The Bioconcentration of Naphthalene in Tissues of Juvenile Olive flounder, Paralichthys olivaceus (넙치(Paralichthys olivaceus) 치어조직의 나프탈렌(Naphthalene) 생물농축)

  • Polycyclic aromatic hydrocarbons (PAHs) such as naphthalene, are released into the aquatic environment and have been identified as endocrine effects on marine organisms. Naphthalene makes up the highest fraction of PAHs in oil and shows the strong toxicity to aquatic organisms. I analyzed 24h-median lethal concentration (24h-$LC_{50}$) for juvenile olive flounder Paralichthys olivaceus to study the acute toxicity of naphthalene. Bioconcentrations in the gill, liver, kidney and muscle of olive flounder were analyzed after to 6 naphthalene concentrations of 0, 1000, 1800, 3200, 5600 and 10000 ${\mu}g/L$. Olive flounder has 24h-$LC_{50}$ value of 2410.76 ${\mu}g/L$ in Large group, and 2230.67 ${\mu}g/L$ in Samll group. Naphthalene concentration was varied with tissues. The concentrations of naphthalene were much higher in liver and kidney than in gill and muscle.

Acute and Subchronic Inhalation Toxicity Evaluation of Methyl Formate in Rats (Methyl formate의 랫드를 이용한 급성 및 아만성 흡입독성 평가)

  • We performed the tests of acute and subchronic inhalation toxicity of methyl formate, which has limited toxicological data in spite of its widespread use and enhanced hazard consequent on its high volatility. The median lethal concentration ($LC_{50}$) was evaluated to be above 5,000ppm(12.27 mg/L). In the test with subchronic inhalation, there are no deaths, but with reduction of body weight, food intake, organ weight by exposure to 400 (0.98 mg/L) and 1,600 (3.92 mg/L) ppm, dose-dependently. There were statistical differences in some hematological and blood biochemical parameters as compared to control (e.g. neutrophile and lymphocyte in the 1,600 ppm group, calcium and A/G in 1,600 ppm group). Methyl formate under the exposure of 1,600 ppm showed the respiratory findings with nasal, it was confirmed that the chemical has respiratory hazard with 1,600 ppm inhalation exposure, induces nasal epithelial atrophy, olfactory cell degeneration/regeneration and the contraction of olfactory cells, etc. According to the notification with Ministry of Labor (No. 2009-68) for classification, labeling and MSDS of chemicals, it is suggested for methyl formate to be classified as category 4 in acute (10.0$4\leq20.0$ mg/L), category 2 (0.2$\leq$1.0 mg/L/6h, 90 days) in specific target organ-repeated exposure.

Tolerance and Histological Responses to Formalin of Black Seabream Acanthopagrus schlegelii Juveniles (감성돔(Acanthopagrus schlegelii) 치어의 포르말린에 대한 내성 및 조직학적 반응)

  • Black seabream juveniles Acanthopagrus schlegelii held at $20^{\circ}C$ were exposed to formalin at concentrations of 50 to 400 ppm, and tolerance values were determined by calculating median lethal concentration values (LC50) through probit analysis. The 12-, 24, 48, 72- and 96-h LC50 values for formalin were 297, 233, 171, 162 and 157 ppm, respectively. The histological effects of formalin on gill and liver tissues in this fish were determined. No histological effects were observed in the control group. The intensity of cell damage increased with the concentration of, and duration of exposure to, formalin. Hyperplasia, separation and epithelial necrosis, epithelial lifting, lamellar synechiae and collapsed lamellae were observed in gill tissues exposed to formalin. Hepatic lesions in liver tissues of fishes exposed to formalin were characterized by cloudy swelling of hepatocytes, necrosis, cytoplasmic vacuolization, deposition of pigments, spongiosis hepatis, nuclear hypertrophy, dilation of sinusoids and bile stagnation. The LC50 values and histological results obtained in this study will aid in designing treatment regimens to minimize toxic side effects and increase efficacy.

Toxicity, antibacterial and parasiticidal effects of hydrogen peroxide for israel carp (Cyprinus carpio) (과산화수소의 이스라엘잉어 (Cyprinus carpio)에 대한 독성 및 세균과 기생충 제거 효과)

  • To investigate the effect of hydrogen peroxide in israel carp, Cyprinus carpio, toxicity and microbial activities were determined. For hydrogen peroxide toxicity test, the median lethal concentration ($LC_{50}$) to israel carps Cyprinus carpio (average weight 0.44 g) by acute toxicity was determined after 24 hour treatment. All israel carp were alive in 24 hours treatment at 80 $\mu\ell/\ell$ concentration and $LC_{50}$ value was 148.9 $\mu\ell/\ell$. For biocidal activities of hydrogen peroxide, remove of parasite and growth inhibition of pathogenic bacteria were determined. The parasite Trichodina sp. infected on the skin and gills of israel carps (average weight 0.1 g) was completely eliminated at 40 $\mu\ell/\ell$ of hydrogen peroxide treatment for 24 hour. Most of the minimum inhibitory concentrations (MICs) against 30 fish pathogenic bacteria were less than 40 $\mu\ell/\ell$.

Lethal Toxicity and Hematological Changes Exposed to Nitrate in Flatfish, Paralichthys olivaceus in Biofloc and Seawater (바이오플락 및 일반 해수에서 질산염의 넙치 (Paralichthys olivaceus)에 미치는 급성 독성 및 혈액학적 성상의 변화)

  • Juvenile Paralichthys olivaceus (mean length $19.8{\pm}2.6cm$, mean weight $97.8{\pm}15.8g$) were exposed for 96 hours to different nitrate concentrations of 0, 62.5, 125, 250, 500, 1,000, and $1,500mg\;L^{-1}$ in biofloc and 0, 62.5, 125, 250, 500, and $1,000mg\;L^{-1}$ in seawater. Median lethal concentration values ($LC_{50}$, the concentration at which 50% of mortality occurred after 96 hours of exposure) of nitrate to P. olivaceus in biofloc and seawater were 1,226 and $597mg\;NO_3L^{-1}$ (P<0.05), respectively, revealing a higher toxicity of nitrate to P. olivaceus in seawater than in biofloc. In hematological parameters, hematocrit level in P. olivaceus exposed to nitrate was significantly increased only at a concentration of $1,000mg\;L^{-1}$ in biofloc and at concentrations higher than $250mg\;L^{-1}$ in seawater, but no significant changes in hemoglobin were found in biofloc and seawater. In plasma parameters, aspartate aminotransferase (AST) and alanine aminotransminase (ALT) were significantly increased by nitrate exposure in biofloc and seawater, but no significant changes in alkaline phosphatase (ALP) were found in biofloc and seawater. Results of this study indicate that nitrate exposure to P. olivaceus have a lethal toxic effect and alter hematological and plasma constituents of flatfish P. olivaceus. Given relatively lower toxicity of nitrate in biofloc than in seawater, the use of biofloc in aquaculture may reduce potential toxic effect caused by nitrate in feces and feed residue.

Applicability of CATIS as a Postharvest Phytosanitation Technology against the Peach Fruit Moth, Carposina sasakii Matsumura (복숭아심식나방(Carposina sasakil)의 수확 후 소독 처리로서 CATTS 기술의 적용 가능성)

  • As an environment-friendly phytosanitary measure, CATTS (controlled atmosphere temperature treatment system) has been developed to kill several quarantine insect pests infesting subtropical agricultural commodities. This study tested any possibility to apply CATTS to apples to effectively eliminate the peach fruit moth, Carposina sasakii, which has been regarded as a quarantine insect from the imported countries. When the larvae of C. sasakii were directly exposed to $46^{\circ}C$ (an installed lethal temperature of CATTS), they showed a median lethal time at 14.66 min. Addition of high carbon dioxide to the temperature treatment enhanced the thermal limit susceptibility of C. sasakii to $46^{\circ}C$. CATTS device was constructed to automatically control $CO_2$ concentration and temperature with real-time monitoring both in the chamber and in the fruit. The larvae internally infesting apples were tested using the CATTS device and showed 100% lethality after 60 min exposure to a treatment of $46^{\circ}C$ under 15% $CO_2$ in the chamber. Relatively long exposure may be due to the deviation between the ramping temperature ($0.35^{\circ}C$/min) of the chamber and the ramping temperature (0.12-$0.23^{\circ}C$/min) inside apple fruit, where the tested larvae were located. This study suggests a possibility that CATTS can be applied as a quarantine measure to kill the larvae of C. sasakii locating inside the apples.

Insecticidal Effect of an Entomopathogenic Fungus, Beauveria bassiana ANU1 to Spodoptera exigua and Plutella xylostella by Different Temperature and Humidity Conditions (파밤나방과 배추좀나방에 대한 곤충병원성 곰팡이 Beauveria bassiana ANU1의 온도와 습도조건에 따른 살충효과)

  • Entomopathogenic fungi have been studied to develop for biological control agents as an alternative to chemical control agents in insect pest management. Two Lepidopteran insects, Spodoptera exigua and Plutella xylostella, are serious insect pests infested various crops, but not effectively controlled by commercial chemical pesticides due to its high insecticide resistance. A fungal isolate was isolated from S. exigua larvae collected from green onion field in Andong, Korea. To identify the fungal isolate, 18srRNA sequence for internal transcribed spacer (ITS) and ${\beta}$-tubulin regions were sequenced. The ITS and ${\beta}$-tubulin sequence were highly matched to Beauveria bassiana and morphological characteristics also was fit to known B. bassiana. Finally, isolated fungus has identified as B. bassiana and named B. bassiana ANU1. The result of bioassay, median lethal concentrations were $2.7{\times}10^3$ and $0.9{\times}10^3conidia/ml$ and medial lethal times were 65.6 and 60.8 h to S. exigua and P. xylostella, respectively. B. bassiana ANU1 showed high pathogenicity to two insect pests from $20^{\circ}C$ to $30^{\circ}C$ at 50% relative humidity (RH) and more than 40% RH at $25^{\circ}C$ with $10^7conidia/ml$ of concentration.

Antimicrobial Effects of Chemical Disinfectants on Fish Pathogenic Bacteria

  • This study was to examine the potential disinfection efficiencies of 10 compounds by determining their antimicrobial capacity and ichthyotoxicity. Antimicrobial effects against Vibrio sp., Edwadsiella tarda, Streptococcus sp., and Staphylococcus sp. were tested using 10 different disinfectants; hydrogen peroxide, sodium hypochlorite, chlorine dioxide, povidon iodine, formaldehyde, glutaraldehyde, quaternary ammonium compounds (QACs), didecyl dimethyl ammonium chloride (DDAC), ortho-dichlorobenzen, and copper sulfate. Chlorine dioxide ($ClO_2$) containing 5% $ClO_2$ and copper sulfate had no effects on bactericidal activity, while the other disinfectants resulted in 99.99% bactericidal activity against 4 strains of fish pathogenic bacteria. The ichthyotoxicity of the 10 disinfectants was investigated using 3 kinds of fish species; flounder (Paralichthys olivaceus), rockfish (Sebastes pachycephalus), and black sea bream (Acanthopagrus schlegelii). Median lethal concentration ($LC_{50}$) values of the 10 disinfectants were estimated to determine toxicity ranges of the doses within 24 hr. Among test disinfectant solutions, hydrogen peroxide showed the highest $LC_50$ in flounder (201.3), rockfish (269.7), and black sea bream (139.3 ppm). DDAC revealed the lowest $LC_{50}$ in flounder (2.1), rockfish (1.0), and black sea bream (1.5 ppm). These results suggest that DDAC, quaternary ammonium compounds, glutaraldehyde, and sodium hypochlorite are effective disinfectants for fish and bacterial species examined in this study.

Study on the Acute and Sub-Acute Inhalation Toxicity of 1-Bromopropane in SD Rats (Rat를 이용 1-Bromopropane의 급성 및 아급성 흡입독성 연구)

  • The purpose of this study was to investigate the acute(4 hrs) and repeated-dose(6 hrs a day, 5 days a week, 8 weeks) toxic effects of 1-bromopropane(1-BP) on Sprague-Dawley (SD) rats which were treated by inhalation. The results were as follows ; 1. The median lethal concentration($LC_{50}$) was estimated 14,374 ppm(confidence limit 95% ; 13,624~15,596 ppm) in acute inhalation. Abnormal clinical signs related to the 1-BP were not observed with the acute inhalation dose. Gross findings of necropsy revealed no evidence of specific toxicity related to the 1-BP. 2. By sub-acute inhalation the body weights of male and female were significantly reduced(p<0.001) by the dose of 1,800 ppm compared with control group, while the relative weights of liver were significantly increased(p<0.001) in both sexes. However there were no significant variation in food consumption, urine biochemistry, hematology and blood biochemistry for the exposed rats compared with the control rats. Abnormal clinical signs and gross findings of necropsy related to the 1-BP were not shown. No toxicologic lesions were observed by the histopathological test.

Effects of antioxidant enzymes and bioaccumulation in eels (Anguilla japonica) by acute exposure of waterborne cadmium

  • This study was conducted to evaluate the acute effects of waterborne cadmium exposure on bioaccumulation and antioxidant enzymes in eels (Anguilla japonica) and to determine the median lethal concentration (LC50). Fish were exposed to different cadmium concentrations (0, 0.15, 0.30, 0.61, 1.83, 3.08, 3.67, 4.29, and 5.51 mg L-1) for 96 h. The LC50 of A. japonica to cadmium was 3.61 mg L-1. Cadmium accumulation generally increased in tissues with increasing waterborne cadmium concentrations. At ≥ 1.83 mg L-1 exposure, all tissues accumulated significant cadmium concentrations compared with the control group, in the order of kidney > liver > gill > spleen > muscle. Measurements of variation in actual cadmium concentrations showed that a reduction of the metal in experimental water was related to cadmium accumulation in tissues. As activity alteration of antioxidant enzymes for reactive oxygen species, superoxide dismutase and catalase activities increased at ≥ 0.61 mg L-1 significantly, glutathione peroxidase and glutathione S-transferase activities were not significantly changed. The results of this study suggest that acute exposure to waterborne cadmium is potentially fatal to A. japonica due to the metal's major accumulation in various tissues and the effect of antioxidant enzyme activity.
