천년초 분말 첨가 파스타의 품질특성 및 제조조건 최적화 (The Antioxidative Characteristics of Opuntia humifusa and its Optimal Conditions for Pasta Production)

  • The purpose of this study was to investigate the antioxidative effects of Opuntia humifusa and determine the optimal mixing ratio of Opuntia humifusa powder and eggs for the preparation of pasta. In regard to its antioxidant effects, Opuntia humifusa powder had a total phenolic content and DPPH free radical scavenging activity of 6.64 mg/g and 90.12%, respectively. We used response surface methodology to obtain ten experimental points (including two replicates for Opuntia humifusa powder and egg) and Opuntia humifusa pasta formulation was optimized using rheology. Yellowness (p<0.001) displayed a linear model pattern, whereas lightness (p<0.01) and redness (p<0.01) were described by a quadratic model pattern. Mechanical properties showed significant values in hardness (p<0.5), chewiness (p<0.5), gummiess (p<0.5), and cohesiveness (p<0.5). The sensory evaluation parameters showed significant differences in color (p<0.5), appearance (p<0.5), flavor (p<0.5), texture (p<0.5) and overall quality (p<0.5). The optimum formulations processed by numerical and graphical optimization were found to be 7.30 g of Opuntia humifusa powder and 31.55 g of egg.

  • The purpose of this study was to investigate the antioxidative effects of Opuntia humifusa powder and determine the optimal composite recipe by three different amounts of Opuntia humifusa powder, sugar and butter in cookies prepared with Opuntia humifusa powder. In regard to its antioxidant effects, Opuntia humifusa powder had a total phenolic content and DPPH free radical scavenging activity of $6.64{\pm}0.54mg/g$ and $90.12{\pm}0.42%$, respectively. response surface methodology was used to obtain sixteen experimental points(including three replicates for Opuntia humifusa powder, sugar and butter) and Opuntia humifusa cookie formulation was optimized using rheology. The results of the sensory evaluation produced very significant values for color (p<0.05), flavor (p<0.05), texture (p<0.05), sweetness (p<0.05), appearance (p<0.05), overall quality (p<0.05), and the results of instrumental analysis showed significant values in sweetness (p<0.001), hardness (p<0.001), lightness (p<0.001), redness (p<0.001), yellowness (p<0.01). As a results, the optimum formulations obtained by numerical and graphical methods were found to be 3.11 g of Opuntia humifusa powder, 35.00 g of sugar and 50.54 g of butter.

3종의 버섯균사체로 발효한 천년초 에탄올 추출물의 생리활성 (Biological Activity of Ethanol Extracts from Fermented Opuntia humifusa with 3 Different Mushroom Mycelia)

  • The present study was carried out to investigate the biological activity of ethanol extracts from fermented Opuntia humifusa with 3 different mushroom mycelia: Phellinus linteus, Lentinula edodes, and Pleurotus ostreatus. Fermented Opuntia humifusa by Phellinus linteus (FOP) and Lentinula edodes (FOL) showed more DPPH and ABTS radical scavenging activities than non-fermented Opuntia humifusa (NFO) and fermented Opuntia humifusa by Pleurotus ostreatus (FOPO). At a concentration of 250 ppm, the ABTS radical scavenging activities of the FOP and FOL were similar to that of BHA, a synthetic antioxidant. The total polyphenol content had a similar tendency to that of the radical scavenging activity. However, the flavonoid content was increased in the order of NFO, FOL, FOPO, and FOP. At all concentrations, the tyrosinase inhibitory activity of FOP and FOPO were significantly higher than that of kojic acid. During adipocyte differentiation, NFO and FOL showed no significant difference in lipid accumulation in 3T3-L1 cells. FOP and FOPO showed a higher fat accumulation inhibitory effect than NFO and FOL. These results provide baseline data for Opuntia humifusa as a novel functional food.

천년초 열매 분말을 첨가한 절편의 품질특성 (Quality Properties of Jeolpyun supplemented with Cheonnyuncho (Opuntia humifusa))

  • This study was undertaken to investigate the effect of Opuntia humifusa on the improvement of quality properties of Korean Jeolpyun. Chemical and mechanical properties of unsupplemented Jeolpyun and Jeolpyun supplemented with 1%-4% O. humifusa. Also, a sensory evaluation by a trained panel was conducted. Concerning color difference, the L value decreased, but a and b values increased as O. humifusa content increased. Texture analysis revealed that hardness, cohesiveness and adhesiveness were the highest in the unsupplemented control group. The texture parameters decreased as the O. humifusa addition ratio increased. In sensory evaluation, color and taste scores were highest score in 2% O. humifusa supplemented samples, with no difference in flavor being evident. Chewiness and adhesiveness scores were highest score in 4%-supplemented O. humifusa samples. Overall quality score was highest in the 2% O. humifusa samples. The addition of $2{\sim}3%$ O. humifusa optimizes Korean Jeolpyun.

천년초 씨 추출물이 난소를 절제한 흰쥐의 혈소판 응집과 혈청 내 지질 조성에 미치는 영향 (The Effect of Opuntia humifusa Seed Extracts on Platelet Aggregation and Serum Lipid Level in Ovariectomized Rats)

  • Estrogen의 감소는 다양한 생리적 변화를 초래하며 특히 심혈관계 질환을 빠르게 진행시킨다고 보고되고 있다. 본 연구에서는 실험동물인 흰쥐의 난소를 절제하여 인위적으로 갱년기를 유도한 후 선인장류인 천년초 씨 추출물의 혈중 지질 함량 및 혈류 개선에 대한 영향을 검토하였다. 7주 동안 천년초 씨 추출물을 섭취한 OHS 2%, OHS 6%군은 체중이 다소 증가하여 체중감소에는 효과를 나타내지 않았지만 섭취 농도가 증가 할수록 식이효율은 낮아져 비만조절 효과의 가능성이 기대된다. 총 콜레스테롤 함량은 천년초 추출물 투여로 인한 유의성 있게 감소하는 결과가 나타나지 않았으나, 혈청 중 중성지방 함량의 경우 천년초 씨 추출물이 2%와 6% 함유된 사료를 섭취시킨 OVX-OHS 2%군 및 OVX-OHS 6%군의 경우, OVX-CON군과 비교하여 중성지방 함량이 유의적으로 감소하는 결과가 나타났다. 혈청 중 LDL-콜레스테롤 함량은 OVX-CON군의 경우 SHAM군에 비해 유의적으로 증가하였으나, 천년초 추출물 투여군인OVX-OHS 2%군과 OVX-OHS 6%군의 경우 OVX-CON군에 비해 유의적으로 감소하는 결과가 나타났다. 동맥경화지수(atherogenic inde, AI)는 난소 절제한 OVX-CON군에 비하여 난소 절제 후 천년초 씨 추출물 첨가식이를 섭취한 OVX-OHS 6%군에서 유의적으로 감소되었다. 또한 심혈관위험지수를 나타내는 CRF역시 천년초 씨 추출물 첨가식이 섭취로 인해 감소하는 결과를 나타내었으며, 특히 OVX-OHS 6%군에서 유의적으로 감소하는 경향을 나타냈었다. 또한 천년초 씨 추출물을 섭취로 인해 난소절제군과 비교하여 혈류 흐름을 개선시키고 혈소판 응집을 저해하는 경향을 나타내었다. 따라서 천년초 씨 추출물이 갱년기 여성의 심혈관계 질환에 유효한 효과를 나타낼 것으로 사료된다.

  • 세 개 유전자의 DNA 분자서열을 이용해서 전라도 지역에서 재배되고 있는 신품종 왕가시천년초(O. humifusa (Raf.) Raf. f. jeollaensis E. J. Kim & S. S. Whang)의 분류학적 지위와 한국산 손바닥 선인장들의 분류학적 유연관계가 조사되었다. 신품종 왕가시천년초는 형태학적으로나 생육 지역이 천년초와 비슷하지만, 노란색 꽃의 안쪽 중심에 붉은 색이 있고, 3 cm 길이의 강모가 많이 발달되어서 차이가 있다. 분자적인 연구결과는 왕가시천년초가 O. humifusa complex에서 Macrocentra 계열에 포함되어 Humifusa 계열에 포함되는 한국산 O. humifusa와 다르게 나타났다. 왕가시천년초는 천년초와 마찬가지로 주로 농장에서 재배되고 있어서 품종으로 지위를 주었다. 한국산 손바닥선인장 3종에 대한 분류학적 유연관계가 자세히 논의되었다.

  • The objective of this study was to determine the optimum mixing ratio of Opuntia humifusa powder and sugar in the preparation of muffin. The optimal sensory composite recipe was determined by following the central composites for muffin with different levels of Opuntia humifusa powder (A) and sugar (B). The analysis was performed using the response surface methodology, and the sensory evaluation was performed with the data. Ten experimental recipes, including two with reference points in the composition, were selected. The compositional and functional properties were measured, and the physical and sensory values were applied to the mathematical models. Perturbation plots showed the effects of each ingredient on the final product. Measurements showed significant values in lightness (p<0.05), sweetness (p<0.05), hardness (p<0.05), gumminess (p<0.05) and cohesiveness (p<0.01). The sensory measurements showed significant values in color (p<0.05), flavor (p<0.05), appearance (p<0.05), softness (p<0.05) and overall quality (p<0.05). Based on the numerical and graphical methods, the optimal formulation was determined to be 6.10 g of Opuntia humifusa powder and 82.61 g of sugar.

Inhibitory Effects of Opuntia humifusa on 7, 12-Dimethyl-benz[a]anthracene and 12-O-tetradecanoylphorbol-13-acetate Induced Two-stage Skin Carcinogenesis

  • Opuntia humifusa, member of the Cactaceae family, was previously demonstrated to have radical scavenging, anti-inflammatory and anti-proliferative effects in in vitro models. It was suggested that O. humifusa could function in the prevention of carcinogenesis. To investigate the in vivo chemopreventive effect of O. humifusa, mice were fed a diet containing either 1% or 3% following 7, 12-dimethylbenz[a] anthracene (DMBA) and 12-O-tetradecanoylphorbol-13-acetate (TPA) induction of skin carcinogenesis. Significant decrease in the numbers of papilloma and epidermal hyperplasia were observed in mice fed with O. humifusa, compared to the control group. O. humifusa also upregulated high total antioxidant capacity and level of phase II detoxifying enzyme such as superoxide dismutase and glutathione S-transferase activity in the skin. Lipid peroxidation activity level was measured in skin cytosol and significantly inhibited in 3% OH fed group compared to the control group. These results suggest that O. humifusa exerts chemopreventive effects on chemical carcinogenesis in mouse skin and that prevention effects are associated with reduction of oxidative stress via the modulation of cutaneous lipid peroxidation, enhancing of total antioxidant capacity especially in phase II detoxifying enzyme system and partial apoptotic influence.

An Analysis of Design Factors for Developing Opuntia Humifusa Spines Removal Device

  • Purpose: Opuntia Humifusa has been used in the food and beauty industry after removing spines and glochids clearly. This study compared the methods used in removing spines and analyzed the design factors for developing a spine removal device. Method: This study compared the spine removal ratios in accordance with the length of brush, water spray pressure, the number of water spray, and the size of Opuntia Humifusa in a rotating brush device and a water spray device. In addition, this study compared the reversal ratios according to the inclination angle of a conveyor, the drop height of Opuntia Humifusa, and the speed of the conveyor to analyze the reversal factors. Results: The spines were not removed clearly in the rotating brush method, and the glochids were nailed deeply. The spine removal ratio was 96.9% with the water spray pressure of 20 $kgf/cm^2$ and the conveyor speed of 10 cm/s in the water spray method. The number of water spray was correlated with the spine removal ratio, and the average spine removal ratio was 95.6% with three cycles of water spray. The reversal ratio was 97% with the inclination angle of the conveyor $20^{\circ}$, the drop height of 380 mm, and the conveyor speed of 10 cm/s. Conclusions: In order to develop a Opuntia humifusa spine removing device, this study compared the rotating brush and water spray methods. As a result, each spine removal performance of the rotating brush and water spray methods was 96.9% and 95.6%, respectively. Although the performance of the rotating brush method was slightly higher than that of the water spray method, the water spray method was suitable for removing spines from stem because the epidermis of stem was damaged and the glochids were nail deeply in the rotating brush method. Further studies on appropriate arrangement of spray nozzles, maintaining the optimal water spray pressure, the speed and angle control of the feeding conveyor, and devices for inducing the stem to the center will be needed in combining the water spray device and the reversal device.

석류와 천년초 분말을 첨가한 젤리의 품질특성 (Quality Characteristics of Jelly Containing Added Pomegranate Powder and Opuntia humifusa Powder)

  • 본 연구에서는 석류와 천년초의 기능성을 살린 젤리를 실용화시키고 효과적인 배합비를 알아보기 위해 석류와 천년초 분말의 첨가 비율을 달리해서 젤리를 제조한 후 pH, 산도, 당도, 색도, 투명도, 탁도, 물성, 관능검사를 실시하여 젤리의 품질특성을 알아보았다. 석류 젤리와 천년초 젤리 모두 첨가량이 증가할수록 pH가 유의적으로 낮아졌고(p<0.001), 산도와 당도는 유의적으로 증가하였다(p<0.001). 색도는 석류 젤리의 경우 석류 분말 첨가량이 증가할수록 명도(L)값과 황색도(b)값이 유의적으로 감소하였고, 적색도(a)값은 유의적으로 증가하는 경향을 보였다(p<0.001). 그리고 천년초 젤리는 천년초 분말의 첨가량이 증가할수록 명도(L)값은 감소했고, 적색도(a)값과 황색도(b)값은 유의적으로 증가하는 경향을 보였다(p<0.001). 투명도는 석류 젤리와 천년초 젤리 모두 분말 첨가량이 증가할수록 유의적으로 감소하는 경향을 보였으나, 탁도는 유의적으로 증가하였다(p<0.001). 물성은 석류 젤리의 경우 석류 분말 첨가량이 증가함에 따라 견고성이 대조군보다 약간 증가하는 경향을 보였고, 부착성과 탄력성은 낮아졌다. 응집성과 검성도 대조군보다 낮아졌으며, 씹힘성은 대조군보다 현저하게 낮아졌다. 또한 천년초 젤리는 천년초 분말 첨가량이 증가할수록 견고도는 대조군보다 약간 증가하였으나 유의적인 차이는 없었다. 부착성은 유의적으로 낮아지는 경향을 보였고(p<0.05), 응집성과 검성도 모두 낮아졌으나 유의적 차이는 없었다. 탄력성과 씹힘성은 천년초 분말의 첨가량 증가에 따라 유의적으로 낮아졌다. 관능검사는 석류 분말 첨가 젤리의 경우 색상은 P4(2%)가 가장 높은 값을 보였고(p<0.05), 외관은 P2(1%)가 가장 높았다. 단맛, 씹힘성, 탄력성, 견고성, 투명도, 전반적인 수용도는 대조군이 모두 높았으나 그 다음으로 석류 분말이 첨가되면서 P1(0.5%)가 가장 높았다. 또한 천년초 분말 첨가 젤리의 경우도 색상, 외관, 단맛, 씹힘성, 탄력성, 견고성, 투명도가 대조군이 모두 높았으나 그 다음으로 천년초 분말이 첨가되면서 O1(0.5%)가 높은 값을 나타냈으며, 전반적인 수용도도 O1(0.5%)가 가장 높았다. 따라서 석류와 천년초의 기능성을 살린 젤리를 실용화시키는 효과적인 배합비는 석류 분말과 천년초 분말 모두 0.5% 첨가가 적절한 것으로 나타났다. 그러므로 기능성 식품 소재로서 젤리에 석류와 천년초 분말을 첨가 하는 것은 활용할만한 가치가 있다고 사료된다.
