Isolation and Cultural Characteristics of a Phosphate-solubilizing Fungus, Penicillium sp. PS-113 (인산가용화 사상균, Penicillium sp. PS-113 균주의 분리 및 배양특성)

  • A fungus with high phosphate solubilizing activites was isolated from soil using potato dextrose agar-calcium phosphate medium and identified to Penicillium sp. PS-113, based on the morphological characteristics of conidiophore and conidia; flask shape of phialide, simple branching type of conidiophore, and columnar shape of conidial head, in malt extract agar and potato dextrose agar media. The optimum temperature ad initial pH to solubilize rock phosphate in potato dextrose broth-rock phosphate medium were 30$^{\circ}C$ and pH 8.0, respectively. In these conditions phosphate solubilizing activities of Penicillium sp. PS-113 against four types of insoluble phosphate like tricalcium-phosphate, aluminium phosphate, hydroxyapatite and rock phosphate were quantitatively determined. As results, this fungus highly produced free phosphates to the culture broth with the concentrations of 1,283 ppm against tricalcium-phosphate, 585 ppm against rock phosphate, 528 ppm against aluminium phosphate, and 242 ppm against hydroxyapatite, respectively.

Characteristics of Growth, Pigment and Monacolin K Production by Monascus strains in Liquid Culture (액체배양에서의 홍국균의 생장, 색소 및 monacolin K 생산 특성)

  • The hyphal growth, production of color pigments and monacolin K by Monascus strains were investigated in liquid medium. Thirty five different strains were collected and cultured in potato dextrose yeast extract broth (PDYB), potato dextrose broth (PDB) and malt extract broth (MEB) medea at $25^{\circ}C$ for 7 days. The growth rates of most of strains were highest in PDYB medium. Growth rate as well as pigment production were influenced by suspension conditions of mycelia during liquid cultivation. Most of strains producing monacolin K corresponded to strains producing red pigment highly and showing more pH changes of liquid media. Monacolin K produced from strains was detected in culture broth as well as mycelia. Any citrinin was not detected in monacolin K producing strains. These results imply that the selection of the strains producing red pigment highly and showing more pH changes in liquid cultivation could be applied for primary screening of Monascus strains for preparation of red mold rice.

Fundamental Cultural Characteristics of Chlorociboria spp. Native to Korea (한국에 자생하는 녹청균류의 기초 배양 특성)

  • Members of Chlorociboria are soft-rot ascomycetes that produce blue-green pigment. We investigated the growth characteristics of two Korean species of Chlorociboria, eight strains of Chlorociboria aeruginascens and Chlorociboria poutoensis, under various culture conditions (solid media, temperature, pH) and screened them for extracellular enzyme activity. Although the growth rate was slow, all tested strains of Chlorociboria spp. grew well on potato dextrose agar (PDA; 16.3~42.6 mm after 60 days) or Sabouraud dextrose agar (SDA), but not on malt extract agar (MEA). Compared with C. aeruginascens strains, C. poutoensis strains exhibited higher expression of blue-green pigments on both PDA and SDA media. The optimal temperature for mycelial growth was $20{\sim}25^{\circ}C$, and mycelial growth was lower at $30^{\circ}C$ than at $10^{\circ}C$. All strains tended to have increased mycelial growth as the incubation temperature increased in the range of 10 to $20^{\circ}C$. The optimal pH of potato dextrose broth (PDB) for mycelial growth varied according to the strain under static culture conditions. Maximum biomass production was obtained at pH 6.0 for NIFoS 579 ($114.3{\pm}5.1mg/60days$), but it maintained a stable pigment expression under a broad pH spectrum. The activities of both cellulase and laccase were observed in all tested strains of Chlorociboria spp. Enzyme activities of NIFoS 579 were remarkably higher than those of the other strains. From these results, we suggest that C. poutoensis NIFoS 579 is a potential candidate for use as a source of natural blue-green dye.

Cultural Characteristics of Shimizuomyces paradoxus Collected from Korea

  • This study investigated the cultural characteristics of Shimizuomyces paradoxus in different nutritional and environmental conditions. The highest mycelial growth was observed in Schizophyllum (mushroom) genetics complete medium plus yeast extract agar medium, and the optimal temperature and pH were $25^{\circ}C$ and pH 8.0, respectively. The optimal carbon and nitrogen sources were 1% dextrose and 1% peptone in agar. However, in liquid culture the highest dry mycelium weight was found for the potato dextrose agar and potato sucrose agar broths. The optimum inoculum size was five mycelial discs (5 mm) per 100 mL of broth, and the optimum liquid culture period was 25 days. This is the first ever report of S. paradoxus cultural characteristics.

Effect of Sunlight on the Reduction of Mycelia and Aflatoxins (태양광선에 의한 Aflatoxin의 감소 효과)

  • This study was performed to investigate the possible effect of sunlight on the reduction or degradation of mycelia and aflatoxins. The mycelia and aflatoxins were produced by Aspergillus parasiticus ATCC 15517 in a yeast-extract sucrose broth (YES) and potato-dextrose agar (PDA) and then exposed to sunlight. The weight of mycelia was decreased to 76.8% in 8 hours and to 66.7% in 168 hours(p<0.05). The total aflatoxin level was significantly decreased to below 50% (46.3% in the YES broth and 49.6% in the PDA) in 8 hours (p<0.05). After 168 hours, a 90.4% degradation of aflatoxin in the YES broth and a 77.2% degradation of aflatoxin in the PDA was observed, respectively (p<0.01). The results showed that the degradation ratios of total aflatoxin level increased with increased exposure time to sunlight. These results indicate that sunlight could be an effective factor in aflatoxin degradation although its effect on mycelia was less pronounced.

Morphological Characteristics of Chlamydospores of Cylindrocarpon destructans Causing Root-rot of Panax ginseng (인삼 뿌리썩음병균 Cylindrocarpon destructans 후막포자의 형태적 특성)

  • Chlamydospore formation from mycelia and conidia of Cylindrocarpon destructans isolated from the root rot lesion of the Panax ginseng was investigated by scanning electron and light microscopy. Typical chlamydospores were formed only from hyphae but not from conidia on culture media. However, immature chlamydopspore-like cells were formed from microconidia after 12 days of incubation at 20$^{\circ}C$ on Czapek Dox broth (CDB) adjusted to pH 4.0. Chlamydospores were yellowish or reddish brown in color, and produced singly or in chain with the hyphal intercalary or terminal position on potato-dextrose agar, V-8 juice agar and CDB with no addition of nitrogen sources after 16∼20 days of incubation at 20$^{\circ}C$. They were 11.3 to 11.9 $\mu\textrm{m}$ in diameter, having many lumps-like warts on their surface with the length of 1.5 to 1.8 $\mu\textrm{m}$.

  • 옥수수 깜부기병균 Ustilago maydis 동포자를 풍건한 후 모래나 silica gel을 보존매체로 해서 4$^{\circ}C$나 그 이하 온도에서 24개월 두었을 때 동포자 생존율은 40~45%였다. 동포자는 28$^{\circ}C$ PDA상에서 쉽게 발아하여 많은 소생자를 형성하였다. U. maydis 동포자로부터 소생자 형성의 최적배지는 corn meal broth였고, carrot broth와 potato-dextrose broth의 순으로 좋았다. 소생자 형성에 가장 적합한 조건은 온도 28~3$0^{\circ}C$, pH5~7이었다. 소생자 형성은 암조건에서 보다 좋았다.

Solubilization of Insoluble Phosphates by Aspergillus sp. PS-104 Isolated from Soil (토양에서 분리한 Aspergillus sp. PS-104 균주에 의한 난용성 인산염 분해)

  • Phosphate-solubilizing microorganisms were isolated from soil around Kyungnam and Kyungbook regions using potato dextrose agar-calcium phosphate medium. A fungus with the greatest phosphate-solubilizing activity was selected and identified to Aspergillus sp. PS-104, based on the morphological characteristics of conidiophore and conidia; unbranching type of conidiophore, terminally swelling of conidiophore and septate of mycelium, in malt extract agar and potato dextrose agar media. The optimum temperature and initial pH to solubilize rock phosphate in potato dextrose broth-rock phosphate medium were $30^{\circ}C$ and pH 7.0, respectively. In these optimum conditions, phosphate-solubilizing activities of Aspergillus sp. PS-104 against four twos of insoluble phosphate, tricalcium phosphate, aluminium phosphate, hydroxyapatite and rock phosphate, were quantitatively determined. As result, the maximum phosphate-solubilizing activity was obtained with tricalcium-phosphate (1,900 ppm) while minimum activity was obtained with hydroxyapatite (320 ppm). Futhermore, phosphate-solubilizing activity of Aspergillus sp. PS-104 was found higher when treated with nitrates as compared to the ammonium salts as a nitrogen sources.

Screening New Antihypertensive Angiotensin I-Converting Enzyme Inhibitor -Producing Yeast and Optimization of Production Condition (항고혈압성 안지오텐신 전환효소 저해제를 생산하는 새로운 효모의 선별 및 저해물질 최적 생산조건)

  • Forty eight strains of yeast were cultured in potato dextorse(PD) broth at $30^{\circ}C$ for 24 hr and centrifuged with 12,000 rpm for 20 min. After concentrated the cultures, antihypertensive angiotensin I-converting enzyme(ACE) inhibitory activities of its concentrates were investigated. Among them, the concentrates from Saccharomyces cerevisiae Y183-3 showed the highest ACE inhibitory activity of 71.8%. The ACE inhibitor from Saccharomyces cerevisiae Y183-3 was maximally produced when Saccharomyces cerevisiae Y183-3 cultured in PD broth at $30^{\circ}C$ for 36 hr.
