AN ANALYSIS OF SELECTED MOLECULAR LINES IN SUNSPOTS

  • Theoretical profiles of selected rotational lines of $C_2$ CH, CN, TiO and MgH are computed by using the current models of sunspot unbrae and penumbrae. It is found that the lines of the diatomic carbides are enhanced in penumbrae relative to umbrae, while MgH lines are more strongly enhanced in umbrae than in penumbrae and the quiet photosphere. The results are discussed with respect to selecting lines suitable for studying the structure of sunspots.

Asymmetries in Chickens from Lines Selected and Relaxed for High or Low Antibody Titers to Sheep Red Blood Cells

  • Wattle length, width, and area were measured to classify bilateral asymmetries in four lines of chickens. The lines were the S26 generation of White Leghorns selected for high (HAS) or low (LAS) response to sheep red blood cells and sublines in which selection had been relaxed for three generations (high antibody relaxed [HAR] and low antibody relaxed [LAR]). Antibody titers (AB) were greater for HAS than for HAR with both greater than for LAS and LAR which while different for males did not differ for females. The low antibody lines were heavier and reached sexual maturity at younger age than the high antibody lines. In general, wattle length, width, and area were greater in the low than high antibody lines. In 24 comparisons for bilaterality 18 exhibited fluctuating asymmetry and 6 exhibited directional asymmetry with 5 of the 6 being for wattle length. There was not a clear pattern for changes in degree of asymmetry when selection was relaxed for 3 generations. For females, the relative asymmetry (RA) of wattle area was larger ($p{\leq}0.05$) for HAR than for LAR and not different from the selected lines and relaxed lines. There were no differences among lines for RA of wattle length and width of females and wattle length, width, and area of males.

Development of Near Isogenic Lines and Genetic Analysis for Alkali Digestibility of Rice Grain

  • In order to develop near isogenic lines (NILs) the heterozygous rice plants for alkali digestibility value(ADV) were selected and tested in every generation from $F_5$to $F_9$ of a cross, Seratus malam/Suwon 345. Finally several sets of NILs, which were six low ADV lines and four medium-high ADV lines, were selected among $F_{10}$ lines. No differences of the plant growth characteristics, amylose content and protein content of rice grain were found between low and medium-high ADV lines. Rice flour of low ADV lines showed longer gel length in gel consistency test than medium-high ADV lines, and also showed different gelatinization characteristics measured by rapid visco analyzer compared with medium-high ADV lines. The result of genetic analysis indicated that ADV-NILs developed were differed in a pair of major gene controlling ADV, and low ADV was dominant over medium-high ADV.

Selection of Active Grow Hairy Root Lines in Ginseng (고생장 인삼 모상근의 선발)

  • These studies were carried out to select the active grow hairy root lines induced from various ginseng(Panax ginseng C. A. Meyer) parts. Hairy roots were induced in root explants, stem and petiole in vitro by A. rhizogenes R1000 or A. rhizogenes $A_4$. These hairy roots could be grown on the phytohormone free medium, and PCR analysis of rol C and vir C gene fragments confirmed that hairy roots were transgenic tissues. We have selected 11 hairy root lines with active growing characters among 300 hairy root lines selected based on growth and morphological characteristics on 1/2MS solid media with 250 mg/L carbenicillin. Morphological characteristics of selected 11 hairy root lines were thickness and thiness of main roots, and many projection for lateral roots, active grow of lateral roots. Among selected 11 hair root lines prominent characteristics of hairy roots with active growing characters were thiness of main roots and active grow of lateral roots. But characteristics of low growing hairy roots were thickness of main roots and low grow of lateral roots. Finally we have selected actively growing hairy roots, KGHR-1, KGHR-5, KGHR-8 among 11 hairy root lines.

Selection of Short Stem Astragalus membranaceus Lines by Assessing Agronomic Characteristics and Biological Activity (농업형질 및 생리활성 평가를 통한 황기 단간 계통 선발)

  • Background: Astragalus membranaceus belonging to the Leguminosae family is often utilized as a traditional medicine. The aim of this study was to elucidate the basic breeding information required to develop short stem A. membranaceus cultivars. Methods and Results: Roots of A. membranaceus advanced yield trial (AYT) lines were harvested in late October 2017. Root yield of six AYT lines were increased in a range of 8.9 - 74.8% compared with 'Aseong' as control (check variety). The height of seven AYT lines were shorter than that of 'Aseong'. In addition, stem diameter of nine AYT lines was thicker than that of 'Aseong'. Consequently, 1502-56, 1503-90, and 1510-80 were selected as elite lines for the development of short stem cultivars. HPLC analysis was perfromed to identify lines with high level active components such as calycosin-7-O-${\beta}$-D-glucoside and calycosin. The levels of both active components were higher in 1502-56, and 1503-90, but not in 1510-80 compared to 'Aseong'. In addition, 2,2-Diphenyl-1-picryl hydrazyl (DPPH) radical scavenging activity was higher in the 1502-56 compared to 'Aseong'. Considering these results, two AYT lines, 1502-56 and 1503-90 were selected as short stem lines with high calycosin-7-O-${\beta}$-D-glucoside and calycosin content. Conclusions: Taken together, Two short stem lines were identified in this study. In our future study, regional yield trial (RYT) will be conducted using these selected lines to develop new cultivars.

Development of a Dike Line Selection Method Using Multispectral Orthoimages and Topographic LiDAR Data Taken in the Nakdong River Basins

  • Dike lines are important features for describing the detailed shapes of dikes and for detecting topographic changes on dike surfaces. Historically, dike lines have been generated using only the LiDAR data. This paper proposes a new methodology for selecting an appropriate dike line on various dike surfaces using the topographic LiDAR data and multispectral orthoimages taken in the Nakdong River basins. The fi rst baselines were generated from the given LiDAR data using the modified convex hull algorithm and smoothing spline function, and the second baselines were generated from the given orthoimages by the Canny operator. Next, one baseline was selected among the two baselines at 10m intervals by comparing their elevations, and the selected baseline at 10m interval was defined as the dike line segment. Finally, the selected dike line segments were connected to construct the 3D dike lines. The statistical results show that the dike lines generated using both the LiDAR data and multispectral orthoimages had the improved horizontal and vertical accuracies than the dike lines generated only using the LiDAR data on the various dike surfaces.

Selection of Salt Tolerant Rapeseed (Brassica napus L.) Mutant Lines Induced by Irradiation of Proton Ion Beams and Gamma Ray (양성자 및 감마선처리에 의한 유채 내염성 변이계통 선발)

  • This study was carried out to develop salt tolerant varities of rapeseed (Brassica napus L.) which can be grown in the high salty reclaimed land. The seeds of three varieties 'Naehan', 'Tammi', and 'Halla' were treated by proton ion beams and gamma rays with 0 to 2,000 Gy. For the selection of salt tolerant lines, emergence and survival rate, and growth characteristics of $M_2$ to $M_4$ generations were investigated in the Saemangeum reclaimed fields with the different salt concentrations. The lines with potential salt tolerance were selected in the $M_4$ generation and tested indoor for their growth characteristics. There was no significant changes in the soil pH for $M_4$ generation during growth period. However, soil EC was higher in early spring than sowing period (mid October). In $M_4$ generation test, the seeds of original and selected line showed high rates of emergence and survival, as determined one month after sowing. After wintering, however, the original varieties showed the significant reduction in the survival rate, while the selected lines showed a higher survival rate and good growth, leading to the completion of their life cycle. Consequently we selected 9 lines from $M_4$ generation with better performance in growth and yield. Soil EC was $2.8{\sim}4.3dS{\cdot}m^{-1}$ during $M_4$ generation growth period. The laboratory test of the lines selected from $M_4$ generation was made for their salt tolerance potential. The selected lines showed higher chlorophyll and proline contents than the original varieties. There was also no significant difference in the emergence rate of seed between the original and selected varieties. In 200 mM natural sea salt, the $N{\gamma}600-21-1-641$ line derived from 'Naehan' was the highest in growth rate, leaf chlorophyll and proline contents. $T{\gamma}800-20-2-461$ line derived from 'Tammi' didn't show significant difference in growth rate compared to original variety in 200 mM and withered in 250 mM like other lines as time passed. $H{\gamma}200-7-1-740$ line showed similar growth and chlorophyll content compared to its original variety.

Breeding of High Yield and Elite Lines for Development 'Ssam' Lettuce (쌈용 상추 품종 개발을 위한 다수성, 우수 계통 육성)

  • More than 90 percent of domestic consumption of the lettuce occupied leaf lettuce, and the farmer preferred high yield, late bolting and stable expression of anthocyanins. So, this research focused on making resistant to disease, late bolting, and good expression of anthocyanin for lettuce. Based on these characteristic's parents, 11 crosses were created. So we advanced the generation and investigated each generation from F1 to F7. In this year, we selected 90 combination 221 lines which was F2 42 combination 137 lines, F3 4 combination 17 lines, F4 11 combination 27 lines, F5 7 combination 30 lines, F6 1 combination 2 lines, F7 3 combination 8 lines. Among the selected 17 lines, 5007, 5014, 6004, 6005, 6021, 6023, 7001, 7002, 7006, 7007, 7010, 7011, 7015, 7018, 7019, and 7021 which were late bolting and good expression of anthocyanins were investigated their yield. Finally, we selected 5 lines 7001, 7002, 7011, 7018 and 7022 in this year.

Breeding of Korean Ginseng (Panax ginseng C. A. Meyer)

  • A lot of individual ginseng plants were selected in the farmer's fields to develop new ginseng varieties with desirable traits. Among them, a promising line has been developed through comparative cultivation of several lines selected with pure line separation of local raced. It was then designated as "KC (denotes Korean Ginseng)" and tested in the regional yield and adaptation trials for 10 years. KG lines grew vigorously after 4 years of age. Especially, KG 102 line arson운 them showed traits of multiple and short stems. As for the root characters, the length of taproot of KG 101 line was longer than that of local race, Jakyung-jong, or other lines ailed the root weight of KG 102 line waIn 15 % higher than that of local race. In general, KG 101, KG 103, KG 104, and Hwangsauk-jong had good root shape. Total amount of ginsenosides of ginseng taproot was the highest in KG 103 line than in local race and other lines. In these studies, we, elected three superior lines, KG 101, KG 102, and KG 103, having characteristic of good root shape, high yield, and large amount of ginsenoside, respectively.

Development of Clubroot Resistant Doubled-Haploid Inbred Lines in Kimchi Cabbage (Chinese Cabbage) (Brassica rapa L.)

  • Kimchi cabbage (Chinese cabbage), radish and Cabbage are major Brassicaceae vegetables in Korea. Especially, we can easily develop whole plant from one microspore in Kimchi cabbage. To develop clubroot resistant doubled-haploid (DH) inbred lines, we pollinated a clubroot resistant turnip of 'IT 033820' with a Kimchi cabbage (Chinese cabbage) inbred of 'BP 079'. More than 85 DH inbred lines were developed from this combination. We screened about 400 materials including these DH inbred lines, commercial cultivars and breeding materials during 3 years using hydroponic system after inoculating single spore isolation race 4(SSI-04) inoculate. One inbred line derived from this combination selected as clubroot resistant and registered as 'Wonkyo20036ho'. We inoculated 26 DH inbred lines derived from 'Zoong-baek 2ho' using SSI-4, the percent of resistant plants varied from 0 to 83%. However the horticultural traits of highly resistant DH inbred line was poor. Thus we selected one DH line showing 77% resistant with yellow inner leaf and maid good head, was registered as 'Wonkyo20034ho'. Another DH inbred line derived from Korean variety of 'Wol-dong' showing 86% resistant was registered as 'Wonkyo20037ho'. Other DH inbred lines were derived from Chinese cultivar of 'Choon-hi-go-hang-wang' and 'Hwang-shim-zo48' showed 80 and 71% resistant, respectively, was also selected for registration. Even though DH inbred lines derived from turnip showed highly resistant to SSI-04 and provincial inoculate, they showed poor characteristics in horticultural traits. However, commercial seed companies showed interesting for adapting these DH inbred lines in commercial breeding.
