Flavonoids analysis in leaves and fruits of Korean mulberry cultivar, Baekokwang having white fruits

  • Morus alba has white and/or purple fruits with a very sweet taste and low acidity. Most Korean mulberry trees have purple fruits. However, Baekokwang is a unique mulberry genetic resource in Korea with white fruits. In this study, flavonoids contents of Baekokwang mulberry leaf and fruit were analyzed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS) technique. UPLC-DAD-QTOF/MS chromatogram showed that 15 flavonoids and 9 flavonoids were isolated and identified from the mulberry leaf and fruit. Total flavonoids contents of Baekokwang leaves and fruits were 812.7 mg and 35.0 mg, respectively. Baekokwang leaves had 4 major flavonoids including quercetin 3-O-(6"-O-malonyl) glucoside, 235.3 ppm, kaempferol 3-O-(6"-O-malonyl) glucoside, 132.3 ppm, kaempferol 3-O-rutinoside (nicotiflorin), 108.1 ppm, and quercetin 3-O-rutinoside (rutin), 103.8 ppm. Baekokwang fruits had 3 major flavonoids including quercetin 3-O-(6"-O-malonyl) glucoside, 13.0 ppm, quercetin 3-O-rutinoside (rutin), 7.8 ppm, and kaempferol 3-O-rutinoside (nicotiflorin), 5.7 ppm. From the above results, mulberry leaves have rich flavonoids compared to its fruits.

Isolation and Identification of Postharvest Spoilage Fungi from Mulberry Fruit in Korea

  • BACKGROUND: Spoilage fungi can reduce the shelf life of fresh fruits and cause economic losses by lowering quality. Especially, mulberry fruits have high sensitivity to fungal attack due to their high water content (> 70%) and soft texture. In addition, the surface of these fruits is prone to damage during harvesting and postharvest handling. However, any study on postharvest spoilage fungi in mulberry fruit has not been reported in Korea. This study aimed to examine the spoilage fungi occurring in mulberry fruits during storage after harvest. METHODS AND RESULTS: In this study, we isolated postharvest spoilage fungi from mulberry fruits stored in refrigerator (fresh fruits) and deep-freezer (frozen fruits) and identified them. In the phylogenetic analysis based on comparisons of the ITS rDNA sequences, the 18 spoilage fungi isolated from mulberry fruits and the 25 reference sequences were largely divided into seven groups that were subsequently verified by high bootstrap analysis of 73 to 100. Alternaria spp. including A. alternate and A. tenuissima, were the most frequently isolated fungi among the spoilage isolates: its occurrence was the highest among the 18 isolates (38.9%). CONCLUSION: The findings of this study will be helpful for increasing the shelf life of mulberry fruits through the application of appropriate control measures against infection by spoilage fungi during storage.

뽕나무 식재거리 및 원가지길이가 오디 수량과 생육에 미치는 영향 (Effects of Spacing and Main Branch Length on the Productivity of Mulberry Fruits and Growth of Mulberry)

  • 오디 생산을 위한 재배법 개발을 위하여 원줄기 길이 1m로 하여 원가지를 3~5개 정도로 유지하는 중간만들기 수형으로 유지하였다. 이 때 뽕나무 재배에 적합한 식재거리는 2${\times}$1 m(500주/10a)로 하는 것이 2${\times}$1.5 m(333주/10a)에 비하여 주당수량은 약간 적으나 식재 주수가 많아 단위면적당 수량은 높은 것으로 나타났다. 원가지 길이를 30, 50, 70 및 90 cm로 한 경우 90 cm 처리구에서 오디의 수량이 가장 많았다. 원가지 길이별 주당 가지수는 원가지 길이가 길수록 가지수가 많았으며, 10a당 가지량 및 엽량은 일정한 경향을 보이지 않았다. 식재 거리별 주당 가지수는 식재거리가 넓을수록 가지수가 많았고, 10a당 가지량 및 엽량은 일정한 경향을 보이지 않았다.

Optimization of Drying Conditions for Quality Semi-dried Mulberry Fruit (Morus alba L.) using Response Surface Methodology

  • Mulberry fruits were semi-dried using hot air ($60-100^{\circ}C$) or cool air ($20-40^{\circ}C$), and the effects of the drying temperature and processing time on the quality of the final dried mulberry fruits were investigated. Response surface methodology was employed to establish a statistical model and predict the conditions resulting in minimal loss of the total phenolic content (TPC) and ascorbic acid. Thus, using overlapped contour plots, the optimal conditions for producing semi-dried mulberry fruits, which reduced the moisture residue to 45% and minimized the nutrient losses of TPC and ascorbic acid, were determined for the hot-air process ($60.7^{\circ}C$ for 5.4 h) and cool-air process ($34.8^{\circ}C$ for 23.3 h). Plus, a higher drying temperature was found to lead to a faster loss of moisture and ascorbic acid, while the TPC was significantly decreased in the cool-air dried mulberry fruits due to the higher activity of polyphenol oxidase between 30 and $40^{\circ}C$.

오디와 포도의 안토시아닌 색소 물질 조성 비교 및 C3G 정량 (Quantification of Cyanidin-3-glucoside(C3G) in Mulberry Fruits and Grapes)

  • 최근 안토시아닌 색소의 생리활성에 대한 관심이 고조됨에 따라 천연색소 자원으로서 유망시되는 뽕나무 오디의 이용성을 높이고 생리활성물질로서 C3G(cyanidin-3-glucoside) 색소를 다량 함유한 뽕나무의 품종육성을 위하여 유전자원으로 보존되고 있는 뽕 계통에 대하여 오디를 수확한 후 색소를 추출, 분리 및 정량 분석을 실시하였다. 1. 오디 함유 anthocyanin 색소의 추출은 1% HCl-MeOH 용액을 사용하였으며, 분석 기기로는 Shimadzu LC, Nova-pack $C_{18}$ Column(300${\times}$3.9 mm)을 사용하였다. 2. 오디와 포도에 존재하는 안토시아닌 색소의 조성 패턴을 비교한 결과, 오디는 C3G(cyanidin-3-glucoside) 단일 물질로 존재하는 반면, 포도는 여러 가지 안토시아닌 색소가 혼합되어 존재하는 것으로 나타났다. 3. 오디와 포도의 계통별 C3G(cyanidin-3-glucoside) 함량을 분석한 결과, 포도에 비해 오디의 C3G함량이 상대적으로 매우 높았다. 4. 이상의 결과 뽕나무 열매인 오디는 안토시아닌 색소의 존재형태, 이용부위 및 함량에 있어 천연색소 자원으로서 유망시되며, 색소자원으로 이용시 타작물보다 유리한 경제수종으로 판단된다.

오디의 항당뇨 효능에 관한 연구 (A Study on the Antidiabetic Effect of Mulberry Fruits)

  • 인슐린 의존형 당뇨병 및 인슐린 비의존형 당뇨명 모델동물을 이용하여 오디의 항당뇨 효과를 검정해 보았다. 오디의 투여에 의해 정상 ICR 마우스 및 STZ 투여로 제1형 당뇨병을 유발한 ICR 마우스에서 체중과 혈당치의 유의성 있는 변화를 관찰할 수 없었다. 뿐만 아니라 STZ-유도성 당뇨병 유발억제 효과에 대한 실험에서도 역시 유의성 있는 억제 효과를 나타내지 않았으나 STZ로 유발한 당뇨병에서 용량의존적으로 혈당이 강하되는 경향을 관찰할 수 있었다. 2일령의 랫트에 STZ를 투여하여 제2형 당뇨병을 유발한 실험에서 오디 투여군은 혈당을 유의성 있는 감소를 나타내었으며, 간장의 무게 역시 대조군에 대해 유의성 있게 감소되었다. 그리고 자연발생 제2형 당뇨병 모델동물인 KK 마우스에서 오디 투여군은 대조군에 비해 내당능이 유의성 있게 회복되는 경향을 관찰 할 수 있었다. 오디는 제1형당뇨병에 대해서는 유의한 영향을 미치지 못하는 반면, 제2형당뇨병에 대해서는 유의성 있는 향당뇨 효능을 나타내었다. 이러한 사실은 오디를 이용하여 인슐린 비의존성 당뇨병에 대하여 항당뇨효과를 가진 기능성 식품을 개발 할 수 있는 가능성을 시사하고 있다.

Characteristics of newly bred mulberry variety, 'Saealchan' resistant to sclerotial disease

  • We bred a mulberry cultivar named 'Saealchan' through local adaptability test, which is under registration as a new cultivar for fruit production. Local adaptability test had been carried out at four local places (Jeonju, Gongju, Jangseong, and Sangju) for five years from 2013. This variety 'Saealchan' belongs to Morus alba L. selected from seedlings collected from mother mulberry 'Shimheung'. Saealchan was resistant to popcorn disease and high yielding variety in fruit productivity by 194% compared to control cultivar 'Shimheung (Morus alba L.)' for three years. Although fruits of 'Saealchan' was smaller in size and lower in sugar content of mulberry fruits compared to control variety 'Shimheung', it showed strong resistance to sclerotial disease compared to 'Shimheung'.

Mulberry popcorn disease occurrence in Korea region and development of integrative control method

  • Mulberry fruits also have tremendous potential for providing various valuable industrial products of very high economic value for human beings. Nevertheless, through global warming, the popcorn disease caused by sclerotia forming fungi reduces the productivity of mulberry fruits in worldwide. So, in this study, we investigated damage ratio of mulberry popcorn disease in mulberry fruit production farm (Buan, Jeongueb, Sangju, Gochang in Korea). In Jeonbuk Buan, popcorn disease rate was the highest about 30%, on the other hand, in case of Gyungbuk Sangju and Jeonbuk Gochang, not damage. Also, we investigated about popcorn disease prevention by various of chemical treatment methods.

  • Anthocyanin and polyphenolic compounds present in fruits of mulberry (Morus alba L.) were determined and the influence of temperature and UV irradiation on stability of the anthocyanin-copigment complex were investigated. The copigmentation substance selected in non-anthocyanin fraction from mulberry for the study included: phenolic acid (hydroxybenzoic acid) and flavonoid (quercetin-3-O-$\beta$-D-glucopyranoside). The copigmentation effect increased with the copigment content. UV irradiation had a stronger degradation effect on the copigmentation complex than heating at $80^{\circ}C$. The non-anthocyanin fraction of mulberry and isolated flavonoid (quercetin-3-O-$\beta$-D-glucopyranoside) from mulberry fruit predominated over other copigment substances.

  • This work was preliminary report on the shape of mulberry fruits and mulberry flowers. The shape of mulberry was varied with mulberry cultivars as bended oval shape (Daeshim), oval shape (Gwasang No. 2), oval-cylindrical shape (Sagyeppong), uneven heart shape (Suwonppong), and roundish shape (Cheonggangsang). The style of female flowers was also varied with cultivars as no style (Palchung), short style (Gwasang No. 2), cone style (Susungppong), and rectangular style (Daedangsang). This observation suggested that the shape and flower of mulberries are different among mulberry cultivars. It needs to further systematic study to know the shape and flower of mulberry cultivars and to elucidate the relationship between the morphological characters and mulberry cultivars.
