Synthesis, Characterization, and Catalytic Properties of Gp 6 Metal Complexes of 1-N,N-dimethylaminomethyl-$1^{\prime}$,2-bis(diphenylphosphino)ferrocene (FcNPP). X-ray Crystal Structure of W$(CO)_4({\eta}^2-FcNPP(O)-P,N)$

  • Reactions of 1',2-bis(diphenylphosphino)-l-(N,N-dimethylaminomethyl)ferrocene (FcNPP) with $M(CO)_6$ (M=Cr, Mo, W) in the presence of TMNO (Trimethylamine oxide) in a stoichiometric ratio of 1 : 1.5 : 3.5 produced a series of Gp 6 metal carbonyl derivatives with a variety of coordination modes: M(CO)$_4({\eta}^2$-FcNPP-P,P) (M=Cr, Mo, W), $M(CO)_5({\eta}^1-FcNPP-P) (M=Mo, W)\;,\; M_2(CO)_9({\eta}^1\;,\;{\eta}^2-FcNPP-P,P,N) (M=Cr, Mo)\;,\;M_2(CO)_{10}({\eta}^1\;,\;{\eta}^1-FcNPP-P,P) (M=Cr, Mo, W)\;, and\;W(CO)_4({\eta}^2-FcNPP(O)-P,N)$. All these complexes were characterized by microanalytical and spectroscopic techniques. In one case, the structure of W(CO)$_4({\eta}^2$-FcNPP(O)-P,N) was determined by X-ray crystallography. Crystals are monocinic, space group P$2_{1/C}$, with a=10.147(2), b=19.902(3), c=19.821(4) ${\AA},\;{\beta}=96.88(2)^{\circ},\;V=3974(l){\AA}^3$, Z=4, and $D_{calc}=1.64 g cm^{-3}$. The geometry around the central tungsten metal is a distorted octahedron, with the nitrogen and phosphorus atoms being cis to each other. Some of these complexes exhibited catalytic activities in the allylic oxidation and epoxidation of cholesterly acetate. Other oxidation products were also formed with the different chemical yields and product distribution depending upon the ligand and the central metal.

Effect of Fermented Chlorella Supplementation on Growth Performance, Nutrient Digestibility, Blood Characteristics, Fecal Microbial and Fecal Noxious Gas Content in Growing Pigs

  • A total of 96 growing pigs ((Landrace${\times}$Yorkshire)${\times}$Duroc; BW = $26.58{\pm}1.41$ kg) were used in a 6-wk feeding trail to evaluate the effects of fermented chlorella (FC) supplementation on growth performance, nutrient digestibility, blood characteristics, fecal microbial and fecal noxious gas content in growing pigs. Pigs were randomly allotted into 1 of 4 dietary treatments with 6 replicate pens (2 barrows and 2 gilts) per treatment. Dietary treatments were: i) negative control (NC), basal diet (without antibiotics); ii) positive control (PC), NC+0.05% tylosin; iii) (fermented chlorella 01) FC01, NC+0.1% FC, and iv) fermented chlorella 02 (FC02), NC+0.2% FC. In this study, feeding pigs PC or FC01 diets led to a higher average daily gain (ADG) and dry matter (DM) digestibility than those fed NC diet (p<0.05), whereas the inclusion of FC02 diet did not affect the ADG and DM compared with the NC group. No difference (p>0.05) was observed on the body weight, average daily feed intake (ADFI), gain:feed (G:F) ratio, the apparent total tract digestibility of N and energy throughout the experiment. The inclusion of PC or FC did not affect the blood characteristics (p>0.05). Moreover, dietary FC treatment led to a higher (p<0.05) lactobacillus concentration and lower E. coli concentration than the NC treatment, whereas the antibiotic supplementation only decreased the E. coli concentration. Pigs fed FC or PC diet had reduced (p<0.05) fecal $NH_3$ and $H_2S$ content compared with those fed NC diet. In conclusion, our results indicated that the inclusion of FC01 treatment could improve the growth performance, nutrient digestibility, fecal microbial shedding (lower E. coli and higher lactobacillus), and decrease the fecal noxious gas emission in growing pigs when compared with the group fed the basal diet. In conclusion, dietary FC could be considered as a good source of supplementation in growing pigs because of its growth promoting effect.

  • Camellia sinensis, Green tea, contains phenolic compounds that act to scavenge reactive oxygen species (ROS), such as catechin, epicatechin, etc. In contrast with the tea leaf, the bioactivity of its fruit and the fruit peels remains still unclear. This study focused on the effects of fruit and fruit peels of C. sinensis (FC and PC) against oxidative DNA damage in NIH/3T3 cells. The scavenging effects of FC and PC on ROS were assessed using 1,1-diphenyl-2-picryl hydrazyl or 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid radicals. The measurement of ROS in cellular levels was conducted by DCFDA reagent and the protein expression of γ-H2AX, H2AX, cleaved caspase-3, p53, and, p-p53 was analyzed by immunoblotting. The gene expressions of p53 and H2AX were assessed using polymerase chain reaction techniques. The major metabolites of FC and PC were quantitatively measured analyzed and the amounts of phenolic compounds and flavonoids in PC were greater than those in FC. Further, PC suppressed ROS production, which protects the oxidative stress-induced DNA damage through reducing H2AX, p53, and caspase-3 phosphorylation. These results refer that the protective effects of FC and PC are mediated by inhibition of p53 signaling pathways, probably via the bioactivity of phenolic compounds. Thus, FC and PC can serve as a potential antioxidant in DNA damage-associated diseases.

  • Foaming capacity (FC) and stability (FS) of protein recovered from red crab (Chitinonecetes opilio) processing in water and soybean protein isolate were determined at pH 2.0~10.0 in water and NaCl solution. The FC values for both proteins showed the lowest values at the isoelectric point (pH 4.0) and increased nth an increase in pH above the isoelectric point. FC of RCP was higher than that of SPI at pH 10.0 in water and both NaCl solutions. FC of SPI increased with an increase in NaCl concentration at pH 4.0 and 6.0, but FC of RCP was not affected. The highest FS values for both proteins were obtained at pH 4.0 in water. At pH 2.0, FC of RCP decreased with NaCl concentration increase, but FS increased. NaCl concentration had little effect on FS of RCP at pH 4.0 and 6.0, but the FS decreased at pH 10.0. FS of SPI was similar to that of RCP at pH 2.0 and increased with NaCl concentration Increase from 0.1 to 0.5M NaCl at pH 10.0.

Quality characteristics of in vitro luwak coffee produced using enzyme and microbial complexes (효소 및 미생물 복합체를 사용한 인비트로 루왁 커피의 품질 특성)

  • In vitro luwak coffee was produced using enzyme­microbial complexes. The coffee quality of non-fermented coffee beans (NFC) and fermented coffee beans (FC) was compared. The total free amino acid content was higher in FC than in NFC. The levels of glutamic acid and γ-amino-n-butyric acid in NFC were higher than those in FC; however, the contents of essential amino acids, such as lysine, leucine, and valine, in FC were higher than in NFC. During fermentation, the sucrose content decreased, whereas the fructose and glucose contents increased (p<0.001). The chromaticity of the coffee extract showed higher lightness (L), redness (a), and yellowness (b) values in FC than those in NFC. The caffeine content was significantly lower in FC (696.94±0.04 ㎍/mL) compared to that in NFC (1,130.22±1.55 ㎍/mL) (p<0.001). Conversely, the polyphenol and chlorogenic acid contents were significantly higher in NFC than in FC (p<0.001). Electronic nose analysis indicated considerable differences between the volatile aromatic components in NFC and FC. Sensory scores were significantly higher for FC than those for NFC. Therefore, the fermentation of coffee beans using enzyme­microbial complexes altered the chemical components, which promoted the Maillard reaction during the coffee bean roasting process. These results suggest the possibility of producing in vitro luwak coffee with better flavor and lower caffeine content.

Effects of Supplemental Phosphorus and Iron in the Fish Meal Diets on Growth in Juvenile Rockfish (Sebastes schlegeli) (단백질원이 어분인 조피볼락 치어 사료에 인(P)과 철(Fe)의 보충 효과)

  • In order to investigate the effects of phosphorus and iron levels in the diets containing fish meal as only protein source on the growth, feed conversion, body composition and blood characteristics of juvenile Korean rockfish, either graded monobasic potassium phosphate (MPP) of 1% (MPP-1), 2% (MPP-2), 3% (MPP-3) and 4% (MPP-4), or Fe-citrate (FC) of 0.022% (FC-1), 0.044% (FC-2), 0.066% (FC-3) and 0.088% (FC-4) was added to fish meal-based diet (MMP-0 or FC-0), respectively. Duplicate groups of 40 fish weighing 7g were fed the diets for 21 weeks. Weight gain, feed efficiency and nutrient retention of fish fed diets with different MPP levels were not significantly different (P>0.05). Lipid content of whole body decreased with an increase in dietary MMP (P<0.05). No significant differences were found in the Ht, Hb, total protein, or total glucose in serum among diets containing different MMP levels (P>0.05). Weight gain of fish fed the FC-0, FC-1 adn FC-3 diets was significantly higher than that of FC-4 diet (P>0.05), whereas feed efficiency, nutrient retention, hematological values (Ht, Hb, total protein, and total glucose), and whole body composition were not affected by dietary FC levels (P>0.05). The present results revealed that phosphate or iron supplementation to the diet containing 65% fish meal for juvenile Koran rockfish was not necessary.

  • This experiment was performed to compare the techniques of the metabolizable energy determination and to, examine the problems associated with metabolizable energy determination of poultry feedingstuffs. The feedingstuffs evaluated herein, were corn, sorghum, barley. fish meal and soybean meal The results obtained were as followings : 1. When comparing AME and AMEn values of five feedingstuffs(measured) by Total Collection ana Index Methods there was no significant difference (P>.05) between two methods except fish meal. 2. The AME$_{F}$ values of feedingstuffs were not different (P>.05) from AME values. which implied that bioassay of AME$_{F}$ be applicable to rapid measure AME value of feedingstuffs for poultry and the problems connecting to AME$_{F}$ determination were discussed. 3. The AME$_{FC}$ and TME values of corn, sorghum and barley were significantly higher (P<.05) than AME and AME$_{F}$ values but those of fish meal and soybean meal were similar or lower than AME and AME$_{F}$ values. This fact was indicative that the techniques measuring AME$_{FC}$ and TME has some depressing factors when fish meal and soybean meal were determined. 4. Comparing of TME, AME$_{FC}$ and T-AME values tile TME values of corn sorghum and barley were significantly higher(P<.05) than AME$_{FC}$ and T- AME values. But that of fish meal and soybean meal was much lower(P< .05)comparing with AME$_{FC}$ and T-AME values. According]y, it night be concluded that the relationship among the values of TME. AME$_{FC}$ and T-AME was shown great variation according to the feedingstuffs, especially in the protein diets.

  • 연구배경 IgG에 대한 $Fc{\gamma}$ 수용기는 치주병인균에 대한 숙주 반응에 있어서 중요한 역할을 하는데, 이 중 $Fc{\gamma}RIIIa$는 NK 세포, 대식세포, 단핵구, ${\gamma}{\delta}T$세포에서 발현되며, EC2 도메인에서 158 아미노산 부위의 valine (V)-phenylalanine (F)의 유전자다형성을 보인다. $Fc{\gamma}RIIIb$는 특이적으로 중성구에 발현되는데, extracellular (ECl) Ig-like 도메인 내 4개의 아미노산 치환(substitutions)에 의한 NA1-NA2 유전자다형성을 보인다. 이 연구의 목적은 한국인에서 급진성 치주염 환자와 $Fc{\gamma}III$ 수용기의 유전자다형성과의 관련성을 알아보는 것이다. 연구방법 및 재료 치주적으로 건강한 90명 (대조군, 남자 64명, 여자 26명)과 서울대학교 치과병원 지주과에 내원하여 급진성 치주염으로 진단된 환자 43명 (aggressive periodontitis patients: AgP, 남자 30명, 여자 13명)을 대상으로 하였다. 모든 실험 대상자는 임상 실험에 대해 동의 하였고, 초진 시 전자 탐침(Florida Probe(R) Co. Gainesville, FL)을 이용하여 탐침 시 치주낭 깊이 (PPD), 임상부착수준 (CAL), 치태지수(PI), 탐침 후 출혈지수 (BOP)를 측정하였다. 또한 이들의 정맥혈에서 추출한 DNA를 PCR법, 전기영동법 등을 이용하여 $Fc{\gamma}RIIIa$, $Fc{\gamma}RIIIb$의 대립 유전자의 존재여부를 확인하였다. 이를 바탕으로 $Fc{\gamma}RIII$ 복합 유전형을 확인하여 각 군 간을 비교하였다. 연구 결과 1. $Fc{\gamma}RIIIa$에 대한 유전자다형성 연구 결과 대조군과 급진성 치주염 환자 군(AgP)사이에서는 대립 유전자 분포가 서로 유의성 있는 차이를 나타내었고 (P<0.05), $Fc{\gamma}RIIIb$에서는 유의성 있는 차이를 발견할 수 없었다. (P>0.05) 2. $Fc{\gamma}RIIIa$ 158F 대립형질이 급진성 치주염 환자에서 유의성 있게 많이 발견되어졌다. (P<0.05) 결론 이 연구를 통하여 $Fc{\gamma}RIIIa$ 유전자의 분석이 한국인의 급진성 치주염에 대한 감수성의 위험요소의 표지자로 활용할 수 있을 것으로 사료되며, 향 후 더 많은 환자를 대상으로 히는 추가 연구가 필요하다고 생각된다.

Fecal Calprotectin Assay at an Early Stage of Treatment Can Be Used as a Surrogate Marker to Predict Clinical Remission and Mucosal Healing in Pediatric Crohn's Disease

  • Purpose: This study evaluated the predictive role of fecal calprotectin (FC) measured at an early stage of treatment for monitoring clinical remission (CR) after six months and endoscopic remission (ER) after one year of treatment in pediatric Crohn's disease (CD). Methods: This retrospective study included 45 patients who simultaneously underwent ileocolonoscopy and FC testing during follow-up. FC levels were measured before and after six weeks of treatment. CR was assessed after six months of treatment using Pediatric Crohn' s Disease Activity Index and acute-phase reactants. ER was assessed after one year using the Simple Endoscopic Score for Crohn's Disease. Results: Twenty-nine (64.4%) patients used oral prednisolone for remission induction and 16 (35.6%) patients used anti-tumor necrosis factor-alpha. Thirty (66.7%) patients achieved CR, while 24 (53.3%) achieved ER. The FC level measured after six weeks of treatment could predict CR (χ2=9.15, p=0.0025) and ER (χ2=12.31, p=0.0004). The δFC could predict CR (χ2=7.91, p=0.0049), but not ER (χ2=1.85, p=0.1738). With a threshold of ≤950.4 ㎍/g, FC at week six could predict CR with 76.7% sensitivity and 73.3% specificity. The area under the curve (AUC) was 0.769 (standard error 0.0773, p=0.0005). The same threshold predicted ER with 87.5% sensitivity and 71.4% specificity. The AUC was 0.774 (standard error 0.074, p=0.0002). Conclusion: FC assay at an early stage of treatment can be used as a surrogate marker to predict CR and mucosal healing in pediatric CD.

Fecal Calprotectin and Phenotype Severity in Patients with Cystic Fibrosis: A Systematic Review and Meta-Analysis

  • Inflammation plays an important role in the outcome of patients with cystic fibrosis (CF). It may develop due to cystic fibrosis transmembrane conductance regulator protein dysfunction, pancreatic insufficiency, or prolonged pulmonary infection. Fecal calprotectin (FC) has been used as a noninvasive method to detect inflammation. Therefore, the aim of the current meta-analysis was to investigate the relationship between FC and phenotype severity in patients with CF. In this study, searches were conducted in PubMed, Science Direct, Scopus, and Embase databases up to August 2021 using terms such as "cystic fibrosis," "intestine," "calprotectin," and "inflammation." Only articles published in English and human studies were selected. The primary outcome was the level of FC in patients with CF. The secondary outcome was the relationship between FC and clinical severity. Statistical analysis was performed using Comprehensive Meta-Analysis software. Of the initial 303 references, only six articles met the inclusion criteria. The mean (95% confidence interval [CI]) level of FC was 256.5 mg/dL (114.1-398.9). FC levels were significantly associated with pancreatic insufficiency (mean, 243.02; 95% CI, 74.3 to 411.6; p=0.005; I2=0), pulmonary function (r=-0.39; 95% CI, -0.58 to -0.15; p=0.002; I2=60%), body mass index (r=-0.514; 95% CI, 0.26 to 0.69; p<0.001; I2=0%), and Pseudomonas colonization (mean, 174.77; 95% CI, 12.5 to 337.02; p=0.035; I2=71%). While FC is a reliable noninvasive marker for detecting gastrointestinal inflammation, it is also correlated with the severity of the disease in patients with CF.
