Study on Antioxidant and Cytotoxic Activities in Ethanol Extract from Prunus mume (오매 에탄올 추출물의 항산화 활성 및 항암 활성 연구)

  • Prunus mume is well known contain many functional materials that play beneficial roles in the human body. Studies have found that many organic acids and polyphenol compounds exist in Prunus mume. In this study, content of total polyphenols and flavonoids, antioxidative activity and cytotoxicity of ethanol extract from Prunus mume were measured Contents of total polyphenolic and total flavonoid compounds in ethanol extract from Prunus mume were 16.92 mg/g and 59.95 mg/g, respectively. The $IC_{50}$ of ethanol extract from Prunus mume were $237.72 {\mu}g$/assay and $239.58 {\mu}g$/assay by DPPH and ABTS radical cation scavenging test, respectively. Additionally, ferric ion reducing antioxidant power (FRAP) value of ethanol extract from Prunus mume was 0.94 mM ($FeSO_4$ eq.) by $800 {\mu}g$/assay. Cytotoxicity of Prunus mume against five kinds of cancer cell lines increased as the extract concentration increased Especially, cytotoxicity of the ethanol extract against A-549 (lung cancer line) was higher than that against any other cancer cell line by both MTT and SRB assay. These results show that ethanol extract of Prunus mume has considerably high antioxidative and cytotoxic activities.

Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium (오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours.   • Objectives : Prunus mume has been used for the folk medicine from old times. The purpose of the study was to investigate the antimicrobial activity of prunus mume extract to various oral microbes. Methods : This study was carried out to examine the antimicrobial effect of Prunus mume extract against oral microbes. Data were collected using a Dentocult SM Strip mutans and Dentocult LB Strip mutans from April 5 to May 4, 2013. A total of 36 experimental and 32 control group were selected for this study. Results : The MIC of Prunus mume extract was tested for 0.39% in S. mutans, S. salivarius and S. auerus, 0.78% in S. mitis, S. equi and E. coli. In vivo, experimental group showed significantly the lower Streptococcus mutans levels by the use of the gum contained Prunus mume extract from 15 days compared with control group(p=0.012). The reduction was more significant in Lactobacilli level of the experimental group than the control group(p=0.022). Conclusions : These findings suggest that the oral products containing Prunus mume extracts is effective in preventing oral diseases.

Effect of Ethyl Acetate Extract of the Dried Prunus mume on the Growth of Staphylococcus aureus (오매의 에틸아세테이트 추출물이 Staphylococcus aureus의 증식에 미치는 영향)

  • The dried Prunus mume, an alkaline food abundant in organic acids (citric acid, malic acid and tartaric acid), has been largely used in both folklore remedies and Chinese herbal medicine for a long time. This study was performed to investigate the antimicrobial activity of the dried Prunus mume. The fractionation of the methanol extracts from Prunus mume was conducted using petroleum ether, chloroform, ethyl acetate and butanol, respectively. The antimicrobial activity of the Prunus mume extracts was then determined against food-borne pathogens using a paper disc method. The ethyl acetate extract showed the strongest antimicrobial activity against the eigth food-born pathogens used in this present study. Diaion HP 20 column chromatography was performed to remove some sugars that might inhibit the antimicrobial activity of Prunus mume. The strongest antimicrobial activity of ethyl acetate fraction of Prunus mume was shown against Staphylococus aureus. The growth inhibition curve was determined using ethyl acetate extracts of Prunus mume against Staphylococus aureus, which showed the growth inhibition up to 72 hours at 1,000 ppm concentration.

Preparation of Chlorella Drinks and Its Quality Characteristics (클로렐라 음료의 제조와 그 품질 특성)

  • This study was performed to evaluate the quality characteristics of the drink with chlorella and Prunus mume. The chlorella drinks were prepared in four different ratios such as chlorella extract 5%, 10% and Prunus mume extract 0.2%, 0.4% added in chlorella extract 5%. In the color of the chlorella drinks, chlorella extract 10% had lower Hunter's color values(L, a, b) than chlorella extract 5%. The hunter's L(lightness) and a(redness) values were lower for Prunus mume extract 0.4% compared to Prunus mume extract 0.2%, however, there was no difference in the b(yellowness) values. A safety test showed that the drinks are clear from heavy metals and total bacteria and colifom bacteria. In the sensory test of the drinks, the sensory scores(color, taste, flavor, overall acceptability) show the best result on chlorella extract 5%(with Prunus mume extract 0.4% added), followed by chlorella 5%(with Prunus mume extract 0.2% added). The score for chlorella extract 10% was the lowest.

Antioxidative Activity of the Extracts of Japanese Apricot (Prunus mume Siebe. et Zucc.)

  • In order to discern the possibility of functional food product or ingredient of a new medicine, the leaf parts and fruit parts of Prunus mume was partitioned with various solvents and their antioxidative activity was measured. When the antioxidative activity of MeOH extracts of leaf parts and fruit parts of Korea and China was compared, all of them showed the highest antioxidative activity in EtOAc fraction. In case of Korean Prunus mume leaf parts showed that quantity required for $RC_{50}$ to be $27.04{\mu}g$ in EtOAc fraction and in case of China Prunus mume leaf parts, it was $23.31\;{\mu}g$ which is similar to that of ${\alpha}$-tocopherol ($22.14\;{\mu}g$) and showed the highest activation. In case of Prunus mume fruit parts MeOH extract, Korean fruit showed $29.16\;{\mu}g$, and Chinese fruit showed $31.21\;{\mu}g$ in EtOAc fraction and thus Korean fruit extract showed a higher activity of antioxidant than the Chines fruit extract. When the antioxidative activity between the fruit parts and leaf parts of Prunus mume was compared, the leaf parts showed a higher antioxidative activity.

Effect of Prunus mume Extract on Gastric Secretion in Rats and Carbon Tetrachloride Induced Liver Damage of Rabbits (매실추출물이 흰쥐의 위액분비 및 사염화탄소로 유발시킨 가토의 간장장애에 미치는 영향)

  • The aim of this study is to investigate effects of Prunus mume extract on gastric secretion in rats and experimentally carbon tetrachloride-induced liver damage of rabbits. The groups of Prunus mume extract showed excellent increasing effect in total acidity and peptic activity and decreasing effect on gastric volume than the control group whereas clear differences were not found between sample adminstered group and control group in gastric pH. In liver function test Prunus mume extract exhibited rapid recuperation of liver function that is sGPT activity showed an apparant decreasing effect from the 6th day total cholesterol and alkaline phosphatase level from the 10th day respectively compared to the control group in carbon tetrachloride-intoxicated rabbits. It is, is suggested that Pruns mume extract can be used as the stomachics to promote the gastric digestion by increasing the peptic activity and health food to shorten the recovery time from liver diseases.

Effect of Prunus mume Extract on Dongchimi Fermentation (매실농축액의 첨가가 동치미 발효 숙성에 미치는 영향)

  • The physicochemical and microbiological studies were conducted to examine the effect of Prunus mume extract on the Dongchimi fermentation. Dongchimi with the addition of various levels (0, 0.05, 01, 0.2 or 0.3%) of Prunus mume extract was fermented at $10^{\circ}C$ for 30 days. The pH of the product decreased in the course of fermentation and it was between 3.8~4.2 at 6~8 days fermentation. Total acidity was decreased by the addition of Prunus mume extract. A more rapid decrease in pH and an increase in total acidity were observed in the control Dongchimi than in that with added Prunus mume extract. The content of vitamin C increased for up to 10 days of fermentation and then gradually decreased. Reducing sugar content increased for up to 10 days, and the highest of sugar content was shown in the Dongchimi with 0.3% of added Prunus mume extract. As Prunus mume extract levels increased, the total viable cell number and the Lactobacillus cell number decreased. Dongchitni in which Prunus mume extract was added gained it,j optimum taste after 10 days of fermentation. The results of this study indicate that Prunus mume extract causes a delay in the fermentation of Dongchimi by slowing down pH drop and inhibiting the Lactobacillus cell growth.

Effects of Ethanol Extract of Prunus mume on the Antioxidative System and Lipid Peroxidation on Ethanol-Induced Hepatotoxicity in Rat Liver (매실 추출물이 알코올 투여 흰쥐의 항산화계 및 지질과산화에 미치는 영향)

  • To investigate the antioxidative effects of Prunus mume ethanol extract on the ethanol-induced hepatotoxicity in rat liver, Sprague-Dawley rats weighing 120∼160 g were divided into 5 groups; normal group(NOR), Prunus mume ethanol extract 200mg/kg treated group(PME), ethanol(10 mL/kg, 35%) treated group(ETH), Prunus mume ethanol extract 200 mg/kg and ethanol treated group (PML) and Prunus mume ethanol extract 400 mg/kg, and ethanol treated group(PMF), respectively. The antioxidative activity in vitro was reduced in order of EtOAC>n-hexane>water> chloroform fraction. The growth rate and feed efficiency ratio decreased by ethanol administration were gradually increased to the adjacent level of NOR by administering Prunus mume ethanol extract. It was observed that activities of catalase, superoxide dismutase(SOD), xanthine oxidase and glutathione peroxidase(GSH-Px) of liver and alanine aminotransferase(ALT) and asparate aminotransferase(AST) of serum were elevated by ethanol administration. Besides, Prunus mume ethanol extract markedly decreased elevated activites of catalase, GSH-Px, ALT and AST, except in activites of SOD and xanthine oxidase compared to ETH. Also, the depleted content of GSH by ethanol was increased similar to NOR level by administering Prunus mume ethanol extract. These results suggested that Prunus mume ethanol extract has a possible protective effect on the ethanol-induced hepatotoxicity in rat liver.

Effect of Prunus mume Extract Containing Beverages on the Proliferation of Food-borne Pathogens (매실 추출물을 함유한 음료가 식중독 유발균의 성장에 미치는 영향)

  • Prunus mume has been used for the folk medicine by many old civilizations to treat food-borne diseases or enteric disorders. The purpose of this study was to investigate the antimicrobial activity of beverages containing Prunus mume extracts against Staphylococcus aureus, Shigella dysenteriae, Shigella flexneri, Shigella sonnei, Escherichia coli and Pseudomonas aeruginosa. Seven different types of Prunus mume extracts containing beverages have been prepared for the test in which minimum inhibitory concentration for each microorganism has been determined by serial dilution method using either TSA or TSB medium. Pseudomonas aeruginosa showed least resistance and 0.3g/$m\ell$ concentrations of 5% Prunus mume extracts containing beverage had antimicrobial property against the organism. Beverages containing more than 15% of Prunus mume extracts showed antimicrobial activity against all tested microorganisms at the MIC value of less than 0.25g/$m\ell$. The effect of Prunus mume on the growth of food-borne pathogens has been also studied using a spectrophotometer. In the growth assay, each of the Prunus mume extracts containing beverage was added to the medium at the concentration of 25% (v/v). Beverage containing 20% Prunus mume extracts showed inhibitory effect on the growth of all tested microorganisms.
