Streptozotocin-Induced Pro-Inflammatory Response in Rat Blood and its Attenuation by External Albumin Administration

  • Previous report has suggested that the albumin levels were reduced in the peripheral blood mononuclear cells (PBMCs) and consequently oxidative stress was elevated in streptozotocin (STZ)-induced diabetic rats as albumin is the predominant antioxidant in plasma. In this study, we suggest that the levels of pro-inflammatory cytokine such as interleukin-6 (IL-6) and tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$) were increased by approximately 3.4- and 2.9-fold, respectively, in the serum of STZ-diabetic rats, compared to those of normal rat. In addition to the cytokines, the levels of C-reactive protein (CRP) were also about 3.6-fold higher, indicating that STZ induced a pro-inflammatory response in rat blood. However, when purified rat albumin was externally co-administrated with STZ through the tail vein, the serum levels of IL-6, $TNF-{\alpha}$, and CRP were markedly reduced, although the values were still higher than those of normal (non-diabetic) rats. Albumin administration also decreased STZ-induced oxidative stress in serum and PBMCs. Moreover, the decrease in cytokine and CRP levels was dependent on the dose of injected albumin. These results suggest that STZ-induced pro-inflammation and oxidative stress in rat blood might be attenuated by treatment with exogenous albumin.

Beneficial Effect of Samgijiwhang-Tang on Diabetic Nephropathy Rats (삼기지황탕이 Streptozotocin에 의한 흰쥐의 당뇨병성(糖尿病性) 신증(腎症)에 미치는 영향)

  • Objectives: The present study was carried out to investigate the preventive effects of Samgijiwhang-Tang(SJT) on streptozotocin(STZ)-induced diabetic nephropathy. Methods: SIT was given to rats through oral administration. The experimental animals were divided into a normal group of rats, a control group of STZ-induced diabetic rats, and a sample group with SIT administration. Rehmanniae Radix Preparat is combined in the original prescription of SIT, but in this experiment, Rehmanniae Radix was combined instead of Rehmanniae Radix Preparat to compare the effects of anti-diabetic nephropathy. Experimental diabetic nephropathy was induced by the injection of STZ(60mg/kg) in the rat via the peritoneum. The effect of SIT on STZ-induced diabetic nephropathy was observed by measuring the serum level of creatinine and BUN, in addition to urine secretion of albumin for 24 hours and the level of glucose found in the urine. Anti-oxidative stress of SIT administration in a living body was estimated by measuring lipid peroxide and GSH content in the cortex of kidneys. Results: STZ induced an increase of serum creatinine, urine glucose and renal cortical lipid peroxidation was lowered by Rehmanniae Radix Preparat combined SIT administration. Conclusions: The SIT treatment showed a protective effect on the rat diabetic nephropathy model, and action mechanism of the effect was thought to be concerned with anti-oxidative stress.

  • Objectives : The present study was carried out to investigate the beneficial effect of Coicis Semen extract(CSe) on streptozotocin(STZ)-induced diabetic nephropathy rats. CSe was given to rats with oral administration. Methods : The experimental animals were divided into 3 groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with CSe treatment. Experimental diabetes was induced by the injection of STZ(60 mg/kg) to the rat via the peritoneum. The effects of CSe on STZ-induced diabetic nephropathy were observed by measuring the serum level of creatinine, BUN and uric level of glucose. Kidney level of lipid peroxidation and the activities of reduced glutathione(GSH) were also examined Results : STZ-induced increase of serum creatinine was lowered by CSe treatment, but BUN and uric level of glucose did not show significant changes. CSe oral administration showed statistical decrease of lipid peroxidation in renal cortical tissues, but it has no effect on the activities of GSH. Conclusion : CSe treatment showed protective effect on rat diabetic nephropathy model, but action mechanism of the effect was still not dear. We thought to be concerned with anti-oxidative stress.

Changes of insulin like growth factor-I, IGF-I carrier protein in streptozotocin-induced diabetic rat (Streptozotocin에 의해 유도된 당뇨쥐의 IGF-I, IGFBPs 및 IGF-I carrier protein의 변화)

  • This study was conducted to investigate the effects of streptozotocin-induced (STZ) diabetes on insulin-like growth factor-I (IGF-I), insulin-like growth factor binding proteins (IGFBPs), and IGF-I carrier proteins in serum, liver, and kidney. The levels of total and free IGF-I were measured by radioimmunoassay (RIA). The patterns of IGFBPs were determined by western ligand blotting (WLB) analysis. The profiles of IGF-I carrier proteins in serum were determined by column chromatography. The levels of total and free IGF-I in serum were lower in STZ-induced diabetic rat than normal rat (p<0.01). Similarly, the levels of total IGF-I in liver was lowered in STZ-induced diabetic rats. On the other hand, the levels of total IGF-I in kidney were increased in STZ-induced diabetic rats compared with normal rats (p<0.01). In serum and liver from STZ-induced diabetic rats, the amount of IGFBP-3 was decreased and the amount of IGFBP-2 was increased compared with normal rats. There was a not difference in amount of IGFBP-4 in serum between STZ-induced diabetic rats and normal rats. The serums of normal rats have higher 150kDa carrier proteins than in STZ-induced diabetic rats, whereas, most of 50kDa carrier proteins were found in STZ-induced diabetic rats. These results demonstrate that in STZ-induced diabetic rats, IGF-I/IGFBPs system that included functional bioactivity was changed in serum as well as tissues, and these changes may play an important role in pathogenesis of diabetes.

Methods : AD-1 consists of eleven herbs that have an effect on diabetes mellitus. AD-1 was given to rats with the combination of oral administration and herbal-acupuncture stimulation. The experimental animals were divided into 3 groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with AD-1 treatment. Experimental diabetes was induced by the injection of STZ(60mg/kg) to e rat via the peritoneum. The effect of AD-1 on STZ-induced diabetic nephropathy was observed by measuring the serum level of creatinine and BUN.   • The purpose of this study was to investigate the effect of pretreatment with Saegchinyanghyoltang(SYT) on the serum lipid composition and atherosclerotic index in streptozotocin(STZ)-induced diabetic rats. SYT pretreatment in STZ-induced diabetic rats inhibited the rise of serum glucose concentration. Serum total lipids and triglyceride levels in the STZ -induced diabetic rats were significantly higher than those in the control group. But in the group pretreated with SYT, triglyceride and lipid levels were significantly lower compared with those of STZ -induced diabetic rat group without STZ. However, the serum phospholipid levels were not statistically different among treatment groups. In the STZ-induced diabetic group, the serum total cholesterol, VLDL-, LDL-cholesterol levels and atherosclerotic index Were higher and HDL-cholesterol level was lower compared to the control group. However, these changes were prevented by SYT pretreatment Pretreatment with SYT significantly increased the activities of serum lipase compared to the STZ-treated group.

The Effects of Bojungikgi-tang on Streptozotocin-induced Diabetic Gastroparesis Rat Model (보중익기탕(補中益氣湯)이 streptozotocin 유발 당뇨병성 위부전마비 백서에 미치는 영향)

  • Objective : Diabetic gastroparesis is a complication that is defined as delayed gastric emptying and upper gastrointestinal symptoms and often occurs in long-standing diabetic patients. Bojungikgi-tang (BJT) is a traditional oriental herbal formula that has long been used for the treatment of digestive disorders. The purpose of this study was to investigate the effects of BJT on streptozotocin (STZ)-induced diabetic gastroparesis rat model. Methods : Sprague-Dawley (SD) male rats (250-270g) were divided into 13 groups including normal group, STZ-induced diabetic control group, BJT diet (7 various concentrations), and insulin-, glibenclamide-, metformin-treated group were used for the experiments for the comparison. Diabetic gastroparesis was induced by intraperitoneal injection of STZ. The water intake, food intake, body weights and fasting blood glucose levels were measured. After 4 weeks the animals were sacrificed and gastrin, leptin, insulin, hemoglobin A1C (HbA1c), lactate, lactate dehydrogenase (LDH), bilirubin, creatinine, albumin and lipid levels were evaluated. Results : Intraperitoneal injection of BJT for 4 weeks resulted in increased levels of gastrin in blood and decreased leptin and lactate concentration in STZ-induced diabetic gastroparesis rat model. BJT did not affect insulin, fasting glucose, HbA1c, and lipid levels in STZ-induced diabetic gastroparesis rat model. Conclusion : These results indicated that BJT would have protect effect on diabetic gastroparesis through the improvement effect of gastric motility and fatigue syndrome in STZ-induced diabetic rats. This study shows that BJT might be effective for treatment of diabetes and its complications such as gastroparesis.

The Effects of Nicotinamide on the Serum Lipid Composition in Streptozotocin-Induced Diabetic Rats (Nicotinamide가 Streptozotocin 당뇨성 쥐의 혈중 지질 성분에 미치는 영향)

Aminotranferase (ALT, AST) activities were not significantly different in any of the groups.

Silymarin, a flavonoid antioxidant, protects streptozotocin-induced lipid peroxidation and β-Cell damage in rat pancreas

  • The present study is aimed at finding the influence of silymarin (a flavonoid) (25 mg/kg & 50 mg/kg) in streptozotocin (STZ)-induced diabetic rats. Type 2 diabetes was induced by single intraperitoneal injection of STZ (100 mg/kg) to 3 days old rat pups. Silymarin was administered for 15 days after the animals were confirmed diabetic (75 days after STZ injection). Blood glucose, glycosylated hemoglobin ($HbA_{1c}$), lipid peroxides (LPO) levels and reduced glutathione (GSH) contents in pancreas and liver were estimated following the established procedures. Biochemical observations were further substantiated with histological examination of pancreas. Blood glucose and $HbA_{1c}$ levels, which were elevated by STZ, were lowered to physiological levels by the administration of silymarin. The levels of LPO were significantly increased in STZ-induced diabetic rats. Silymarin reduced the LPO levels in both pancreas and liver. GSH contents which were reduced significantly in pancreas and liver of STZ-induced diabetic rats were brought back to near normal levels by silymarin treatment. Multifocal necrotic and degenerative changes of pancreas in STZ-diabetic rats were minimized to near normal morphology by administration of silymarin as evident by histopathological examination. Silymarin showed a dose dependent protective effect on STZ-induced $\beta$-cell damage. It could be attributed to the antioxidative and free radicals scavenging properties of the flavonoid. Thus, it may be considered as a natural antioxidant with potential therapeutic application in the treatment of type 2 diabetes.
