  • Two unrecorded taxa of Polygonatum (Liliaceae), P. odoratum var, odoratum from IsI. Pungdo, Ansan-shi, Kyonggi Province and P. acuminatifolium from Mishiryong, Kangwon Province were reported. The morphological difference between P. odoratum var. odoratum and P. odoratum var. pluriflorum includes distribution patterns of papillae on the filament surface.

Effects of Polygonatum odoratum on In vivo Insulin Activity in Streptozotocin-Induced Diabetic Rats (둥글레 섭취가 Streptozotocin 유발 당뇨병 쥐의 In vivo 인슐린 작용에 미치는 영향)

  • This study investigated the in vivo insulin function of Polygonatum odoratum in normal and diabetic male Sprague-Dawley rats. Diabetes mellitus was induced by an i.p. injection of streptozotocin. Normal and diabetic rats were assigned to the diet groups of the control basal diet and Polygonatum odoratum diet. The animals were fed the diet and water ad libitum for 15 days. Initial and final body weights, total food intake and serum glucose and insulin levels were measured. An insulin suppression test was performed to elucidate the insulin function in the peripheral tissues. The results showed that the final serum glucose levels significantly decreased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The final serum insulin levels were increased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The in vivo function of the insulin increased in the diabetic rats on the Polygonatum odoratum compared with the diabetic rats on the control diet. These data indicate that Polygonatum odoratum may be beneficial in improving the in vivo insulin function in streptozotocin-induced diabetic rats.

Effect of Polygonatum Odoratum on Lowering Lipid and Antioxidation (옥죽(玉竹)의 지질강하 및 항산화효과)

  • Objectives : This study was designed to examine the effects of Polygonatum odoratum EtOH ext. on lowering lipid and anti-oxidation using hyperlipidemic rat. Methods : Male rats weighting $195.21{\pm}4.93g$ were divided into 4 groups and fed high fat diet for 8 weeks. Each of 7 rats was divided into a control and sample group. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats basal diet and administered an extract of Polygonatum odoratum(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of anti-oxidative activity and tumor necrosis factor-$\alpha$($TNF-{\alpha}$). Results : 1. Concentration of plasma free fatty acid, low density lipoprotein-cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of plasma total cholesterol, triglyceride showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. However, concentration of plasma high density lipoprotein-cholesterol was not significantly different in all the treatment groups. 2. Concentration of liver total cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of liver triglyceride(TG) showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. 3. Concentration of plasma thiobarbituric acid reactive substance, and liver thiobarbituric acid reactive substance showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 4. The values of glutathione peroxidase activity showed a significant increment in all Polygonatum odoratum EtOH ext. groups than that of control group. The values of superoxide dismutase activity and catalase activity showed a significant increment in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 5. The values of plasma aspartate aminotransferase and alanine aminotransferase activities were not significantly different in all treatment groups. 6. Concentration of liver $TNF-{\alpha}$ showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Conclusions : Based on the results in this study, the Polygonatum odoratum EtOH ext. showed a positive effect in lowering lipid and anti-oxidation.

Hypoglycemic & Hypotensive Effects of Polygonatum odoratum Consumption in Non-Insulin Dependent Diabetes Mellitus Patients (둥굴레(Polygonatum odoratum) 섭취가 인슐린비의존헝당뇨병(NIDDM) 환자의 혈당과 혈압에 미치는 영향)

  • The hypoglycemic and hypotensive effects of Polygonatum odoratum in non-insulin dependent diabetes metlitus (NIDDM) patients were investigated in this study. Sixty five NIDDM patients were divided into two groups: hospital diet (HD), Polygonatum odorat um diet (PD). HD group was provided with the diabetic diet used in a hospital and PD group consumed the Polygonatum odoratum added to the HD. The index of obesity, the level of fasting blood pressure (systolic and diastolic), fasting and postprandial blood glucose of NIDDM patients of the PD group were significantly lowered after the experimental period of 4 weeks. The levels of the plasma glucose, lipid and index of obesity was measured. The tendency of elevating the level of HDL-cholesterol and lowering the levels of triglyceride, LDL-cholesternl and total cholesternl were also noticed in NIDDM patients of the PD group. The results showed that Polygonatutn odoratum could be an important dietary source to control hyperglycemia and hypertension and that the plant could be recommended as a preventive or therapeutic agent for NIDDM patients.

The Effect of Butanol Fraction of Polygonatum odoratum with Vitamin E on Blood Glucose Levels and Lipid Peroxidations in Streptozotocin-Induced Diabetic Rats (둥굴레(Polygonatum odoratum)분획물과 비타민 E 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당과 지질과산화에 미치는 영향)

  • The hypoglycemic effects of butanol(BuOH) fraction of Polygonatum odoratum with vitamin E in streptozotocin(STZ)-induced diabetic rats were investigated. Sprague-Dawley rats weighing 200-230g were devided into five groups, and four groups induced diabetes mellitus by the STZ injection(45mg/kg b.w.) into the tail vein : Normal, diabetic-control, and three diabetic experimental groups(p. odoratum group, P. od-vit. E group and Vit. E group). All groups were fed on a AIN-76 diet, and the experimental groups were orally administered with the BuOH fraction of Polygonatum odoratum(500mg/kg b.w.) and vitamin E(10mg/kg b.w.) for 14 days. The body weight, diet intake and organ weights were monitored. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol and aspartate and alanine aminotransferase activities were analyzed. The levels of glycogen in liver and muscle, cholesterol in liver were determined. The malondialdehyde(MDA) levels in liver, kidney and lung were assayed. The body weight loss was seen in P. odoratum group, P. od-vit. E group, Vit. E group and diabetic control group, while the loss in P. odoratum group was much less than that in the diabetic control group. The plasma glucose levels were significantly lowered in P. odoratum group compared to diabetic control group. The plasma insulin levels were noticeably higher in P. odoratum and Vit. E groups. The rats in P. odoratum and P. od-vit. E group showed higher liver glycogen levels than in the diabetic control group. The MDA levels in liver, kidney and lung were also significantly reduced in P. od-vit. E and Vit. E groups compared to the diabetic control group. The results suggest that the administration of BuOH fraction of Polygonatum odoratum along with vitamin E reduced blood glucose levels and peroxidative tissue damage in STZ-induced diabetic rats, showing the possibility of preventive and therapeutic use of the wild edible plant to the diabetes mellitus. Adventitious Bud Formation and Mass Propagation by Root and Propagation Medium of Wild Polygonatum odoratum Druce (야생 둥굴레의 뿌리와 배양토에 따른 대량번식과 부정아 발생)

  • To improve new propagation method, I try to propagate wild polygonatum odoratum roots which formate adventitious bud without bud. The obtained results of size of roots and adventitious bud to formate for multiple propagation of wild polygonatum odoratum as the follow. One year grown wild polygonatum odoratum root in 5cm diameter formate 66.5% adventitious buds. One year grown wild polygonatum odoratum root in 15cm in diameter germinate 113% adventitious bud. Two years grown root in 15cm diameter formate adventitious bud three times more then one year root. Wild polygonatum odoratum in 15cm diameter without any bud in experiment plot makes 4 times of propagation. Sand and Perlite media propagaton plot was increased 3.1∼3.3times more than common culture medium. The best result of mass propagation wild polygonatum odoratum is 10mm diameter root and 5cm cut in length to plant sand media plot.

Color Changes and Optimization of Orgsnoleptic Properties of Roasted Polygonatum odoratum Tea (둥굴레의 볶음처리에 따른 차의 색도 변화와 관능적 특성의 최적화)

  • Response surface methodology(RSM) was applied in roasting processes of Polygonatum odoratum roots in order to monitor Hunter color parameters and to optimize organoleptic properties of the tea. In roasting processing based on the central composite design with variations in roasting temperature (110~19$0^{\circ}C$) and roasting time(15~75min), coefficients of determinations($R^{2}$) of the models were above 0.9827(p<0.01) in Hunter color parameters and above 0.7748 in organoleptic properties. Hunter color L and $\Delta$E values of the powdered samples decreased with the roasting processes, whereas Hunter color a and b values increased up to around 59 min at 136$^{\circ}C$ and thereafter showed a decreased tendency. Organoleptic qualities on color, taste and aroma of Polygonatum odoratum tea were improved along with roasting processes, thereby estimating the optimum roasting conditions by RSM for the maximized qualities of organoleptic properties as a range from 127$^{\circ}C$ to 135$^{\circ}C$ and from 60min to 66 min. Hunter color a value(redness) was found to generate a similar response surface to that of organoleptic properties, showing the possibility of quality index in roasting processes of Polygonatum odoratum roots.

A taxonomic review of Korean Polygonatum (Ruscaceae) (한국산 둥굴레속(Polygonatum, Ruscaceae)의 분류학적 재검토)

  • Polygonatum taxa of Korea were classified as two sections, three series and 14 species and 2 varieties and reviewed taxonomically. Among the examined taxa, endemics in Korea were recorded as two species; P. grandicaule Y. S. Kim, B. U. Oh & C. G. Jang and P. infundiflorum Y. S. Kim, B. U. Oh & C. G. Jang. The habitat of Korean unrecorded species, P. acuminatifolium Kom. and P. odoratum var. odoratum were confirmed in this study. Keys to the sections, series and species including description of each taxon were provided here.

Micropropagation and Mass Production of Adventitious Roots of Polygonatum odoratum via the Culture of Seedling Explnnts

  • Micropropagation and adventitious root production via the culture of Polygonatum odoratum were performed. Stem segments of seedlings of Polygonatum odoratum were the most efficient explants for adventitious shoot formation compared to leaf and root segments. Exogenous cytokinin treatment was required for adventitious shoot formation. Among the cytokinin (BA, Kinetin and Zeatin) tested, BA was most effective for shoot formation from stem segments. Auxin (NAA or IBA) in combination with cytokinin significantly enhanced adventitious shoot formation. Twenty five percent of explants produced adventitious shoots on medium with 2.0 mg/L BAP alone, while 83% of explants produced adventitious shoots on medium with the combination of 2.0 mg/L BAP and 0.1 mg/L IBA. Rooting of adventitious shoots was achieved after transferring to 112 MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L zeatin. When stem segments were cultured on MS medium with various kinds of auxin (IBA, NAA and 2,4-D), adventitious roots were formed from callus. frequency of adventitious root formation was highest in 2,4-D than NAA and IBA. When roots were in clusters together with parental stem segments, growth of roots actively occurred in hormone-free MS liquid medium. The above results represent that possible application for the mass production of roots and plantlets through in vitro culture system of Polygonatum odoratum.

The Effect of BuOH Fraction of Polygonatum odoratum with Selenium on Blood Glucose Level and Lipid Peroxidation in Streptozotocin Induced Diabetic Rats (둥굴레 분획물과 Selenium이 Streptozotocin 유발 당뇨 흰쥐의 혈당수준과 지질과산화에 미치는 영향)

  • The purpose of this study was to investigate the effect of butanol(BuOH) fraction of Polygonatum odoratum with selenium tr-eatment on blood glucose levels and lipid peroxidations in streptozotocin(STZ) induced diabetric rats. Male Sprague-Dawly rats weighing(180-200g) were divided into five groups : normal STZ-control and three expreimental groups(P, odoratum group P, odo-Se group and Se group) Diabetes mellitus was induced by injection STZ in the tail vein at the dose of 45mg/kg B.W The BuOH fraction of Polygonatum odoratum(500mg/kg. B,W) given orally administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na2Seo3(2mg/kg diet). Diabetic rats showed the lower weight gain compared to the normal rats. the plasma glucose levels of the P. odo-Se group were significantly lower than the other experimental groups. The plasma cholesterol levels were higher in STZ-control and Se groups compared toP.odoratum and P. odo-Se groups and HDL-cholesterol levels were increased in the diabetic experimental groups fed on BuOH fraction of P. odoratum with Se supplementation. The liver and muscle glycogen levels were not significantly differ among all groups. The plasma free fatty acid levels were lower in diabetic experimental groups fed on BuOh fraction of P. odoratum or Se sup-plementation than STZ-control and Se groups. Diabetics rats showed the higher levels of triglyceride in plasma andlower levels in liver compared with the normal group. Supplementation with Se decreased significantly the liver triglyceride level. The MDA levels in liver and kidney were significantly reduced in all the experimental groups. In conclusion administration of BuOH fraction of Polygonatuum odoratum with selenium supplementation reduced blood glucose levels and peroxdative tissue damage in STZ induced diabetic rats showing the possibility of preventiave and therapeutic use of the wild edible plant to the diabetes mellitus.
