Rhizoctonia Black Stem Rot of Saururi Herba Caused by Rhizoctoia solani (Rhizoctonia solani에 의한 삼백초 검은줄기썩음병(가칭))

  • Rhizoctonia stem rot of Saururui herba [Saururus chinensis (Lour) Baill] was observed during plantations from 1996 to 1997 in Chungbuk area. Infected plant showed damping-off and stem rot at soil line. The causal fungus of stem rot isolated from the infected plant was identified as Rhizoctonia solani according to the criteria based on the cultural and morphological characteristics. The stem rot of Saururi herba caused by Rhizoctonia solani was first described in Korea, and the name the“Rhizoctonia black stem rot”is proposed.

  • To confirm the antifungal activity to plant pathogenic fungi, Rhizoctonia solani, Rhizoctonia solani AG2-2, Fusarium oxysporium, Phytophthora dreschler, Alternaria sp. were selected. Helicosporium has the antifungal activity to Rhizoctonia solani, Rhizoctonia solani AG2-2, while Fusarium oxysporium, Phytophthora dreschler did not affected even though Alternaria made a feeble response to that antifungal compounds. From $^1H-NMR$ spectrum of antifungal compound, this compound was guessed to be a structure corresponding to cholesterol.

Response of Systemic Fungicides of Rhizoctonia spp. Causing Rhizoctonia Blight on Turfgrass (잔디에 Rhizoctonia 마름병을 유발하는 Rhizoctonia spp.의 침투성 살균제에 대한 반응)

  • The Rhizoctonia blight causing by Rhizoctonia spp. is an important disease of turfgrass, requiring fungicide application to maintain acceptable conditions for turfgrass good qualities in the golf course. The experiment was conducted to determine the mean 50% effective concentration inhibiting mycelial growth ($EC_{50}$) value of to flutolanil, pyraclostrobine and hexaconazole to Rhizoctonia solani AG-1 IB, Rhizoctonia cerealis and Rhizoctonia solani AG2-2IV isolated from Gyeongbuk province of Korea in vitro. Five discriminatory concentrations of each fungicide were used to detect in vitro sensitivity. The mean of $EC_{50}$ values to three systemic fungicides was the lowest isolate of R. solani AG-1 IB. However, the sensitivity of fungicides to Rhizoctonia solani AG2-2IV were higher mean $EC_{50}$ value of 0.026 ${\mu}g\;a.i.\;ml^{-1}$ of pyraclostrobine and 0.044 ${\mu}g\;a.i.\;ml^{-1}$ of flutolanil. R. cerealis was the lowest sensitivity to hexaconazole which was an average $EC_{50}$ value of 0.022 ${\mu}g\;a.i.\;ml^{-1}$. Inhibition of mycelial growth rate (%) by three combine fungicides using the $EC_{50}$ value of each fungicide was the highest R. solani AG2-2IV. Results of this study were may confirmed in vitro response fungicide of three Rhizoctonia species for control of Rhizoctonia blight in the field.

Control Activities of Natural Compound, TGR-N1, against Turfgrass Fungal Diseases in vitro and in Field (식물유래 천연물, TGR-N1의 잔디 진균병에 대한 방제효과 검정)

  • This study was carried out to investigate control efficacy of natural compound, TGR-N1 against turfgrass fungal diseases. TGR-N1 showed antimicrobial activity in vitro. Rhizoctonia solani AG 1-1, Rhizoctonia solani AG2-2, Rhizoctonia cerealis, Sclerotinia homoeocarpa and Colletotrichum graminicola were inhibited by 500 times and 1,000 times of diluents of TGR-N1. Pythium spp. was not inhibited by the same diluents of TGR-N1. The TGR-N1 also showed a remarkable antifungal activity against Rhizoctonia solani AG 1-1, Rhizoctonia solani AG2-2 in field. This study proved the antifungal activity of natural compound, TGR-N1 and showed its potential as a natural pesticide.

Stem Rot of Carnation Caused by Fusarium oxysporum and Rhizoctonia solani (Fusarium oxysporum과 Rhizoctonia solani에 의한 카네이션 줄기썩음병)

  • Incidence of stem rot on carnation (Dianthus caryophyllus L.) ranged 11 to 29% in Namwon and Chongup area during the growing seasons from 1993 to 1994. Among 129 isolates from carnations in Namwon, 77 isolates were identified as Rhizoctonia solani, 38 isolates were Fusarium oxysporum and 14 isolates were not identified. Among 169 isolates in Chongup, 19 isolates were identified as R. solani, 106 isolates as F. oxysporum and 44 isolates were not identified. Among 77 isolates of R. solani isolated from the specimens of Namwon, 52 isolates were classified as anastomosis group AG 2-2 by anastomosis test, 14 isolates as AG 2-1 and 11 isolates as AG 4. Among 19 isolates from specimens of Chongup, 14 isolates were classified as anastomosis group AG 2-2 and 5 isolates as AG 4. Pathogenicity tests revealed that isolates of F. oxysporum and R.solani AG 2-2 were highly virulent and isolates of R.solani AG 2-1 and AG 4 were mildly virulent.

Screening of Potent Biofungicide for the Growth Inhibition of Soilborne Pathogenic Fungi, Rhizoctonia solani (잔디 뿌리병 병원균인 Rhizoctonia solani의 성장을 저해하는 미생물 선발)

  • Various Trichoderma spp. were evaluated for the development of biofungicides to control soilborne pathogen, Rhiztonia solani, Various Trichoderma spp. were initially tested for their ability to inhibit growth of R. solani by inhibition zone test. Inhibition zones of 3∼5 mm toward R. solani were detected on PDA agar plates. The parasitic activity of strains, the activities of cell-wall-degrading enzymes such as glucanases and chitinases, were also evaluated. Highest activities of glucanase and chitinase were 3.5 U/ml and 0.9 U/ml, respectively, Isolated Trichoderma spp. also exhibited good growth with currently used agrochemicals, which represents that the isolated biofungicides can be mutually used with agrochemicals.

Differentiation of Intraspecific Groups within isolates of Rhizoctonia solani Using PCR-RFLP of Ribosomal DNA (Ribosomal DNA의 PCR-RFLP에 의한 국내산 Rhizoctonia solani 균주들의 종내그룹의 구분)

  • Genetic diversity among 27 isolates of Rhizoctonia solani, which were obtained from diseased crops in Korea and classified into 9 intraspecific groups by anastomosis test and cultural characteristics, was studied by PCR-RFLP. Gene regions of nuclear 17S ribosomal DNA and internal transcribed spacers including 5.8S rDNA of the isolates were amplified with polymerase chain reaction and digested with 12 restriction enzymes. Differences of restriction patterns were not shown among isolates within each intraspecific groups, however, each anastomosis group and culturala type sowed unique restriction fragment length polymorphisms by restriction patterns using HaeIII, Cfr13I and MspI. The results suggest that PCR-FRLP of rDNA using three restriction enzymes could be used to differentiate intraspecific groups of Rhizoctonia solani in Korea.

Seedling Rot of Kamchatka Goatsbeard Caused by Rhizoctonia solani AG-1(IB)

  • In July 2021, we surveyed diseases affecting wild vegetables grown in Hongcheon, Gangwon Province, Korea. During this survey, we observed severe seedling rot symptoms in Kamchatka goatsbeard (Aruncus dioicus) grown in a vinyl greenhouse. The incidence of the disease in the plant seedlings was 1-10%. Diseased seedlings from this population were collected, and fungi were isolated from leaf and petiole lesions. Rhizoctonia sp. was consistently isolated from the lesions. We examined the morphological and cultural characteristics and anastomosis groups of nine Rhizoctonia sp. isolates obtained from the lesions. The results revealed that all isolates corresponded to Rhizoctonia solani AG-1(IB). Three isolates of R. solani AG-1( IB) were evaluated to determine their pathogenicity towards Kamchatka goatsbeard seedlings through artificial inoculation. The tested isolates caused rot symptoms on the inoculated plant seedlings. The symptoms were similar to those observed in plant seedlings from the vinyl greenhouse. We found that R. solani AG-1(IB) caused seedling rot of Kamchatka goatsbeard.

Damping-off of Edible Amaranth Caused by Rhizoctonia solani AG-4

  • Damping-off symptoms were frequently observed on young plants of edible amaranth (Amaranthus mangostanus) grown in vinyl greenhouses by farmers located in Goyang and Yangpyeong in Korea during disease surveys carried out in 2019 and 2020. The incidence of the disease was 1-20%. A total of eight isolates of Rhizoctonia sp. were obtained from the stems of the diseased plants. All the isolates were identified as Rhizoctonia solani AG-4, based on the morphological characteristics and anastomosis test. Three isolates of R. solani AG-4 were tested for pathogenicity on edible amaranth by artificial inoculation. All the tested isolates of R. solani AG-4 induced damping-off symptoms on the inoculated plants; these symptoms were similar to those observed on the vinyl greenhouse plants surveyed. This is the first report of R. solani AG-4 causing damping-off in edible amaranth.

First Report of Damping-Off of Ovate-Leaf Atractylodes Caused by Rhizoctonia solani AG-5 in South Korea

  • In May to July 2019, ovate-leaf atractylodes seedling and plant with Damping-off symptoms were observed in farmer field at Sangju and Mungyeong, Korea. Seven fungal isolates have been retrieved from diseased root tissue and identified as Rhizoctonia solani AG-5 based on morphological and molecular characteristics. To the best of our knowledge, this is the first report on damping-off of ovate-leaf atractylodes caused by R. solani AG-5 in South Korea.
