Extract Yields and Saponin Contents of Red Ginseng Extracts prepared with various Concentrations of Ethanol (홍삼의 알콜 농도별 추출 엑스의 수율과 사포닌 함량)

  • Red ginseng extracts were prepared with various concentrations of ethanol. Extract yields were examined and saponins in the extracts were identified and determined by TLC and HPLC, respectively. Yields of the extracts, $19.7{\sim]50.3%$, were the highest in water extract and showed significant decrease with the increase of ethanol concentration used for extraction. Contrary to the extract yields, saponin yields from red ginseng were conspicuously increased with the increase of ethanol concentration and were $3.47{\sim}5.13%$ of crude saponins and $1.28{\sim}1.93%$ of six major ginsenosides. Saponin contents in the red ginseng extracts were $6.9{\sim}24.2%$, of crude saponin and $2.57{\sim}9.22%$, of six major ginsenosides.

  • Red ginseng tails were extracted with ethanol solutions over a range of concentrations and temperature conditions. Investigations were carried out to study the effects of treatments on yields, soluble solids, saponin and precipitate occured in red ginseng extract beverage during storage. It was found that: (1) Higher concentration of ethanol at low temperature resulted in less yield of crude extract (2) The amount of precipitate in the non-purified extract beverage were less with decrease in ethanol concentration used (3) The treatment for purification of extracts and storage of purified extract at 37$^{\circ}C$ for 6 months had no effect on HPLC chromatogram pattern of saponins (4) The amount of purified extract decreased by purification treatment and more decrease was found as the temperature and concentration of ethanol increased. For Preparation of red ginseng extract beverage, the treatment of extracts with ethanol at low temperature was found to be more effective to minimize precipitation in tile beverage.

Anti-oxidant and Hepatoprotective Effect of White Ginsengs in H2O2-Treated HepG2 Cells

  • The antioxidant activity of white ginseng was not recorded in Korea Functional Food Code, while its activity of red ginsengs was recorded. The aim of this study was to evaluate the antioxidant and hepato protective effect of different ginsengs in H2O2-treated HepG2 cells. White and red ginseng were prepared from longitudinal section of the same fresh ginseng (4-year old). The whole parts of white and red ginsengs were separately extracted with 70% ethanol and distilled water respectively, at 70 ℃ to obtain therapeutic ginseng extracts namely, WDH (distilled water extract of white ginseng), WEH (70% ethanol extract of white ginseng), RDH (distilled water extract of red ginseng) and REH (70% ethanol extract of red ginseng). In this work, we have investigated the DPPH, hydroxyl radical, Fe2+-chelating activity, intracellular ROS scavenging capacity and lipid peroxidation of different ginsengs. All these extracts showed a dose dependent free-radical scavenging capacity and a ROS generation as well as lipid peroxidation was significantly reduced by treatment with bioactive extracts of white ginsengs (WDH) than red ginsengs. Additionally, white ginseng extracts (WDH) has dramatically increased intracellular antioxidant enzyme activities like superoxide dismutase and catalase in H2O2-treated HepG2 cells. All these results explain that administration of white ginseng is useful as herbal medicine than red ginseng for chemoprevention of liver damage.

Effect of Panax ginseng Extracts on the Immunotoxicity of Ethanol (Ethanol의 면역독성에 대한 인삼엑기스의 영향)

  • Experiments were performed on mice to investigate the effect of panax ginseng extracts on the immunotoxicity of ethanol. Immune response were evaluated by antibody production, Arthus reaction, delayed type hypersensitivity (DTH), Rosette froming cell (RFC) and macrophage activity in mice, sensitized and challenged with sheep red blood cells. The weight of liver, spleen and thymus were measured. Following results obtained in this experiment. The exposure of ethanol decreased humoral and cellular immune response, the body weight and macrophage activity. Ginseng extracts such as ethanol extract, petroleum ether extract and n-butanol fraction were significantly increased the body weight. The administration of ginseng ethanol extract and ginseng petroleum ether extract were restored or increased humoral and cellular immune response. Macrophage activity was decreased by ethanol, but restored by the ginseng extracts.

  • This study was undertaken to determine whether the 9 herbal complex induces apoptosis in human breast cancer MCF-7 cells and adriamycin-resistant MCF7/adR cells. Ethanol extracts of Bojungbangamtang (BBTE) and acidic polysaccharide of Red Ginseng (GIN) induced cell death in both MCF-7 and MCF7/adR cells. Ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng also induced $G_2/M$ cell cycle arrest and increased TUNEL positive cells in MCF7/adR cells. In addition, flow cytometric analysis revealed the decreased expression of P-glycoprotein (P-gp) in ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng treated MCF7/adR cells. Similarly, decreased protein levels of P-glycoprotein and multidrug resistance associated proteins-1 were also determined by immunocytometry in ethanol extracts of Bojungbangamtang treated MCF7/adR cells. Taken together these data indicate that ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng inhibit the function of ABC transporters such as multi drug resistance associated proteins (MRPs) and P-glycoprotein as well as induce apoptosis in MCF7/adR cells. Thus, these data suggest that ethanol extracts of Bojungbangamtang and polysaccharide of Red Ginseng can be candidates for the treatment of multidrug-resistant MCF7/adR cells.

Comparative Studies on the Effects of Fresh, White and Red Ginseng Their Effects on the Content of Several Components in Rat Serum and the Production of Lipid Peroxide in Rat Liver (수삼, 백삼 및 홍삼의 효과에 대한 비교 연구 흰쥐의 혈청내 몇가지 성분 함량 및 과산화지질생성에 미치는 영향)

  • Seventy percent ethanol extracts were prepared from 6-year-old fresh, white and red ginseng cultivated in Kangwha, and administered orally into rats for the comparison of their effects. And their effects on the production of lipid peroxide in the livers of ethanol-administered rats were measured. Red ginseng administered group showed the largest body weight increase. However, fresh, white, red ginseng extract administered group showed no significant change in concentration of total lipid, triglyceride and free cholesterol in serum. White and red ginseng extract administeration decreased blood-sugar levels significantly. Lipid peroxide content in livers of white and red ginseng administered groups was decreased significantly. Red ginseng administered group showed greater decrcase in lipid peroxide content than that of white ginseng administred group. The increase of lipid peroxide content in the livers of ethanol-administered group was inhibited by administration of fresh, white, and red ginseng extract. And the strongest-inhibitory action was observed in red ginseng administered group. Therefore, it is supposed that red ginseng has the most powerful antiaging effect.

Detection of Estrogen-like Activities of Hydrothermal and Ethanol Extracts of Oriental Medicines (한약재 열수 및 에탄올 추출물의 여성호르몬 유사활성 검증)

  • The purpose of this study was to investigate the estrogen-like activities of eight oriental medicines, including red ginseng, fenugreek, and dandelion. Hot water and ethanol extracts were prepared from Cervus nippon temminck (Nokgaksang), Cynanchum wilfordii (Baeksuo), Lespedeza cuneata (Yagwanmun), Panax ginseng (red ginseng), Smilax china (Toryeong), Taraxacum platycarpum (Mindeulre, dandelion), Tribulus terresteis (Namgase), and Trigonella foenum-graecum (Horopa, fenugreek). Then, estrogen-like activities were verified by the in vitro transcriptional activity assay. The extracts showing estrogen-like activities were red ginseng, Baeksuo, fenugreek, Yagwanmun, and dandelion in the ethanol extracts, and red ginseng, fenugreek, and Baeksuo in the hydrothermal extracts. Red ginseng extract showed a higher activity than the standard $10^{-8}M$ $17{\beta}$-estradiol in both the hydrothermal and ethanol extract at $500{\mu}g/ml$, while the $50{\mu}g/ml$ of the red ginseng ethanol extract and the $500{\mu}g/ml$ of the Baeksuo hydrothermal extract showed estradiol activities between $10^{-9}$ and $10^{-8}M$ $17{\beta}$-estradiol. This shows that it would be possible to contribute to the development of functional materials using red ginseng, Baeksuo, fenugreek, and dandelion.

Antioxidant Activity of Yogurt Supplemented with Red Ginseng Extract (홍삼 추출물을 첨가한 요구르트의 항산화능)

  • The antioxidant properties of red ginseng extracts prepared under different extraction conditions were evaluated by a variety of antioxidant assays, including $DPPH^{\cdot}$ radical scavenging, $ABTS^{+\cdot}$ radical scavenging, superoxide anion scavenging, nitrite scavenging and reducing power activities. The contents of total phenolic compounds and flavonoids were also determined. The various antioxidant activities were compared to positive controls such as Trolox, tannic acid and ascorbic acid. The antioxidant activities of all of the extracts were shown to be the highest in the ethanol extract. The antioxidant activities of the red ginseng powder were the lowest among the samples. The amounts of total phenolic compounds and flavonoids were at a maximum in the ethanol extract. Correlation analysis demonstrated the existence of a linear relationship between free radical scavenging activities and the phenolic compounds contents of extracts. The antioxidant activity of yogurt was increased as the result of the addition of red ginseng extract. The quality characteristics of the yogurt to which red ginseng extract was added were similar to those of yogurt without red ginseng extract. The overall sensory score and color of yogurt made from 0.5% red ginseng was the best of the tested yogurts. In accordance with the antioxidant activity and quality characteristics, the optimal concentration of red ginseng extract was approximately 0.5%.

Biological Activities and Cell Proliferation effects of Red Ginseng Ethanol Extracts (홍삼 에탄올 추출물의 생리활성과 세포증식 효과)

  • Objectives: Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical, ROS such as hydrogen peroxide($H_2O_2$), nitric oxide(NO) increases with age. Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. In this study, we have conducted to investigate the pharmacological effects of red ginseng for the development possibility to pharmacopuncture drug sources or healthy aid foods. Methods: For our aims, it was investigated the biological activities of Red Ginseng ethanol extracts (RGEE) by measuring total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity, ABTS radical scavenging activity and cell viability of MCF 10A and SK-MEL-2 in vitro with MTT assay method. Results: The total polyphenol contents of RGEE was 3.06${\pm}$0.11mg/g in 10mg/ml, the total flavonoid contents of RGEE was 1.35${\pm}$0.01mg/g in same concentration. The ABTS radical scavenging activity was about 80% and that of DPPH activity was 65% in 50mg/ml of RGEE. The cell viability of SKMEL-2, skin cancer cell line was decreased and that of MCF 10A, skin normal cell line was increased. Conclusions: We conclude that RGEE may be useful as potential functional foods or pharmacopuncture drug sources on the diseases induced by oxidant stress.

Effect of White and Red Ginseng Extracts on the Immunological Activities in Lymphocytes Isolated from Sasang Constitution Blood Cells (백삼 및 홍삼 추출물의 사상체질별 면역세포 활성효과)

  • The effects of white ginseng and red ginseng ethanol extracts on the proliferation, nitric oxide (NO) production and tumor necrosis factor-$\alpha$ (TNF-$\alpha$) in human Band T lymphocytes isolated from blood cell of sasang constitution were examined in vitro. White ginseng extract known as medicine suitable for soeumin constitution showed the highest immune activities such as proliferation, NO production and TNF-$\alpha$ in soeumin constitution, and followed by taeeumin and soyangin, respectively (P<0.05). However, there were immunologically insignificant differences among sasang constitution lymphocytes treated by red ginseng ethanol extract. Both white and red ginseng ethanol extracts showed significantly higher immunological activities than LPS-induced sasang constitution lymphocytes on NO production and TNF-$\alpha$ release (P<0.05). The different immunological activities of white and red ginseng extracts in sasang constitution lymphocytes might be due to the component changes during the processing of ginseng. Thus, the findings of components suitable for all constitutions from red ginseng extract could be applied as a new constitution marker.
