Biochemical Properties and Antimicrobial Drug Susceptibility of Pasteurella haemolytica Isolated from Pneumonic Calves (호흡기질병 감염 송아지에서 분리한 Pasteurella haemolytica의 생화학적 특성 및 약제 감수성)

  • The present study was conducted to investigate biochemical properties and antimicrobial drug susceptibilities of 36 strains of Pasteurella haemolytica(P haemolytica) isolated from pneumonic calves in Kyongbuk province during the period from January 1990 to December 1990. P haemolytica was isolated from 36 of 111(32.4%) pneumonic calves of 1 to 6 months of age. The majority of biochemical and cultural properties of P haemolytica isolated from calves were identical to those of the reference strains employed. All isolated were susceptible to baytril, gentamicin, and kanamycin, some of them were resistant to tetracycline, amikacin, streptomycin, and ampicillin.

  • The biovars and serovars of 57 strains of Pasteurella haemolytica(P haemolytica) isolated from pneumonic calves and goats were investigated. The biovars were determined some biochemical and cultural properties and susceptibility to penicillin. All 57 isolates were considered to correspond to biovar A. Among 57 P haemolytica, 39 isolates of them(68.4%) were serovar 1.2(3.5%) were serovar 5 and 2(3.5%) were serovar 7, the remaining 14 isolates(24.6%) were untypable.

  • The present study was conducted to investigate biochemical properties and antimicrobial drug susceptibilities of 57 strains of Pasteurella haemolytica (P haemolytica) isolated from pneumonic calves and goats in Youngnam province during the period from September 1989 to February 1991. P haemolytica was isolated from 95 of 142(31.7%) pneumonic calves of 1 to 6 months of age and from 9 of 24(37.5%) pneumonic goats of 2 to 10 months of age. Seasonal isolation frequency of P haemolytica in calves and goats varied from 16.6 to 35.7%, and it was higher in winter. The majority of biochemical and cultural properties of P haemolytica isolated from calves and goats were identical to those of the reference strains employed. All isolates were susceptible to ceftiofur, erythromycin, kanamycin, gentamicin, and chloramphenicol (MIC: <$25{\mu}g/ml$), some of them were resistant to amikacin, sulfadimethoxin, and streptomycin (MIC: >200IU or ${\mu}g/ml$).

  • Pasteurella haemolytica is well known to cause severe pneumonia, consolidation and oedema of the lung, and fibrinous pleurisy under the stress and infection of virus in the cattle. In the course of our screening for antimicrobial agents against P.haemloytica, compound 51086 has been isolated from the fermentation broth of Streptimyces sp. 51086. The compound 51086 was purified by SiO2, Sephadex LH-20 and ODS column chromatographies and HPLC, subsequently. The structure of compound 51086 was determined as hygromycin A by combination of 1H NMR, 13C NMR, HMBC, and ESI-MS. This compound showed significant antibacterial activity against P.haemolytica and P.multocida.

  • The present study was conducted to investigate isolation and antimicrobial drug susceptibility of Pasteurella spp from pneumonic calves and cows (pneumonic lungs) in Gyeongnam south area from January to November 2000. Pasteurella multocida and P haemolytica were isolated 136 (53.8%) and 22(8.7%), respectively, from 158 of 253 pneumonic calves of 1 to 12 months of age and from pneumonic lungs and simultaneous isolation rate of P mutocida and P haemolytica was 4 (1.6%). Seasonal isolation rate of Pasteurella spp in calves and pneumonic lungs varied from 3 to 65.2%, and it was higher in Fall. The majority of biochemical and cultural properties of P multocida and P haemolytica isolated from calves and pneumonic lung were identical to those of the reference strains employed. Pneumonic lesions of the examined lungs were histologically identified as purulent bronchopneumonia and fibrinous pneumonia. All isolates were resistant to penicillin, amikacin, lincomicin, colistin, cloxacilling, sulfonamide, kanamycin, sulfamethoxasoletrimetoprim, and gentamicin(>80%), but some of them were susceptible to amoxicillin, norfloxacin, cephalexin, cefazolin, orbifloxacin, enrofloxacin, and ciprofloxacin(>60%).

  • A 3-year-old female jaguar (Panthera onca onca) died after having 1 day history of respiratory failure. At necropsy, the pericardial sac contained large amounts of cloudy fluid enriched with fibrin. Numerous yellowish nodules, which are variable in size and often confluent, are randomly scattered throughout the myocardium. Pasteurella haemolytica was isolated from the pericardial sac and myocardium. In the lung, severe pulmonary congestion, edema and vasculities with intralesional presence of heartworm were found. Therefore the cause of death in this jaguar is believed to be due to respiratory failure following concurrent heartworm infection and bacterial pericarditis and myocarditis.

  • The Pasteurella haemolytica-induced pneumonic lesions are frequently occurred with stress and infection of virus in the cattle. In the course of screening for antimicrobial activity against Pasteurella haemolytica, compound 51086 has been isolated from the fermentation broth of the strain streptomyces sp. The compound 51086 was purified by column chromatography and HPLC, subsequently. The structure of compound 51086 was determined as a hygromycin A by a combination of $^1$H NMR, $\^$l3/C NMR, HMBC, and ESI -MS. This compound showed significant antibacterial activity against P. haemolytica and P. multocidn in Gram-negative bacteria.

  • The eleven strains of Pasteurella haemolytica var. ureae were isolated from discharges of respiratory tract and the eyes. Three strains were isolated in pure cultures from the diseased conditions(chronic dacryocystitis), but further clarifications for their pathogenicities to man were urged.

  • Bovine Pneumonic Pasteurellosis는 수송열(輸送熱)로 일반적으로 알려져 있는 질병으로서, 여러가지 요인의 복합적(複合的)인 작용에 의해 발병하는 것으로 알려져 있으나, Pasteurella haemolytica A1이 가장 주요(主要)한 인자(因子)로 밝혀져 있다. P haemolytica A1은 leukotoxin(LKT), lipopolysaccharide(LPS), capsular polysaccharide 등 여러가지의 병원성인자(病原性因子)을 생성한다. 이들 인자중 LKT가 가장 중요한 병원성인자로 밝혀져 있다. 이에 본 실험은 P haemolytical A1의 LKT 유전자를 Bacillus subtilis에서 발현(發現)시킴으로서 LPS에 오염(汚染)되지 않은 LKT을 대량으로 생산할 목적으로 실시되었다. 실험의 첫 단계(段階)로서 pLKT52 plasmid을 Sau3 A1의 제한효소을 이용하여 부분소화(部分消化)시킨 후 이 부분 소화(消化)된 유전자들로부터 3~5kb 크기의 유전자들을 순수분리하여 pUC18와 결합시킨 후 E coli NM522에 형질전환(形質轉換)시켰다. 이때 형질전환된 균주들은 LKT에 대한 단크론 항체인 MAb601을 이용하여 colony blot 법에 의해서 LKT 유전자 보유 및 발현여부(發現與否)을 조사하였다. 이들 양성 clone들은 제한효소분석(制限酵素分析), 염기서열분석(鹽基序列分析) 및 Western blot 등에 의해서 재확인(再確認)하였다. 총 9개의 양성 clone중 위의 방법에 의해서 한 clone을 선택(選擇)하여 lktCA insert를 재분리하여 shuttle vector에 subcloning 하였다. Subcloning된 LKT 유전자들은 shuttle vector의 종류(種類)(pHPS9, p602/20, pHPS9-Sac)와 각기(各其) 다른 종류(種類)의 B subtilis(spoO12A, BR121, WB3O, Raj1105) 숙주내(宿主內)에서 발현정도를 Western blot 법에 의해서 비교(比較)하였다. 이때 최적발현조건(最適發現條件)은 p602/20와 pBL1의 dual plasmid system을 이용하여 B subtilis spoO12A에서 2시간동안 IPTG로 발현을 유도(誘導)하는 것이었다. B subtilis에서 발현된 LKT을 visual 법과 neutral red uptake 법을 이용하여 소 폐포(肺胞) 대식구(大食求)에 대한 biological activity를 확인하였다. 발현된 LKT에 대한 LPS 오염은 LKT을 SDS-PAGE 후 silver stain에 의해서 확인하였다. 본 실험을 통해서 볼 때에 lktCA 유전자를 보유(保有)하고 있는 p602/20는 B subtilis에서 매우 불안정(不安定)하였고, 발현된 LKT는 세균자체(細菌自體)에서 생성되는 protease들에 의해서 파괴(破壞)됨으로서 농도(濃度)가 매우 낮았다. 이러한 문제점들은 다음 단계(段階)의 실험에서 해결되어야할 문제들이다.

Prevalence of bacterial respiratory diseases in cattle (소 세균성 호흡기 질병 분포도 조사)

  • The objective of this study was to determine the prevalence of pneumonic bacteria in lungs and nasal swabs of cattle with respiratory diseases. From 95 pneumonic lungs of slaughtered cattle, 41 (43.2%) positive lungs were yielded with 54 pneumonic bacteria, which corresponded to P. multocida (n = 34), A. pyogenes (n = 14) and P. haemolytica (n = 6). One-hundred sixty seven pneumonic bacteria were isolated from 195 nasal swabs in calves, 64.7% (108 isolates) belonged to P. multocida, 16.2% to A. pyogenes, 13.8% to P. haemolytica and 5.4% to H. somnus. Fifty percents (n = 6) of isolates from pneumonic lungs of calves were identified as P. multocida. All isolates of P. multocida belonged to type A according to hyaluronidase test. Antimicrobial susceptibility tests showed that most isolates of P. multocida and P. haemolytica were sensitive to amoxicillin/clavulanic acid, cephalothin, ciprofloxacin, enrofloxacin, fluorophenicol and norfloxacin. The emergence of antimicrobial resistant Pasteurella spp. observed in this study, however, might limit such application. According to histopathological examination, pneumonia by mycoplasma or/and bacteria accounted for 92.8% among 69 pneumonic lungs of slaughtered cattle.
