Control Effects of Microbial Products on Pythium Blight, Brown Patch and Dollar Spot of Creeping Bentgrass (Creeping Bentgrass에서 미생물제에 의한 Pythium Blight, Brown Patch 및 Dollar Spot 방제 효과)

  • 길항곰팡이 3종(Aspergillus sp. A101, Penicillium sp. B202, Trichoderma sp. C303), 길항세균 3종(Arthrobacter sp. AN303)또는 이들 모두의 혼합균주를 배양하여 creeping bentgrass 양묘장에 처리하여 Pythium blight, brown patch 및 dollar spot의 방제효과를 조사하였다. 또한 길항미생물 6종의 배양액을 살균제와 병용하여 그린에 살포하여 이들 토양병에 대한 방제효과를 조사하였다. 양묘장에서는 미생물의 종류에 관계없이 Pythium blight 억제효과가 커 3회 이상 미생물제 처리시 병이 전혀 발생하지 않았다. 그러나 brown patch와 dollar spot은 미생물제에 의한 방제효과가 크지 않았으며, 살균제에 의해 효과적으로 방제되었다. 미생물제와 살균(Pythium blight 방제용 살균제 제외)를 병용하여 처리한 그린에서는 살균제 단독처리와 비교할 때 brown patch는 유의적으로 억제되었고 Pythium blight와 dollar spot은 차이가 없었다.

Effect of a Microbial Product on the Control of Soilborne Diseases of Turfgrasses (미생물제에 의한 잔디의 토양전염병 방제 효과)

  • A microbial product composed of three antagonistic fungal isolates (Aspergillus sp., Penicillium sp. and Trichoderma sp.) and three bacterial isolates (Arthrobacter sp., Bacillus sp., and Pseudomonas sp.) was tested for the control of Pythium blight caused by Pythium sp., brown patch by Rhizoctonia solani (anastomosis group(AG) 1-1) and large patch by R. solani (AG 2-2) of turfgrasses. Cultures of the antagonistic fungi and bacteria varied in the effectiveness in reducing disease severity of Pytium blight and brown patch on bentgrass. The antagonistic fungal and bacterial isolates were mixed and cultured at 20-$25^{\circ}C$ for 3 days in a growth medium, and the diluted solution of the microbial culture was applied under the field conditions after inoculation of the above turfgrass pathogens. The treated turfgrass was incubated at 28$^{\circ}C$ in a growth chamber. In this experiment, Pythium blight was almost completely controlled and brown patch was slightly decreased by the microbial product, while no control was observed in large patch of zoysiagrass. In zoysiagrass treated with the microbial culture, thatch accumulation was notably reduced.

  • The rose is one of the most important commercial cut flowers in the world. Recently stem blight symptoms have often found in hydroponics and soil cultured roses. These symptoms are appeared by four diseases; gray mold caused by Botrytis cinerea, common canker by Coniothyrium fuckelii, anthracnose by Colletotrichum gloeosporioides, and Pythium root rot by Pythium sp. Each symptom and its causal pathogen is described.

Occurrence of severe soybean-sprout rot caused by Pythium deliense in the recirculated production system

  • Severe soybean-sprout rot was found at the mass productive factory in 2000 and 2001 and it caused 10-20% loss of the production. Pythium sp. was isolated almost 90% by potato dextrose agar from rotted root and hypocotylsof the sprouts. And the pathogencity tests using test tubes with 2% water agar and small containers (30 ${\times}$ 30 ${\times}$ 50 cm, WxLxH) cultivation were shown a similar rot on roots and hypocotyls. The fungal mycelium grew rapidly on the water agar and it prevented the seed germination. Density of the Pythium sp. in the recycled water system at the factory was periodically measured using a selective medium, corn meal agar with Pimaricin 10 mg, Rifampicin 10 mg, Ampicillin 100 mg per 1 liter in order to check the contamination of recycled water. After fitering step using 5 and 1 ml in the recycled system was applied and it was effectively controlled Pythium rot. The daily yield of sprout was stable and the occurrenceof Pythium in the recycled water was much less after filtering. The fungal isolates were identified as Pythium deliense Meurs based on various mycological characteristics on corn meal agar and sucrose-asparagine bentgrass leaf culture medium. P. deliens oogonia were spherical, smooth, 19-23 urn in diameter, and their stalk bending toward antheridia. Antheridia were straw hat-shaped, curred club-shaped, therminal or intercalary, monoclinous, occasionally diclinous, 12∼15 ${\times}$ 8∼11 um, 1(∼2) per oogonium.

Isolation, Cultivation, and Antifungal Activity of a Lichen-Forming Fungus

  • A lichen-forming fungus was successfully isolated by discharged spore method from Korean lichen (Hetero-dermia sp.) and cultivated in pure culture. The isolate JR0012 inhibited mycelial growth of several plant-pathogenic fungi. Mycelial growth of the four Pythium spp. tested was completely inhibited. Potato dextrose broth was found to be the medium favorable for large-scale production of antibiotics from the isolate. Anti-fungal substances produced in axenic culture were partially purified. This is the first report in Korea of lichen-forming fungus successfully isolated and which exhibited strong antifungal activity against plant-pathogenic fungi, especially the four Pythium spp..

  • This study was carried out to isolate of antagonistic bacterium against Pythium ultimum and Rhizoctonia solani AG-4, causal pathogens of vegetables damping-off. Total of 600 strains were isolated from soil and plait roots. The isolates were screened for antagonism against Pythium ultimum and Rhizoctonia solani AG-4. One strain, named YJ-37, was sellected for detained study among those microoganisms screened. It was identified as Bacillus ehimensis based on morphological and physiological characterisitics according to the Bergey's mannual of systematic bacteriology, Sherlock system of Microbial ID Inc. and 16S rDNA sequences methods. Furthermore Bacillus ehimensis YJ-37 showed antifungal activities against Alternaria altrata, Collectotrichum gloeosporioides, Didymella bryoniae, Fusarium moniliforme, Fusarium oxysporum, F. oxysporum cucumerinum, F. oxysporum niveum, Gloeosporium sp., Glomerella sp., G. cingulata, G. lagenaria, Penicillium digitatum, P. italicum, Phytophthora capsici, Sclerotinia sclerotiorum, and Stemprhylium solani.

  • The effect of calcium cyanamide (China-made) soil fumigation on the growth of the ginseng pathogen Rhizoctonia, Pythium), soil microbes, and seed germination of lettuce and radish was investigated. At twice the recommended level (2S0-ppm $CaCN_2$), the growth of Rhizoctonia and Pythium, and the seed germination, were not inhibited. Rather, the effective level was 10,000 ppm. The powder form was more effective than the granular form in inhibiting pathogen growth and seed germination. The lettuce seed was also more sensitive than the radish seed. Calcium cyanamide appearedto decrease the fungi population and to increase Actinomycetes in the soil.

  • The mycelial growth of Pythium graminicola, P. ultimum, P. rostratum was best at -3 osmotic potential. P. monospermum was best grown at -8 bars osmotic potential. Mycelial growth by Pythium spp. was prevented at -42 bars for P. graminicola, P. ultimum and P. monospermum. Sporangial formation by P. graminicola and P. ultimum was influenced by water potential in much the same way as mycelial growth: -3 bars was maximum and -42 bars or below was minimal. Pythium spp. tested was grown faster on PDA than other media including oat meal agar. P. ultimum was best grown on any media among Pythium spp. tested.

Root Rot of Codonopsis pilosula Caused by Pythium myriotylum (Pythium myriotylum에 의한 만삼 뿌리썩음병)

  • A Pythium species was isolated from roots of Codonopsis pilosula showing wilt symptoms in a field at Yonchon, Kyonggido of Korea from late June to early August in 2000. The Pythium species was identified as Pythium myriotylum Drechsler based on various mycological characteristics. The isolate was strongly pathogenic when inoculated to root of C. pilosula plants in pot. The inoculated plants showed typical symptoms of root rot, resulting in reduced growth of root and consequently wilting of above ground part of plants. Pythium root rot of C. pilosula caused by P. myriotylum has not been reported previously in Korea.
