Antimicrobial Characteristics of Scutellariae Radix Extract (황금추출물의 항균특성)

  • It was carried out for research and development of natural antimicrobial on Scutellariae Radix extract against food-borne infection bacteria .Scutellariae Radix extract showed remarkable antimicrobial activites against Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus when examined by disk method, it was very stable on the wide rane of temperature and pH,.The growth rates of Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus were decreased at the concentration of more than 500 ppm Scutellariae Radix extract, Indicating that the minimum inhibitory concentration (MIC)of the Bacillus cereus, Listeria monocytogenes, Escherichia coli, and Vivro parahaemolyticus to Scutellariae Radix extract were around 500 ppm . The morphological changes were observed by transmission electron microscope and scanning electron microscope and the microbial cells membran was destroyed by Scutellariae Radix extract. It was identified that the membrane integrity of the sensitive cells was disrupted by exposure to Scutellariae Radix extract as the $\beta$-galactosidase test on experimental substrate ONPG(o-nitrophenyl-$\beta$-D-galacto-pyranoside)

  • The aim of this experiment was to investigate the effects of a mixture of Scutellariae Radix, Coptidis Rhizoma and Coptidis Rhizoma water extracts on the blood pressure, plasma renin activity, plasma levels of aldosterone and atrial natriuretic peptide (ANP) in spontaneously hypertensive rats (SHR). The results of this study were as follows: 1. Systolic blood pressure decreased significantly after the administration of a mixture of Scutellariae Radix, Coptidis Rhizoma, and Coptidis Rhizoma water extract 0.2 ml/200g. 2. Plasma renin activity and plasma levels of ANP were not changed after the administration of a mixture of Scutellariae Radix, Coptidis Rhizoma and Coptidis Rhizoma water extracts, 3. Plasma levels of aldosterone was increased significantly after the administration of a mixture of Scutellariae Radix, Coptidis Rhizoma and Coptidis Rhizoma water extracts 0.1 ml/200g.

Effect of Scutellariae Radix Water Extract on Hydrogen Peroxide Production in LPS-induced RAW 264.7 Mouse Macrophages (황금(黃芩) 물추출물이 LPS로 유발된 마우스 대식세포의 hydrogen peroxide 생성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate effects of Scutellariae Radix water extract on hydrogen peroxide production in LPS-induced RAW 264.7 mouse macrophages. Methods : Scutellariae Radix produced from South Korea (SK) and Scutellariae Radix produced from China (SC) were extracted by hot water. Effects of SK and SC on hydrogen peroxide production in LPS-induced RAW 264.7 were measured by dihydrorhodamine 123 assay after 20, 24, 28, 44, 48, and 52 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL. Results : SK significantly increased hydrogen peroxide production in LPS-induced RAW 264.7 cells for 20, 24, 28, 44, 48, and 52 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL (P < 0.05). But SC did not represent any significant effect on hydrogen peroxide production in LPS-induced RAW 264.7 cells. Conclusions : These results suggest that Scutellariae Radix, especially produced from South Korea, has the immune-enhancing property related with its increasement of bacteriocidal hydrogen peroxide production in LPS-induced macrophages.

Effects on the secretion of Gastrin and Uropepsin of rats treated with Coptidis Rhizoma, Scutellariae Radix and Phellodendri Cortex (황련 황금 황백이 백서의 GASTRIN 및 UROPEPSIN 분비에 미치는 영향)

  • Each decoction of Coptidis Rhizoma, Scutellariae Radix and Phellodendri Cortex was orally administrated to the rates. And the effects on the Gastrin and Uropepsin secretions were observed. The results were as follows: 1. The effects on the secretio of Gastrin in the experimental groups administrered each with Coptidis Rhizoma and Scutellariae Radix comparing with the normal group, showed signficant values P<0.05, P<0.02, P<0.01 and P<0.001 separately. However the group medicated with Phellodendri Cortex showed significant values only 30 minutes after the administration. 2. Uropepsion level in the experimental group administered each with Coptidis Rhizoma and Phellodendri Cortex, comparing with the normal group, showed highly significant statistic values (P<0.001). Another experimental group medicated with Scutellariae Radix showed significantly statistical values (P<0.001) 240 minutes after the administration. 3. According to the above mentioned results it was onculuded tat the secretion of Gastrin in plasma and Uropepsin content of urine in albino rats treated with Coptiodis Rhizoma decoction were remarkaby increased as comparing with Scutellariae Radix and Phellodendri Cortex decoction.

For 2 h incubation, SC significantly increase hydrogen peroxide production in RAW 264.7 cells at the concentrations of 10, 25, and 100 ug/mL (P < 0.05). For 44 h incubation, SC significantly increase hydrogen peroxide production in RAW 264.7 cells at the concentrations of 10, 25, and 50 ug/mL (P < 0.05). Conclusions : These results suggest that Scutellariae Radix has the immune - enhancing property related with its increasement of hydrogen peroxide production in macrophages.

The Effective Preparation of Flavonoids from Scutellaria baicalensis GEORGI by Diaion HP-20 Resin

  • Scutellaria baicalensis $G_{EORGI}$ (Scutellariae Radix) has been used to clear heat and to dry dampness in the stomach or intestines, which manifests as diarrhea or dysenteric disorder. In this study, we investigated the effective preparation of active components in Scutellariae Radix using the methods of solvent extraction and absorption fractionation for the development of new functional food or pharmaceuticals. The marker substances, baicalin, baicalein, wogonoside, and wogonin were directly isolated from the Scutellariae Radix. There chemical structures were elucidated by spectroscopic analysis. The Scutellariae Radix was extracted with hot water. To enhance yield of flavonoids in Scutellariae Radix, the hot water extract was dissolved in ethanol with concentration dependent manner. The precipitates were separated using centrifugal techniques at 10,000 rpm. Supernatant liquid was applied to the HPLC for quantification of major compounds. Separately, the hot water extract was absorbed on Diaion HP-20 resin. And then, the absorbed fraction was eluted with methanol for HPLC. The contents of baicalin, baicalein, wogonoside and wogonin in different treatment methods were analyzed by HPLC. Total amount of four major components were 16.9% in 50% ethanol extract, 21.7% in 70% ethanol extract, 20.5% in 90% ethanol extract, and 39.3% in absorbed fraction of Diaion HP-20 resin. In these results, we found that resin absorption method is suitable for the extraction of enriched flavonoids from Scutellariae Radix.

Inhibitory Activity of Advanced Glycation Endproducts (AGEs) Formation and Antioxidant Activity of Processed Korean Medicines (포제한약재의 최종당화산물 생성 억제 활성 및 항산화 효과)

  • Objectives : Advanced glycation end product (AGEs) is combine formation of glucose and protein. AGEs and reactive oxygen species are potential therapeutic targets for the various disease such as diabetic complications, renal injury, skin damage. The aim of this study was investigated the AGEs inhibitory activity and antioxidant activity of water extracts from 40 Korean medicines and 5 heating-processed Korean medicines. Methods: AGEs formation inhibitory activities of Korean medicines measured using bovine serum albumin (BSA), glucose, and fructose. Then, five effective Korean medicines were selected and heated with 30% ethanol. The AGEs inhibitory activities of heated Korean medicine were measured compared with not-heated Korean medicines. The antioxidant activities were evaluated through radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals. Furthermore, we examined total phenol and flavonoids contents. Results: Scutellariae Radix, Corni Fructus, Persimmon Fruit, Paeoniae Radix, Mori Folium respectively reduced AGEs production. Morever, heating-processed Scutellariae Radix has AGEs inhibitory activities better than not-processed Scutellariae Radix. Heating- processed Scutellariae Radix scavenged DPPH and ABTS effectively and $IC_{50}$ of DPPH and ABTS radical scavenging activity of Heat processed Scutellariae Radix were $15.47{\pm}0.26{\mu}g/m{\ell}$ and $12.07{\pm}1.23{\mu}g/m{\ell}$. It caused heat processing methods of Scutellariae Radix up regulated total phenol and flavonoids contents ($26.68{\pm}0.01$ to $46.15{\pm}0.10$, $20.30{\pm}0.38$ to $64.20{\pm}0.52$). Conclusion: It has AGEs inhibitory activities that 20 kind of medicinal plants of 40 medicinal plants. Especially, heat processed Scutellariae Radix has excellent AGEs inhibitory activities and antioxidant effect.

Effects of Scutellariae Radix-acupuncture solution On Anti-inflammatory in RAW 264.7 macrophages (황금약침액(黃芩藥針液)이 LPS로 유도(誘導)된 RAW 264.7 대식세포(大食細胞)에서의 항염증효과(抗炎症效果))

  • Objective : Recently, Herbal-acupuncture therapeutics has been used for the treatment of inflammatory diseases such as rheumatoid arthritis. Especially, we have been interested in chemical mediators concerned with inflammation such as prostaglandin, cytokine, nitric oxide. The purpose of this study is investigated that the effect of Scutellariae Radix-acupuncture solution in lipopolysaccharide-stimulated RAW 264.7 macrophages, performed several expeimental items : those are prostaglandin E. nitric oxide and cyclooxygenase-2. Method : The cytotoxicity of Scutellariae Radix-acupuncture solution in RAW 264.7 macrophages were measured by MTT-based cytotoxicity assay. In order to observe cyclooxygenase-2 mRNA expression in lipopolysaccharide-stimulated RAW 264.7 macrophages, RT-PCR was used. Prostaglandin $E_2$ formation and nitric oxide production was measured by competitive enzyme immunoassay and Griess assay. Results : 1. The MTT assay demonstrated that cytotoxic effect of Scutellariae Radix-acupuncture solution in RAW 264.7 macrophages were not appeared before concentration of 5mg/mL 2. Scutellariae Radix-acupuncture solution inhibited cyclooxygenase-2 mRNA expression in lipopolysaccharide-treated RAW 264.7 macrophages. 3. Scutellariae Radix-acupuncture solution inhibited nitric oxide production in lipopolysaccharide-treated RAW 264.7 macrophages. 4. Scutellariae Radix-acupuncture solution inhibited prostaglandin $E_2$ formation in lipopolysaccharide-treated RAW 264.7 macrophages. Conclusion : On the basis of these results, It was shown that Scutellariae Radix-acupuncture solution is significantly able to inhibit the production of $PGE_2$ and NO, as well as COX-2 mRNA expression. Our results may provide new mechanism by which Scutellariae Radix-acupuncture solution accounts for its beneficial effect on accelerating wound healing and anti-inflammation.

Analysis of Scutellaria baicaleinsis Georgi (Scutellariae Radix) by LC-DAD and LC-ESI/MS

  • In this study, baicalin, as a marker substance of Scutellariae Radix, was quantitatively analyzed by a high performance liquid chromatography-photodiode array detector (HPLC-DAD). We identified wogonoside, baicalein, and wogonin in the Scutellariae Radix by a high performance liquid chromatography-electrospray ionization-mass spectrometer (HPLC-ESI-MS). The baicalin was separated on a Xterra C18 column ($5{\mu}m$, $4.6{\times}250mm$) using mobile phase consisting of 38% acetonitrile in 0.68% phosphoric acid. The baicalin spectrum in the Scutellariae Radix extracts was coincided by comparing with UV-visible spectrum (200-550 nm) of baicalin standard in the library. The amount of baicalin in Scutellariae Radix was 10.46%, which is higher than KFDA's guideline. The marker substances of Scutellariae Radix showed a strong base peak $[M]^+$ in the positive detection mode following as; baicalin (m/z; $271[MH^+-sugar]^+$, $447[M+H]^+$), wogonoside (m/z; $285[MH^+-sugar]^+$, $461[M+H]^+$), baicalein (m/z; $271\;[M+H]^+$), wogonin (m/z; $285[M+H]^+$). These results are consistent with the fragment pattern and molecular weight of standard components from literature.

Identification of Index Components of Scutellariae Radix and Quantitative Determination of Baicalin from Curde Drug Preparation Containing Bupleuri Radix (시호(柴胡)함유 한방제(漢方劑)중 황금(黃芩) 지표성분(指標成分)의 확인(確認) 및 Baicalin 정량(定量))

  • As a part of studies on the quality control of crude drug preparation, So-Shi-Ho-Tang, index components of Scutellariae Radix were identified by TLC and quantified by HPLC. Wogonin and baicalein were identified in ethyl ether fraction on silica gel plate with benzene/ethyl acetate (1 : 1, v/v). The content of baicalin was determined by HPLC on ${\mu}-Bondapak\;C_18$ column with 0.5%-phosphoric acid/acetonitrile (73 : 27, v/v). Its recovery rate in the extract granules, compared to the content in the Scutellariae Radix, was $52.1\;{\pm}\;0.7%$.
