Effect of LDL in Combination with Taurine, Hypotaurine and Trehalose as a Antioxidant on Freezing Thawed Semen Function in Korean Jeju Black Bull (제주흑우 동결정액 제조에 있어 Low Density Lipoproteins (LDL)과 항산화제로서 Taurine, Hypotaurine 그리고 Trehalose 조합이 동결 융해 후 정자의 성상에 미치는 영향)

  • This study was designed to determine whether low-density lipoproteins (LDL) from egg yolk and taurine, hypotaurine and trehalose as antioxidant in extender improve the freezability and fertility of Korean Jeju Black Bull semen. The semen was cryopreserved with tris egg yolk extenders containing 7% glycerol and treated 4% LDL, 20 mM taurine, hypotaurine and trehalose. Frozen-thawed sperm were evaluated motility, viability, membrane, and acrosome integrity and sperm penetration ability. The results were compared to semen cryopreserved in tris egg yolk extender only as control. Frozen-thawed semen evaluation cleary indicated that the addition of LDL and LDL-antioxidants (taurine, hypotaurine and trehalose) combination were significantly improved (p<0.05) the viability (%; with staining test using eosin-Y) compared to control spermatozoa. Also, in membrane integrity (%; with supravital hypo-osmotic swelling test), not only LDL-antioxiants combination but also LDL were significantly increased (p<0.05) the swelled sperm using HOST compared to control. Sperm acrosome integrity state was classified by CTC (chlortetracycline) staining test. F pattern was significantly increased in LDL-antioxidant combination than control (p<0.05) and B pattern was not significantly differences among all treatments and control. However, AR pattern was significantly decreased in LDL-antioxidants combination than control (p<0.05). Pronucleus formation and sperm penetration index (SFI) were significantly increased in LDL and LDL-antioxidants combination than control (p<0.05). Especially, LDL-taurine significantly improved pronucleus fomation and SFI than LDL (p<0.05). It was concluded that LDL and LDL-antioxidants in extender improved the freezability and fertility of Korean Jeju Black bull spermatozoa.

The Usefulness of Beavers Systems Model on Adolescent Adjustment (청소년의 적응에 대한 Beavers의 Ssytems 모델의 유용성)

  • The purpose of this study was to examine the usefulness of Beavers Systems Model on adolescent adjustment. For this study adolescents' perceptions of family health/competence family style and adolescent adjustment were utilized. The 443 subjects were selected randomly from the second grade of middle schools and high schools in the city of Taegu. The instruments of measurement were SFI II State-Trait Anxiety Inventory Depression Scale Self-Esteem Scale and Delinquency Scale. Factor Analysis Cronbach's a, MANOVA and Scheffe test were conducted for the data analysis. The conclusion of this research findings was as follows: There is significant differences I nadolescents' anxiety depression global self-esteem social-peer self esteem property predatory behaviors and school-norm avoidant behaviors by the family functioning types of BSM. Therefore the group classification by BSM was generally useful in evaluating adolescent adjustment.

Restriction Fragment Fingerprint of an Alkaliphilic Micrococcus sp. Y-1 Genome by Pulsed-field Gel Electrophoresis

  • A genomic DNA of alkaliphilic bacterium, Micrococcus sp. Y-l, was analysed using the physical mapping method of pulsed-field gel electrophoresis (PFGE). Five restriction enzymes of Sspl, Hpal, Xbal, Ndel or EcoRI, which recognize the Adenine-Thymine-rich sequences of genomic DNA, were used for the generation of few (7 to 20) distinctly separate fragments, with average sizes in the range of 200~500 kb. However, the sites for Notl and SfiI, 8 base-recognizing enzymes, were highly frequent. The genome size of this strain was determined to be 4 mega base pairs (Mb) from restriction fragments separated by PFGE. This is the first case of restriction mapping in alkaliphilic bacterium.

Soil Properties in Two Forest Sites in Cox's Bazar, Bangladesh

  • Soil samples were collected from three depths (0-10 cm, 10-40 cm and 40-80 cm) of two forest sites including one plantation dominated by teak with some other minor species and another degraded natural forest in Cox's Bazar, Bangladesh to compare their soil properties. Some vegetation parameters were also studied. For this study $10{\times}10\;m$ and $2{\times}2\;m$ quadrats were used for the tree and undergrowth parameters, respectively. Soil samples were also collected from these quadrats. Between the two forest types, the highest levels of organic carbon, total nitrogen, available phosphorus, exchangeable bases and cation exchange capacity (CEC) were found in soils of the plantation. The soils were acidic in nature and exchangeable Al concentrations were low. Teak dominated forest plantation had higher soil fertility index (SFI) than the degraded natural forest site. Steps for reforestation and appropriate protection are needed to improve the situation.

An Exploratory Study of Developing Model on Family System Related to Adolescent Adjustment (청소년의 적응에 영향을 미치는 가족체계모델개발에 관한 연구)

  • The purpose of this study was to identify the relative influence of OCM and BSM's family functioning dimensions and develop a new family system model related to adolescent adjustment. The 443 subjects were selected randomly from the second grade of middle and high schools in the city of Taegu. The survey instruments were FACESⅢ, SFI-Ⅱ, State-Trait Anxiety Inventory, Depression Scale, Self-Esteem Scale, and Delinquency Scale, Factor Analysis, Cronbach's α, Multiple Regression, MANOVA, Scheffe test were conducted for the data analysis. The major findings of this study were as follows: First, OCM's and BSM's family functioning dimensions respectively had different relative influence that affected adolescent adjustment level. In anxiety and depression. BSM's family health/competence dimension had superior influence to any other family functioning dimensions and in self-esteem and delinquency, OCM's cohesion dimension was superior to any other family functions. Second, family system classification method by a new family system model using family cohesion(OCM's relationship dimension) and family health/competence(BSM's change dimension) was more useful than OCM in evaluating adolescent adjustment.

Enzymatic Interesterification and Melting Characteristic for Asymmetric 1,2-Distearoyl-3-Oleoyl-rac-Glycerol Triacylglycerol Enriched Product (효소적 반응을 이용한 비대칭형 1,2-Distearoyl-3-Oleoyl-rac-Glycerol 혼합물의 생성 및 융점 특성)

  • Asymmetric 1,2-distearoyl-3-oleoyl-rac-glycerol (SSO) triacylglycerol (TAG) is used as a cocoa butter replacer (CBR). In this study, it was produced by lipase-catalyzed interesterification of fully hydrogenated soybean oil (FHSBO) and oleic ethyl ester (OEE) in a batch type reactor at $75^{\circ}C$, 250 rpm. Different molar ratios (FHSBO : OEE=1:1, 1:2 and 1:3, w/w) and various reaction times (1, 2, 3, 4, and 5 hr) were also tested. The optimized condition for SSO was a FHSBO : OEE molar ratio of =1:1 at reaction times of 2, 3, 4, and 5 hr. Enzymatic synthesis generated SSO/SOS, as well as the other TAGs (e.g., PSO/POS, SOO/OSO, SSS), ethyl esters, monoacylglycerol (MAG), and diacylglycerol (DAG). After scale-up, fractionation by solvent (methanol and acetone) fractionation and column chromatography was applied. To reduce ethyl esters, high-melting TAGs (e.g., SSS), and SOO/OSO in reactants, solvent fractionation was applied. Using a silica gel column (sample : silica gel=2:1, wt%), MAG and DAG were removed at $25^{\circ}C$. The major fatty acid composition of the final products (with a high SSO/SOS content) was palmitic acid (C16:0, 10.9~12.9 area%), stearic acid (C18:0, 52.2~54.9 area%), and oleic acid (C18:1, 34.2~35.5 area%). In reversed-phase HPLC analysis, the major TAG species of the final product (FHSBO : OEE=1:1, 2 hr) were SSO/SOS (82.31 area%) and PSO/POS (14.51 area%). Based on the $[SS]^+$ : $[SO]^+$ ratio obtained by RP-HPLC/APCI-MS, the final product had a higher SSO (AAB type TAG) content than cocoa butter (CB). The solid fat index (SFI) of CB and the final product obtained were similar with a narrow melting point range around ~32 to $35^{\circ}C$.

PERIPHERAL NERVE REGENERATION USING A THREE-DIMENSIONALLY CULTURED SCHWANN CELL CONDUIT (삼차원 배양된 슈반세포 도관을 이용한 말초 신경 재생)

  • The use of artificial nerve conduit containing viable Schwann cells is one of the most promising strategies to repair the peripheral nerve injury. To fabricate an effective nerve conduit whose microstructure and internal environment are more favorable in the nerve regeneration than existing ones, a new three-dimensional Schwann cell culture technique using $Matrigel^{(R)}$. and dorsal root ganglion (DRG) was developed. Nerve conduit of three-dimensionally arranged Schwann cells was fabricated using direct seeding of freshly harvested DRG into a $Matrigel^{(R)}$ filled silicone tube (I.D. 1.98 mm, 14 mm length) and in vitro rafting culture for 2 weeks. The nerve regeneration efficacy of three-dimensionally cultured Schwann cell conduit (3D conduit group, n=6) was assessed using SD rat sciatic nerve defect of 10 mm, and compared with that of silicone conduit filled with $Matrigel^{(R)}$ and Schwann cells prepared from the conventional plain culture method (2D conduit group, n=6). After 12 weeks, sciatic function was evaluated with sciatic function index (SFI) and gait analysis, and histomorphology of nerve conduit and the innervated tissues of sciatic nerve were examined using image analyzer and electromicroscopic methods. The SFI and ankle stance angle (ASA) in the functional evaluation were $-60.1{\pm}13.9$, $37.9^{\circ}{\pm}5.4^{\circ}$ in 3D conduit group (n=5) and $-87.0{\pm}12.9$, $32.2^{\circ}{\pm}4.8^{\circ}$ in 2D conduit group (n=4), respectively. And the myelinated axon was $44.91%{\pm}0.13%$ in 3D conduit group and $13.05%{\pm}1.95%$ in 2D conduit group to the sham group. In the TEM study, 3D conduit group showed more abundant myelinated nerve fibers with well organized and thickened extracellular collagen than 2D conduit group, and gastrocnemius muscle and biceps femoris tendon in 3D conduit group were less atrophied and showed decreased fibrosis with less fatty infiltration than 2D conduit group. In conclusion, new three-dimensional Schwann cell culture technique was established, and nerve conduit fabricated using this technique showed much improved nerve regeneration capacity than the silicone tube filled with $Matrigel^{(R)}$ and Schwann cells prepared from the conventional plain culture method.

Enzymatic Synthesis of Diacylglycerol Oil from Glyceryl Mono-oleate and Conjugated Linoleic Acid Using a Stirred-Batch Type Reactor (회분식 반응기를 이용한 Glyceryl Monooleate와 Conjugated Linoleic Acid로부터 효소적 반응을 통한 디글리세롤 유지의 합성)

  • Diacylglycerol(DAG) was produced by enzymatic esterification of glyceryl mono-oleate(GMO) and conjugated linoleic acid(CLA) in a stirred-batch type reactor. The reaction was catalyzed by lipozyme RMIM(an immobilized lipase from Rizomucor miehei). DAG was isolated by a short-path distillation process and decolorized. DAG oil was composed of 87.3% DAG, 11.4% triacylglycerol(TAG), and 1.5% monoacylglycerol(MAG)(all w/w). Major fatty acids in DAG oil were oleic acid(54%), CLA(31.1%), and linoleic acid(7%). DAG oil iodine,and acid values were 108.8, 2.57, and 1, respectively. The DAG oil solid fat index(SFI) and thermograms were obtained using differential scanning calorimetry.

Effects of Electro Acupuncture on ST36 and LR3 on the Pain and Nerve Regeneration in Sciatic Nerve Injured Rats (좌골신경손상 백서에서 족삼리와 태충 전침자극이 통증 및 신경재생에 미치는 효과)

  • The present study was conducted with the object of examining how ST36 and LR3 electro acupuncture affects the injured side and the intact side in rats with sciatic nerve injury. For this study, we divided rats into a control group that were injured but not treated, experimental group I that were injured and had electro acupuncture on the intact side, experimental group II that were injured and had electro-acupuncture on the injured side, and experimental group III that were injured and had electro-acupuncture on both the intact side and the injured side, and performed behavioral and immunohistochemical tests. The results of this study are as follows. In the results of hot plate test, on Day 1 of experiment, experimental group I did not show a statistically significant difference but experimental group II and III showed a statistically significant difference, and on Day 2 and 3, all of experimental group I, II and III showed a statistically significant difference. In the results of SFI test, on Day 7 of experiment, experimental group I did not show a statistically significant difference but experimental group II and III showed a statistically significant difference, and on Day 14 and 21, all of experimental group I, II and III showed a statistically significant difference. In the results of immunohistochemical test, the expression of c-fos decreased gradually on Day 1, 2 and 3 in all the experimental groups, and the decrease was larger in the experimental groups that had electro-acupuncture than in the control group. In the results of immunohistochemical test, the expression of BDNF increased gradually on Day 7, 14 and 21 in all the experimental groups, and the expression was higher in the experimental groups than in the control group. Summing up the results, ST36 and LR3 electro acupuncture was effective when it was applied to the injured side and to both sides after sciatic nerve injury, but it was also effective in pain relief and nerve regeneration when it was applied to the intact side.

Molecular Typing of Legionella pneumophila Isolated in Busan, Using PFGE (부산지역에서 분리한 레지오넬라균에 대한 PFGE를 이용한 molecular typing)

  • In this study, we did the molecular typing of 39 environmental Legionella pneumophila serogroup 1 isolates collected from 2001-2003 in Busan using the pulsed-filed gel electrophoresis (PFGE). PFGE of SfiI fragments were divided into 10 pulsotypes $(A\~J)$, corresponding to $<65\%$ similarity and a subtype within each pulsotype was characterized by $>84\%$ similarity. The major cluster was pulsotype E $(46.2\%)$, which included 18 isolates and was divided into 4 subtypes $(E1\~E4)$. PFGE of NotI fragments were divided into 8 pulsotypes $(a\~h)$, corresponding to $<60\%$ similarity and a subtype within each pulsotype was characterized by $100\%$ similarity. The major cluster was pulsotype f $(38.5\%)$, which included 15 isolates. The ATCC type strain L. pneumophila serogroup 1 was identified as a different molecular pulsotype compare to the Busan isolates. It is possible that L. pneumophila serogroup 1 isolated in Busan with specific DNA pattern is comparable with those isolation in other cities in Korea.
