A Study on the Relationships Between the Epiphytic Microbes and the Blight of Porphyra Species from the Coastal Waters of the Yellow Sea, Korea -II.   • To study the seasonal variation of heterotrophic bacteria near the surrounding seawater of Porphyra forming area, samples were collected in the intertidal waters of Mokp'o of the Yellow Sea from February to December, 1990. Annual distribution of heterotrophic bacteria ranged from $7.5\times10^2\;to\;1.1\times10^5\;cfu/ml,$ annual distribution of physiological characteristic bacteria ranged from $5.0\times10\;to\;4.34\times10^4\;cfu/ml$ for proteolytic bacteria, from 0 to $1.35\times10^4\;cfu/ml$ for lipolytic bacteria and from 0 to $1.2\times10^4\;cfu/ml$ for amylolytic bacteria. Sixty-five percent of isolates from seawater were rods, and $76.7\%$ of isolates were Gram-negative. Most isolates were mesophiles and showed utilization of various carbon sources such as glucose, maltose, lactose, sucrose and arabinose. Most isolates also showed tolerance to a broad range of salt concentration. Dominant genus in seawater were Flavobacterium spp., in February, Moraxella spp., Acinetobacter spp. in March, Bacillus spp., Chromobacterium spp., Micrococcus spp., Vibrio spp. in July and Chromobacterium spp., Micrococcus spp., Bacillus spp. in November.

Fungal Distribution and Varieties Resistance to Kernel Discoloration in Korean Two-rowed Barley (국내 육성 2조 겉보리 변색 종실에서의 곰팡이 분포와 품종 저항성)

  • Barley kernel discoloration (KD) leads to substantial loss in value through downgrading and discounting of malting barley. The objective of this research is to investigate fungal distribution and varieties resistance to KD in Korean two-rowed barley. Several fungal organisms including Alternaria spp., Fusarium spp., Aspergillus spp., Epicoccum spp. and Rhizopus spp. were isolated from Korean two-rowed barley representing KD. The symptoms of KD were brown and black discolorations of the lemma and palea. The most frequently detected fungal species was Alternaria spp. which exhibited 69.1% and 72.2% in 2011 and 2012, respectively. Epicoccum spp., Fusarium spp., and Aspergillus spp. were also detected. Fusarium spp., primary pathogen of barley head blight, were rarely occurred in the 2011 and their occurrence increased to 4.7% in 2012. Twenty cultivars of Korean two-rowed barely were evaluated to KD. The average percentage of KD was 8.0-36.0% in 2011 and 5.2-36.6% in 2012. Two cultivars ('Sacheon 6' and 'Dajinbori') showed KD of 6.2% to 8.8% and determined resistant, however 'Samdobori' and 'Daeyeongbori' demonstrating KD of 22.2-36.6% were highly susceptible. 'Jinyangbori', 'Danwonbori', 'Sinhobori' and 'Kwangmaegbori' showing KD of less than 15% were moderately resistant cultivar.

  • In the present study, we investigated the actions of sphingosine-I-phosphate (SPP) in Mel-Ab melanocytes. We observed the cytoprotective effect of SPP on UVB-induced cell death. Following exposure of cells to UVB, a significant protective effect was seen in cultures pretreated with SPP. Since SPP is well known as a mitogenic agent, it is possible that the mitogenic effect of SPP may contribute to cell survival. Surprisingly, we found that SPP inhibited DNA-synthesis significantly. We were next interested in the regulation of the extracellular signal-regulated protein kinase (ERK) and Akt pathways by SPP. We clearly observed that SPP potently stimulated the phosphorylation of both ERK and Akt against UVB-induced cell death. Based on these results, we conclude that SPP may show its cytoprotective effect through ERK and Akt activation.

Survey of Poisonous Plants Distributed in Jeju-Island (제주도(濟州道)의 유독식물(有毒植物)에 대(對)한 조사연구(調査硏究))

  • Because of very little information available regarding the poisonous plants distributed in Jeju-Island, the author made a survey of such plants from March, 1974 to Dec., 1977. The results obtained were as follows: 1. If not all, 68 families, 137 genera and 190 species of poisonous plants distributed in Jeju-island were classified. And 190 species were divided into 137 spp. of herbs, 45 spp. of shrubs, 3 spp. of molds and 5 spp. of toadstools. 2. Relatively, a large number of poisonous plants were distributed in wild field. Jungle area, inhabited region, seashore and high mountainous area took next place gradually by the number of plants. 3. And according to their toxic chemical components or poisoning types, they were classified as 16 spp. of cyanogenetic plants, 14 spp. of oxalate-producing plants, 14 spp. of nitrate. producing plants, 10 spp. of photosensitization and 136 spp. of the other activities.

Characteristics of Nanoscale Modes Guided by the Total External Reflection of Surface Plasmon-Polaritons (표면 플라즈몬-폴라리톤의 외부-전반사에 의해 도파되는 나노 크기 모드의 특성)

  • Total external reflection (TER), which does not occur on a dielectric interface, is a unique feature of surface plasmon-polaritons (SPP). We propose an SPP-TER waveguide structure consisting of low-index dielectric nanocore covered with high-index dielectric on a flat metal surface. The SPP mode confined in the nanocore by the TER effect has a mode size much smaller than wavelength scale. Numerical comparison of mode characteristics between the SPP-TER waveguides and other total-internal-reflection-based waveguides such as metal or high-index dielectric nanowires show that the SPP-TER structures can possess higher modal gain for applications of nanocavity lasers.

  • Ticks cause economic losses to the deer industry by decreasing the growth and production of the farmed animals. The mediation of ticks affects humans and animals by causing contagious disease both directly and indirectly. Blood from farmed deer from the areas near Jangsu branch was collected for screening of infectious protozoa and rickettsial disease. Seventy deer blood samples were collected from 30 different deer farms located in Jinan, Jangsu and Muju. This blood samples were used for blood slide smear examination and hematological analysis. DNA from these samples was extracted and was used for PCR analysis for detection of gene fragments of Theileria spp, Babesia spp, Anaplasma spp and Ehrlichia spp. In the blood slide smear examination and PCR analysis all samples did not show presence of protozoal and rickettsial diseases. Eight blood samples showed anemia, 1 sample showed iron deficiency and 7 samples showed regenerative anemia. Results for PCR analysis showed 2 samples were positive for T orientalis. All DNA samples were negative for Babesia spp, Anaplasma spp, and Ehrlichia spp.

  • In order to fine some proper bacterial food for Tigriopus japoncus, bacterial flora of the tide pool inhabited by the copepod has been isolated and tested as bacterial food. Food effect and optimum density of the bacteria in terms of survival rate of the copepod was measured in the larval and the adult stages. Among the 264 strains of isolated bacteria, Acinetobacter spp. Moraxella, spp., Flavobacterium spp. and Pseudomonas spp. were certified as effective food for the copepod. According to the experimental results, Acinetobacter spp. AG-3 was the most effective food for all the stages from nauplius to adult, while Moraxella spp. and Flavobacterium spp. were effective for copepodite stage, and Pseudomonas spp. for the adult stage only. The optimum density of bacteria for the food was about $10^6\;cell/ml$, which was the same average density of bacteria in the tide pool.

Physiological Characteristics of Green Mold(Trichoderma spp.) Isolated from Oyster Mushroom(Pleurotus spp.)

  • This study was conducted to investigate physiological characteristics of Trichoderma spp. isolated from Pleurotus spp. Damage tests of Pleurotus spp. and mycotoxins tests of Trichoderma spp. were also done. The optimal growth temperature of Trichoderma spp. was $27{\sim}30^{\circ}C$. Although, T. longibrachiatum was able to grow at $37^{\circ}C$ and grew $30{\sim}40$ times faster than Pleurotus. The colony colour on PDA medium of T. cf. virens was yellowish green, T. longibrachiatum was yellow, and T. harzianum was turning to bright green. In damage tests of Pleurotus by Trichoderma, T. cf. virens caused the most severe damage to Pleurotus. T. longibrachiatum and T. harzianum caused less damage on Pleurotus but were able to cause greater damage to P. eryngii. One of the mushroom cultivars, P. ostreatus 8 was the most resistant to all Trichoderma spp.. Chitinolytic mycotoxin released by Trichoderma spp. caused 52.7% damage to Pleurotus. Mycotoxins released by T. longibrachiatum caused the greatest damaged(78.6%) on P. eryngii.

  • To investigate the epidemiological trait of listeriosis, Listeria spp were isolated from poultry meat, products and environmental specimens in chicken slaughterhouse. Also determined were isolation rates by the different enrichment procedures, the biochemical properties of isolates. In a total of 307 samples including poultry meat, liver, feathers, feces, chiller water, scalding water overflow and slaughterhouse floor, Listeria spp were isolated predominantly from scalding water overflow (90.0%), body skin before washing (66.7%), liver (20.0%) and feathers(15.0%) However, few Listeria spp were isolated from body skin after washing and feces. The higher isolation rates were obtained in the secondary enrichment procedure (7.2%) than in the primary enrichment (3.9%); after stored the secondary enrichment cultures for 2 weeks at $4^{\circ}C$, Listeria spp were present in 9.8%. The majority of the isolated Listeria spp were identical to those of the standards strains in biochemical and cultural properties. Overall, Listeria spp were present in 13.4% of the specimens tested, and were in order of prevalence of L innocua(11.1%), L monocytogenes(3.3%), L grayi(0.7%) and L murrayi(0.3%).

Antimicrobial Susceptibility of Bacterial Isolates from Domestic Dogs with Urinary Tract Infection (세균성 요로 감염증 애완견의 세균 분포 및 항생제 감수성)

  • Bacterial pathogens were isolated from dogs with urinary tract infection (UTI) in local animal hospitals between August 2003 and December 2009. Bacteria were isolated from urine of 47 dogs. The isolated pathogens were Escherichia coli (n = 27), Streptococcus spp. (n = 7), Staphylococcus spp. (n = 5), Enterobacter spp. (n = 3), Proteus spp. (n = 2), other species were 3 strains, respectively. E. coli were susceptible to imimpenem, polymyxin B, amikacin, cephalosporins, aztreonam, amoxicillin clavulate, cephalosporins, tricarcillin, and amoxicillin clavulate, while were resistant bacitracin, erythromycin, lincomycin, oxacillin, penicillin, and novobiocin. Streptococcus spp. were susceptible to bacitracin, imimpenem, and trimethoprime-sulfa, while were highly resistant amikacin, cefotaxim, cefoxitin, cloxacillin, gentamicin, lincomycin, oxacillin, penicillin, streptomycin, and tobramycin. Staphylococcus spp. were susceptible to cefoxitin, doxycycline, enrofloxacin, imimpenem, and tobramycin, but were resistant aztreonam and tetracycline.
