Fabrication and Application of Micro Polymer Chip Platform for Rare Cell Sample Preparation (희귀 세포 샘플 준비를 위한 마이크로 폴리머 칩 플랫폼 제작 및 활용)

  • In this paper, a new micro polymer chip platform and protocol were developed for rare cell sample preparation. The proposed platform and protocol overcome the current limitation of the dilution method which is based on statistics and the FACS method which expensive and requires fluorescence staining. It allows collecting exact number of target cells simply and selectively because the cells are visually confirmed during the collecting process. The collected cells can be transported or spiked into a desired locations, such as a microchamber, without cell loss. This research may applicable not only to a rare cell sample preparation for Lab on a Chip cancer diagnosis, but also to a single/double/multiple cell sample preparation for a cell analysis field. To verify this platform and protocol, five human breast cancer cells (MCF-7) were collected and transported into a hemocytometer chamber.

Jackknifed Cochran-Mantel-Haenszel Test for Conditional Independence in Sparse $2\tims2\tims$K Tables

  • We are interested in the conditional independence in sparse $2\tims2\tims$K tables with very rare cell counts. The most popular test is Cochran-Mantel-Haenszel statistic when sample sizes are moderately large enough to guarantee the chi-square approximation. We will consider jackknifing the CMH test and also suggest an approximate normal distribution for the standardized jackknifed CMH statistic. The main focus of this paper is to improve the chi-squared approximation to the CMH test by using the asymptotic normality of the jackknifed CMH test when sample sizes are very sparse but K and N$\infty$. The performance of the proposed jackknifed test, in the sense of significance level control and power, will be compared with that of the CMH test through a Monte Carlo study.

Basal cell carcinoma misdiagnosed as trichoepithelioma

  • Trichoepithelioma is a benign hair follicle tumor that can undergo malignant transformation into basal cell carcinoma in rare cases. Due to the similar clinical and histological features of trichoepithelioma and basal cell carcinoma, distinguishing between these types of tumors can be a diagnostic challenge. Punch biopsy obtains only a small sample of the entire lesion, and thus inherently involves a risk of misdiagnosis between histologically similar diseases. Therefore, if the possibility of misdiagnosis can reasonably be suspected, clinicians should conduct an excisional biopsy or immunohistochemical staining (e.g., CD10 and Bcl-2) to ensure an exact diagnosis. Although trichoepithelioma is benign, the surgical excision of solitary trichoepithelioma should be considered in order to avoid the possibility of malignant transformation, which has occasionally been documented for multiple familial trichoepitheliomas. Herein, we report a case that was initially misdiagnosed as trichoepithelioma before ultimately being diagnosed as basal cell carcinoma through excision and immunohistochemical staining.

Common Docking Domain Mutation E322K of the ERK2 Gene is Infrequent in Oral Squamous Cell Carcinomas

  • Background: Mutations in the MAPK (Mitogen Activated Protein Kinase) signaling pathway - EGFR/Ras/RAF/MEK have been associated with the development of several carcinomas. ERK2, a downstream target of the MAPK pathway and a founding member of the MAPK family is activated by cellular signals emanating at the cell membrane. Activated ERK2 translocates into the nucleus to transactivate genes that promote cell proliferation. MKP - a dual specific phosphatase - interacts with activated ERK2 via the common docking (CD) domain of the later to inactivate (dephosphorylate) and effectively terminate further cell proliferation. A constitutively active form of ERK2 carrying a single point mutation - E322K in its CD domain, was earlier reported by our laboratory. In the present study, we investigated the prevalence of this CD domain E322K mutation in 88 well differentiated OSCC tissue samples. Materials and Method: Genomic DNA specimens isolated from 88 oral squamous cell carcinoma tissue samples were amplified with primers flanking the CD domain of the ERK2 gene. Subsequently, PCR amplicons were gel purified and subjected to direct sequencing to screen for mutations. Results: Direct sequencing of eighty eight OSCC samples identified an E322K CD domain mutation in only one (1.1%) OSCC sample. Conclusions: Our result indicates that mutation in the CD domain of ERK2 is rare in OSCC patients, which suggests the role of genetic alterations in other mitogenic genes in the development of carcinoma in the rest of the patients. Nevertheless, the finding is clinically significant, as the relatively rare prevalence of the E322K mutation in OSCC suggests that ERK2, being a common end point signal in the multi-hierarchical mitogen activated signaling pathway may be explored as a viable drug target in the treatment of OSCC.

The Effects of Sochungyong-tang plus Okbyeong-san Extract on Allergic Rhinitis of Rats (소청룡탕합옥병풍산(小靑龍湯合玉屛風散)이 백서(白鼠)의 알레르기 비염(鼻炎)에 미치는 효과)

  • Background and Objectives : Although the incidence of allergic rhinitis has increased specific therapeutics with a certain medicine is not well developed yet. In therapeutics of allergic rhinitis, limited medicines just have been used in the clinin. Sochungyong-tang has been a widespread medicine for allergic rhinitis. However, okbyeongpung-san has been taken for raising immunity. In medical treatment of allergic rhinitis, raising immunity is a necessary part of the process. In this reason, this study aimed to find out therapeutic effects of combination of sochungyoung-tang and okbyeongpung-san extract on allergic rhinintis of nasal tissue in rats. Materials and Methods : The Sprague-Dawley male rats, about 200g of body weight, were divided into three groups: the normal group, the control groups, and the sample groups. With the Levine and Vaz method, the allergic reaction was induced with ovalbumin as antigen in the intraperitoneal route. Rats of the sample groups were treated with 2cc of sochungyoung-tang plus okbyeongpung-san extract once a day for 28days. I observed historical changes in nasal tissue. I also found changes in the segment of lympocyte and neutrophil of Leukocyte and erythrocyte. I used the statistical methods of post Hoc test(p<0.05) Results : The increase of the goblet cell and mucous secretion in the sample groups was rare when compared to the control groups. Effects of sochungyoung-tang plus okbyeongpung-san extract on the liver function were also studied in rats. Treatment of sochungyoung-tang plus okbyeongpung-san extract did not affect GOT and GPT. The number of lymphocyte was decreased, while the number of erythrocyte was increased in sample groups. Conclusion : This study shows that sochungyoung-tang plus okbyeongpung-san plus okbyeongpung-san extract has considerable effects on allergic rhinitis.

Comparative Analysis of Screening Results from Various ELISA Formats Used for Detection of Anti-Erythropoietin Antibodies in Korean Patients

  • Clinical cases of pure red cell aplasia (PRCA) have been reported during the recombinant human erythropoietin (EPO) therapy for the anemia patients. PRCA is a rare hematological disorder leading to a severe anemia due to an almost complete stop of red blood cell production. Antibody (Ab)-associated PRCA is caused by the EPO-neutralizing Abs that eliminate the biological activity of EPO. In order to detect anti-EPO Abs in human sera, we performed conventional ELISA, directly coated bridging ELISA, and streptavidin coated bridging ELISA, and compared their sensitivity and specificity. Some false positive results were obtained in the conventional ELISA. One positive sample was detected successfully by streptavidin coated bridging ELISA, which was not appeared in the directly coated bridging ELISA. In conclusion, streptavidin coated bridging ELISA was substantially sensitive and specific format and one out of sixty-eight serum samples was proved to be anti-EPO positive.

Exploratory Investigation of Genetic Associations with Basal Cell Carcinoma Risk: Genome-Wide Association Study in Jeju Island, Korea

  • Aim: Little is known about the genetic associations with Basal cell carcinoma (BCC) risk in non-Caucasian populations, in which BCC is rare, as in Korea. We here conducted a pilot genome-wide association study (GWAS) in 12 patients and 48 standard controls. Method: A total of 263,511 SNPs were analyzed with the Illumina HumanOmni1 Quad v1.0 DNA Analysis BeadChip for cases and Korean HapMap 570K for controls. Results: SNP-based analyses, based on the allele genetic model with adjustment for sex and age showed suggestive associations with BCC risk for 6 SNPs with a P-value (P < 0.0005). However, these associations were not statistically significant after Bonferroni correction: rs1040503, rs2216491, rs13407683, rs4751072, rs9891263, and rs1368474. In addition, results from gene-based analyses showed suggestive associations with BCC risk for 33 candidate genes with a P-value (P <0.0005). Consistent with previous GWAS and replication studies in Caucasian populations, PADI6, RHOU and SLC45A2 were identified as having null associations with BCC (P > 0.05), likely due to the smaller sample size. Conclusions: Although this was a small-scale negative study, to our knowledge, we have conducted the first GWAS for BCC risk in an Asian population. Further large studies in non-Caucasian populations are required to achieve statistical significance and confirm these findings.

IMMUNOHISTOCHEMICAL STUDY ON EXPRESSION PATTERNS OF TUMOR GROWTH RELATED FACTORS IN SALIVARY GLAND TUMORS (타액선 종양에서 종양증식 관련인자 발현에 관한 면역조직화학적 연구)

  • Objective : Lots of papers have revealed that tumor growth related factors such as EGF, EGFR, c-erbB-2 play an important role in tumorigenesis and proliferation. These factors are found in most tumors of ectodermal origin. But, documentations of tumor growth related factors on salivary gland tumors were rare. Therefore, we determined expressions of tumor growth related factors; PCNA, p53, EGF, EGFR, cerbB2(HER-2), Maspin, DMBT-1, N-Ras in representative salivary gland tumors. Materials and methods : A few types of salivary tumors were examined by immunohistochemical assays. Each antibody was applied to specimens of tumors. Specimens were composed of 5 pleomorphic adenomas (PA), 3 mucoepidermoid carcinomas (MEC), 2 adenoid cystic carcinomas (ACC) and 2 squamous cell carcinomas (SCC) from 12 patients. One specimen was selected randomly as negative control. For evaluation of staining intensity, each stained sample was divided into 5 grade; no staining, obscure, weak staining, moderate staining, strong staining. Results : Strong expressions of PCNA were found in all tumors except of PA. EGF was expressed strongly in SCC, ACC sequently. But in both PA and MEC, EGF expression was weak. EGFR and c-erbB-2 expression showed similar patterns in all salivary gland tumor tissues. P53 showed weak expression generally in all salivary gland tumors. DMBT-1 was expressed in SCC rather than in ACC or in MEC. N-Ras showed weak expressions in all salivary gland tumors except of squamous cell carcinoma. Conclusion : Taken together, tumor growth related factors were expressed in salivay tumors as well as mucosal squamous cell carcinoma. Especially EGFR and c-erbB-2 could be candidates as diagnostic markers for estimating clinical grade of salivary gland tumors. But further studies with reliable methods will be needed to confirm the results of this study.

A case of polypoid cystitis in a dog

  • Among benign proliferation of the urinary bladder, polypoid cystitis is a rare disease in dogs. It is characterized by epithelial proliferation, chronic inflammation in lamina propria, and development of a polypoid mass or masses without evidence of neoplasia. This report describes histopathologic features of polypoid cystitis in dog. A 10-year-old spayed female shihtzu-dog was presented with two-month history of hematuria. Abdominal ultrasonography confirmed the thickened bladder wall and calculi in both kidneys. Surgical biopsy sample was taken from the thickened bladder mucosa for the histopathologic examination. The mass was covered with irregular hyperplastic transitional epithelium with the projection into the lumen in multifocal areas as well as many Brunn's nests in lamina propria. Many inflammatory cells such as lymphocyte, plasma cell, and macrophage and few neutrophils were occupied in lamina propria and submucosa. Proliferated fibrous tissues in lamina propria were clarified by using special staining methods. These collagens were stained blue with Masson's trichrome and red with van Gieson, but negative for alcian blue. Based on the clinical, gross, and histopathologic examinations, this case was diagnosed as polypoid cystitis in a dog. In our best knowledge, this is the first report of polypoid cystitis in dog in Korea.

New record of Pleuronema marinum Dujardin, 1841 (Protozoa, Ciliophora) from South Korea

  • During a field survey of Korean marine ciliates, we collected Pleuronema marinum from a brackish water sample. It is characterized by the presence of a contractile vacuole in mid-body, rather than the subterminal/ terminal contractile vacuole as in other congeners. The cells were examined in vivo and based on protargol and 'wet' silver nitrate impregnation. In addition, the nuclear 18S rRNA gene was sequenced using a single cell. The Korean population morphologically and molecularly resembles a Chinese population of P. marinum. Historical review of the species concludes that 1) two or more species have been assigned into P. marinum, 2) the position of contractile vacuole (e.g., in mid-body) is a valid character state, and 3) P. marinum is probably a rare species. Here we provide a monographic treatment of P. marinum to clarify the issue and for further studies of relevant species. Considering there are about 40 nominal species and complex nomenclatural acts in the genus Pleuronema, further studies should provide descriptions based on protargol and 'wet' silver impregnation with marker gene(s).
