α-Glucosidase Inhibitory Effects for Solvent Fractions from Methanol Extracts of Sargassum fulvellum and Its Antioxidant and Alcohol-Metabolizing Activities (참모자반 메탄올 추출 분획물의 항산화 및 숙취해소능과 α-glucosidase 활성저해효과)

  • We investigated the physiological activity and solvent-partitioned fractions of methanol extracts from the green seaweed Sargassum fulvellum. The methanol extract from S. fulvellum was sequentially fractionated with n-hexane (SFMH), methanol (SFMM), buthanol (SFMB), and water (SFMA). We investigated the antioxidant activities of solvent fractions from S. fulvellum by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity and an SOD activity assay. DPPH radical scavenging capacity of SFMM was 79.5% at 10 mg/ml. SOD activity of SFMM was 79.9% at 10 mg/ml. Nitrite scavenging activities of solvent fractions from S. fulvellum were investigated under different pH conditions and showed the most remarkable effect at pH 1.2. In particular, the activity of SFMB was higher than the other fractions. ADH activity and ALDH activity of SFMM were 177.0% and 167.4% at 10 mg/ml, respectively. ${\alpha}$-Glucosidase inhibitory activity of SFMH increased in a dose-dependent manner and was about 94.1% at 2 mg/ml. Elastase inhibitory activity was 93.2% at 2 mg/ml. These results revealed that S. fulvellum extracts have strong antioxidant and alcohol dehydrogenase activities and ${\alpha}$-glucosidase inhibitory activity, suggesting that S. fulvellum extracts have potential as a source of natural products for health and beauty.

Effects of pH and Salinity on the Cultivated Brown Alga Sargassumfulvellum and Associated Animals (갈조류 양식 모자반(Sargassum fulvellum)과 해적생물에 대한 pH와 염분의 효과)

  • The effects pH and salinity on the brown alga Sargassum fulvellum were investigated in the context of the removal of two major associated animals, Caprella scaura and Gammaropsis utinomi. Optimum quantum yield (Fv/Fm) of S. fulvellum was also examined in the same experimental conditions as an index of stress. Experiments on pH and salinity tolerances of the two animal species indicated that mortality was more than 80% at extremes of pH (2, 3, 4, 10, 11, 12) and salinity (0, 3.5, 7, 10, 44 psu) after a 5 min treatment. Lethal time (LT50) from pH 2 to pH 4 was less than 90 sec in C. scaura, and less than 70 sec in G. utinomi. From 0 to 10 psu, LT50 was less than 20 sec in C. scaura and less than 60 sec in G. utinomi. The quantum yield of S. fulvellum was not significantly different from controls within the pH range 4~10, and within the salinity range of 7-40 psu. When exposed to pH and salinity conditions outside these ranges, the effect of these factors on the removal of two animal species was higher, but quantum yield was highly reduced. These results indicate that the optimal conditions for removing the animal species without affecting optimum quantum yield were pH 4-10, and salinities 7-10 psu and 44 psu.

Anti-inflammatory Activity of the Water Extract of Sargassum fulvellum (참모자반 (Sargassum fulvellum) 물 추출물의 염증 억제 활성)

  • The anti-inflammatory effects of Sargassum fulvellum water extracts (SFWE) were investigated using lipopolysaccharide (LPS)-induced inflammatory response in this study. To examine the potential anti-inflammatory properties of SFWE, the NO, TNF-${\alpha}$, IL-6, IL-$1{\beta}$, and cell proliferation were measured. It was confirmed that the NO and TNF-${\alpha}$ secretion were significantly suppressed when SFWE was added to LPS-stimulated RAW 264.7 cells. Moreover, the expression of IL-6 and IL-$1{\beta}$ cytokines was suppressed by SFWE in a dose-dependent manner. Especially, IL-6 inhibition activities were over 50% at 1% of SFWE. The cytotoxicity of SFWE and the proliferation of macrophages was measured by MTT assay. As a result, there was no cytotoxicity in the macrophage proliferation treated with SFWE compared to the control. In conclusion, these results suggested that the SFWE may have significant effects on inflammatory factors and can be a potential anti-inflammatory therapeutic materials.

Production of Total Reducing Sugar and Levulinic Acid from Brown Macro-algae Sargassum fulvellum (거대 갈조류 모자반으로부터 환원당과 레불린산의 생산)

  • Recently, many biofuels and chemicals converted from renewable resources have been introduced into chemical industries. Sargassum fulvellum is a brown macro-algae, which is found on the seashores of Korea and Japan. In this work, the production of total reducing sugar and levulinic acid from S. fulvellum, using dilute-acid catalyzed hydrothermal hydrolysis and statistical methodology, was investigated. As a result, 15.28 g/l total reducing sugar was obtained under the optimized conditions of $160.1^{\circ}C$, 1.0% sulfuric acid, in 20.2 min. Furthermore, 2.65 g/l levulinic acid was obtained at $189.5^{\circ}C$, 2.93% sulfuric acid, in 48.8 min.

Chemical Properties of Fucoidans from Hizikia fusiformis and Sargassum fulvellum (톳과 모자반 fucoidan의 화학적 특성)

  • Fucoidans were isolated from Hizikia fusiformis and Sargassum fulvellum and characterized on their chemical properties. Crude fucoidans were extracted at $65^{\circ}C$ for 1hr with acid solution of pH 2.0, and cetylpyridinum chloride was used for partial purification. The yields of partially purified fucoidans were $2.51\%$ for H. fusiformis, and $65\%$ for S. fulvellum, respectively. The partially purified fucoidans were separated into 3 fractions by DEAE-Sephadex A-25 ion exchange column chromatography and the major fractions were refractionated with fractional preripitation with ethanol. $60-70\%$ ethanol precipitated fraction(P-70) of H. fusiformis and $60-65\%$ ethanol precipitated fraction(P-65) of S. fulvellum turned out to be homogeneous by cellulose acetate electrophoresis and gel filtration chromatography. The molar ratios of fucose, galactose, and sulfate In the purified fucoidans were 1 : 0.66 2.74 for H. fusiformis and 1 : 0.24. 1.46 for 5. fulvellum. The averaged molecular weights of the purified fucoidans from H. fusiformis and S. fulvellum were 26,000 and 105,000, respectively.

Inhibitory effect of Mojaban (Sargassum fulvellum) extract on ultraviolet B-induced photoaging in hairless mouse skin (무모 마우스 피부에서 모자반 추출물의 자외선비(ultraviolet B)에 의해 유도된 광노화 억제 효과)

  • Ultraviolet radiation is a major cause of photoaging, characterized by wrinkle formation and skin dehydration. Mojaban (Sargassum fulvellum), belonging to the brown algae of the Sargassaceae family, exerts anti-inflammatory, antioxidant, and anti-atopic activities. Particularly, Mojaban extract alleviated ultraviolet B (UVB)-induced inflammatory responses in human keratinocytes and BALB/c mice. Here, the dorsal skin of hairless mice was exposed to UVB for photoaging. The mice were administrated 300 mg/kg Mojaban extract (S. fulvellum extract; SFE) daily. Phenotypically, SFE reduced wrinkle formation and epidermal thickness and increased skin elasticity. SFE increased hydroxyproline content by upregulating the expression of collagen-related genes and downregulating matrix metalloproteinase expression. Furthermore, SFE increased skin moisture content and inhibited transepidermal water loss. SFE upregulated the expression of involucrin, loricrin, and transglutaminase for cornified envelope formation as well as the expression of filaggrin and caspase-14 for natural moisturizing factors. Collectively, SFE may be employed as a nutraceutical ingredient for anti-photoaging.

Characteristics of Lead Biosorption by Biosorbents of Marine Brown Algae (해양 갈조류를 생물흡착제로 이용한 납흡착 특성 연구)

  • Lead sorption performances by biomass of nonliving, dried marine brown algae Undaria phnaunda, Hlzikia hsyormls. and Sugassum fulvellum used as biosorbent materials were investigated. As the amount of biosorbent materials added was increased, the lead removal by biosorbent materials Increased but the lead biosorption capacities decreased. However, with increasing Initial lead concentration the lead biosorption capacities by the biosorbent materials Increased but lead removal efficiencies decreased. In the range of Initial lead concentration(Co) 10-500 mg/L the lead biosorption capacities and removal efficiencies by the biosorbent materials Increased with increasing pH. Among the biosorbent materials used in this study, the lead biosorption capacities decreased in the following sequence: U. plilnaunda > H. fusiformis > S. fulvellum. The lead biosorption by biosorbent materials were expressed by the Langmuir Isotherm better than the Freundlich Isotherm. The biosorption rate could be expressed by the first order reaction rate equation for initial lead concentration like that rad : 0.288Co for U. phanda, rad = 0.255Co for H. fusiformis, and rad : 0.161Co for S. fulvellum. Anti-Inflammatory Effects of Grasshopper Ketone from Sargassum fulvellum Ethanol Extract on Lipopolysaccharide-Induced Inflammatory Responses in RAW 264.7 Cells

  • This study evaluated the anti-inflammatory potential of a grasshopper ketone (GK) isolated from the brown alga Sargassum fulvellum on lipopolysaccharide (LPS)-induced RAW 264.7 murine macrophage cell line. GK was isolated and purified from the n-hexane fraction and its structure was verified on the basis of NMR spectroscopic data. GK up to $100{\mu}g/ml$ is not cytotoxic to RAW 264.7, and is an effective inhibitor of LPS-induced NO production in RAW 264.7 cells. The production of pro-inflammatory cytokines, including IL-6, $IL-1{\beta}$, and $TNF-{\alpha}$ was found significantly reduced in $0.1-100{\mu}g/ml$ dose ranges of GK treatment (p < 0.05). We confirmed the dose-dependent and significant inhibition of iNOS and COX-2 proteins expression. In addition, it has been shown that GK induces anti-inflammatory effects by inhibiting MAPKs (ERK, JNK, and p38) and $NF-{\kappa}B$ p65 phosphorylation. Our results show that the anti-inflammatory properties of GK may be due to the inhibition of the $NF-{\kappa}B$ and MAPKs pathways, which are associated with the attenuation of cytokine secretion.
