaves and its triterpenoid constituents might be beneficial in the prevention or treatment of diabetic disease.

New dammarane-type triterpenoids from the leaves of Panax notoginseng and their protein tyrosine phosphatase 1B inhibitory activity

  • Background: Panax notoginseng has been used as a general tonic agent to invigorate human body for millennia in China and continued to be used until present. Methods: Some chromatographic methods were performed to isolate pure triterpenoids, and their structures were determined by nuclear magnetic resonance (NMR) experiments. Anti-diabetes activities of isolated compounds were evaluated through their inhibitory activity of protein tyrosine phosphatase 1B (PTP1B) enzyme. Results and Conclusion: Three new dammarane-type triterpenoids, notoginsenoside-LX (1), notoginsenoside-LY (2), and notoginsenoside-FZ (3) together with eighteen known compounds were isolated from the Panax notoginseng leaves. The structure-activity relationship of the compounds with dammaranetype triterpenoids and their PTP1B inhibitory activity were also reported. Results showed that compounds 2, 15, 20, and 21 can significantly inhibit the enzyme activity of PTP1B in a dose-dependent manner, with inhibitory concentration 50 ($IC_{50}$) values of $29.08{\mu}M$, $21.27{\mu}M$, $28.12{\mu}M$, and $26.59{\mu}M$, respectively. The results suggested that Panax notoginseng leaves might have potential as a new therapeutic agent for the treatment of diabetes.
