Optimization of Extraction Process to Increase the Antioxidant Activity of Sargassum serratifolium Extracts (톱니모자반(Sargassum serratifolium) 추출물의 항산화 활성 증가를 위한 추출공정 최적화)

  • Sargassum serratifolium is a rich source of antioxidant meroterpenoids. The optimal extraction temperature and time for obtaining maximal antioxidant yield, antioxidant activity, and phenolic content from Sargassum serratifolium were determined using response surface methodology (RSM). The ranges of the independent variables for extraction temperature and time were 30-70℃ and 12-36 h, respectively. With increasing temperature and time, the yield increased significantly, while DPPH (2,2-dipheny-1-picrylhydrazyl) radical-scavenging activity and total phenolic content decreased significantly. The optimal extraction temperature and time obtained by RSM were 54℃ and 7 h, respectively, providing a yield of 8.2%, DPPH radical-scavenging activity of 60%, and total phenolic content of 163 mg GAE/g. The findings of this study provide useful information for the development of Sargassum serratifolium extraction processes for the food and cosmetic industries.

Effect of Sargassum serratifolium Extracts on β-Amyloid Production (β-아밀로이드 단백질 생성에 대한 톱니모자반(Sargassum serratifolium) 추출물의 효과)

  • Alzheimer's disease (AD) is a progressive neurodegenerative disorder of insidious onset that causes gradual loss of memory and cognitive function, and it is the most common form of dementia in the elderly. AD is characterized by neuritic plaques and neurofibrillary tangles in the brain, together with loss of neuronal cells. The major neuropathological hallmark of AD is the accumulation of extracellular neurotoxic ${\beta}-amyloid$ ($A{\beta}$) peptides, such as $A{\beta}1-42$, in the brain. In the present study, we investigated the effect of sargachromenol (SCM), sargaquinoic acid (SQA) and sargahydroquinoic acid (SHQA) isolated from Sargassum serratifoilum ethanol extract (SSE) on $A{\beta}$ production in vitro using APP751-transfected Chinese hamster ovary cells (CHO-751). CHO-751 cells were treated with various concentrations of SSE, SCM, SQA and SHQA, and the level of extracellular $A{\beta}1-42$ was evaluated by enzyme-linked immunosorbent assay. SSE and SHQA reduced the production of $A{\beta}1-42$ in CHO-751 cells. Therefore, SHQA isolated from S. serratifolium has potential as an inhibitor of neurotoxic $A{\beta}$ peptide production.

Effect of a Sargassum serratifolium Extract on Neuroinflammation Induced by Lipopolysaccharides in Mice (LPS로 유도한 마우스의 급성신경염증에 대한 톱니모자반(Sargassum serratifolium) 추출물의 효과)

  • The common hallmark of several neurodegenerative disorders, including Alzheimer's disease (AD), is the presence of chronic neuroinflammation, which contributes to the loss of neuronal structure and function. This study investigated the effects of an ethanolic extract of Sargassum serratifolium (SSE) in a lipopolysaccharides (LPS)-induced murine neuroinflammation model. Mice were administered SSE (100 mg/kg body weight) or vehicle for 5 days by oral gavage, and then treated with LPS or saline by intraperitoneal injection. Thereafter, the brain tissues were collected, and the expression of pro-inflammatory cytokines was analyzed by quantitative real-time RT-PCR. There was a marked increase in the spleen weight index in the LPS-treated groups, which indicated the induction of acute systemic inflammation. Based on significant increases in the levels of IL-1 and IL-6 expression, the induction of neuroinflammation was also evident in the cortex and hippocampus of the LPS-treated groups. The overall expression of IL-1 and IL-6 was decreased slightly by SSE administration, compared with the LPS group, and a marked change in IL-1 was observed in the cortex of the SSE-treated (SSE/LPS) group. These results suggest that SSE has potential as an anti-neuroinflammatory nutraceutical.

Hexane fraction from the ethanolic extract of Sargassum serratifolium suppresses cell adhesion molecules via regulation of NF-κB and Nrf2 pathway in human umbilical vein endothelial cells

  • Sargassum serratifolium ethanolic extract has been known for strong antioxidant and anti-inflammatory properties. We prepared hexane fraction from the ethanolic extract of S. serratifolium (HSS) to improve biological activities. In this study, we investigated the effects of HSS on the inhibition of tumor necrosis factor (TNF)-${\alpha}$-induced monocyte adhesion to human umbilical vein endothelial cells (HUVECs). We found that HSS suppressed the production of cell adhesion molecules such as intracellular adhesion molecule-1 and vascular cell adhesion molecule-1 in TNF-${\alpha}$-induced HUVECs. Moreover, TNF-${\alpha}$-induced production of monocyte chemoattractant protein 1 and keratinocyte chemoattractant was inhibited by HSS treatment. HSS suppressed TNF-${\alpha}$-induced nuclear factor kappa B ($NF-{\kappa}B$) activation via preventing proteolytic degradation of inhibitor ${\kappa}B-{\alpha}$. HSS induced the production of heme oxygenase 1 via translocation of Nrf2 into the nucleus in TNF-${\alpha}$-treated HUVECs. Overall, HSS alleviated vascular inflammation through the downregulation of $NF-{\kappa}B$ activation and the upregulation of Nrf2 activation in TNF-${\alpha}$-induced HUVECs. These results indicate that HSS may be used as therapeutic agents for vascular inflammatory disorders.

  • Eight species of Sargassum subgenus Bactrophycus section Halochloa and Repentia were collected from the coasts of Korea: Sargassum coreanum J. Agardh, Sargassum sagamianum Yendo, Sargassum micracanthum (Kützing) Endlicher, Sargassum macrocarpum C. Agardh, Sargassum autumnale Yoshida, Sargassum siliquastrum (Mertens ex Turner) C. Agardh, Sargassum serratifolium (C. Agardh) C. Agardh, Sargassum yezoense (Yamada) Yoshida et Konno. These species were investigated on morphological variations and geographical distributions in Korea. The valuable taxonomic characters between similar species of these section were discussed with special reference to types of holdfasts, shape of main branch, leaves and vesicles.

Synergistic Antimicrobial Effect of Sargassum serratifolium (C. Agardh) C. Agardh Extract against Human Skin Pathogens (피부 병원균에 대한 톱니모자반 추출물의 항균 시너지 효과)

  • The object of this study was to develop an alternative way to treat human skin pathogens using marine algae. During this study, we observed that the ethanolic extract of the edible brown algae [Sargassum serratifolium (C. Agardh) C. Agardh] exhibited potential antimicrobial activity against pathogenic commensal bacteria related with acne vulgaris (Propionibacterium acnes, Staphylococcus epidermidis, Staphylococcus aureus and Pseudomonas aeruginosa), and Candida albicans which causes cutaneous candidiasis. Among the solvent-soluble fractions from the ethanolic extract, a hexane-soluble fraction showed the strongest antimicrobial activity against all tested human skin pathogens with MIC values ranging from 32 to $512{\mu}g/mL$. In addition, the hexane fraction exhibited a synergistic antimicrobial activity with commercial antibiotics used in the treatment of acne vulgaris or cutaneous candidiasis. Thus, this study suggests that S. serratifolium extract could be a potential source of natural antimicrobial agents or a pharmaceutical component against human skin pathogens.

Studies on Bioactive Substances and Antioxidant Activities of Marine Algae from Jeju Island (제주도 연안 서식 해조류에 대한 주요 활성성분 및 항산화효능 연구)

  • The objective of this study was to investigate the useful components and potential antioxidant activities of 30 marine algae, collected from Jeju Island in Korea. The contents of bioacitve substance of extracts from marine algae, such total polyphenol and carbohydrate, were determined. The extract of Distromium decumbens had the highest amount of total polyphenol content (21.27%), and that of Gracilaria incurvata Okamura had the highest amount of total carbohydrate content (10.18%). The antioxidant activities of extracts obtained from algae were tested through the evaluation of DPPH radical and hydroxyl radical scavenging activity. The extracts of Distromium decumbens, Sargassum hemiphyllum (Turner) C.Agardh, Sargassum serratifolium (C. Agardh) C. Agardh and Acrosorium yendoi Yamada were found to have more than 80% DPPH radical scavenging activity and that of Dictyota okamurae (Dawson) $H{\ddot{o}}tning$, Schnetter, et Prud'homme van Reine, Myagropsis myagroides (Martens ex Turner) Fensholt, Sargassum serratifolium (C. Agardh) C. Agardh and Cladophora wrightiana Harvey showed more than 50% hydroxyl radical scavenging activity. These results suggest that algae collected from Jeju Island would be good raw materials for antioxidant.

Community Structure of Benthic Marine Algae at Daejin and Jukbyeon on the Mid-East Coast of Korea (동해 중부 대진과 죽변 연안의 저서 해조류 군집구조)

  • The species composition and variation of benthic marine algae at Daejin and Jukbyeon on the mid-east coast of Korea were investigated seasonally from August 2006 to April 2007. As a result, the total of 85 species, including 8 chlorophytes, 26 phaeophytes, 50 rhodophytes and 1 sea grass were identified. 52 species were found at Daejin and 74 species at Jukbyeon. Dominant species in importance value were Corallina pilulifera, Chondrus ocellatus, melobesioidean algae, Sargassum horneri, Sargassum yezoense and Phyllospadix japonica at Daejin, and Corallina pilulifera, melobesioidean algae, Phyllospadix japonica, Acrosorium polyneurum, Sargassum siliquastrum and Hizikia fusiformis at Jukbyeon, respectively. The vertical distribution of algae were characterized by Corallina pilulifera, Hizikia fusiformis, Sargassum spp. (S. siliquastrum, S. yezoense) and Symphyocladia latiuscula at intertidal zone, Sargassum spp. (S. horneri, S. serratifolium, S. yezoense), melobesioidean algae, Phyllospadix japonica and Corallina pilulifera at 1 m depth, melobesioidean algae, Phyllospadix japonica and Sargassum spp. (S. horneri, S. yezoense) at 5 m depth and melobesioidean algae, Phyllospadix japonica and Sargassum spp. (S. horneri, S. micracanthum, S. yezoense) at 10 m depth.

  • The Fucales plants have very homogenous taxonomic characteristics at order level, but show many morphological variatations at species or subspecies levels. This study is one of the serial works to obtain more taxonomic data than morphotaxonomic ones and to clarify the taxonomic ranks and characteristics of Korean Fucales plants through physiological and biochemical analyses. Total lipid compositions of Korean Fucales plants were analyzed by the 3-dimensional TLC method. Major components of phospholipids (PA, PC, PG, PE, DPG, LPE, LPC), neutral fat TG and sterols were commonly contained in each species. Unknown lipids X1, X2 and X3 were contained in genus Sargassum, but Hizikia was lacking X3 and Myagropsis was lacking both X2 and X3. The latter contained unique X8. From the view-point of the phylogeny of lipid metabolism, Sargassum might be more closely related to Hizikia than to Myagropsis. A variant of S. patens from Seongsan showed minute metabolic difference from that of the typical plant; but a variant of S. serratifolium did not show any significant difference from that of the typical plant.
