Antithrombotic Efficacy of Protaetia brevitarsis Extract (흰점박이꽃무지(Protaetia brevitarsis) 추출물의 항혈전 효능)

  • We aimed to evaluate antithrombotic efficacy of Protaetia brevitarsis extract during 21 days. Rats (SPF rat, weight 240~260 g) were divided into 16 groups (5 rats per group), they were: control group and Protaetia brevitarsis extract groups with dose of 0.1, 0.5, 1, 2.5, 5, 10, 25, 50, 75, 100, 200, 250, 500, 750, 1,000 mg/kg kg of body weight. Thromboplastin time (PT) and activated partial thromboplastin time (aPPT) as antithrombotic efficacy were tested in this animal experiment (at 7, 14 and 21 days). Overall, the admistration dose of Protaetia brevitarsis extract over 50 mg/kg at 7, 14 and 21 days for PT and over 25 mg/kg at 7, 14 and 21 days for aPPT tented to be longer than that of other groups. In addition, the optimal admistration doses of Protaetia brevitarsis extract to improves antithrombotic efficacy were 75, 100, 200 and 250 mg/kg at 7, 14 and 21 days for PT (p<0.05) and 50 and 100 mg/kg at 7 days, 75 mg/kg at 14 days, or 50, 100, 200 and 250 mg/kg at 21 days for aPPT (p<0.05). It can be concluded that Protaetia brevitarsis extract at optimal levels have antithrombotic efficacy.

Preventive Effect of A Chafer, Protaetia brevitarsis Extract on Carbon Tetrachloride-Induced Liver Injuries in Rats (사염화탄소로 유발된 랫드의 간 손상에 대한 흰점박이꽃무지 (Protaetia brevitarsis) 추출물의 예방효과)

  • The hepato-protective effects of the extract from Protaetia brevitarsis against hepatotoxicity by carbon tetrachloride ($CCI_4$) were studied in rats. The rats were orally treated with $CCI_4$ (50% in corn oil) at initial dose of $1\;m{\ell}/kg$ followed by $0.5m{\ell}/kg$ four times during 2-week period. The extract of P. brevitarsis (50, 100 or 200 mg/kg) or its vehicle was administered day after day from 1 week before $CCI_4$ Injection during five weeks. $CCI_4$ induced hepato-celluar degeneration and necrosis induced to increase in serum aspartate amintransferase (AST) and alanine aminotransferase (ALT) levels. In biochemical analyses, thiobarbituric acid-reactive substances (TBARS) and antioxidant enzymes such as superoxide dismutase (SOD) and catalase in hepatic tissues were remarkably increased by $CCI_4$ treatment. Not only increases in serum AST and ALT, but also induction of lipid peroxidation and antioxidant enzymes in hepatic tissues caused by $CCI_4$ were significantly attenuated by the P. brevitarsis extract in a dose-dependent manner. Such hepato-protective effects of P. brevitarsis extract were confirmed by histopathological examinations, wherein only mild hepatocytic vacuolations were observed in the liver of rats treated with a high dose (100 mg/kg) of P. brevitarsis extract in comparison with severe hepatocytic degenerations administered with $CCI_4$ alone. From these results, it is suggested that the extract of Protaetia brevitarsis could be a promising candidate for the protection of liver injury, based on the preventive effects against morphological cellular injuries, lipid peroxidation and serum biochemical parameters.

  • Protective effects of the water extract of Protaetia brevitarsis larva against $CCl_4-induced$ toxicity were investigated in primary cultures of adult rat hepatocytes. The extract used in these studies contained several minerals, fatty acids and amino acids. Treatment of hepatocyte cultures with the extract provided a significant protection from the increased LDH activity induced by $CCl_4$. The results demonstrated that the extract may have the protective effect against $CCl_4-induced$ toxicity in hepatocyte cultures.

Immune-enhancing effects of Protaetia brevitarsis seulensis larvae extracts on RAW264.7 macrophages

  • Protaetia brevitarsis seulensis larva is well-known as an edible insect. The present study aimed to explore the immune-enhancing effect of 30% ethanol extract of Protaetia brevitarsis seulensis larvae (PBE) in RAW264.7 macrophage cells. PBE were not cytotoxic to RAW264.7 cells and nitric oxide production increased on PBE treatment in a dose-dependent manner. Furthermore, PBE significantly promoted the expression of immune-related mediators (Inos and COX-2) and cytokines (TNF-α, IL-6, and IL-1β) and the phosphorylation of MAPKs (ERK, p38, and JNK). Taken together, the immune-enhancing effects of 30% ethanol extract of PBE in vitro were identified. These findings can be used as data for the development of edible insect-based functional foods that improve immune function.

  • The white-spotted flower chafer (Protaetia brevitarsis) larva is one of the insects widely-used for edible and medicinal purposes in Eastern Asia. As a result of increasing demand for P. brevitarsis, massrearing systems in domestic farms have become necessary. However, the mass-rearing of larvae under confined rearing conditions could provide conditions unsuitable for preventing entomopathogenic diseases. Metarhizium anisopliae is the strongest fatal entomopathogenic fungus against P. brevitarsis. For inhibition of M. anisopliae, we used a Rhus verniciflua Stokes extract that has antifungal components. We investigated the inhibitory effect of the R. verniciflua extract at 1%, 5%, and 10% concentrations. The results showed that a 1% R. verniciflua extract added to sawdust produced a significantly low P. brevitarsis mortality rate. Moreover, extract-treated groups were heavier and had a shorter larval period than those of the untreated group. Consequently, we suggest that using an R. verniciflua extract can reduce the P. brevitarsis fatality rate from entomopathogenic fungi (e.g. M. anisopliae), resulting in more effective mass-rearing systems for P. brevitarsis.

Radiation Protection Effect of Protaetia Brevitarsis Larvae Extracts on Blood and Prostate in Male Rats Irradiated with Co-60 Gamma-ray (흰점박이꽃무지 유충 추출물이 Co-60 감마선에 조사된 수컷 흰쥐의 혈구 및 전립선에 미치는 방사선 방호효과)

  • This study is desinged to examine for radiation protection effect of Protaetia Brevitarsis Larvae extracts on the blood and prostate of male rat as a natural radiation protection agent. 5 groups were classified using 90 male rat as experimental animals. Each group was clssified as normal control group (NC Group), the group administered protaetia brevitarsis larvae extracts (PBE Group), irradiated group (IR Group), irradiated group after administration of protaetia brevitarsis larvae extracts (PBE+IR Group), the group administered protaetia brevitarsis larvae extracts after irradiaton (IR+PBE Group). In IR Group, 7 Gy/h of Co-60 gamma ray was irradiated to SD rats. In PBE+IR Group, protaetia brevitarsis larvae extacts wewe injected at 200 mg/kg/day for 14 days before irradiation, In IR+PBE Group, protaetia brevitarsis larvae extract was injeted after irradiation. On the 1, 7 and 21 days after irradiation, the experimental animals were sacrificed to evaluate the changes in blood cell component, superoxide dismutase (SOD) activity, histopathological evaluation of the liver and prostate gland. As a result, the PBE+IR Group and IR+PBE Group showed a significantly recovery of white blood cell (p<0.01, p<0.01), platelet (p<0.01, p<0.01) than the IR Group. It was also confirmed that SOD activity of PBE+IR Group (p<0.01) and IR+PBE Group (p<0.01) was significantly increased than the IR Group. Also PBE+IR Group and IR+PBE Group showed less inflammatory reactions of cystoplasm in the prostate gland than the IR Group. In conclusion, the protaetia brevitarsis larvae have radioprotection effect against blood and prostate gland. It is expected to be useful for research of radiation protection agent.

Antioxidant Activities of Protaetia brevitarsis Larvae Fermented by Lactobacillus acidophilus (유산균 발효가 흰점박이꽃무지(Protaetia brevitarsis) 유충의 항산화활성에 미치는 영향)

  • This study was carried out to evaluate the effect of fermentation by Lactobacillus acidophilus on the antioxidant activity of Protaetia brevitarsis larvae fed with mushroom substrates (king oyster mushroom). The total polyphenol content of the P. brevitarsis larvae extracts (PLEs) (93.33±0.98 mg GAEs/extract g) was higher than that of the fermented P. brevitarsis larvae extracts (FPLEs) (65.02±1.32 mg GAEs/extract g). The flavonoid contents of the PLEs and FPLEs were 18.3±1.57 QEs mg/extract g and 17.69±0.95 QEs mg/extract g, respectively. The DPPH radical scavenging activity showed no significant difference between the PLEs and FPLEs at a concentration of 2-4 mg/ml. However, at a concentration of 8 mg/ml or more, the DPPH radical scavenging activity of the FPLEs was higher than that of the PLEs. The reducing power of the FPLEs was also higher than that of the PLEs, and more than twice as high at a concentration of 1.6 mg/ml or more. The ORAC value of the FPLEs (79.22±0.72 μM TEs/extract g) was higher than that of the PLEs (74.34±0.37 μMTEs/extract g). A WST-1 assay of the RAW 264.7 cells indicated that the PLEs and FPLEs showed no cytotoxicity.

  • Hepatoprotective effects of the water extract of Protaetia brevitarsis larva (PB) was investigated in carbon tetrachloride ($CCI_{4}$) treated male Sprague-Dawley rats. PB administration protected rats against ALT, AST and LDH elevations induced by $CCI_{4}$, as well as the severity of liver damage. PB recovered the decrease in serum level of high-density lipoprotein-cholesterol and the increase in serum level of low-density lipoprotein-cholesterol induced by $CCI_{4}$. In histopathological observation, massive fatty change and necrosis in the centrilobular area, degenerative change including pyknosis of nucleus and swelling of parenchymal cell induced by $CCI_{4}$ were clearly protected by PB. These histopathological findings paralleled with the serum biochemical results. The present results demonstrated that the water extract of PB may have the hepatoprotective effect against $CCI_{4}$-induced liver damage in vivo.

Influence of Protaetia brevitarsis Extract on Liver Damage Induced by Carbon Tetrachloride and Ethanol in Rats (에탄올 및 사염화탄소의 긴 손상에 미치는 흰점박이꽃무지 추출물의 영향)

  • Protaetia brevitarsis has been utilized as an ingredient of the description for the treatment of patients with chronic hepatic diseases in oriental medicine. This study was attempted to investigate whether Protaetia brevitarsis extract(PBE) protects or modulates the liver injuries induced by carbon tetrachloride or ethanol in Sprageue-Dawley rate. The liver injuries of rats induced by the treatment of carbon tetrachloride or ethanol were manifested by the observation of the significant changes in liver weight, serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities, serum thiobarbituric acid reactive substances (TBARS), and microsomal detocification enzymes(cytochrome P_450), cytochrome b$_{5}$, and cytochrome b$_{5}$ reductase).The effect of PBF on the liver damage induced by the chemicals was evaluated with the extent modulated in change of biochemical parameters above. Exposure to ethanol alone resulted a significant change in the ration of liver per body weight, ALT activity, and microsomal detoxification enzymes (cytochrome P_450, cytochrome b$_{5}$, and cytochrome b$_{5}$ reductase), but did not significantly changes in the levels of serum AST activity and TBARS. Pretreatment coith PBE did not modulate the alteration of the ratio of liver to body weigth, and the activities of serum aminotransferascs (AST. ALT), TBARS, and micro somal detoxification enzyme (cytochrome p_450, cytochrome b$_{5}$,and cytochrome b$_{5}$ reductase. These result suggested that PBE has not appreciable therapeutic effect on carbon tetrachloride or ethanol induced hepatotoxicity.
