Comparison of the Contents of Phenolic Compounds of Sea Buckthorn(Hippophae rhamnoides) Cultivated in Korea and Mongolia (비타민 나무(사극)의 페놀성 성분 분석)

  • The purpose of this research is to provide basic informations to discriminate between sea buckthorn (Hippophae rhamnoides) cultivated in the Republic of Korea and Mongolia. The phenolic compounds of sea buckthorn, were measured by the HPLC analysis. Catechin, rutin, quercetin, isorhamnetin were found in methanol extracts of sea buckthorn. Total phenolic compound of Korean sea buckthorn leaves (1.852%) was about five times higher than those of Mongolian sea buckthorn berries (0.338%). As a result, the order of the total phenolic compound and Catechin content was 1) sea buckthorn leaves, 2) sea buckthorn stems, 3) sea buckthorn roots, and 4) sea buckthorn berries. Statistically no big differences in levels of phenolic compounds were consistently found in sea buckthorn cultivated in the Republic of Korea and Mongolia investigated in this work.

  • Sea buckthorn (Hippophae rhamnoides L.) is deciduous shrubs in the genus Hippophae, mainly cultivated in Europe and Asia. Sea buckthorn berries have a high vitamin C, vitamin E, carotenoids, carbohydrates, protein, organic acids, dietary minerals, triterpenoids, polyphenolic acids and amino acids. Extracts of sea buckthorn berries have anti- obesity, anti-oxidantive, anti-microbial, anti-ulcerogenic, anti-diabetic and nutritional effects. Sea buckthorn used as a traditional medicine for the treatment of cough, aid digestion, invigorate blood circulation and alleviate pain. Extracts of sea buckthorn branches and leaves was administered to humans and animals to treat gastrointestinal distress in Mongolia. This paper briefly reviews the most relevant experimental data on the pharmacological effects and isolated component of sea buckthorn. And, we also describe the importance of sea buckthorn as the environmental-friendly crops.

Antioxidative Activity of Sea Buckthorn and Quality Characteristics of Brown Rice Sulgidduk (비타민나무 잎 첨가 현미설기떡 제조 및 항산화 활성)

  • This study was performed to investigate the antioxidative properties of sea buckthorn (Hippophae rhamnoides L.; leaf, fruit and stem) and the quality characteristics of brown rice sulgidduk prepared using a powder of sea buckthorn leaves. First, the antioxidative activities of sea buckthorn were measured to choose the most effective part of this plant. By analyzing the measured values, we concluded that the effective part of sea buckthorn was its leaves. The $IC_{50}$ value of the DPPH radical scavenging activity and the ABTS radical scavenging activity in sea buckthorn leaves were $7.78{\mu}g/mL$ and $264.04{\mu}g/mL$, respectively. The total polyphenol and total flavonoid contents of sea buckthorn leaves were 3.80 mg/mL and .19 mg/mL, respectively. Therefore, the brown rice sulgidduk was prepared using a powder of sea buckthorn leaves in the weight ratio of 0%, .5%, 1.0%, 1.5%, 2.0% and 2.5%. For analyzing the quality characteristics of the prepared sulgidduk, proximate compositions, color and texture profiles were measured and a sensory evaluation was conducted. With an increase in the added content of the sea buckthorn leaf powder (SBLP), the L-value significantly decreased while the a-value and the b-value increased. In the case of texture profiles, the control group (control A) had a higher score for hardness than the case groups in which the SBLP was added. However, springiness, chewiness and adhesiveness were not significantly different among the groups. In the sensory evaluation, the sample containing 1.5% SBLP yielded the best results. Therefore, we suggest that the powder of sea buckthorn leaves is a good ingredient for increasing the consumer acceptability and functionality of sulgidduk.

The Effect of Extract from Sea Buckthorn on DNCB-induced Atopic Dermatitis in NC/Nga Mice

  • Sea Buckthorn (Hippophae rhamnoides L.) has been used in traditional medicine for the treatment of cough, indigestion, circulatory problems and pain. The associated anti-inflammatory effect of this agent is achieved via the inhibition of Nf-${\kappa}B$ signaling, a property that has been demonstrated to effectively control the symptoms of various skin disorders, including atopic dermatitis. Accordingly, the purpose of this study was to assess the efficacy of Sea Buckthorn in reducing the production of lipopolysaccharide (LPS) activated nitric oxide (NO) by inhibiting the Nf-${\kappa}B$ pathway, as measured by the symptoms of atopic dermatitis (AD) occurring secondarily to inflammation and immune dysregulation. Our data demonstrate that Sea Buckthorn significantly decreased the LPS-induced production of NO (p<0.001). Atopic dermatitis was induced by repeated application of 2,4-dinitrochlorobenzene to the dorsal skin of mice. Topical application of 5% Sea Buckthorn extract improved the symptoms of AD, specifically reducing disease severity scores, scratching behaviors and epidermal thickness. When compared to the control group, animals treated with Sea Buckthorn exhibited increased serum IL-12 levels and decreased serum TNF-${\alpha}$, IL-4 and IL-5 levels. Such a modulation of biphasic T-helper (Th)1/Th2 cytokines may result in a reduction in serum IgE levels. Our findings suggest that mechanism of action of Sea Buckthorn in the treatment of AD is associated with a marked anti-inflammatory effect as well as an inhibition of Th2-mediated IgE overproduction via the modulation of biphasic Th1/Th2 cytokines. Such results suggest that topical Sea Buckthorn extract may prove to be a novel therapy for AD symptoms with few side effects.

  • Sea-buckthorn fruits (Hippophae rhamnoides L.) are used in Mongolia as traditional medicine due to the health-benefits associated with its bioactive components. The purpose of this study was to investigate the chemical composition of Mongolian sea-buckthorn fruits. In terms of proximate composition, crude fat content was the highest, whereas its crude ash content was the lowest. In organic acid contents, malic acid content ($6.30{\pm}0.005$ mg/g) was the highest. Free sugars were composed of sucrose, xylose, glucose and mannitol. Vitamin C and carotenoid content were 320 mg/100 g and 715.25 mg/100 g, respectively. The major fatty acids were palmitic (C16:0), palmitoleic (C16:1), caprylic (C8:0) and linoleic (C18:2) acid.

Radical Scavenging Activity of Sea Buckthorn Oils from Different Parts of Sea Buckthorn Berry

  • Antioxidant-rich oils were extracted from different parts of sea buckthorn berry with supercritical $CO_2$ (SC-$CO_2$) and n-hexane. The functional components were analyzed and the extracts were screened for their potential as radical scavengers in 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), 2,2'-azinobis-3-ethylbenzotiazoline-6-sulphonic acid (ABTS), galvinoxyl systems. Minor differences were found in fatty acid composition of oils extracted by the two methods. Seed oil contains the highest content of tocopherols while pulp oil and whole berry oil possessed higher concentration of carotenoids. Whole berry oil, pulp oil, and seed oil extracted by SC-$CO_2$ showed 91.7, 90.9, and 93.5% radical scavenging activity (RSA) at 6 mg/mL towards DPPH and 74.3, 54.3, and 74.8% towards galvanoxyl radical at 10 mg/mL. The hexane-extracted oils showed similar scavenging ability. However, the oils obtained by hexane showed significantly higher RSA (p<0.05) than those obtained by SC-$CO_2$ while whole berry oil has the highest RSA towards ABTS among 3 oil samples.

Effect of Sea Buckthorn (Hippophae rhamnoides L.) Seed Supplementation on Egg Quality and Cholesterol of Rhode Island Red×Fayoumi Laying Hens

  • The present trial was carried out to study the effect of sea buckthorn seed supplementation on egg quality of laying birds. A total of 160 Rhode Island Red${\times}$Fayoumi layers was divided into four groups of 40 birds each, which was further replicated four times with 10 hens per replicate. Group one was kept as a control, while other three groups were supplemented with sea buckthorn seed powder at a dose rate of 1 (T1), 2 (T2) and 3 (T3) g/kg of feed. The results showed that egg production was significantly (p<0.05) higher in T3 at the end of the study. Egg weight was significantly (p<0.05) high in T2 and T3 during week 39 and 40. Egg yolk weight was significantly (p<0.05) in T3 compared to the control. Significantly (p<0.01) lower egg cholesterol was recorded in T2 and T3. From the results of the present study, we concluded that laying hens supplemented with sea buckthorn at the rate of 2 and 3 g/kg improved the egg quality parameters and egg cholesterol.

A Study on Anti-oxidant Activity and Anti-inflammatory Action of Sea Buckthorn Seed Extract (비타민 나무 Seed 추출물의 항산화 활성 및 항염증에 관한 연구)

  • This study aimed to observe the effects of Sea Buckthorn seed extracts (SEB) on anti-oxidation and antiinflammation. SEB were found to strongly depress ROS production depending on concentration, and 46.5% anti-oxidation effects were found in the top concentration of 100 ${\mu}g/mL$. Histamine and arachidonic acid release were measured to observe the effects of SEB on anti-inflammation. SEB inhibited histamine release and arachidonic acid release on dose dependent manner. Based on the results above, the conclusion is drawn that SEB have the potential to be used as natural materials that are effective for anti-oxidation and antiinflammation.

Optimization for Decolorization and UV-Absorbility of Refined Sea Buckthorn Oil Using CCD-RSM (CCD-RSM을 이용한 시벅턴 오일의 탈색공정 최적화 및 자외선 흡수능력 평가)

  • In this study, the adsorption decolorization process of sea buckthorn oil was carried out to verify the possibility of the sea buckthorn oil as a natural UV absorber. The optimization was carried out by using the central composite design model-response surface methodology (CCD-RSM). The response values of CCD-RSM were selected as the decolorization effect through the process, acid value after decolorization, and UV absorbance of the decolored oil at 290nm. The amount of adsorbent, temperature and time were selected as the process variables for the experiments. According to the results of CCD-RSM, the results of optimization were all consistent. The optimal conditions, which satisfy CCD-RSM statically and mathematically, were 4.32 wt.%, 134.90 ℃, and 19.8 min for the amount of adsorbent, temperature and time, respectively. The estimated response values expected under these optimal conditions values were 94.78%, 2.08 mg/g KOH, and 2.91 for the decolorization effect, acid value and UV absorbance at 290 nm, respectively. Also the average error from actual experiment for verifying the conclusions was smaller than 2%. Therefore, it was confirmed that the application of CCD-RSM to the adsorption decolorization process of sea buckthorn oil showed a very high level of acceptable results and that the sea buckthorn oil has high possibility to be used as a natural UV absorber.

Arbuscular Mycorrhizal Fungi Enhance Sea Buckthorn Growth in Coal Mining Subsidence Areas in Northwest China

  • Land subsidence induced by underground coal mining leads to severe ecological and environmental problems. Arbuscular mycorrhizal fungi (AMF) have the potential to improve plant growth and soil properties. We aimed to assess the effects of AMF on the growth and soil properties of sea buckthorn under field conditions at different reclamation times. Inoculation with AMF significantly promoted the survival rate of sea buckthorn over a 50-month period, while also increasing plant height after 14, 26, and 50 months. Crown width after 14 months and ground diameter after 50 months of inoculation treatment were significantly higher than in the uninoculated treatment. AMF inoculation significantly improved plant mycorrhizal colonization rate and promoted an increase in mycelial density in the rhizosphere soil. The pH and electrical conductivity of rhizosphere soil also increased after inoculation. Moreover, after 26 and 50 months the soil organic matter in the inoculation treatment was significantly higher than in the control. The number of inoculated soil rhizosphere microorganisms, as well as acid phosphatase activity, also increased. AMF inoculation may play an active role in promoting plant growth and improving soil quality in the long term and is conducive to the rapid ecological restoration of damaged mining areas.
