Comparison of the characteristics of Al(III) hydrolyzed species by improved ferron assay test (개선된 Ferron 분석 비교를 통한 Al(III) 가수분해종 특성 연구)

  • In this study, newly improved Ferron assay test haved on timed spectrometry was used for the determination of hyolrolytic Al species presented in PACl coagulant. The color development reagent ferron was prepared by using conventional method and two newly developed methods. Then the ferron assay test was used to compare and analyze the distribution of Al(III) hydrolyzed species presented in the prepared PACl and alum. The preparing method of reagent A required an aging period of 7 days by adding a hydroxylamine hydroxide and a 1,10-phenanthroline monohydrate reagent, whereas the preparing method of reagent B was used as a coloring agent immediately without aging time. The regression analysis between UV absorbance and Al concentrations of conventional method and newly developed method of ferron reagents in low-concentration aluminum solutions and high-concentration aluminum solutions, showed the correlation coefficients of 0.999 or higher, as showing high correlations of conventional method and newly developed method. Applying Ferron assay test, Al species in the PACls and alum were classified as Ala(monomeric Al), Alb (polymeric Al), and Alc (colloidal and precipitated Al). Distribution of Al(III) hydrolyzed species according to the preparation of ferron colorimetric reagents was similar.

Characterization of Synthetic Polymeric AI(III) Inorganic Coagulants for Water Treatment (상수처리용 합성 무기고분자 Al(III)계 응집제의 화학적특성)

  • This research explored the feasibility of preparing and utilizing a prefonned polymeric solution of Al(III) for coagulation in water treatment. Slow base(NaOH) injection into supersaturated aluminum chloride and aluminum sulfate solutions did produce high yields of Al polymers useful to water treatment applications. The method of characterization analysis was based on timed spectrophotometer with ferron as a color developing reagent. The hydrolytic Al species were divided into $monomeric(Al_a),\;polymeric(Al_b),\;and\;precipitate(Al_c)$ from the difference in reaction kinetics. The analysis of PACl's characteristics showed that the quantity of polymeric Al produced at value of$ r(OH_{added}/AI)=2.2$ was $83\%$ of the total aluminum in solution, as showing maximum contents and precipitated Al was dramatically increased when r was increased above 2.35. In addition, the characteristics of polyaluminum sulfate (PAS) showed that polymeric Al contained at r = 0.75 was $18\%$ of the total aluminum in solution. The synthesized PACI and PAS were stable during storing period, as indicating negligible aging effect. The effect of sulfate ion on PACI was dependent on the concentration of sulfate ion. That is, polymeric species decrease and precipitate species increase as sulfate ion concentration increased. It can be concluded that the sulfate cause the formation of $Al(OH)_{3(S)}$ at low pH. However, The effect of calcium ion was negligible for distribution of Al species.

  • The adsorption behavior of aromatic acids on Amberlite XAD-4 resin was investigated by measuring the distribution coefficient by batch method. The adsorption of solutes on XAD-4 was affected by the several factors such as, analyte concentration, the pH of solution and concentration of pairing ion. The enhanced adsorption of solutes on XAD-4 in the presence of tetraalkylammonium salt as an ion pairing reagent, referred to as ion interaction, was suggested to follow a double layer model where the pairing ion occupies a primary layer at the adsorbent while the solute anion and other anions in the system comlpete for the secondary layer. Therefore, the ability of an ion pairing reagent to enhance solute adsorption depended significantly on the type and concentration of counter-ion and co-anion accompanying the ion pairing reagent or salt used for ionic strength control. In addition, a good linear relationship between the logarithm of capacity factors measured by batch and elution method as a function of the concentration of ion pairing reagent and methanol can be used to predict the retention in elution method on the basis of capacity factors measured by batch method.

  • 1-Ethyl-6-fluoro-1, 4-dihydro-4-oxo-7-(1-piperazinyl)quinolinea-3-car-boxylic acid-dextran was synthesized by the reaction of 1-ethyl-6-fluoro-1, 4-dihydro-4-oxo-7-(1-piperazinyl )quinoline-3-acryloyl chloride with dextran. Polymeric drug was tested for antimicrobial activity in vitro against ten species of microorganisms. Polymeric drug revealed good antibacterial activity against Bacillus subtillis ATCC 6633, Staphyloccus aureus ATCC 25923, Mycrobactertum phlei IFO 3158, Salmonella typhimurium KCTC 1925, Escherichia coli KCTC 1039, Escherichia coli ESS, Klebsiella puemouiae KCTC 1560 and Pseudomonas aeruginosa IFO 13130. Polymeric drug have no antimicrobial against Candida albicans ATCC 10231, but moderately active Micrococcus luteus ATCC 9341.

Polymers for Microfluidic Chips

  • Microfluidic systems have attracted much research attention recently in the areas of genomics, proteomics, pharmaceutics, clinical diagnostics, and analytical biochemistry, as they provide miniaturized platforms for conventional analysis techniques. The microfluidic systems allow faster and cheaper analysis using much smaller amounts of sample and reagent than conventional methods. Polymers have recently found useful applications in microfluidic systems due to the wide range of available polymeric materials and the relative ease of chemical modification. This paper discusses the fundamentals of microfluidic systems and the roles, essential properties and various forms of polymers used as solid supports in microfluidic systems, based on the recent advances in the use of polymers for microfluidic chips.

PAMAM Dendrimers Conjugated with L-Arginine and γ-Aminobutyric Acid as Novel Polymeric Gene Delivery Carriers

  • In this study, we synthesized functional dendrimer derivatives as nonviral gene delivery vectors. Poly(amidoamine) dendrimer (PAMAM, generation 4) was modified to possess functional amino acids to enhance gene transfection efficiency. PAMAM G4 derivatives conjugated with L-arginine (Arg) and ${\gamma}$-aminobutyric acid (GABA) showed higher transfection efficiency and lower cytotoxicity compared to the native PAMAM G4 dendrimer. The polyplex of the PAMAM G4 derivative/pDNA was evaluated using an agarose gel retardation assay and Picogreen reagent assay. Additionally, the MTT assay was performed to examine the cytotoxicity of synthesized polymers. All PAMAM G4 derivatives showed lower cytotoxicity than PEI25kD. Particularly, PAMAM G4-GABA-Arg displayed enhanced transfection efficiency compared to the native PAMAM G4 dendrimer.

Mechanistic Aspects in the Grignard Coupling Reaction of Bis(chloromethyl)dimethylsilane with Trimethylchlorosilane

  • The Grignard reactions of bis(chloromethyl)dimethylsilane (1) with trimethylchlorosilane (2) in THF give both the intermolecular C-Si coupling and intramolecular C-C coupling products. At beginning stage, 1 reacts with Mg to give the mono-Grignard reagent ClCH2Me2SiCH2MgCl (1) which undergoes the C-Si coupling reaction to give MC2Si(CH2SiMe3)2 3, or C-C coupling to a mixture of formula Me3SiCH2(SiMe2CH2CH2)nR1 (n = 1, 2, 3, ..; 4a, R1I = H: 4b, R1 = SiMe3). In the reaction, two reaction pathways are involved: a) Ⅰ reacts with 2 to give Me3SiCH2SiMe2CH2Cl 6 which further reacts with Mg to afford a Me2SiCH2Mel-SiCH2MgCl (Ⅱ) or b) I cyclizes intramolecularly to a silacyclopropane intermediate A, which undergoes a ring-opening polymerization by the nucleophilic attack of the intermediates I or Ⅱ, followed by the termination reaction with H2O and 2, to give 4a and 4b, respectively. As the mole ratio of 2/1 increased from 2 to 16 folds, the formation of product 3 increased from 16% to 47% while the formation of polymeric products 4 was reduced from 60% to 40%. The intermolecular C-Si coupling reaction of the pathway a becomes more favorable than the intramolecular C-C coupling reaction of the pathways b at the higher mole ratio of 2/1.

Development of HPLC Determination Method for Trace Levels of 1-, 2-Nitropyrenes and 2-Nitrofluoranthene in Airborne Particulates and Its Application to Samples Collected at Noto Peninsula

  • 1-Nitropyrene (1-NP), 2-NP and 2-nitrofluoranthene (2-NFR) are useful markers for studying the atmospheric behaviors of polycyclic aromatic hydrocarbons (PAHs) and nitropolycyclic aromatic hydrocarbons (NPAHs). However, present methods for measuring trace levels of these compounds are lesssensitive and laborious. Here we describe several improvements to a previously reported high-performance liquid chromatography-chemiluminescence detection system that allows it to determine trace levels of 1-, 2-NPs and 2-NFR. The proposed system was equipped with a reducer column packed with Pt/Rh instead of zinc whose life-time was limited. The combination of Cosmosil MS-II (monomeric ODS) and AR-II (polymeric ODS) columns was used instead of polymeric ODS columns as the separator column to improve the separation. An ethanol mixture with acetate buffer (pH 5.5) was used in place of an acetonitrile mixture with the same buffer to activate the reducer column. The same ethanol mixture was used as the mobile phase for the clean-up column. The switching time of the column switching valve was optimized to concentrate the amino-derivatives of above NPAHs quantitatively on the concentrator column. The concentrations of bis(2,4,6-trichlorophenly) oxalate and hydrogen peroxide in the chemiluminescence reagent solution were optimized to 0.4 mM and 30 mM, respectively, to increase the sensitivity. Under the above conditions, the detection limits (S/N=3) of 1-, 2-NPs and 2-NFR were 1 fmol (0.25 pg), 10 fmol (2.5 pg) and 4 fmol (1 pg), respectively. The proposed system was effectively used to determine trace levels of 1-, 2-NPs and 2-NFR in airborne particulates collected at Noto Peninsula. The atmospheric concentrations of 1-, 2-NPs and 2-NFR were not more than sub pg $m^{-3}$ levels. They were higher in winter (January) than in summer (July). In both seasons, the concentrations were in decreasing order, [2-NFR]>[1-NP]>[2-NP].

Characteristic of Al(III) Hydrolysis Specie Distribution on Coagulation Process (응집공정에서 발생하는 알루미늄 가수분해종 분포특성)

  • The overall objective of this research was to find out the role of rapid mixing conditions in the species of hydrolyzed Al(III) formed by Al(III) coagulants and to evaluate the distribution of hydrolyzed Al(III) species by coagulant dose and coagulation pH. When an Al(III) salt was added to water, monomeric Al(III), polymeric Al(III), precipitate Al(III) was formed by Al(III) hydrolysis. The method of hydrolyzed Al(III) species characterization analysis was based on timed spectrophotometer with ferron as a color developing reagent. The hydrolytic species were divided into monomer, polymer, precipitate from the reaction kinetics. And then, the color intensity for monomeric Al(III) was read 3 min after mixing. With standard Al solution containing monomeric Al(III) only, the Al-ferron color intensity slightly increased with until about 3 min. During the rapid mixing period, for purewater, formation of dissolved Al(III) (monomer and polymer) was similar to rapid mixing condition, but for raw water, the species of Al(III) hydrolysis showed different result. During the rapid mixing period, for high coagulant dose, Al-ferron reaction increases rapidly. The kinetic constants, Ka and Kb, derived from Al-ferron reaction. The kinetic constants followed very well the defined tendencies for coagulation condition. For pure water, when the rapid mixing time increased, the kinetic constants, Ka and Kb showed lower values. Also, for raw water, when the rapid mixing time increased, the kinetic constants, Ka and Kb showed lower values.
