Distribution of N-Methyl-D-Aspartate Receptor Subunits NR2A and NR2B in Chum salmon Brain (연어 뇌에서 N-Methyl-D-Aspartate 수용체 아단위 NR2A와 NR2B의 분포)

  • We carried out immunoblot analyses to study expression and subcellular distribution of the N-methyl-D-aspartate receptor(NR) subunits in salmon (Chum Salmon, Oncorhynchus keta). We prepared subcellular fractions such as brain homogenates, synaptosomes, and postsynaptic density (PSD) from salmon brains, and analyzed protein compositions by SDS-polyacrylamide gel electrophoresis (SDS-PAGE). In a Coomassie-stained 6% SDS-gel, about 20 distinct major protein bands could be identified in the PSD fraction. Immunoblot analyses using antibodies against rat NR subunit 2A and 2B antigens (NR2A and NR2B, respectively) showed weak but evident signals at the 180 kDa positions in the salmon PSD fractions. However, in contrast to rat NRs, the salmon NR2A and NR2B are not recognized by a phosphotyrosine-specific antibody suggesting that the salmon NRs are regulated differently from those of the rat by protein tyrosine kinases. Our results indicate that NR2A and NR2B subunits are expressed in the salmon PSD fraction but not regulated by tyrosine phosphorylation.

Effect of Intracellular Calcium Chelator on Phosphorylation of Spinal N-Methyl-D-Aspartate Receptor following Electroacupuncture Stimulation in Rats (칼슘 저해제가 전침자극에 의한 척수 N-Methyl-D-Aspartate 수용체 인산화에 미치는 영향)

  • Objectives : We investigated the role of intracellular calcium chelator, bis-(2-aminophenoxy)-ethane-N,N,N',N'-tetraacetic acid(BAPTA), on the modulation of phosphorylation of the spinal N-methyl-D-aspartate receptor(NMDAR) NR1 and NR2B subunits following electroacupuncture(EA). Methods : Bilateral 2 Hz EA stimulation with 1.0 mA was delivered at those acupoints corresponding to Zusanli(ST36) and Sanyinjiao(SP6) in man via needles for 30min. Results : EA analgesia was reduced by intra-peritoneal injection at a higher dose of BAPTA from termination of EA stimulation. At 60 min after EA treatment, the total number of c-fos-immunostained neurons in each regions of the dorsal horn in the $L_{4-5}$ segments was decreased by BAPTA injection, especially in nucleus proprius. The mean integrated optical density (IOD) of NR1 and NR2B subunits were increased only in superficial laminae of EA-treated rats when compared with normal rats. However, the mean IOD of pNR1 was significantly decreased by BAPTA injection in both the superficial laminae and neck region and pNR2B in the superficial laminae. Western blot analyses confirmed the decreased expression of pNR1 and pNR2B. Conclusions : We concluded that intracellular calcium may well play an important role in EA analgesia by modulating the phosphorylation state of spinal NMDAR subunits.

High Cytoplasmic Expression of the Orphan Nuclear Receptor NR4A2 Predicts Poor Survival in Nasopharyngeal Carcinoma

  • Objective: This study aimed at investigating whether the orphan nuclear receptor NR4A2 is significantly associated with clinicopathologic features and overall survival of patients with nasopharyngeal carcinoma (NPC). Methods: Immunohistochemistry was performed to determine NR4A2 protein expression in 84 NPC tissues and 20 non-cancerous nasopharyngeal (NP) tissues. The prognostic significance of NR4A2 protein expression was evaluated using Cox proportional hazards regression models and Kaplan-Meier survival analysis. Results: We did not find a significant association between total NR4A2 expression and clinicopathological variables in 84 patients with NPC. However, we observed that high cytoplasmic expression of NR4A2 was significantly associated with tumor size (T classification) (P = 0.006), lymph node metastasis (N classification) (P = 0.002) and clinical stage (P = 0.017). Patients with higher cytoplasmic NR4A2 expression had a significantly lower survival rate than those with lower cytoplasmic NR4A2 expression (P = 0.004). Multivariate Cox regression analysis analysis suggested that the level of cytoplasmic NR4A2 expression was an independent prognostic indicator for overall survival of patients with NPC (P = 0.033). Conclusions: High cytoplasmic expression of NR4A2 is a potential unfavorable prognostic factor for patients with NPC.

Identification of a Potential Tyrosine Phosphorylation Site on the NR2B Subunit of the N-methyl-D-aspartate Receptor (NMDA 수용체 아단위 2B의 Tyrosine 인산화 위치의 동정)

  • The 2B subunit of N-methyl-D-aspartate (NMDA) receptors (NR2B) is the major phosphotyrosine-containing pro-tein in the postsynaptic density (PSD). In order to identify the site for tyrosine phosphorylation on NR2B, a mass spectrometry was applied on tryptic and endolys-C peptides. The NR2B subunit was isolated from N-octyl glucoside (NOG)-insoluble PSD fraction through SDS-PAGE and electroelution. The eluted protein was confirmed to be NR2B and phosphorylated on tyrosine by its cognate antibody and phosphotyrosine-specific antibody. By matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry of the peptides generated by digesting the eluted NR2B with trysin or endolys-C, a potential site for tyrosine phosphorylation could be identified as Tyr-1304.

  • Effects of the ratio of rubber composition on covulcanization and ozone resistance were studied in this study. Specimens used in this study were rubber compounds(specimen-A) blended with various ratio of NR, SBR, BR, IIR, and EPDM, unsaturated rubber compounds(specimen-B) with NR/BR/SBR, and saturated rubber compounds(specimen-C) with NR/IIR/EPDM. PAD adhesion specimen was prepared from vulcanizing specimen-A and B, and specimen-A and C, respectively. Using same adhesion specimen, peel strength was measured and tested ozone resistance. In specimen-A, peel strength was higher with increasing NR ratio for NR and BR contained blends. In other specimen-A containing NR and SBR, the peel strength was also increased with increasing SBR ratio. NR/BR/IIR/EPDM rubber compounds had also better adhesion property than NR/SBR/IIR/EPDM compounds. As more unsaturated rubber was blended, the peel strength was higher but ozone resistance was worse. Optimum ratio of unsaturated and saturated rubbers for the peel strength and ozone resistance was 60/40.

  • T0901317 is a potent synthetic ligand for liver X receptor (LXR, NR1H2/3), a member of the nuclear receptor superfamily that functions as a transcription factor. However, T0901317 has been also reported to modulate the activity at least four other nuclear receptors (NRs), acting as agonists for farnesoid X receptor (FXR, NR1H4) and pregnane X receptor (PXR, NR1I2) and as antagonists for androgen receptor (AR, NR3C4) and retinoid-related orphan receptor-${\alpha}$ (ROR-${\alpha}$, NR1F1). We report here that T0901317 can also function as an inhibitor for constitutive androstane receptor (CAR, NR1I3). Since CAR is a major player of xenobiotic and cholesterol metabolism in the liver, along with PXR, FXR and LXR, which are reported to be regulated by T0901317, this further complicates the interpretation of potential results with T0901317 in liver cells.

Optimal Conditions of Protoplast Formation of Aspergillus coreanus NR 15-1 and Aspergilus oryzae NR 2-5 (Aspergillus coreanus NR 15-1 과 Aspergillus oryzae NR 2-5의 원형질체 형성의 최적조건)

  • Aspergil-lus coreanus NR-15 and Aspergilus oryzae NR-2-5 from traditional Korean Nuruk were selected as parental strains producing starch hydrolysis enzyme. Xll(Arginine-) mutant from A. coreanus NR 15-1 showed high glu-doamylase activity and total acid productivity. Z6(Adenine-) mutant from A. oryzae NR2-5 showed the highest $\alpha$-amylase activity. Therefore, both XII and Z6 mutants were selected and investigated for the optimal conditions of protoplast formation for protoplast fusion. Mixture of equal amount of cellulase and driselase(10mg/ml each) was the most effective as lytic enzymes. The optimal pH and temperature for protoplast formation were 5.0 and $30^{\circ}C$, respectively. The most effective reaction for protoplast formation time was 4 hours. The maximum of protoplst for- mation of Xll mutant and Z6 mutant were $6.54$\times$10^{7}$ protoplasts/ ml and $3.04$\times$10^{ 7}$ protoplasts/ml, and the regen-eration frequencies of the protoplasts were 11.3% and 11.6%, respectively. The size of the protoplasts from X11 and Z6 mutants were 3~6 $\mu\textrm{m}$ and 4~9$\mu\textrm{m}$, respectively.

Effect of Precured EPDM on the Property of Magneto-rheological Elastomer Based on NR/EPDM Blend

  • Magneto-rheological elastomers (MREs) are smart materials in which the inherent stiffness and damping properties can be changed by the influence of an external magnetic field. The magneto-rheological (MR) effect depends on the orientation characteristics of the dispersed magneto-responsible particles (MRPs) in the matrix. In this study, natural rubber (NR) and ethylene propylene diene rubber (EPDM) were blended and used as a matrix of an MRE. EPDM was pre-cured before blending with NR. The Mooney viscosity, curing characteristics, and mechanical properties were analyzed with various pre-curing conditions of EPDM and the NR/EPDM blend. The results show that excellent mechanical properties of the NR/EPDM blend-based MRE were obtained when the pre-curing time of EPDM was 60 min. The aging property of the NR-based MRE was improved by the introduction of pre-cured EPDM. Also, the anisotropic MRE showed a higher MR effect than that of the isotropic MRE.

Nutrient requirements in Hanwoo cows with artificial insemination: effects on blood metabolites and embryo recovery rate

  • Here, we investigated the effects of different nutrient requirements (NR) on blood metabolites, transferable embryo number after multiple superovulations with artificial insemination (AI), body condition score (BCS), and estrus cycle in Hanwoo cow. Nineteen Hanwoo cows were randomly divided into three groups (80%, 100%, and 120% NR, containing 6, 8, and 5 individuals, respectively) and fed based on the NR. In experiment 1, glucose, total cholesterol, triglyceride, blood urea nitrogen (BUN), aspartate aminotransferase (AST), alanine aminotransferase (ALT), non-esterified fatty acids (NEFA), albumin (ALB), and total protein (TP) were analyzed. In experiment 2, total number of recovered embryos and transferable embryos was examined after embryo recovery and multiple superovulations with AI. In experiment 3, body weight, BCS, and estrus cycle were examined. In experiment 1, total cholesterol was significantly different among the 80%, 100%, and 120% NR groups (126.5 ± 3.3, 152.6 ± 2.4, and 177.4 ± 1.8 mg/dL, respectively, p < 0.05). The triglyceride and BUN levels in the 120% NR group were significantly higher than those in the 80% and 100% groups (p < 0.05). The NEFA levels were significantly different among the 80%, 100%, and 120% NR groups (440.5 ± 18.2, 318.5 ± 23.1, and 195.1 ± 8.5 ЧEq/L, respectively, p < 0.05). The AST and TP levels in the 80% NR group were significantly lower than those in the 100% and 120% NR groups (p < 0.05). In experiment 2, the 120% NR group showed a higher percentage of transferable embryos than the 80% and 100% groups (p < 0.01). The mean body weight and BCS among the 80%, 100%, and 120% NR groups were significantly different (p < 0.05). The estrus cycle in the 80% NR group was delayed compared with the 100% and 120% NR groups (20.8 ± 0.2 and 21.2 ± 0.5 days, respectively). In conclusion, the blood metabolic tests proved that Hanwoo cows with 120% NR can produce a large number of transferable embryos. Thus, 120% NR is the appropriate feeding level for this type of cows as it results in the production of a large number of transferable embryos by multiple superovulations with AI.

Noise Reduction Method in consideration of bandwidth of Low Pass Filter (저역통과 필터의 대역폭을 고려한 잡음 제거 방법)

  • Most digital cameras apply a Charge Coupled Device(CCD) Sensor or a Complementary Metal Oxide Semiconductor(COMS) Sensor. And the images captured these sensors include unwanted noises. In this paper, we proposed a method of the Noise Reduction(NR) about noise reduction or recovery in the image. The proposed NR method is related to Bandwidth of the Low Pass Filter(LPF). For improvement of NR, we were changing the filter coefficient of the LPF. The results of simulations with various filter coefficients, [1 2 1] in the LPF and [-1 2 -1] in the High Pass Filter(HPF) have ideal frequency bandwidth and high performance. We proposed a filter coefficient [1 2 1] and [-1 2 -1] in the LPF and the HPF respectively.
