다당류를 이용한 중금속 이온의 흡착특성 비교

  • Polysaccharides are very effective adsorbents for heavy metals. In this study, the adsorption characteristics of various polysaccharides for heavy metal adsorption were investigated. Tested polysaccharides were homogeneous polysaccharides such as curdlan, chitin, starch, cellulose, Avicel, and Solka floc and heterogeneous polysaccharides such as zooglan, locust bean gum, ghatti gum, pectin, and xylan. Lead(II) adsoption characteristic on these polysaccharides followed Freundlich isotherm and the isotherm parameters were calculated. For adsorption of lead(ll), Avicel, starch, and zooglan were found to be good adsorbents.

  • The polysaccharides were extracted from fruiting body, mycelia, and cell-free broth of Agaricus blazei Murill. The crude polysaccharides were obtained by the ethanol addtion. They were further purified using ion-exchange chromatography and gel chromatography. Ion-exchange chromatography using DEAE-cellulose column separated neutral and acidic polysaccharides. Neutral polysaccharides were then purified with gel filtration chromatography. For single peak obtained from gel filtration chromatography was molecular weight was measured with Sepharose CL-6B. The same procedure with acidic polysaccharides were performed to get the purified polysaccharides.

Analysis of the Extraction Condition of Soluble Acidic Polysaccharides from Ginseng Marc (인삼박으로부터 수용성 산성다당체의 추출 조건 분석)

  • This study was carried out to investigate the optimum conditions for extraction of soluble acidic polysaccharides from ginseng marc. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in ginseng marc. The amounts of soluble acidic polysaccharides in water extract of ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite the extraction time increasing from 0.5 hours to 6 hours. To estimate the rehydration rate of the freeze dried polysaccharide, the extracted acidic polysaccharide fraction powder was determined the amount of soluble acidic polysaccharides by carbazole-sulfuric acid method again. The rehydration rate of acidic polysaccharides from water-extract of red ginseng marc at room temperature was 100%. On the other hand, the rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 50%. The rehydration rate of acidic polysaccharides from water-extract of white ginseng marc at room temperature was 50%. The rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 40%. The rate of soluble acidic polysaccharide of Red Ginseng is higher than that of White Ginseng. We can find out the maximum extraction method of soluble acidic polysaccharide from ginseng marc.

Immuno-stimulatory Effects of Sulfated Polysaccharides Isolated from Codium fragile on Interleukin-1β Gene Expression in Olive Flounder, Paralichythys olivaceus

  • Sulfated polysaccharides are known to be immune-stimulators in mammals and can be used as food additives to enhance immunity. In this study, the immune-stimulating activity of water-soluble anionic macromolecules F2 fractionation isolated from Codium fragile using ion-exchange chromatography was tested in olive flounder, Paralichythys olivaceus, in vitro and in vivo. The gene expression of interleukin (IL)-1β was adopted to check the immune-affection. As a result, in vitro study revealed that the expression of IL-1β was significantly upregulated in head kidney cells by 1 and 5 ㎍/ml of polysaccharides 4 h and by 5 ㎍/ml of polysaccharides at 24 h. In vivo, IL-1β gene expression in head kidney was significantly upregulated by 20 and 100 ㎍ of the polysaccharides at day 1 post-i.p. injection, while downregulated at day 3 but not significant. Meanwhile, in peritoneal cells, it was upregulated by 20 ㎍ of the polysaccharides at day 1 but the upregulation was sustained until day 3 though it was not significant. These results indicate that the sulfated polysaccharides from C. fragile are an immune-stimulator and might be potential feed additives for olive flounder.

Extraction and Component Sugar Analysis of Polysaccharides from Buckwheat (메밀의 다당류 추출과 구성당 분석)

  • This study was conducted to extract and isolate the polysaccharides from buckwheat. Also the sugar composition of the polysaccharides was investigated. The soluble and indigestible polysaccharides were isolated from supernatant and residue after enzyme treatment of raw, roast and steam buckwheat. The yields of low molecular weight soluble polysaccharides(LMS-P: MW<10 Kda) and high molecular weight soluble polysaccharides(HMS-P: MW>10 Kda) were $74.9{\sim}84.2%$ and $5.5{\sim}9.4%$, respectively. The yields of indigestible polysaccharides were low molecular weight insoluble polysaccharides; $0.8{\sim}4.2%$, crude hemicellulose; $3.2{\sim}9.6%$, alcohol insoluble hemicellulose; $0.9{\sim}1.7%$, residue; $2.0{\sim}2.4%$, respectively. The free sugars were detected in the soluble polysaccharides and low molecular weight insoluble polysaccharides but were not detected in the crude hemicellulose, alcohol insoluble hemicellulose and residue. The protein of all fraction were detected and the content was $1.0{\sim}18.9%$. The main sugar of soluble polysaccharides was glucose and the indigestible polysaccharides were composed of glucose, arabinose, rhamnose, xylose, mannose and galactose.

  • Prebiotic oligosaccharides, with a degree of polymerization (DP) of mostly less than 10, exhibit diverse biological activities that contribute to human health. Currently available prebiotics are mostly derived from disaccharides and simple polysaccharides found in plants. Subtle differences in the structures of oligosaccharides can cause significant differences in their prebiotic properties. Therefore, alternative substances supplying polysaccharides that have more diverse and complex structures are necessary for the development of novel oligosaccharides that have actions not present in existing prebiotics. In this review, we show that structural polysaccharides found in plant cell walls, such as xylans and pectins, are particularly potential resources supplying broadly diverse polysaccharides to produce new prebiotics.

Inhibition of Metastasis and Invasion of Ovarian Cancer Cells by Crude Polysaccharides from Rosa Roxburghii Tratt in Vitro

  • Background: Rosa Roxburghii Tratt is a promising wild fruit crop in Southwest China. Its extracts have been used as traditional Chinese medicine, which benefit immune responses and cure various health disorders. However, whether Rosa Roxburghii Tratt polysaccharides could inhibit metastasis and invasion of ovarian cancer cells remains unknown. Materials and Methods: Effects of crude polysaccharides from Rosa Roxburghii Tratt on the viability of ovarian cancer A2780 cells were detected by MTT assay. Ovarian carcinoma cell migration and invasion after exposure to Rosa Roxburghii Tratt polysaccharides were quantified by wound healing and Transwell assays, respectively. Western blotting was applied to assess protein levels of MMP-9. Results: The results indicated that Rosa Roxburghii Tratt polysaccharides significantly reduced wound closure rate of A2780 cells, inhibited their migration and invasion, and suppressed the expression of MMP-9. Conclusions: Our findings indicated that Rosa Roxburghii Tratt polysaccharides have potential for develop as anti-metastatic cancer drug preparations for ovarian cancer patients.

Protective Effects of Some Phytobased Polysaccharides on the Acute Hepatic Damages of ICR-Mice Induced by the Administration of Carbon Tetrachloride and D-Galactosamine (수종 식물성 다당류의 사염화탄소 및 D-Galactosamine 유발 급성 간손상 보호작용)

  • Polysaccharides obtained from Sappan Lignum, Mori Radicis Cortex and Olibanum were examined for their liver protective effects against carbon tetrachloride-and D-galactosamine intoxication in ICR-mice. Hexobarbital sleeping time and serum transaminases (S-GOT and S-GPT) were measured as parameters for the evaluation of liver protective effects. All polysaccharides tested in this experiment showed remarkable positive effects on the prevention of hepatic intoxication with carbon tetrachloride-and D-galactosamine. Much better liver protective effects of polysaccharides were observed in D-galactosamine-liver injuries than in carbon tetrachloride-injuries.

  • Low molecular weight molecules and high molecular weight substances were found to have anti-tumor and immuno-modulating activity. Previously polysaccharides have been received much attention because of adhesives, food additives or animal foods (Whistler et al., 1976). In effort of developing new anti-tumor substances with low toxicity, numerous polysaccharides from yeast, algae, bacteria, higher plants and especially fungi have been investigated for anti-tumor and immuno-modulating activities. Thus the high molecular weight molecule was reported to have anti-tumor activity through host mediated immunity. In this brief article, attention will be paid to polysaccharides which is especially fungal origin.

  • To find antitumor components in liquid cultured cell-free broth and the hot water extract from the fruiting body of Hericium erinaceus, polysaccharides were purified and fractionated by DEAE-cellulose ion exchage column chromatography and Sepharose CL-6B gel filtration chromatography. Among various crude polysaccharides and purified polysaccharides the fractions showing immune-activity were determined by NO assay.
