The CsSTE50 Adaptor Protein in Mitogen-Activated Protein Kinase Cascades Is Essential for Pepper Anthracnose Disease of Colletotrichum scovillei

  • Anthracnose, caused by the ascomycete fungus Colletotrichum scovillei, is a destructive disease in pepper. The fungus germinates and develops an infection structure called an appressorium on the plant surface. Several signaling cascades, including cAMP-mediated signaling and mitogen-activated protein kinase (MAPK) cascades, are involved in fungal development and pathogenicity in plant pathogenic fungi, but this has not been well studied in the fruit-infecting fungus C. scovillei. Ste50 is an adaptor protein interacting with multiple upstream components to activate the MAPK cascades. Here, we characterized the CsSTE50 gene of C. scovillei, a homolog of Magnaporthe oryzae MST50 that functions in MAPK cascades, by gene knockout. The knockout mutant ΔCsste50 had pleiotropic phenotypes in development and pathogenicity. Compared with the wild-type, the mutants grew faster and produced more conidia on regular agar but were more sensitive to osmotic stress. On artificial and plant surfaces, the conidia of the mutant showed significantly reduced germination and failed to form appressoria. The mutant was completely non-pathogenic on pepper fruits with or without wounds, indicating that pre-penetration and invasive growth were both defective in the mutant. Our results show that the adaptor protein CsSTE50 plays a role in vegetative growth, conidiation, germination, appressorium formation, and pathogenicity in C. scovillei.

Anti-oxidant, Anti-inflammatory, and Wound Healing Activities of Selaginella tamariscina Leaf Extract (부처손 잎 추출물의 항산화, 항염 및 피부재생 효능)

  • The leaves of Selaginella tamariscina were used for the treatment of many diseases in traditional medicine. In the study, antioxidant, anti-inflammatory, and wound healing activities of the hot-water extract(STW) and 80%ethanol extract(STE) obtained from S. tamariscina were evaluated. As a result, the polyphenol content of STW and STE were 38.108±0.766 mg/g and 17.927±1.064 mg/g, respectively. The DPPH and ABTS radical scavenging activities with the IC50 values of the STW were over 2 times lower than that of the STE. In the MTT assay, RAW264.7 cell viability of two extracts was decreased by about 6% at 1 mg/mL, whereas for HaCaT cell viability increased by 18% at 50 ㎍/mL. In addition, STW and STE suppressed the production of nitric oxide(NO), Tumor-necrosis(TNF)-��. COX-2 and PGE2 in lipopolysaccharide(LPS) induced RAW264.7 cells. Furthermore, the STE showed wound healing effect through the promotion of skin cell migration in TNF-�� stimulated human keratinocytes. These results indicated that the STW and STE have the potential to be used as a new cosmetic active ingredients in skin care.

Effect of the Selaginella tamariscina Extract on Antioxidation and Inhibition of Matrix Metalloproteinase-1 in Human Skin Fibroblasts (권백 추출물의 인간 피부 섬유아세포에서의 항산화와 Matrix Metalloproteinase-1 발현저해효과)

  • The production of matrix metalloproteinases (MMPs) by the UV irradiated skin fibroblast and the degradation of extracellular matrix (ECM) by these enzymes is known as one of the main reasons of photoaging. In this paper, to investigate the relationship between aging and Selaginella tamariscina extract (STE), we investigated the effects of antioxidant and expression of UVA-induced MMP-1 in human dermal fibroblasts. STE was found to show scavenging activities of radicals and reactive oxygen species (ROS) with the $IC_{50}$/ values of 65.1 $\mu\textrm{g}$/$m\ell$ against 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and 40.9 $\mu\textrm{g}$/$m\ell$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. UVA induced MMP expression was reduced 75.5% by treatment with STE, and MMP-1 mRNA expression was reduced in a dose-dependent manner. Therefore STE was able to significantly inhibition of MMP expression in protein and mRNA level. All these results suggested that STE may act as an anti-aging agent by antioxidation and reducing UVA-induced MMP-1 production.

Protective Effect of Schizonepeta tenuifolia Briquet Extracts on Oxidative DNA Damage in Human Leucocytes and on Hydrogen Peroxide-induced Cytotoxicity in PC12 Cells

  • The present study was conducted to examine the antioxidant activities and neuroprotective effects of methanolic extracts from Schizonepeta tenuifolia Briquet (STE). STE ($100\;{\mu}g/mL$) showed $43.33\;{\mu}M$ of total phenolic content, 64.43% of radical scavenging activity, and 0.157 of reducing power. In addition, the effect of STE on $H_2O_2$-induced DNA damage in human leucocytes was evaluated by the comet assay, where STE was a dose dependent inhibitor of DNA damage induced by $200{\mu}M$ of $H_2O_2$. The protective effect of STE against $H_2O_2$-induced oxidative damage on PC12 cells was investigated by an 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) reduction assay and lactate dehydrogenase (LDH) release assays. After 2 hr of cell exposure to $H_2O_2\;(500\;{\mu}M)$, a marked reduction in cell survival was observed. However, this reduction was significantly prevented by $1-50\;{\mu}g/mL$ of STE. Therefore, these results suggest that STE could be a new antioxidant candidate against neuronal diseases.

Permeation Efficiency of Sea Tangle (Laminaria japonica) Extract into Egg Using Temperature Change Method and Pressure (온도 변화 방법과 압력을 이용한 계란 내부로 다시마 추출물의 침투 효율)

  • In this study, the permeation efficiency (PE) of sea tangle (Laminaria japonica) extract (STE) into egg using the temperature change method (TCM) and pressure was investigated. STE was prepared in a volume of water equal to 10 times the weight of the sea tangle at $100^{\circ}C$ over a 3 hr period. The PE of iodine as a representative component of the STE as well as the total polyphenol content at ambient pressure were determined as follows: 1) after soaking in the STE at 30, 40, and $50^{\circ}C$ for 3 hr, and 2) after soaking in the STE at $4^{\circ}C$ immediately after soaking in distilled water at $50^{\circ}C$ for 1, 2, and 3 hr (TCM), respectively. The PE of iodine as well as the total polyphenol content improved as the temperature of the STE increased (P<0.05). The permeation of iodine was more rapid, and the total polyphenol content increased with the TCM (121 ${\mu}g/100$ mg and 1.74 mg/mL for 3 hr, respectively) compared to without the TCM (72 ${\mu}g/100$ mg and 1.48 mg/mL for 3 hr, respectively), and the highest PE was observed with the TCM after soaking at $50^{\circ}C$ for 3 hr. At a pressure of 5.0 MPa, the PE was more rapid with the TCM (139 ${\mu}g/100$ mg and 1.74 mg/mL for 90 min, respectively) than without the TCM (105 ${\mu}g/100$ mg and 1.56 mg/mL for 90 min, respectively). Consequentially, these results suggest that the preparation of seasonings and functional eggs using various components and bioactive substances is possible by the TCM.

PET/CT SUV Ratios in an Anthropomorphic Torso Phantom (의인화몸통팬텀에서 PET/CT SUV 비율)

  • The standard uptake values (SUVs) strongly depend on positron emission tomographs (PETs) and image reconstruction methods. Various image reconstruction algorithms in GE Discovery MIDR (DMIDR) and Discovery Ste (DSte) installed at Department of Nuclear Medicine, Seoul Samsung Medical Center were applied to measure the SUVs in an anthropomorphic torso phantom. The measured SUVs in the heart, liver, and background were compared to the actual SUVs. Applied image reconstruction algorithms were VPFX-S (TOF+PSF), QCFX-S-350 (Q.Clear+TOF+PSF), QCFX-S-50, VPHD-S (OSEM+PSF) for DMIDR, and VUE Point (OSEM) and FORE-FBP for DSte. To reduce the radiation exposure to radiation technologists, only the small amount of radiation source 18F-FDG was mixed with the distilled water: 2.28 MBq in the 52.5 ml heart, 20.3 MBq in the 1,290 ml liver and 45.7 MBq for the 9,590 ml in the background region. SUV values in the heart with the algorithms of VPFX-S, QCFX-S-350, QCFX-S-50, VPHD-S, VUE Point, and FOR-FBP were 27.1, 28.0, 27.1, 26.5, 8.0, and 7.4 with the expected SUV of 5.9, and in the background 4.2, 4.1, 4.2, 4.1, 1.1, and 1.2 with the expected SUV of 0.8, respectively. Although the SUVs in each region were different for the six reconstruction algorithms in two PET/CTs, the SUV ratios between heart and background were found to be relatively consistent; 6.5, 6.8, 6.5, 6.5, 7.3, and 6.2 for the six reconstruction algorithms with the expected ratio of 7.8, respectively. Mean SNRs (Signal to Noise Ratios) in the heart were 8.3, 12.8, 8.3, 8.4, 17.2, and 16.6, respectively. In conclusion, the performance of PETs may be checked by using with the SUV ratios between two regions and a relatively small amount of radioactivity.

Efficiency Evaluation of Vegetative Filter Strip for Non-point Source Pollutant at Dense Upland Areas - Focused on Non-point Source Management Area Mandae, Gaa, and Jaun Basins - (고랭지밭 밀집지역 초생대의 비점오염 저감 효율 평가 - 비점오염원 관리지역을 중심으로 (만대지구, 가아지구, 자운지구) -)

  • A vegetative filter strip (VFS) is one of the best management practices (BMPs) to reduce pollutant loads. This study aims to assess the effectiveness of VFS in dense upland field areas. The study areas are agricultural fields in the Maedae (MD), Gaa (GA), and Jaun (JU) watersheds, where severe sediment yields have occurred and the Korean government has designated them as non-point management regions. The agricultural fields were divided into three or four clusters for each watershed based on their slope, slope length, and area (e.g., MD1, MD2). To assess the sediment trapping (STE) and pesticide reduction efficiency (PRE) of VFS, the Vegetative Filter Strip Modeling System (VFSMOD) was applied with three different scenarios (SC) (SC1: VFS with rye vegetation; SC2: VFS with rye vegetation and a gentle slope in VFS range; and SC3: VFS with grass mixture). For SC1, there were relatively short slope lengths and small areas in the MD1 and GA3 clusters, and they showed higher pollutant reduction (STE>50%, PRE>25%). For SC2 and SC3, all clusters in GA and some clusters (MD1 and MD3) in MD show higher pollutant reduction (>25%), while the uplands in JU still show a lower pollutant (<25%). With correlation analysis between geographic characteristics and VFS effectiveness slope and slope length showed relative higher correlations with the pollutant efficiency than a area. The results of this study implied that slope and slope length should be considered to find suitable upland conditions for VFS installations.

A Design of the Fuzzy Neural Network Image Recognizer

  • Neural networks have become more popular recently and are now being applied to numerous fiedls. One of the major applications of neural networks is image recognition. Various image recognition system have been proposed so far, but there is no definite solution yet. In this paper, we propose a design of Fuzzy Neural Network Image Recognizer(FNNIR). Our model uses a fuzzy neural network model, named SONN[KIM90]. This model returns the information of the number of clusters and cluster and cluster center values for a given image data ste. Unlike the well-kinwn backpropagation technique, we do not need retraining for new data. Our newly designed image recongitionsystem FNNIR that uses fuzzy merger is proposed and experimented for a sample color image.

The Optimization of Reconstruction Method Reducing Partial Volume Effect in PET/CT 3D Image Acquisition (PET/CT 3차원 영상 획득에서 부분용적효과 감소를 위한 재구성법의 최적화)

  • Purpose: Partial volume effect (PVE) is the phenomenon to lower the accuracy of image due to low estimate, which is to occur from PET/CT 3D image acquisition. The more resolution is declined and the lesion is small, the more it causes a big error. So that it can influence the test result. Studied the optimum image reconstruction method by using variation of parameter, which can influence the PVE. Materials and Methods: It acquires the image in each size spheres which is injected $^{18}F$-FDG to hot site and background in the ratio 4:1 for 10 minutes by using NEMA 2001 IEC phantom in GE Discovey STE 16. The iterative reconstruction is used and gives variety to iteration 2-50 times, subset number 1-56. The analysis's fixed region of interest in detail part of image and compute % difference and signal to noise ratio (SNR) using $SUV_{max}$. Results: It's measured that $SUV_{max}$ of 10 mm spheres, which is changed subset number to 2, 5, 8, 20, 56 in fixed iteration to times, SNR is indicated 0.19, 0.30, 0.40, 0.48, 0.45. As well as each sphere's of total SNR is measured 2.73, 3.38, 3.64, 3.63, 3.38. Conclusion: In iteration 6th to 20th, it indicates similar value in % difference and SNR ($3.47{\pm}0.09$). Over 20th, it increases the phenomenon, which is placed low value on $SUV_{max}$ through the influence of noise. In addition, the identical iteration, it indicates that SNR is high value in 8th to 20th in variation of subset number. Therefore, to reduce partial volume effect of small lesion, it can be declined the partial volume effect in iteration 6 times, subset number 8~20 times, considering reconstruction time.

  • Heat transfer phenomena during inward freezing process of the water in a horizontal cylinder were experimentally studied. The cooling temperature of a wall more significantly affects the timewise average temperature than the initial superheating temperature of the water. In addition, it was absolved that the timewise average temperature was influenced by the initial volume ratio of the water($V_l/V_{tot}$) at the same temperature conditons. One the other hand, the freezing speed of the upper part in the water-ice interface was quickly progressed due to natural convection. Furthermore, experimental observation showed that the frozen mass fraction($M_s/M_{tot}$) was influenced by the initial volume ratio of the water($V_l/V_{tot}$). It was noted that the frozen mass fraction for each $V_l/V_{tot}$ represented by $Ste^*$ and Fo.
