한국 여수 연안에 출현하는 참문어(Octopus vulgaris)의 위내용물 조성 (Diet Composition of Common Octopus Octopus vulgaris in the Coastal Waters of Yeosu, Korea)

  • The diet composition of octopus Octopus vulgaris was studied using 392 specimens collected from January to December 2020 in the coastal waters of Yeosu, Korea. The body weight ranged from 86.4 to 3,645.4 g. The most important prey item in the diet of O. vulgaris was decapods, which constituted 43.9% in ranking index (%RI). The second largest prey item was Pisces (%RI=27.9%), and cephalopods (%RI=21.8%) constituted the third largest prey item. The average trophic level of O. vulgaris was 3.97±0.59. Ontogenetic changes were significant among size classes (<400 g, 400-800 g, ≥800 g). The small size class (<400 g) mainly fed on decapods, whereas the medium (400-800 g) and large size (≥800 g) classes mainly fed on cephalopods and Pisces, respectively. The dietary composition varied significantly with season.

  • The purpose of this study is to provide information about annual variations in catch size and changes in reproductive biology in the common octopus, Octopus vulgaris, a commercially important species in Jeju Island. Samples were collected from coastal waters of Jeju Island, Korea by coastal trap fishery from January to November 2021. Octopus vulgaris have been decreasing in this region since 1984. The mantle length (ML) ranged from 4.5 to 17.8 cm and body weight (BW) ranged form 88.5 to 2,657.5 g. A χ2-test revealed that the number of males was significantly greater than females (χ2 = 32.712, df = 10, P<0.05). Mature females were found from January to July and male occurred January to August. The gonadosomatic index value was higher from July to August than other months. The relationship between mantle length and body weight was BW = 2.4527ML2.3139 (female) and BW = 2.6785ML2.3159 (male). At 50% group maturity, female and male average weight was estimated to be 554.7 g and 330.6 g, respectively.

  • The purpose of this study was to provide information about annual variations in catch size and changes in reproductive biology in the common octopus Octopus vulgaris, a commercially important species. Samples were collected from coastal waters off Gyeongnam, Korea, by coastal trap and coastal complex fishery from February to December 2018. Octopus vulgaris have been decreasing in this region since 2007 and catch prices have been increasing. The mantle length (ML) of O. vulgaris ranged from 1.2 to 21.2 cm and body weight ranged from 42.6 to 5,537 g. The sex ratio differed significantly from what was expected, at 0.45:0.55 (P<0.05). Mature females were found from February to December, except in October and November, and males occurred from February to December, except in October. The gonadosomatic index value was higher from May to September than in other months. The total number of eggs in fecund individuals ranged from 40,280 in 578.1 g of body weight to 138,994 in 1,666.5 g of body weight. The relationship between body weight and fecundity was F=76.486BW1.0087 (r2=0.7897). At 50% group maturity, average weight was estimated to be 636.35 g, while it was 969.30 g at 75% maturity and 1736.55 g at 97.5% maturity. This study highlights changes in catch size and reproductive biology between 2007 and 2018. These results provide basic evidence that may be used to establish regulatory policy for O. vulgaris fisheries.

  • The maturity and spawning of the common octopus Octopus vulgaris were studied using 317 samples collected monthly from January to December of 2020 from the coastal waters of Yeosu, Korea. The mantle length (ML) of O. vulgaris ranged from 5.1 to 19.7 cm and body weight (BW) ranged from 117.6 to 3,645.4 g. We analyzed monthly changes in sex ratio, gonadosomatic index (GSI), histological maturity stage, and body weight at 50%, 75% and 97.5% group maturity. The sex ratio was 1:1.02, which was not significantly different from the value that expected. Based on histological observation of the gonad development and gonadosomatic index, the spawning periods lasted throughout the year, with peak spawning periods estimated from March to April and July to August. The percentages of sexually mature females estimated using a logistic equation were over 50% at 554.46 g BW, 75% at 1,134.38 g BW and 97.5% at 2,474.22 g BW respectively.

Comparison of the Exopeptidase Activity of Fractions from Crude Extracts of Octopus Octopus vulgaris Cuvier Hepatopancreas Using Different Fractionation Methods

  • This study was performed to identify the optimum fractionation method and conditions to obtain exopeptidase-active fractions from octopus hepatopancreas (HP) crude extracts (CEs) using four techniques: solid ammonium sulfate fractionation, polyethylene glycol (PEG) fractionation, anion exchange chromatography, and gel filtration chromatography. The fractions with the highest total activity toward L-leucine-p-nitroanilide (Leu-pNA) were fraction IV from the ammonium sulfate and PEG fractionation, and fraction II in ion exchange and gel filtration chromatography. The total exoprotease activity of these fractions was highest in fraction IV (4,050.20 U) of ammonium sulfate fractionation, followed by fraction II (3,600.28 U) from gel filtration chromatography, fraction IV (2,861.30 U) from PEG fractionation, and fraction II (2,576.28 U) from ion exchange chromatography. These results suggest that ammonium sulfate fractionation using 60-80% ammonium sulfate was the most efficient method for separating the exoprotease active fractions from CEs of octopus HP.

  • This study aimed to optimize mixing conditions (adding amount of squid skin and sea tangle Saccharina japonica) for concentrates of octopus Octopus vulgaris cooking effluent (COCE) and roasting conditions (temperature and time) of seasoned Laver Pyropia yezoensis with concentrates of octopus cooking effluent (SL-COCE) using response surface methodology (RSM). The results of RSM program for COCE showed that the optimum independent variables ($X_1$, squid skin amount; $X_2$, sea tangle amount) based on the dependent variables ($Y_1$, odor intensity; $Y_2$, amino-N content; $Y_3$, sensory overall acceptance) for high-quality COCE were 0.53% (w/w) for $X_1$ and 0.48% (w/w) for $X_2$ for uncoded values. The results of the RSM program for SL-COCE showed that the optimum independent variables ($X_1$, roasted temp.; $X_2$, roasted time) based on the dependent variables ($Y_1$, burnt odor intensity; $Y_2$, water activity; $Y_3$, sensory overall acceptance) for high-quality SL-COCE were $344^{\circ}C$ for $X_1$ and 8 sec for $X_2$ for uncoded values. The SL-COCE prepared under optimum procedure was superior in sensory overall acceptance to commercial seasoned laver.

  • This study aimed to compare on sensory and nutritional characteristics between seasoned Laver Pyropia yezoensis with concentrates of octopus Octopus vulgaris cooking effluent (SL-COCE) and commercial seasoned laver (C-SL). Proximate composition of SL-COCE was significantly higher in moisture and ash, lower in lipid than those of C-SL. However, there was no differences in protein between seasoned lavers (P<0.05). The SL-COCE was stronger in sensory appearance, taste, flavor and texture than the C-SL. The SL-COCE was higher in mineral (Ca, P, K, Fe and Zn) than the C-SL. Total amino acid content of SL-COCE was slightly higher than that of C-SL. The major amino acids of SL-COCE were aspartic acid, glutamic acid and alanine. Mineral content based on the 100 g of SL-COCE was 272.3 mg in calcium, 392.2 mg in phosphorus, 1,025.8 mg in potassium, 10.6 mg in iron, 4.4 mg in zinc, which was higher than C-SL. Total content of fatty acid per the 100 g of SL-COCE was 35,098 mg, which was lower than C-SL (39,679 mg). The major fatty acids of SL-COCE were 16:0, 18:1n-9 and 18:2n-6.

  • 이 연구는 경남 통영과 사천지역의 연안에서 2007년 2월부터 2008년 1월까지 연안통발어업에 의해 어획된 참문어를 매월 1회 채집하여 성숙과 산란에 관한 연구를 수행하였다. 조직학적 검경을 통한 생식소 성숙도의 월별 발달과정을 분석한 결과 암컷의 경우, 조사 기간 중 미숙, 중숙, 성숙, 완숙기의 개체들이 혼재하여 출현하였으며 산란이 가능한 완숙 개체는 10월과 12월을 제외하고 연중 관찰되었다. 수컷의 경우도 조사기간 중 미숙, 중숙, 성숙, 완숙기의 개체들이 혼재하여 출현하였으며 방정이 가능한 성숙 이상의 개체는 전 기간에 걸쳐 관찰되었다. 생식소 중량지수의 월 변화는 암컷의 경우, 2월 1.26에서 5월 4.84로 증가하다 이후 감소하였으며 8,9월에 다시 상승하였다가 10월에 0.38의 낮은 값으로 감소하였다. 이러한 생식소 성숙도의 월별발달과정과 생식소 중량지수의 월변화를 바탕으로 참문어의 산란기를 추정한 결과 주 산란기는 9월로 나타났다. 수컷의 경우는 연중 1.5이상의 높은 값을 나타냈다. 참문어의 50% 군 성숙체중은 919.6 g로 추정되었다.

  • 본 연구는 경남 통영과 사천지역의 연안에서 2007년 2월부터 2008년 1월까지 매월 1회 연안통발어업에 의해 채집된 참문어를 대상으로 참문어의 성숙과 산란에 관하여 조사하였다. 생식소 성숙도의 월별 발달과정을 분석한 결과 암컷의 경우, 조사 기간 중 미숙, 중숙, 성숙, 완숙기의 개체들이 혼재하여 출현하였으며 산란이 가능한 완숙 개체는 10월과 12월을 제외하고 연중 관찰되었다. 수컷의 경우도 조사기간 중 미숙, 중숙, 성숙, 완숙기의 개체들이 혼재하여 출현하였으며 방정이 가능한 성숙 이상의 개체는 전 기간에 걸쳐 관찰되었다. 난소의 성숙발달에 따른 난경크기 변화를 알아보기 위해 난소 내 난경조성의 월 변화를 조사한 결과 9월에 1.1-1.2 mm로 최고값을 나타냈다. 암수의 비율은 약 0.48:0.52로 나타났지만 암수의 성비(1:1)에 대한 유의한 차는 없었다(p > 0.05).

$CO_2$ 환경에서의 참문어의 급성 독성반응 (Acute Toxic Responses of Octopus vulgaris to $CO_2$ Environment)

  • 이산화탄소($CO_2$) 해양격리처리 방안의 실효성을 검토하기 위해서는 해양에 처리된 $CO_2$가 해양생태계 및 해양생물에 미칠 수 있는 영향에 대해서 광범위하게 정보를 축적할 필요가 있다. 본 연구에서는 두족류인 참문어 Octopus vulgaris를 대상으로 고농도 $CO_2$ 환경(1, 2, 3%-$CO_2$)에서의 폐사율 및 1%-$CO_2$ 환경에서의 생리학적 반응에 대해서 조사하였다. 참문어는 혈액채취를 위하여 복부대동맥에 케뉴레이션을 행한 후 호흡실에서 회복시켰다. 회복된 개체에 대하여 헤모림프의 산염기 조절인자에 대한 측정을 행하였다. 참문어는 3%-$CO_2$ 환경에서 72시간 안에 100% 폐사하였다. 헤모림프의 pH는 1%-$CO_2$ 노출 30분 후 유의하게 감소하였으나 실험종료 때까지 회복되지 않았으며, $[HCO_3^-]$$CO_2$ 노출 후 유의하게 증가하여 8시간에 7.8 mM를 나타내었으나 그 후 점차 감소하는 경향이었다. 헤모림프 이온$([Cl^-],\;[Na^+],\;[K^+])$들은 유의한 변화를 보이지 않았다. 본 연구 결과 참문어는 방어, 넙치와 같은 경골어류 및 별상어와 같은 판새류보다 $CO_2$에 민감한 것으로 사료된다.
