Antioxidative Constituents from Lycopus lucidus

  • Three phenolic compounds, rosmarinic acid (1), methyl rosmarinate (2), ethyl rosmarinate (3), and two flavonoids, luteolin (4), luteolin-7-O-$\beta$-D-glucuronide methyl ester (5) were isolated from the aerial part of Lycopus lucidus (Labiatae). Their structures were determined by chemical and spectral analysis. Compounds 1-5 exhibited potent antioxidative activity on the NBT superoxide scavenging assay. The $IC_{50}$ values for compounds 1-5 were 2.59, 1.42, 0.78, 2.83, and 3.05 $\mu\textrm{g}$/mL respectively. In addition, five compounds were isolated from this plant for the first time.

  • Four flavonoids (1-4), a phenylethyl glycoside (5), and a phenylpropanoid glycoside (6) were isolated from the flowers of Buddleia officinalis (Loganiaceae). Their structures were determined by chemical and spectral analysis. Among the isolated compounds, luteolin (1) and acteoside (6) exhibited the most potent antioxidative activity on the NBT superoxide scavenging assay. In addition, compounds 1-6 revealed weak antifungal activity, and no hemolytic activity.

  • A new phenolic amide, dihydro-N-caffeoyltyramine (1) was isolated from the root bark of Lycium chinense Miller, along with known compounds, trans-N-caffeoyltyramine (2), cis-N-caffeoyltyramine (3), and lyoniresinol $3{\alpha}-Ο-{\beta}-D-glucopyranoside$ (4). Their structures were determined by spectroscopic analysis. A NBT superoxide scavenging assay revealed that three phenolic amides showed potent antioxidative activity.

  • Bromotopsentin (BSM) is a bisindole alkaloid compound, which is recognized as a metabolite of the marine sponge Spongosorites sp. In this study, the antioxidant activity of BSM was investigated. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the trolox equivalent antioxidant capacity (TEAC) assay, the superoxide radical scavenging (NBT) assay, the lipid peroxidation and hydroxyl radical-induced DNA damage assays were carried out to evaluate the antioxidant activity of BSM. It was found that BSM had stronger scavenging activity on the stable free radical DPPH and superoxide radical than L-ascorbic acid with an $IC_{50}$ value of 62 and 64 ${\mu}M$, respectively. The TEAC value which indicated the total antioxidant capacity of BSM was about 0.8, which was also stronger than L-ascorbic acid. About 1.3 ${\mu}M$ of BSM induced 50% inhibition of lipid peroxidation. 60 nM of BSM exhibited a significant protective activity against DNA strand scission by hydroxyl radical on pBR322 DNA. Taken together, we suggest that BSM possesses strong antioxidant activity, and could be a valuable new addition to the list of anti-aging chemotherapeutic agents.

Anti-oxidant Activities of Punica granatum Extracts (석류 추출물의 항산화 작용)

  • Pomegranate (Punica granatum L.) which is very rich in Polyphenols and tannins was recently reported its anti-oxidant activities and phytoestrogenic activities in vivo test and many clinical studies, but the effects of them on the skin have not been reported. The experiments were tarried out in vitro to determine anti-oxidant activities of pomegranate extracts on DPPH radical scavenging assay, NBT/Xanthine Oxidase-superoxide scavenging assay, silica-induced intracellular $H_2O_2$, hydroperoxide and superoxide generation assay in RAW 264.7 cells. It showed that the methanolic extract of dried pomegranate peels have the most significant anti-oxidant activities on free radical scavenging assay and inhibitory activities on silica-induced intracellular free radical generation in RAW 264.7 cells. The concentrated juice of pomegranate showed only DPPH radical scavenging activities and inhibited hyaluronidase activity. Moreover, pomegranate seed oil inhibited specially silica-induced intracellular hydroperoxide generation in RAW 264.7 cells. These results suggest that the methanolic extract of dried pomegranate peels and pomegranate seed oil have more anti-oxidant activity than concentrated juice of pomegranate. Thus the extracts of pomegranate peels and seed oil could be developed cosmetic ingredients for anti-aging.

Antioxidant Effects of Noni (Morinda citrifolia) Extracts Treated with Hel and Trypsin (염산과 트립신으로 처리한 노니(Morinda citrifolia) 추출물의 항산화 효과)

  • To investigate biological activity of noni extracts treated with HCl and trypsin, we measured the antioxidant activity through vitro assay and cellular system. Both water and lipid soluble fraction of noni extracts dose-dependently scav­enged DPPH radical. Superoxide scavenging activity of lipid soluble fraction after treating HCl and trypsin was significantly more potent than those of other fractions in NBT/xanthine oxidase assay, which suggests that antioxidant activity of noni extracts was increased by the treatment with HCl and trypsin. In antioxidant assay using RBL 2H3 cells, water soluble frac­tion of noni extracts had little effect on silica-induced reactive oxygen species generation, whereas lipid soluble fraction inhibited in a dose dependent manner. In non-treated noni extracts, effect of water soluble fraction on silica/$CuSO_4$-induced lipid peroxidation was more potent than that of lipid soluble fraction. However, the effects of noni extracts were reversed in noni extracts treated with HCl and trypsin. These data suggest that water soluble substances may be converted into lipid soluble substances by the treatment with HCl and trypsin. From the above results, it is suggested that lipid soluble fraction of noni extracts contain antioxidant used in vitro assay and RBL 2H3 cellular system. Such an effect of noni extracts may be increased by the treatment with HCl and trypsin.

Lignans Isolated from the Roots of Patrinia scabiosaefolia F. and their IL(Interleukin)-6 Inhibitory Activity (마타리 뿌리로부터 분리한 Lignan 화합물의 IL-6 저해활성)

  • In an ongoing investigation into anti-inflammatory compounds from natural products, the $CH_2Cl_2$ soluble fraction of Patrinia scabiosaefolia F. (Valerianaceae) was found to inhibit IL-6 production in TNF-$\alpha$ stimulated MG-63 cell line. By means of a bioassay-directed chromatographic separation technique, lappaol E (1), and nortrachelogenin (2) were isolated. These compounds have been isolated from this plant for the first time. Compounds $1{\sim}2$ showed potent antioxidative activity using NBT superoxide scavenging assay. Moreover, compound 1 decreased IL-6 production in TNF-$\alpha$ stimulated MG-63 cell line.

Anti-oxidant and Anti-aging Activities of Essential Oils of Pinus densiflora Needles and Twigs (소나무 잎과 가지의 정유와 그 성분들의 항산화 및 항노화 작용)

  • To investigate effects of pine essential oil isolated by steam distillation from Pinus densiflora needles (PN) and twigs (PT) on anti-oxidant and anti-aging activities, the experiments were carried out to determine anti-oxidant and antiaging activities on DPPH radical scavenging activity, NBT/xanthine oxidase-superoxide scavenging activity, silica-induced intracellular $H_2O_2$ and hydroxyl radical generation in RAW264.7 cells, hyaluronidase and elastase activities in vitro. Essential oil of PN and PT were contained 0.225 and 0.176 (w/v) %, respectively. PN was contained with 30 kinds of essential oil and its major constituent is $\alpha$-pinene (21.5%). Further PT was contained with 40 kinds of essential oil and its major constituent is $\beta$-pinene (22.4%) in GC/MS assay. Other essential oils of PN were $\beta$-pinene, camphene, myrcene, limonene, terpinolene, $\alpha$-terpineol, bornyl acetate, $\alpha$-copaene, caryophellene and humulene and PT were $\alpha$-pinene, camphene, phellandrene, limonene, terpinolene, $\alpha$-terpineol, bornyl acetate, $\alpha$-copaene, caryophellene and humulene. The essential oil of PT have more active than that of PN in anti-oxidant activity which has significant DPPH radical and superoxide scavenging activity, and significant inhibitory activities on silica-induced intracellular $H_2O_2$ and hydroxyl radical generation, as well as, significantly inhibited elastase and hyaluronidase activities. Further, phellandrene, myrcene and $\alpha$-pinene have DPPH radical and superoxide scavenging activities, $\beta$-pinene, terpinolene, myrcene and phellandrene inhibited silica-induced intracellular $H_2O_2$ and hydroxyl radical generations. And also phellandrene and $\beta$-pinene inhibited hyaluronidase and elastase activities. In conclusion, the essential oils isolated from PN and PT have anti-oxidant and anti-aging activities.

Biological Activity of Flavonoids Isolated from Aster tataricus L. (자원에서 분리한 플라보노이드의 생리활성)

  • In an ongoing investigation into anti-oxidative compounds from natural products, the EtOAc soluble fraction of Aster tataricus L. (Compositae) showed significant anti-oxidative activity on the NBT superoxide scavenging assay. By means of an activity-guided purification, three flavonoids, kaempferol (1), quercetin (2), astragalin (3) and one monoterpene glycoside, shionoside A (4) were isolated. Their structures were determined spectral analyses. Compounds 2 and 3 showed potent antioxidative activity, while, compounds 1 and 4 were inactive $IC_{50}$>120${\mu}g/mL$. In addition, these compounds were examined for the effect of interleukin-6 (IL-6) production in TNF-${\alpha}$ stimulated MG-63 cell. Compounds 1-3 showed negligible inhibitory activity against IL-6 production in $TNF-{\alpha}$ stimulated $MG-6{\beta}$ cell, and compound 4 was inactive.

Anti-oxidative Activities of Castanea crenata Leaf Extract/Fractions and Application on Cosmetics (밤나무 잎 추출물의 항노화 활성 및 화장품에의 응용( I ))

  • In this study, the antioxidative effects, inhibitory effects on tyrosinase and elastase and components of Castanea crenata leaf were investigated. The free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity ($FSC_{50}$) of extract / fractions of Castanea crenata left was in the order: 50% ethanol extract ($13.6{\mu}g/mL$) < ethyl acetate fraction (6.2) < aglycone fraction (2.1). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$ of extract / fractions from Castanea crenata leaf extract / fractions on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activity was in the order: aglycone fraction (0.8) < 50% ethanol extract (0.5) < ethyl acetate fraction (0.3). The scavenging activity ($IC_{50}$ for ${O_2}^{{\cdot}\;-}$ (superoxide anion radical) generated by NBT method was in the order: ethyl acetate fraction (145.5) < aglycone fraction (65.5). The protective effects on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction exhibited the most prominent cellular protective effect (${\tau}_{50}$, $191.9{\pm}12.2\;min$ at $10{\mu}g/mL$). The inhibitory effect of aglycone fraction ($9.1{\mu}g/mL$) on elastase was higher than oleanolic and ($13.7{\mu}g/mL$). And the inhibitory effect of aglycone fraction ($21.6{\mu}g/mL$) on tyrosinase was higher than arbutin ($226.2{\mu}g/mL$). But 50% ethanol extract rarely exhibited the inhibitory activity on tryosinase and elastase. Flavonoids were contained in Castanea crenata left (96.3 mg / 100 g dried Castanea crenata leaf). And flavonoids contained in ethyl acetate fraction were kaempferol, quercetin, quercitrin, and so on. Quercitrin is the most abundant component. These results indicate that extract / fractions of Castanea crenata can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging free radical and ROS, Castanea crenata leaf extract/ fractions could be used as new cosmeceutical for whitening and anti-wrinkle products.
