A Study of Clinical Effect in Kwi-Bi-Tang and Sa-Mul-An-Shin-Tang (귀비탕(歸脾湯) 및 사물안신탕(四物安神湯)의 임상효과(臨床效果)에 관(關)한 연구(硏究))

  • In order to observe clinical effects of Kwi-Bi-Tang(歸脾湯) and Sa-Mul-An-Shin-Tang(四物安神湯), I reached following conclusion through the physiochemical investigation the following results obtained. 1)In serum Lipid, only Total cholesterol is significantly decresed in medication group of Kwi-Bi-Tang(歸脾湯), but is not in the medicaton of Sa-Mul-An-Tang(四物安神湯). 2). The value of Cardiac enzyme is remarkably decreased in the medication group of Kwi-Bi-Tang(歸脾湯), Only AST is significantly decresed in the medication group of Sa-Mul-An-Tang(四物安神湯). 3) In determination of LDH isoenzyme, LDI is significantly decrease both in the medication group of Kwi-Bi-Tang(歸脾湯) and Sa-Mul-An-Shin-Tang(四物安神湯). Especially, LDI is remarkably decreased in the medication group of Kwi-Bi-Tang(歸脾湯). In view of the results so far achieved, we knew that Kwi-Bi-Tnag(歸脾湯) and Sa-Mul-An-Shin_Tang(四物安神湯) had improved ischemic condition of cardiac muscle, specially, Kwi-Bi-Tnag(歸脾湯) was significant compared to Sa-Mul-An-Shin-Tang(四物安神湯).

Suppressive Effect of SaengRyoSaMulTang on Activated RBL-2H3 Mast Cells (RBL-2H3 비만세포의 알레르기 염증 반응에 미치는 생료사물탕(生料四物湯)의 영향)

  • Objectives: SaengRyoSaMulTang is a herbal formula in Oriental Medicine, known anti-allergens. However, its mechanism and cellular targets have not been found yet. Thus the study has developed to investigate the suppressive effect of SaengRyoSaMulTang. Methods: In the study, cellular viability, IL-4, IL-13 mRNA expression, IL-4, IL-13 production, manifestations of GATA-1, GATA-2, NF-AT1, NF-AT2, AP-1 and NF-${\kappa}$B p65 transcription factors were examined by Real-Time PCR, ELISA analysis and western blotting. Results: As a result of treating with SaengRyoSaMulTang extract(SRSMT), the study has shown that the amount of Th2 cytokines, which include PI induced IL-4 and IL-13, plays a significant role in suppressing effect. RBL-2H3 mast cells significantly suppressed the PI-induced Th2 cytokine production including IL-4 and IL-13 in a dose dependent manner. PI-induced IL-4 and IL-13 production was significantly suppressed by SRSMT intervention. Western blot analysis of transcription factors involving IL-4 and IL-13 expression also revealed a prominent decreases of mast cell's specific transcription factors including GATA-1, GATA-2, NF-AT2, c-Jun and c-Fos, but NF-${\kappa}$B p65. Conclusions: The study suggests that the anti-allergenic activities of SRSMT may regulate the transcription factors GATA-1, GATA-2, NF-AT2, c-Jun and c-Fos inhibiting Th2 cytokines IL-4 and IL-13 in mast cells.

Anxiolytic Effects of the Three Kinds of Traditional Chinese Medicine, Shin-Ki-Hwan, Bo-Jung-lk-Ki-Tang, and Sa-Mul-Tang, Using the Elevated Plus-maze Test (Elevated plus-maze를 이용한 신기환, 보중익기탕 및 사물탕의 항불안 효과)

  • Shin-Ki-Hwan (Shen-Qi-Wan, SKH), Bo-Jung-Ik-Ki-Tang (Bu-Zhong-Yi-Qi-Tang, BJIKT), and Sa-Mul-Tang (Si-Wu-Tang, SMT) have been used for various kinds of deficiency syndromes, such as 'yang', 'qi', and 'blood', respectively. The objects of this study were to determine the effects of water extracts of three different kinds of traditional Chinese medicine (TCM), SKH, BJIKT, and SMT, on the anxiolytic activities in the elevated plus-maze test and to clarify the differences among 'yang', 'qi', and 'blood'. The water extracts of SKH, BJIKT, and SMT were orally administered to male SD rats, at 1.0 g/kg for 10 days. All rats were subjected to behavioral tests for the anxiolytic activity at 10 days. SKH, for the benefiting 'yang'agents, significantly increased the ratio of open arms entry to the total arms entry and time spent in the open arms (p<0.05), suggesting anxiolytic effect. However, both BJIKT and SMT decreased the ratio of open arms entry to the total arms entry and increased times spent in the closed arms (p<0.05). From these findings, it can be speculated that SKH only exhibits anxiolytic effect and that the different anxiolytic effects in the elevated plus-maze test may be come from the meanings of 'yang', 'qi', and 'blood'in oriental diagnostics though the cases are restricted.

The Effect of DanSamSaMul-Tang on Hematopoiesis (단삼사물탕이 조혈작용에 미치는 영향)

  • Objectives : Korean traditional medicinal herbs and some formulas have been used to improve the function of the hematopoietic system. The present research is to examine the effect of DanSamSaMul- Tang(DSSMT) as a biological response modifier related to hemato-potentiating function using bone marrow cell as a testing system. Methods : Mice with leukopenia and thrombocytopenia induced by CTX were treated with DSSMT Extracts. Then we observed cytokines mRNA expression level, the number of CFU-GEMM and BFU-E, and the expression of TPO and SCF in bone marrow cells of mice. Results : In the DSSMT group, the gene expressions of hematopoietic cytokines were increased significantly and the number of CFU-GEMM and BFU-E of this group was significantly increased compared to the group treated only with EPO and IL-3. The expressions of TPO and SCF in the group treated with DSSMT were higher than those of the control group. Conclusions : We showed that DSSMT has a good effect on the hematopoietic system. This could also be used as important research data for Korean oriental medicine about hemato-potentiating.

The effects of Sa-Mul-Tang (Si-Wu-Tang), a Traditional Chinese Medicine, on Phenylhydrazine-induced Anemic Rats

  • Sa-Mul-Tang (Si-Wu-Tang, SMT), a kind of Chinese medicine, has been used for the hemato-deficient disease for hundreds of years. In this work, investigations on the anti-anemic activity of an aqueous extract of SMT were undertaken in order to find the pharmacological basis for the ethnomedical use of the formulation. Three kinds of Angelicase species, such as Angelica sinensis, Angelica acutiloba, and Angelica gigas, were used for preparing the water extracts of SMT. Anemic model rats were induced by the treatment of phenylhydrazine (40 mg/kg/day, i.p.) for 4 days. After the treatment of phenylhydrazine, rats were divided into several groups for their different treatment of three kinds of SMT. Red blood cell (RBC), hemoglobin (Hg), and hematocrit (Hct) were determined on the day 0, 3, 6, 10, 14 after the treatment of SMTs and erythrocytes deformabilities were also determined at the end of experiments. Oral administration of SMT (1g/kg/day) for 14 days did not ameliorate drug-induced anemic states evaluated by RBC counts, Hg contents, and Hct values. However, the erythrocyte deformabilities were improved in phenylhydrazine-treated group by the administragest that SMTs (p<0.05). Especially, these effects were high in the Angelica acutiloba group. These results suggest that SMTs have an ameliorative effect on blood rheology related to the blood stasis syndrome in oriental diagnostics not on the blood deficient states related to the anemic syndrome.

Effects of Sa-Mul-Tang on Immunocytes of L1210 Cells-transplanted or Antitumor Drugs-administered Mice (사물탕이 L1210 세포 이식 및 항암제를 투여한 마우스의 면역세포에 미치는 영향)

  • Sa-Mul-Tang(SMT) consist of Rehmanniae Radix Preparata, Paeoniae Radix Alba, Cnidii Rhizoma and Angelicae Gigantis Radix. In L1210 cells-transplanted BALB/c mice, T-lymphocyte apoptosis, $CD8^+T_C$ cells population in thymocyte and nitric oxide production in macrophage were enhanced, but phagocytic activity was decreased. SMT suppressed T-lymphocyte apoptosis and enhanced CD^4+T_H$ cells population, but did not affect nitric oxide production and phagocytic activity in L1210 cells-transplanted mice. In antitumor drugs-injected mice, T-lymphocyte apoptosis was enhanced, but $CD4^+T_H/CD8^+T_C$, cells population and T-lymphocyte proliferation were decreased. SMT suppressed T-lymphocyte apoptosis, and enhanced $CD8^+T_C$ cells population, T-lymphocyte proliferation and phagocytic activity in vincristine-injected mice. These results suggest that SMT enhances T cell-mediated immunity in L1210 cell-transplanted mice, and enhances T cell-mediated immunity and phagocytic activity in vincristine-injected mice.

Effect of Si-Wu-Tang and Si-Jun-Zi-Tang on the Survival of Jejunal Crypt Cells and Hematopoietic Cells in Irradiated Mice (방사선조사 마우스에서 소장움세포 및 조혈세포 생존에 미치는 사물탕 및 사군자탕의 영향)

  • In order to investigate the radioprotective effect of Si-Wu-Tang (Korean name: Sa-Mul-Tang), a kind of traditional Oriental medicine as a blood-building decoction (Oriental medical concept: Bu-Xie), and Si-Jun-Zi-Tang (Korean name: Sa-Gun-Ja-Tang), one of the widely used Oriental herbal medicines as an energy tonic (Chinese medical concept: Bu-Qi). the jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells were observed in irradiated mice. Jejunal crypts were protected by Si-Wu-Tang pretreated both per os (2 mg/mL of drinking water for 7 days, p<0.05) and intraperitoneally (1 mg/head, single injection at 24 hours before irradiation). Si-Wu-Tang adminstration before irradiation(1 mg/head, single injection at 24 hours before irradiation) resulted in an increase of the formation of endogenous spleen colony (p<0.005). The frequency of radiation-induced apoptosis in intestinal crypt cells was also reduced by pretreatment of Si-Wu-Tang (p<0.01). However, the radioprotective effect of Si-Jun-Zi-Tang was not as significant as that of Si-Wu-Tang. These results suggest that Si-Wu-Tang may be a useful radioprotective food, especially since it is a relatively nontoxic natural product.

  • Among oriental medicine,s literatures, through Hwang-Je-Nae-Kyung(黃帝內徑) to Chung(淸), I extracted contents related to atrophy syndrome(?證). And studied it,s pathophysiology, therapy and treatment. Then, I concluded that result same below 1. The pathophysiologies of atrophy syndrome are lung heat(肺熱) & decreasing of it,s circulation, making liver and stomach weaken & difficulty it's circulation, injurious to liver and kidney & atrophy of bonemarrow and muscle. 2. The most important point of atrophy syndrome therapy is Yang-Myung(陽明). The Priority of therapy is stomach & liver,s balance. And then we must protect acquired human function & clean humidity & temperature of Yang-Myung(陽明). under the principle of decreasing south organ,s function & protecting north organ,s function, we should Ja-Yeum-Chung-Yeul(滋陰淸熱). so remove temperature of lung & protect liver and kidney & make strong stomach. 3. Among the therapy of atrophy syndrome in literatures Yi-Jin-Tang(二陳湯), Sa-Gun-Ja-Tang(四君子湯), Sa-Mul-Tang-Je(四物湯劑), Ho-Jam-Hwan(虎蠶丸), Dong-Won-Geun-Bo-Hwan(東垣健步丸) and Chung-Jo-Tang(淸燥湯) were many. These make strong spleen & dry humidity organ using Sa-Gun-Ja-Tang(四君子湯)and Yi-Jin-Tang(二陳湯) by Dog-Cheu-Yang-Myung(獨取陽明) method. Sa-Mul-Tang(四物湯), Phellodendri cortex(黃柏), Radix sophorae flavescentis(苦蔘), Carapax Testudinis(龜板) bitter taste make strong Yeum(陰) & decrease Yang(陽) so important human muscle powerful. Ho-Jam-Hwan(虎蠶丸), Dong-Won-Geun-Bo-Hwan(東垣健步丸), Chung-Jo-Tang(淸燥湯) make Chung-Yeul-Jo-Seup(淸熱燥濕), protect liver and kidney & strong muscle and bone. Besides Gum-Gang-Hwan(金剛丸), Yi-Myo-Hwan(二妙丸), Nok-Gak-Geu-Hwan(鹿角膠丸)&Ga-Mi-Sa-Geun-Hwan(加味四斤丸) were used in treatment of atrophy syndrome.

Effect of Several Combined Preparation of Crude Drugs on the Adverse Effects of Anticancer Agent-Mitomycin C (항암제 Mitomycin C의 부작용에 대한 수종 복합생약의 영향)

  • The studies were conducted to investigate the combined effects of several combined preparation of crude drugs and mitomycin C(MMC). The combined effects on the proliferation of HepG2, A549, KHOS-Np, A431 and HeLa cells were estimated by MTT colorimetric assays. Sa Kunja Tang(SKT), Boyang Hwanoh Tang(BHT) and Hyulbu Choogo Tang(HCT) inhibited the proliferation of A549 and HeLa cell. The inhibitory action of MMC was increased by the combined treatment of SKT and MMC, and Sa Mul Tang(SMT) and MMC, respectively. When the mice were treated by MMC, the number of leukocyte was decreased significantly at the 3rd day, but recovered at the 7th day. In the groups of MMC treated with SKT or HCT, the number of leukocyte was increased significantly that the group of MMC treated only at the 1st and 3rd day. The combined treatment of SKT, SMT, BHT, HCT and MMC retained the spleen weight of mice at the level of normal mice, but decreased the thymus weight of mice. The combined treatment of SKT, SMT, BHT, HCT and MMC increased the number of PFC significantly than the MMC treated group. The combined treatment of SKT, SMT, BHT, HCT and MMC increased the T cell proliferation significantly thant the MMC treated group.
