Critical Suspension Condition of Particles in a Shaking Vessel of Solid-Liquid System (고-액계 진동교반에서 입자의 부유화 한계조건)

  • Shake mixing has been widely used in cell culture. The mixing performance for shake mixing, however, has not been reported quantitatively. The critical circulating frequency and the power consumption for complete suspension of particles, based on the definition of Zwietering, were measured in a shaking vessel containing a solid-liquid system. The critical suspension frequency was correlated by the equation from Baldi's particle suspension model modified with the physical properties of the particles. Critical suspension frequency was correlated as following ; $$N_{JS}={\frac{0.58\;d{_p}^{0.06}(g{\Delta}{\rho}/{\rho}_L)^{0.004}X^{0.03}}{D^{0.35}d^{0.17}{\upsilon}^{0.04}}}$$ The power consumption at the critical suspension condition in the shaking vessel was less than that in an agitated vessel with impeller.

Amylase Production by Continuous Cultures of Aspergillus oryzae and its Mutants (Aspergllus oryza와 그 변이주의 연적배양에 의한 amylase 생성에 관한 연구)

  • Irradiation with high doses of gamma rays induced the reduction of mycelial weight and anaylase activity, and increased relative amylase activity in surface cultures. Biphase in growth curves was shown in aeration-agitation cultures but the behavior of the first phase of growth could be eliminated by replacing the amylasehydrolysed starch substrates, so that enzyme production was shortened ca. 40 hours and relative amylase activity was increased about 3 times higher before onset of autolysis. In the effect of gibberellin on amylase production, the positive stimulation was appeared to only surface culturs of the liquid medium and the negative effect to shake-cultures in a mutant. Trials of various continuous culture were resulted not only the approalch to the value of amylase activity in surface cultures of liquid medium, but also higher productivity than in batch cultures. The culture-degeneration was observed in two-stage continuous culture, but did not appear in continuous elevation culture.

  • Seeds of neem were collected from different parts of India and analyzed for their azadirachtin content by High Performance Liquid Chromatography (HPLC). In order to assess the effects of genotypic and geographical variation on azadirachtin content in cell cultures, callus development was attempted in the seeds containing high and low concentration of azadirachtin. The concentration of azadirachtin in callus cultures was significantly affected by the explant source. Seed kernels with higher azadirachtin content produced higher azadirachtin content in callus cultures and lower azadirachtin content was seen in callus cultures produced from seed kernels with low azadirachtin content. The protocol for development of elite stock culture of Azadirachta indica was established with the objective of selecting a high azadirachtin-producing cell line. The highest azadirachtin-producing cell line was selected and the effects of different media and illumination conditions on growth and azadirachtin production were studied in shake flask suspension culture. Detailed batch growth kinetics was also established. These studies provided elite starter culture and associated protocols for cultivation of A. indica plant cell culture in the bioreactor.

  • The production of polysaccharide from Paecilomyces japonica was studied in the shake flask culture. For the cell growth and the polysaccharide production, the optimum synthetic medium was glucose peptone(YMP) medium. The flask culture conditions for the polysaccharide production were $27^{\circ}C$ and 200rpm with the initial pH 9 for 8days cultivation.

  • The optimal temperature and pH for both mycelial growth and exe-biopolymer production by Cordyceps millitaris in shake flask culture were found to be $20^{\circ}C$ and 6.0, respectively. Sucrose (4%) and corn steep powder (1%) were the most suitable carbon and nitrogen source for mycelial growth and exo-biopolymer production. The maximum specific growth rate $(0.142h^{-1})$ was achieved when sucrose was used as the sole carbon source. Exo-biopolymer production was increased with the increase in C/N molar ratio concentration, probably due to the facilitated carbon uptake. Under the optimal culture conditions, the maximum mycelial growth exe-biopolymer concentration were reached to around 13.3 g dry cell weigh/l and 3.33 g/l, respectively.

  • The optimal temperature, pH and aeration rate for spore production by Bacillus polyfermenticus SCD in 500 ml shake flask and 5-1 jar fermenter were found to be $32^{\circ}C$, 7.0 and 1.0 vvm. respectively. When batch culture processes was performed under optimized culture conditions. viable cells were $3.3{\times}10^{10}$ CFU/ml and spore cells were $3.3{\times}10^{10}$ CFU/ml. Fed-batch culture processes were also examined with regard to higer maximum viable cell and spore production. The highe viable cells and spores were obtained in 5-1 jar fermenter at 72 h cultivation time by strategy in an intermediate feeding mode with 60% glucose solution 150 ml and 5% soybean flour solution 150 ml fed to the fermenter twice, and the productivity of spore cells was significantly increased. Finally. volumetric productivity of spore cells on fed-batch culture indicated $9.9{\times}10^8$ CFU/ml/h, which was approximately 2 times higher than batch culture. Thus, fed-batch culture show a promise as an industrial production method.

  • To improve biomass and astaxanthin production by wild-type Xanthophyllomyces dendrorhous simultaneously in shake flask culture, physical factors, nutritional factors and carotenogenesis stimulating factors affecting astaxanthin production were studied on base of HPLC quantitative analysis. The results suggested that carotenogenesis precursor composition acetic acid, mevalonic acid, tomato extract, and carrot extract could increase the productivity of astaxanthin markedly based on the optimized temperature, initial pH value, carbon and nitrogen sources conditions.

Antitumor Activity of Kp, a Protein-polysaccharide from the Mycelial Culture of Phellinus linteus (Phellinus linteus 균사 배양물로부터 분리한 단백다당체 Kp의 항암활성)

  • A protein-polysaccaride fraction Kp(53.9% polysaccharide, 14.2% protein) was separated from the shake-cultured mycelia of a basidiomycetous fungus, Phellinus linteus, and its antitumor activity against sarcoma 180 in ICR mice was investigated. When administered after the tumor implantation, Kp exerted antitumor activity by inhibiting the growth of the sarcoma 180 solid tumor by 71.5% and increasing the life span of the sarcoma 180 ascitic mice by 51.5% at 100 mg/kg. In pretreatment tests, in which Kp was administered once daily for 9 days before the tumor implantation, Kp inhibited the growth of the solid and ascites form of sarcoma 180, respectively, by 35.4% and by 80.3% at 100 mg/kg. However, Kp showed no in vitro cytotoxic activity against a murine leukemia L1210 and a human gastric tumor SNU.1 upto the concentration of $200\;{\mu}g/ml$. From these results, it is clear that the antitumor activity of Kp is exerted through its immunomodulating activity on the host's immune system.

Kinetics and Modelling of Cell Growth and Substrate Uptake in Centella asiatica Cell Culture

  • In this study, we have conducted kinetics and modelling studies of Centella asiatica cell growth and substrate uptake, in an attempt to evaluate cell growth for a better understanding and control of the process. In our bioreactor cultivation experiment, we observed a growth rate of 0.18/day, a value only 20% higher than was seen in the shake flask cultivation trial. However, the observed maximum cell dry weight in the shake flask, 10.5g/L, was 14% higher than was achieved in the bioreactor. Ninety seven percentage confidence was achieved via the fitting of three unstructured growth models; the Monod, Logistic, and Gompertz equations, to the cell growth data. The Monod equation adequately described cell growth in both cultures. The specific growth rate, however, was not effectively predicted with the Logistic and Gompertz equations, which resulted in deviations of up to 73 and 393%, respectively. These deviations in the Logistic and Gompertz models may be attributable to the fact that these models were developed for substrate-independent growth and fungi growth, respectively.

  • The gellan was extracellular polysaccharide produced by Pseudomonas elodea A TCC 31461 at aerobic condition. Gellan provides various functionalities such as gelling, suspending, stabilizing, emulsifying and binding properties in aqueous systems. In this study, the effect of pH on the cell growth and the gellan production were evaluated in shake- flasks and in 5 ${\ell}$ batch fermentor. In the shake-flasks culture, maximum gellan production was obtained with 1.66g/ ${\ell}$ when initial pH was 7.0. The batch fermentation was performed in the medium pH control ranged pH 5.5-8.5. The maximum gellan production of 1.97g/l was obtained with constant pH 6.0.
