Functions of the Plant Qbc SNARE SNAP25 in Cytokinesis and Biotic and Abiotic Stress Responses

  • Eukaryotes transport biomolecules between intracellular organelles and between cells and the environment via vesicle trafficking. Soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNARE proteins) play pivotal roles in vesicle and membrane trafficking. These proteins are categorized as Qa, Qb, Qc, and R SNAREs and form a complex that induces vesicle fusion for targeting of vesicle cargos. As the core components of the SNARE complex, the SNAP25 Qbc SNAREs perform various functions related to cellular homeostasis. The Arabidopsis thaliana SNAP25 homolog AtSNAP33 interacts with Qa and R SNAREs and plays a key role in cytokinesis and in triggering innate immune responses. However, other Arabidopsis SNAP25 homologs, such as AtSNAP29 and AtSNAP30, are not well studied; this includes their localization, interactions, structures, and functions. Here, we discuss three biological functions of plant SNAP25 orthologs in the context of AtSNAP33 and highlight recent findings on SNAP25 orthologs in various plants. We propose future directions for determining the roles of the less well-characterized AtSNAP29 and AtSNAP30 proteins.

Immunostimulatory Activity of Solanum nigrum Through TLR4-Mediated JNK Activation in RAW264.7 Cells

  • In this study, we investigated the effect of Solanum nigrum aerial parts (SNAP) on macrophage activation and macrophage autophagy in RAW264.7 cells. SNAP increased the production of immunostimulatory factors and phagocytosis in RAW264.7 cells. TLR4 inhibition blocked SNAP-mediated production of immunostimulatory factors. In addition, the JNK inhibition reduced the SNAP-mediated production of immunostimulatory factors, and the SNAP-mediated JNK activation was blocked by the TLR4 inhibition. SNAP activated macrophage autophagy. TLR4 inhibition blocked SNAP-mediated macrophage autophagy and inhibition of p38 and JNK attenuated SNAP-mediated macrophage autophagy. These findings indicate that SNAP may induce TLR4/JNK-mediated macrophage activation and TLR4/p38 and JNK-mediated macrophage autophagy.

  • It is used to lead to serious structural vulnerability of the system security of security-critical system when we have quickly developed software system according to urgent release schedule without appropriate security planning, management, and assurance processes. The Data Set and Provider of DataSnap, which is a middleware of Delphi XE2 of the Embarcadero Technologies Co., certainly help to develop an easy and fast-paced procedure, but it is difficult to apply security program and vulnerable to control software system security when the connection structure Database-DataSnap server-SQL Connection-SQL Data set-Provider is applied. This is due to that all kinds of information of Provider are exposed on the moment when DataSnap Server Port is sure to malicious attackers. This exposure becomes a window capable of running SQL Command. Thus, it should not be used Data Set and Provider in the DataSnap Server in consideration of all aspects of security management. In this paper, we study on the verification of the security vulnerabilities for Client and Server DataSnap in Dlephi XE2, and we propose a secure coding method to improve security vulnerability in the DataSnap server system.

Exogenous Nitric Oxide Donation During In Vitro Maturation Improves Embryonic Development after Parthenogenesis and Somatic Cell Nuclear Transfer in Pigs

  • Nitric oxide (NO) has an important role in oocyte maturation and embryonic development in mammals. This study examined the effect of exogenous NO donor S-nitroso-N-acetylpenicillamine (SNAP) in a maturation medium on meiotic progression and embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) in pigs. When oocytes were exposed to $0.1{\mu}M$ SNAP for first 22 h of in vitro maturation (IVM) in Experiment 1, SNAP significantly improved blastocyst development in both defined and standard follicular fluid-supplemented media compared to untreated control (48.4 vs. 31.7-42.5%). SNAP treatment significantly arrested meiotic progression of oocytes at the germinal vesicle stage at 11 h of IVM (61.2 vs. 38.7%). However, there was no effect on meiotic progression at 22 h of IVM (Experiment 2). In Experiment 3, when oocytes were treated with SNAP at 0.001, 0.1 and $10{\mu}M$ during the first 22 h of IVM to determine a suitable concentration, $0.1{\mu}M$ SNAP (54.2%) exhibited a higher blastocyst formation than 0 and $10{\mu}M$ SNAP (36.6 and 36.6%, respectively). Time-dependent effect of SNAP treatment was evaluated in Experiment 4. It was observed that SNAP treatment for the first 22 h of IVM significantly increased blastocyst formation compared to no treatment (57.1% vs. 46.2%). Antioxidant effect of SNAP was compared with that of cysteine. SNAP treatment significantly improved embryonic development to the blastocyst stage (49.1-51.5% vs. 34.4-37.5%) irrespective of the presence or absence of cysteine (Experiment 5). Moreover, SNAP significantly increased glutathione (GSH) content and inversely decreased the reactive oxygen species (ROS) level and mitochondrial oxidative activity in IVM oocytes. SNAP treatment during IVM showed a stimulating effect on in vitro development of SCNT embryos (Experiment 7). These results demonstrates that SNAP improves developmental competence of PA and SCNT embryos probably by maintaining the redox homeostasis through increasing GSH content and mitochondrial quality and decreasing ROS in IVM oocytes.

Quality Characteristics of Sugar-snap Cookies by additions of Curcuma Longa L. Powder (강황분말을 첨가한 Sugar-snap cookies의 품질 특성)

  • The purpose of this study is to investigate the quality characteristics of sugar-snap cookies prepared with Curcuma Longa L. powder(0, 3, 6, 9, 12, 15%) substituted for flour. The pH of the dough of sugar-snap cookies ranged from 6.58 to 7.32, and their moisture contents ranged from 9.09% to 10.77%. The width and spread factor of sugar-snap cookies decreased significantly with increase in Curcuma Longa L powder content(p<0.05); Lightness decreased significantly as the Curcuma Longa L. powder content increased while redness and yellowness increased significantly(p<0.05). Hardness increased with more powder added, and sensory evaluation indicated that the samples with 6% of the powder received higher scores in terms of taste, color, flavor, texture and overall tastes. As a result, sugar-snap cookies with 6% of Curcuma Longa L. powder were valued as best.

  • SNAP-25 was first investigated as a neuron-specific protein preferentially expressed in CA3 pyramidal neurons of mouse hippocampus. It is a presynaptic plasma membrane protein in the nerve cell and plays an important role in the synaptic vesicle membrane docking and fusion pathway. We have recently isolated SNAP-25 cDNA from a rat brain cDNA library using a probe of Z2 cDNA. It consisted of 2,101 bp and an open reading frame (ORF) was identified between nucleotides (nt) 209 and 827. The AUG codon (nt 209∼211) was surrounded by CTACCATGG, which corresponded to the consensus sequence of ribosomal binding site. The ORF was terminated by TAA (nt 827∼829) to encode a polypeptide of 206 amino acid residues. The 3'-untranslated region contained two extensive stretches of repeated (CA)28 and (CA)19 at positions 925∼980 and 1645∼1682. It is noteworthy that cysteine residues were clustered in the span of amino acid residues 84∼991 : Cys-Gly-Leu-Cys-Val-Cys-Pro-Cys. Rat SNAP-25 showed 88% and 97% identity in nucleotide sequences to that of human and mouse, respectively. Amino acid sequence of rat SNAP-25 showed 100% identity to that of mouse and human SNAP-21.

  • The use of snap-fit features is highly recommended to reduce overall product cost and manufacturing time by reducing the number of parts and eliminating assembly with conventional fasteners. Application to particular product for integral attachment using snap-fit features is needed. Alternative attachment concept for specific application has been introduced. In this study, the optimal attachment design based on the given design objectives is identified using newly developed alternative attachment concept for the specific application to LCD monitor case assembly. Integral attachment using snap-fit features applies the systematic procedure and the intensity analysis.

  • Glass fiber reinforced composite decks have high-strength, light-weight and high durability. The composite decks having vertical snap-fit connections are designed for pedestrian bridges and their structural behaviour are studied. The existing connection method of the composite decks in horizontal direction is replaced by the developed snap-fit connection method in vertical direction. The section shape of the composite decks having the vertical snap-fit connection is designed. The safety of the vertical snap-fit connection is verified by finite element analysis.

Chaotic Responses of Curved Plate under Sinusoidal Loading

  • In the present investigation, the nonlinear dynamic buckling of a curved plate subjected to sinusoidal loading is examined. By the theoretical analyses, a highly nonlinear snap-through motion of a clamped-free-clamped-free plate and its effect on the overall vibration response are investigated. The problem is reduced to that of a single degree of freedom system with the Rayleigh-Ritz procedure. The resulting nonlinear governing equation is solved using Runge-Kutta (RK-4) numerical integration method. The snap-through boundaries, which vary with different damping coefficient and linear circular frequency of the flat plate are studied and given in terms of force and displacement. The relationships between static and dynamic responses at the start of a snap-through motion are also predicted. The analysis brings out various characteristic features of the phenomenon, i.e. 1) small oscillation about the buckled position-softening spring type motion, 2) chaotic motion of intermittent snap-through, and 3) large oscillation of continuous snap-through motion crossing the two buckled positions-hardening spring type. The responses of buckled plate were found to be greatly affected by the snap-through motion. Therefore, better understanding of the snap-through motion is needed to predict the full dynamic response of a curved plate.

Effect of Black Rice Flour on the Quality of Sugar-snap Cookie (흑미 가루의 첨가가 sugar-snap cookie의 품질 특성에 미치는 영향)

  • The quality of sugar-snap cookie prepared with wheat flour supplemented with black rice flour was investigated. The pH of cookie batter decreased as the amount of black rice flour increased. Increasing proportions of black rice flour resulted in increase of width and spread factor of cookie, whereas thickness and fracturability decreased. L, a, and b values decreased as the amount of black rice flour increased. Sensory evaluation showed that supplements of 20, 25, and 20% black rice flour had the best overall preference in strong, medium, and weak flours, respectively.
