Inhibitory Effect of Persicaria perfoliata (L.) H. Gross on IgE Mediated Allergic Responses in RBL-2H3 Cells

  • Purpose : This study aimed to investigate the anti-allergic effect of Persicaria perfoliata water extract (PPWE) on IgE stimulated rat basophilic leukemia (RBL-2H3) cell line. Methods : P. perfoliata (L.) H. Gross has been used in traditional medicine as an anti-allergic agent, antipyretic, and diuretic and for respiratory disorders. To analyze the anti-allergic activity of PPWE, release of β-hexosaminidase in RBL-2H3 cells was estimated by enzyme linked immunosorbant assay (ELISA). Also, the cytotoxic effect of PPWE was identified by WST assay, and nuclear factor (NF)-κB and its upstream signaling molecules were assessed by western blot analysis. Results : PPWE treatment significantly attenuated β-hexosaminidase release in a dose dependent manner without any cytotoxicity. PPWE inhibited β-hexosaminidase activity by 38.4±1.2, 36.6±0.6, 32.5±2.2 and 26.5±1.2 at 500, 250, 100, and 50 ㎍/㎖ of PPWE, respectively, compared with the control group. In addition, an analysis of the expression level of NF-κB, an inflammation transcription factor, in RBL-2H3 cells upon IgE stimulation provided reults consistent with the results of β-hexosaminidase release. The phosphorylated status of upstream signaling molecules for transcription factor, mitogen activated protein kinases (MAPKs), was also analyzed. The results showed that PPWE treatment dose-dependently inhibited phosphorylation of extracellular regulatory kinase (ERK) and c-Jun N-terminal kinase (JNK). These results show that PPWE had a strong IgE-mediated degranulation inhibitory effect on RBL-2H3 cells. Conclusion : P. perfoliata ameliorated IgE-mediated allergic reaction via the modulation of MAPK and NF-κB signaling pathway in RBL-2H3 cells. These results indicate that P. perfoliata could be a potential candidate for a treatment strategy against various allergic disorders.

Study on the Mechanism of Vascular Relaxation of Ethanol Extract of Persicaria Perfoliata H. Gross (하백초 에탄올 추출물의 혈관이완 기전에 대한 연구)

  • The ethanol extract of Persicaria perfoliata (EPP) induced relaxation of the phenylephrine-precontracted aorta in a dose-dependent manner, which was abolished by removal of functional endothelium. Pretreatment of the aortic tissues with NG-nitro-L-arginine methyl ester (L-NAME) or 1H-[1,2,4}-oxadiazole-[4,3-${\alpha}$)-quinixalin-1-one (ODQ) inhibited the relaxation induced by EPP. However, EPP-induced relaxation was not blocked by pretreatment with indomethacine, glibenclamide, tetraethylammonium (TEA), atropine, or propranolol. Incubation of endothelium-intact thoracic aortic ring with EPP increased the production of cGMP, which was also blocked by pretreatment with L-NAME or ODQ. These results suggest that EPP dilates vascular smooth muscle via endothelium-dependent NO/cGMP signaling.

Vegetation Structure of Hyeonchang Wetland and its Watershed in Nakdong-gang (낙동강 현창늪과 주변 분수계의 식생 구조)

  • Vegetation structure was investigated in the Hyeonchang wetland and its watershed around the Nakdong-gang, Changryeong-county, Gyeongsangnamdo, Korea. from May to August, 2001. The vegetation type of the Hyeonchang wetland was classified into 12 communities based on the actual vegetation map: Phragmites communis community, Zizania latifolia community, Phragmites communis - Persicaria perfoliata community, Salix koreensis community, Persicaria perfoliata - Phragmites communis community, Spirodela polyrhiza community, Persicaria perfoliata community, Cyperus amuricus community, Cyperus amuricus-EchinochJoa crus-galli var. frumentacea community, Phragmites communis-Zizania latifolia community, EchinochJoa crus-galli var. frumentacea community, and Persicaria maackiana community. Among them, Phragmites communis community was the largest (4.3 ha, 24%). The dominant vegetation type were Phragmites communis community, Echinochloa crus-galli var. frumentacea-Persicaria maackiana community, and Cyperus amuricus subcommunity based on the phytosociological method. The vegetation type of the Hyeonchang wetland watershed was classified into five communities based on the actual vegetation map: Pinus densilflora community, Pinus rigida community, Pinus densiflora-Quercus acutissima community, Pinus densiflora-Larix gmelini var. principis-ruprechtii community, and Populus tomentiglandulosa community. Among them, Distribution area of Pinus densiflora community was largest (399.3 ha, 61.8%). And the degree of green naturality of the Pinus densiflora community was 7 and 8 degree.

Effect of Gibberellin on the Adventitious Root Formation from the Leaves-derived Calli in Persicaria perfoliata (며느리배꼽 잎 유래 캘러스의 부정근 형성에 미치는 지베렐린의 작용)

  • This study was carried out to investigate the action of phytohormones which influence the adventitious root formation of calli originating from the leaves of Persicaria perfoliata. The optimal medium condition for callus formation was ½-strength MS, 1% sucrose, and 4.5 μM 2,4-D. In order to determine which phytohormones had an effect on the adventitious root formation, the calluses were cultured in various media with different kinds of phytohormones. As a result, the medium with GA3 or IAA was shown to induce root formation. To deeply investigate the effects of GA3 and IAA, calli were cultured in 0.1, 1, and 10 mg/l levels of phytohormones. Numbers of roots formed per callus were 10.9, 14.2, 22.6 in GA3, 5.8, 3.9, 1.1 in IAA, respectively. Therefore, the higher GA3 or the lower IAA concentration, the more roots formed. To confirm this role of GA3 we tested with inhibitors PBZ and NPA. GA3 with PBZ resulted in reduction by 52.4~69.4% compared to GA3 alone. In contrast, GA3 with NPA resulted in an increase by -8~45.6% compared to GA3 alone in root formation. Also, results were determined on the effect of GA3 with other phytohormones on root formation. Kinetin, 2iP and ABA with GA3 had a negative effect, but IAA with GA3 showed a similar result to GA3 alone. From these results we infer GA plays a key role and auxin has subsidiary activity on adventitious root formation. This is the first report that indicates GA3 promotes adventitious root formation from calli in P. perfoliata.

Antibacterial Activity and Component Analysis of Persicaria perfoliata Extracts (며느리배꼽 추출물의 항균 작용과 성분 분석)

  • In this study, the antibacterial activity and component analysis of Persicaria perfoliata extracts were investigated. MIC (Minimum Inhibitory Concentration) values of ethyl acetate fraction from P. perfoliata on Escherichia coli, Propionibacterium acnes, and Staphylococcus aureus were 0.25%, equally. The results showed that the antibacterial activity of the ethyl acetate fraction was the highest in the S. aureus and P. acnes. Ethyl acetate fraction from P. perfoliata extracts showed 5 bands in TLC and 8 peaks in HPLC (320nm). In HPLC chromatogram of ethyl acetate fraction, first main peak 2 (24.99%) and peak 3 (22.65%) were identified as quercetin-3-o-glucuronide and isoquercitrin in the order of elution time. Also, in HPLC chromatogram of deglycosylated fraction, main peak 2 (76.93%) was identified as quercetin in the order of elution time. And these components are very effective as antioxidants. Thus, these results indicate that extract/fractions of P. perfoliata can be applicable to new functional cosmetics for antioxidant, antibacterial activity.

Moisturizing and Dryness Reduction Effect of Face Cream Containing Persicaria Perfoliata (L.) Extract (며느리배꼽추출물을 함유하는 페이스 크림의 보습 및 건조함 감소 효과)

  • Purpose : This study was aimed to analyze the effects of cosmetics containing Persicaria perfoliata water extract (PPWE) on the skin moisturizing and improvement of skin condition in clinical trials. Methods : Clinical trial was conducted for five weeks after IRB approval at Dong-Eui University. Out of a total of 64 people, 15 people each were assigned to four groups as follows; control group A, B, C and the experimental group A that using cosmetic containing PPWE. Skin condition was measured two times, before and after clinical trial, by a professional skin analyzer, SDM (skin diagnosis system). Moisture and oil value of participants was analyzed twice, each morning and evening, using a portable device on their cheeks. In addition, the survey was investigated subjective satisfaction on change in skin condition and the satisfaction on the use of cosmetics. Result : The experimental group exhibited subjectively significant changes before and after clinical trials on skin its dryness (p=.039), blush (p=.017), and redness (p<.001). In addition, subjective evaluation was also the highest satisfaction in aspects of number of application (p=.003), amount of application (p=.002), moisture maintenance, and skin scratching frequency. The satisfaction on the use of cosmetics was the highest in the intention to repurchase (p=.045), recommendation willingness to others (p=.020), and intention to use various products (p=.001). Skin moisture of the clinical trial participants using the SDM, moisture level and elasticity of the experimental group increased by 12.94 and 10.28. Moisture level, which was measured by a portable device, was the most potently increased in the experimental group. Conclusion : Consequently, PPWE containg cosmetics exhibited the effects of moisturization and attenuated skin dryness in clinical trials, which might be utilized as a fundamental data to develop numerous lines of cosmetics.

Antioxidant Activity of Persicaria perfoliata Extracts (며느리배꼽 추출물의 항산화 활성)

  • In this study, the antioxidative effects, inhibitory effects on tyrosinase, elastase of Persicaria perfoliata extracts were investigated. The deglycosylated fraction of extract ($12.38{\mu}g$/mL) showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of P. perfoliata extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction of extract ($0.35{\mu}g$/mL) showed the most prominent ROS scavenging activity. The protective effects of P. perfoliata extract/fractions on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The P. perfoliata extracts suppressed photohemolysis in a concentration dependent manner ($1{\sim}50{\mu}g$/mL) except the deglycosylated fraction of extract. The inhibitory effect of P. perfoliata extracts on tyrosinase was investigated to assess their whitening efficacy. Inhibitory effects ($IC_{50}$) on tyrosinase were determined with ethyl acetate fraction of P. perfoliata extract ($136.00{\mu}g$/mL) and deglycosylated fraction of extract ($68.10{\mu}g$/mL). Finally, their anti-elastase activities were measured to predict the anti-wrinkle efficacy in the human skin. Inhibitory effects ($IC_{50}$) on elastase were determined with ethyl acetate fraction of P. perfoliata extract ($67.20{\mu}g$/mL) and deglycosylated fraction of extract ($43.50{\mu}g$/mL). These results indicate that extract/fractions of P. perfoliata can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Extract/fractions of P. perfoliata can be applicable to new functional cosmetics for antioxidant, antiaging.

Floristic Composition and Community Dynamics Along the Nakdong River (낙동강변(洛東江邊)의 잡초식생(雜草植生)에 관(關)한 연구(硏究))

  • Weed species investigated along the Nakdong River on September, 1989 were 105 species in 29 families. Vegetation analysis along the Nakdong River using a two dimensional ordination diagram showed the existence of six plant community types, namely, Humulus japonicum, Persicaria thunbergii, Artemisia capillaris, Persicaria blumei-Setaria viridis-Digitaria sanguinalis and Phragmites communis. Especially the largest community type, Phragmites communis had dominant weed species such as Humulus japonocum, Arundinella hirta, Persicaria thunbergii, Setaria viridis, Sasola collina and Persicaria perfoliata.

Screening of Natural Herb Resources for Anti-oxidative Effects in Korea (국내 자생 자원식물들의 항산화 활성 검색)

  • In the present study, the anti-oxidative properties of 80% ethanol extracts from 140 kinds of natural plants were investigated. Persicaria perfoliata had the highest anti-oxidative relative potency(VitaminE, $ORAC_{PE}$=1.0), followed by Sorbaria sorbifolia var. stellipila, Rosa multiflora, Lysimachia vulgaris var. davurica, Quercus aliena, Quercus mongolica and Lespedeza bicolor ($ORAC_{PE}$ ${\geq}$ 1.50). Among them, the anti-oxidative activity values of the n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH, and water fractions of the 80% EtOH extracts of P. perfoliata and S. sorbifolia in the ORAC assay system were $1.13{\pm}0.002$, $1.10{\pm}0.004$, $3.67{\pm}0.232$, $1.62{\pm}0.049$ and $1.05{\pm}0.003$; $1.02{\pm}0.007$, $1.00{\pm}0.016$, $2.94{\pm}0.130$, $1.80{\pm}0.019$ and $1.04{\pm}0.012$, respectively, with marked anti-oxidative activity in the ethyl acetate fraction from selected the 2 plant extracts.

Ameliorative Effect of Persicaria Poliata Etract through the Rgulation of AP-1, PI3K/Akt and MAPK Sgnaling Mlecules in UVB-Iradiated HaCaT Clls (HaCaT 세포에서 며느리 배꼽 추출물의 AP-1, PI3K/Akt 및 MAPK 활성 조절을 통한 광손상 억제 효과)

  • Purpose : Skin is the primary barrier to protect the body from various exogenous factors. Among them, UVB exposure can cause the induction of not only excessive inflammatory responses but also the degradation of extracellular matrix (ECM), including collagen and elastin. This study tried to investigate the ameliorative effect of Persicaria perfoliata ethanol extract (PPEE) on UVB-irradiated photodamage through the regulation of activator protein (AP)-1, phosphoinositide 3-kinase (PI3K)/Akt, and mitogen-activated protein kinase (MAPK) signaling molecules in HaCaT cells. Methods : The cytotoxicity of PPEE on HaCaT cells was evaluated by the WST-1 assay. The 80 mJ/cm2 of UVB (312 nm) was irradiated on HaCaT cells to induce the photodamage. Western blot analysis was conducted to investigate the protein expression levels of cyclooxygenase (COX)-2, matrix metalloproteinase (MMP)-9, and heme oxygenase (HO)-1 for ameliorative status by PPEE treatment in UVB-exposed HaCaT cells. In addition, the activated status of the inflammatory transcription factor, AP-1, as well as upstream signaling molecules, PI3K/Akt, and MAPK, were also evaluated by Western blot analysis. Results : Any cytotoxic effect was not induced at the concentration up to 200 ㎍/ml by PPEE treatment. Protein expression levels of COX-2 and MMP-9 were significantly down- and up-regulated by PPEE treatment. The inflammatory transcription factor AP-1, stimulated by UVB irradiation, was also significantly attenuated by PPEE treatment. The phosphorylated status of PI3K/Akt and MAPK were mitigated by PPEE treatment in UVB-exposed HaCaT cells. Moreover, PPEE treatment potently accelerated the expression of HO-1 and its transcription factor, nuclear factor-erythroid 2-related factor (Nrf)2, which is known for its anti-inflammatory activity. Conclusion : Consequently, PPEE treatment significantly regulated COX-2 and MMP-9 expressions in UVB-irradiated HaCaT cells. The inflammatory transcription factor AP-1, along with upstream signaling molecules PI3K/Akt and MAPKs, were also attenuated by PPEE treatment in UVB-exposed HaCaT cells. Additionally, PPEE treatment exaggerated HO-1 expression and Nrf2 activation, which might have contributed to the anti-inflammatory activity of PPEE. These results indicate that PPEE could be a candidate for attenuating UVB-induced photodamage in human skin.
