We investigated the effects of skin hydration and composition analysis of proteoglycan (chia seed polymer) produced from chia (Salvia hispanica) seed. The result showed that proteoglycan of chia seeds is composed of galactose (46.8 %), glucuronic acid (27.1 %), rhamnose (8.7 %), xylose (7.6 %), glucose (4.9 %), fructose (2.3 %), mannose (1.8 %), arabinose (0.9 %) and the amount of proteins contained is 31.3 mg/g with the constituent amino acid compositions (mg/g) of Asp (1.9), Glu (3.6), Ser (0.9), Gly (3.6), Thr (0.8), Arg (1.0), Ala (2.0), Tyr (0.4), Cys (4.8), Val (1.1), Phe (0.5), Ile (0.6), Leu (0.9). The molecular weight of the proteoglycan measured by GPC (Gel Permeation Chromatography) is the range of 100,000~250,000 Da and the average molecular weight is 170,000 Da. The moisturizing effects and trans-epidermal water loss (TEWL) of chia seed polymer in cosmetic products (O/W emulsion) were studied in vivo. Chia seed polymer showed good skin hydration effects when compared with sodium hyaluronate which is a common moisturizer. Taken all together, chia seed polymer should be a very useful cosmetic ingredient as a moisturizer and a protecting agent from various skin irritations.
Objective: The aim of this study is to elucidate the effects of transforming growth factor-${\beta}$ (TGF-${\beta}$)1 and L-ascorbic acid on proteoglycan synthesis, and the relationship between Sox9, proteoglycan, and TGF-${\beta}1$ in intervertebral disc cells. Methods: Human intervertebral disc tissue was sequentially digested to 0.2% pronase and 0.025% collagenase in DMEM/F-12 media and extracted cells were cultured in $37^{\circ}C$, 5% $CO_2$ incubator. When intervertebral disc cells were cultured with TGF-${\beta}1$ or L-ascorbic acid, the production level of sulfated glycosaminoglycan (sGAG) was estimated by dimethyl methyleneblue (DMMB) assay. The changes of Sox9 mRNA and protein levels via TGF-${\beta}1$ were detected by RT-PCR and Western blot analysis in each. Results: The amount of sGAG was increased with the lapse of time during incubation, and sGAG content of pellet cultured cells was much larger than monolayer culture. When primary cultured intervertebral disc cells in monolayer and pellet cultures were treated by TGF-${\beta}1$ 20 ng, sGAG content of experimental group was increased significantly compared to control group in both cultures. L-Ascorbic acid of serial concentrations (50-300 ug/ml) increased sGAG content of mono layer cultured intervertebral disc cells significantly in statistics. The co-treatment of TGF-${\beta}1$ and L-ascorbic acid increased more sGAG production than respective treatment. After treating with TGF-${\beta}1$, Sox9 mRNA and protein expression rates were significantly increased in disc cells compared with the control group. Conclusion: This study suggests that TGF-${\beta}1$ would increase sulfated glycosaminoglycan (sGAG) and other proteoglycans such as versican by elevating Sox9 mRNA and protein expressions in order.

Osteoarthritis (OA) is an age-related joint disease characterized by degradation of articular cartilage and its association with chronic pain. Purified Bee venom (PBV) has been traditionally used to the treatment for inflammatory diseases. The purpose of the current study is to examine whether PBV regulates the pro-inflammation against a mouse model of knee OA induced by papain. We studied the effect of PBV on papain-induced OA in the knee joints of mice. Mice were split into following groups: normal control, papain induced OA, OA treated with PBV, OA treated with meloxicam as positive control. Proteoglycan deposition was analyzed by safranin O-fast green staining and H&E staining. Papain injection significantly degraded the proteoglycan in OA mice at 42 days. Cartilage proteoglycan density was significantly higher in PBV treated OA group than those of the positive control groups. These results demonstrate that PBV efficiently suppresses pathological processes in an OA model. Thus, PBV could be a potential therapeutic strategy for the treatment of OA.

BACKGROUND: Glucosamine hydrochloride (GlcN HCl) has been shown to inhibit cell growth and matrix synthesis, but not with N-acetyl-glucosamine (GlcNAc) supplementation. This effect might be related to an inhibition of critical growth factors (GF), or to a different metabolization of the two glucosamine derivatives. The aim of the present study was to evaluate the synergy between GlcN HCl, GlcNAc, and GF on proliferation and cartilage matrix synthesis. METHOD: Bovine chondrocytes were cultivated in monolayers for 48 h and in three-dimensional (3D) chitosan scaffolds for 30 days in perfusion bioreactors. Serum-free (SF) medium was supplemented with either growth factors (GF) $TGF-{\beta}$ ($5ng\;mL^{-1}$) and IGF-I ($10ng\;mL^{-1}$), GlcN HCl or GlcNAc at 1mM each or both. Six groups were compared according to medium supplementation: (a) SF control; (b) SF + GlcN HCl; (c) SF + GlcNAc; (d) SF + GF; (e) SF + GF + GlcN HCl; and (f) SF + GF + GlcNAc. Cell proliferation, proteoglycan, collagen I (COL1), and collagen II (COL2) synthesis were evaluated. RESULTS: The two glucosamines showed opposite effects in monolayer culture: GlcN HCl significantly reduced proliferation and GlcNAc significantly augmented cellular metabolism. In the 30 days 3D culture, the GlcN HCl added to GF stimulated cell proliferation more than when compared to GF only, but the proteoglycan synthesis was smaller than GF. However, GlcNAc added to GF improved the cell proliferation and proteoglycan synthesis more than when compared to GF and GF/GlcN HCl. The synthesis of COL1 and COL2 was observed in all groups containing GF. CONCLUSION: GlcN HCl and GlcNAc increased cell growth and stimulated COL2 synthesis in long-time 3D culture. However, only GlcNAc added to GF improved proteoglycan synthesis.
Objectives: This study was carried out to investigate the protective effects of Jeungmiobi-tang on the articular cartilage injuries induced by monosodium iodoacetate in rats. Methods: Twenty four rats were divided into three groups. Rats of normal group (n=8) were injected with 0.1 ml physiological saline into both knee joint cavities. In the rats of control group (n=8) and Jeungmiobi-tang group (n=8), Arthritis was induced by injecting with 0.1 ml monosodium iodoacetate (5 mg/ml) into both knee joint cavities. After the experiment, Gross and histopathological examinations on the knee joint were performed. The content of proteoglycan in articular cartilage and TNF-α and IL-1β in synovial fluid were also analyzed. Results: Grossly, Injuries to the articular cartilage surface was observed weak in the Jeungmiobi-tang group compared to the control group. Proteoglycan content in the articular cartilage was significantly higher in the Jeungmiobi-tang group than in the control group. The chondrocyte score was significantly lower in the Jeungmiobi-tang group than in the control group. Conclusion: According to these results, that Jeungmiobi-tang has protective effects on the articular cartilage injuries induced by monosodium iodoacetate in rats.
$\beta$-Immunan, a proteoglycan, was isolated from the mycelium of Canoderma lucidum which belongs to a medicinal mushroom. The effects of $\beta$-Immunan on cell-cell and cell-matrix interactions mediated by carbohydrate-recognition and the mechanism responsible for the inhibition of experimental metastasis of Bl6-F10 and B16/BL6 murine melanoma were studied. The results showed that $\beta$-Immunan inhibited Bl6-Fl melanoma cell's adhesion to laminin and asialofetuin-induced homotypic aggregation and reduced invasion against Bl6-F10 murine melanoma cells through matrigel in vivo assay. When $\beta$-Immunan was intraperitoneally administrated to C57B/6 mice bearing B16/BL6 murine melanoma cells, it was decreased the number of pulmonary metastatic colony by the dose dependent manner ranging from 20 to 100 mg/kg/day. The results indirectly indicate that clinical treatment with $\beta$-Immunan might be expected to exhibit anti-metastatic effect. In the pulmonary metastasis, the number of pulmonary metastatic colony of melanoma when $\beta$-Immunan was intraperitoneally administrated to C57BL/6 mice bearing B16/BL6 murine melanoma cells by intravenous injection were decreased by the dose dependent manner ranging from 20 to 100 mg/kg/day.
So as to evaluate its use an immune stimulator in humans, the toxicity and action against immune cells by an anti-yeast substance (AYS), a bacterial proteoglycan, were investigated. The AYS did not possess hemolytic activity with human red blood cells (hRBC). Nor did it exhibit cytotoxicity against human peripheral blood mononuclear cells (hPBMC). In addition, the AYS did not induce hPBMC proliferation, but it did agglutinate hPBMCs in vitro. Moreover, hPBMC induced inflammatory cytokines such as IL-6, IL-5, IFN-${\gamma}$ and TNF-${\alpha}$ with the AYS during culture. Compared with alum, the AYS as an adjuvant has an increased antibody production rate against bovine serum albumin (BSA) in mice.
Objectives This study was aimed to offer basic data in long term drug efficacy test using monosodium iodoacetate-induced osteoarthrits model. Methods Sixty male rats were divided into normal and osteoarthritic group. Rats of normal group were injected with 0.1 ml physiological saline, and rats of osteoarthritic group were inected with 0.1 ml monosodium iodoacetate (3 mg/ml) into each left and right knee joint cavities. Gross examination, proteoglycan contents and histopathological examination on the knee joint were performed at 10, 20, 40, and 60 days after injection. Results Grossly, degenerative changes at 10 days, desquamation at 40 days, and ulceration of articular cartilages at 60 days were observed. Proteoglycan contents in articular cartilages were decreased rapidly to 40 days, after than decreased gradually. Osteoarthritic scores were increased rapidly to 20 days, after than increased gradually to 60 days. Conclusions From above results, osteoarthritis model induced by a single intra-articular injection of monosodium iodoacetete is useful model for long term drug efficacy test.
The interactions between the host and microbial pathogen largely dictate the onset, progression, and outcome of infectious diseases. Pathogens subvert host components to promote their pathogenesis and, among these, cell surface heparan sulfate proteoglycans are exploited by many pathogens for their initial attachment and subsequent cellular entry. The ability to interact with heparan sulfate proteoglycans is widespread among viruses, bacteria, and parasites. Certain pathogens also use heparan sulfate proteoglycans to evade host defense mechanisms. These findings suggest that heparan sulfate proteoglycans are critical in microbial pathogenesis, and that heparan sulfate proteoglycan-pathogen interactions are potential targets for novel prophylactic and therapeutic approaches.
Rat hepatocytes were isolated and cultured on the petri dishes treated with various coating materials. Untreated or collagen coated petri dish gave monolayer culture of hepatocyte and proteoglycan, dermatan sulfate, and BSA treated petri dish gave hemispheroid. The untreated Primaria petri dish gave spheroid type of hepatocyte, and heparin and hyaluronic acid treatment gave multilayers. To sustain high cell viability, monolayer cultured hepatocytes was more useful, while it was found that the hemispheroid or spheroid type hepatocytes was more active in the hepatic functions such as ammonia metabolism and albumin synthesis.
