  • Suspected infections of Campylobacter spp and Clostridium spp were observed in three dogs. The diagnosis was based on fecal cytology, Gram's stain, clinical signs and serum chemistry. The rectal swabs of diarrheic dogs were performed to confirm the enteropathogens. Suspected Campylobacter spp were a sea-gull shape and Clostridium spp had a large, clear endospore in rectal cytology. Treatment with appropriate antibiotics resulted in a complete resolution of all clinical abnormalities in three cases. The source of Campylobacter spp and Clostridium spp could not be found clearly in three cases, but gastrointestinal origin was most likely. When detecting the enteropathogens in feces, fecal smear with Wright's and Gram's stain should be made at first and also, if the patients have canine parvoviral enteritis, attention should be paid to confirm the Campylobacter spp and Clostridium spp. In addition, since Campylobacter spp and Clostridium spp as normal bacterial flora exists in canine intestines, it is thought that microbiological isolation should be performed to confirm the suspected Campylobacter spp and Clostridium spp as primary enteropathogens in subsequent study.

  • Lectin activities and chemical characteristics of Escherichia coli, Lactobacillus spp. and Bifidobacterium spp. originating from the porcine cecal mucosal layer were studied based on hemagglutination assay (HA) and hemagglutination inhibition assay (HIA). Although all the bacterial strains were able to agglutinate erythrocytes of porcine or rabbit origin, much higher HA titers were consistently observed for Lactobacillus spp. than for E. coli or for Bifidobacterium spp. A remarkable reduction in HA titers occurred by the treatment of E. coli and Lactobacillus spp. with protease or trypsin and of Bifidobacterium spp. with protease, trypsin or periodate. There were no significant effects on the HA titers of the three groups of bacteria after the treatment with lipase. Hemagglutination of E. coli was strongly inhibited by D (+)-mannose and D (+)-galactose; Lactobacillus spp. by $\alpha$-L-rhamnose and methyl-$\beta$-galactopyranoside; Bifidobacterium spp. by D (+)-alactose, $\alpha$-L-rhamnose, $\alpha$-L-fucose, L (+)-arabinose, D (+)-mannose, D (-)-fructose at a relatively low concentration (1.43 to 3.75 mg/ml). These results, combined with the enhanced HA activities of the three bacterial strains by modification of rabbit erythrocytes with neuraminidase and abolished HA activity of E. coli after treatment with $\beta$-galactosidase, indicate that it might be the glycoproteinous substances surrounding the surface of the bacterial cells that are responsible for the adhesions of these microorganisms by recognizing the specific receptors on the red blood cell.

  • A multiplex polymerase chain reaction (PCR) was developed for the simultaneous detection and differentiation among Vairimorpha spp. and Nosema spp. and identification of Vairimorpha necatrix from Lepidoptera insects. Three sets of primers were selected from different genomic sequences to specifically amplify an 831 bp amplicon within the SSU rRNA gene, specific for both Vairimorpha spp. and Nosema spp. (MSSR primer); a 542 bp amplicon within the SSU rRNA gene, specific for Vairimorpha spp. (VSSU primer); and a 476 bp amplicon within the actin gene, specific for Vairimorpha necatrix (VNAG primer). Using the primers in conjunction with multiplex PCR, it was possible to detect Vairimorpha spp. and Nosema spp. and to differentiate between them. The sensitivity of this PCR assay was approximately 10 spores per milliliter. It is proposed that the multiplex PCR is a sensitive, specific, and rapid tool that can serve as a useful differential diagnostic tool for detecting Vairimorpha spp. and Nosema spp. in Lepidoptera insect.

  • This study was investigated to isolate identify the total bacteria and fungi from the indoor air of work-place of the shoes, paint, stainless steel, and plastic industries. The number of bacterial colonies on the nutrient agar plates were calculated by the open petridish method for 30 minutes in indoor air of work-places at the autumn and winter. The isolated bacteria were identified by Gram stain and biochemical test using API Staph and API 20E kits. The isolated fungal colonies were identified by gross appearance of the giant colonies and microscopic examination of their spore and hyphal characteristics on the slide culture method. The minimum inhibitory concentration (MIC) of several antibiotics against isolated bacteria was determined by the microdilution method with Mueller-Hinton broth. The 70-400 colonies in autumn and 54-236 colonies in winter were isolated from the indoor air of work-places of several industry. The isolation rates of Gram positive cocci, Gram positive bacilli, Gram negative bacilli, and Gram negative cocci were 46.3%, 19.8%, 17.3%, and 16.1%, respectively. In Gram positive cocci, the most strains were identified as Aerococcus spp, Micrococcus spp, and Staphylococcus spp. In Gram positive and negative bacilli, and Gram negative cocci were identified as Bacillus spp, Pseudomonas spp, and Neisseria spp, respectively. The frequently isolated fungi were Aspergillus spp, Penicillium spp and Rhizopus spp, respectively. The frequently isolated Aerococcus spp, Micrococcus spp, and Staphylococus spp were highly resistance against ampicillin, erythromycin, methicillin, and tetracycline. These results arouse our attention to microbiologic pollution in the indoor air of work-places of industries.

  • This study was performed to examine the distribution patterns, ecological characteristics and celluar fatty acid compositions of marine bacteria in Suyeong Bay. During study periods, total cell count (TC) and viable cell count (VC) were $10^7-10^8/m\ell\;and\;10^4-10^6\;cfu/m\ell$, respectively. The temporal variations of TC showed similar patterns between surface and bottom layer, but the VC at bottom decreased gradually from winter to summer. Among the 303 bacterial strains isolated in the study area, which belong to 10 genus types, Pseudomonas spp., $(32.3\%)$, Acinetobader sup. $(19.1\%)$, Vibrio spp. $(11.2\%)$, Flovobacterium spp. $(10.6\%)$ and Bacillus spp. $(7.9\%)$ were dominant. Thirty-one fatty acids were detected from Pseudomonas spp. and Bacillus spp., which were the most predominated among Gram positive and Gram negative bacteria. Celluar fatty acid compositions of Bacillus spp. were relatively simple compared to those of Pseudomonas spp.. Relatively high ratio of monounsaturated forms were detected in Pseudomonas spp. while branched types were dominant in those of Bacillus spp.. Hydroxy and cyclopropane fatty acid were detected only in the cellular fatty acid of Pseudomonas spp.. Cellular fatty acid compositions of Pseudomonas spp. revealed relatively high percentage of $C_{16:1},\;C_{17:1},\;C_{18:1}\;and\;C_{16:0}$, While Bacillus spp. predominated $C_{16:0}\;iso\;C_{16:0}\;anteiso,\;C_{17:0}\;anteiso\;and\;C_{16:1}$.
