Continuous Production of Citric Acid from Dairy Wastewater Using Immobilized Aspergillus niger ATCC 9142

  • The continuous production of citric acid from dairy wastewater was investigated using calcium-alginate immobilized Asrergillus niger ATCC 9142. The citric acid productivity and yield were strongly affected by the culture conditions. The optimal pH, temperature, and dilution rate were 3.0, 30$^{\circ}C$, and 0.025 h$\^$-1/, respectively. Under optimal culture conditions, the maximum productivity, concentration, and yield of citric acid produced by the calcium-alginate immobilized Aspergillus niger were 160 mg L$\^$-1/ h$\^$-1/, 4.5 g/L, and 70.3%, respectively, The culture was continuously perfored for 20 days without any apparent loss in citric acid productivity. Conversely, under the same conditions with a batch shake-flask culture, the maximum productivity, citric acid concentration, and yield were only 63.3 mg L$\^$-1/h$\^$-1/, 4.7 g/L and 51.4%, respectively, Therefore, the results suggest that the bioreactor used in this study could be potentially used for continuous citric acid production from dairy wastewater by applying calcium-alginate immobilized Aspergillus niger.

  • Aspergillus terreus NRRL 1960에 의한 이다콘산의 생산에 관해 연구하였다. pH, 접종량, 배지조성 등의 최적조건을 확립하였다. 이타콘산의 최대생산 량은 37°C, pH 2.5의 조건에셔, 5% (w/v) glu­c cose, 0.5 % (w/v) $NH_4Cl$, 0.2 % (w/v) yeast ex­tract, 0.1 % (w/v) $MgSO_4$, 0.2 % (w/v) NaCI을 포함하는 배지에서 7일만에 $19.18g/\ell$를 얻을 수 있었다. 종균배양 배지로서 2 % malt extract가 적합하였다. 교반식 반응기에서 유리균체에 의한 이타콘 산의 회분식 생산은 플라스크 배양에 비해 비효율적 이었다.

  • 석탄중의 무기황화합물인 pyrite를 생물학적으로 제거하기 위하여 호산성, 독립영양성인 철산화 세균 7 Thiobacillus ferrooxidans를 샤용하였다. 호주산 역 청탄을 대상으로, 석탄 업자의 크기, 농도, 계면활성 제의 영향 등의 공정변수들이 탈황속도에 미치는 영 향을 조사하였고, airlift 생물반응기에셔 탈황특성을 조사하였다. 플라스크 배양의 경우, 탈황률은 슬러리 농도 증가에 따라 감소하였고. 20 % 이하의 슬러리 농도에서 78 % 이상의 탈황이 가능하였지만 슬러리 농도가 30% 이상이 되연 탈황률은 40% 미만으로 저하하였다. 또한, 석딴 업자의 크기가 증가함에 따 라 탈황률은 급격하게 감소하였는데. 0.42mrn이하의 업자크기에서 초기 탈황속도 169rng-SjR day가 얻 어졌다. 슬러리 농도가 50 %로 고농도인 경우, 플라 스크 실험에셔는 15 %의 낮은 탈황률이 얻어진 반 면, 물질전탈 효율이 높은 airlift 생물반응기에서는 탈황률이 50 %로 상대적으로 높은 값을 얻을 수 있 었다. 동일조건하에서 플라스크 실험에서는 계면활 성제의 첨가로 탈황률이 약 10% 정도 향상되었으나, a airlift 생물반응기에서는 탈황률이 약간 감소하였다. 이러한 결과로부터 계면활성제의 역할은 석탄 입자 의 침전을 방지하여 훈합을 용이하게 하고 산소와 탄소원인 이산화탄소의 물질전달속도를 향상시키는 것임을 알 수 있었다. 무연탄이 70% (w/v)의 고농 도에서도 80 % 이상의 탈황률과 비교해서 역청단의 탈황률은 매우 낮았다. 이는 역 청탄 중에는 미생물 의 활성을 저해하는 물질이 포함되어 있으며, 석탄 의 생물학적 탈황은 석탄의 종류에 많은 영향을 받 음을 시사하였다. 318K, 압력 5500psig에서 24.(1 의 이산화탄소를 사용했을때 효율은 유기용매 추출의 약 36%를 얻었고, methan이을 보조용애로 사용하 여 온도 308K, 압력 3000psig에 서 $24\ell$를 사용했을 때 약 81 %를 보였다. 하고 있음을 시사해 주고 있다.only either 'front and back' or 'left and right' and also sa-pok(사폭) of man's trousers seop(섶) of jeo-go-ri(short jacket 저고리) kyut-ma-gi(곁마기) of Sam-hwai-jang jeo-go-ri (삼회장 저고리), mu(무) of jig-ryung trouser wide and ga-rae-ba-dae(가래바대) of dan-cok-kok The formation line of Korean clothes is based on theprinciple of unlimited m bius strips by twisting of turning direction from universal principle and original basic form is not changd even by turning in-side out. Kinetics of Kojic Acid Fermentation by Aspergillus flavus Link S44-1 Using Sucrose as a Carbon Source under Different pH Conditions

  • Kojic acid production by Aspergillus flavus strain S44-1 using sucrose as a carbon source was carried out in a 250-mL shake flask and a 2-L stirred tank fermenter. For comparison, production of kojic acid using glucose, fructose and its mixture was also carried out. Kojic acid production in shake flask fermentation was 25.8 g/L using glucose as the sole carbon source, 23.6 g/L with sucrose, and 6.4 g/L from fructose. Reduced kojic acid production (13.5 g/L) was observed when a combination of glucose and fructose was used as a carbon source. The highest production of kojic acid (40.2 g/L) was obtained from 150 g/L sucrose in a 2 L fermenter, while the lowest kojic acid production (10.3 g/L) was seen in fermentation using fructose as the sole carbon source. The experimental data from batch fermentation and resuspended cell system was analysed in order to form the basis for a kinetic model of the process. An unstructured model based on logistic and Luedeking-Piret equations was found suitable to describe the growth, substrate consumption, and efficiency of kojic acid production by A. flavus in batch fermentation using sucrose. From this model, it was found that kojic acid production by A. flavus was not a growth-associated process. Fermentation without pH control (from an initial culture pH of 3.0) showed higher kojic acid production than single-phase pH-controlled fermentation (pH 2.5, 2.75, and 3.0).

  • 1. Cassava의 전분을 이용하여 검정곰팡이(Aspergillus niger)를 $30^{\circ}C$에서 4일 동안 진탕배양 하였을 때 배지 중 Cassava의 전분의 함량이 4%, 6%, 8% 및 10%일 때 96시간 배양된 각각의 균체량은 13.35mg/ml, 15.78mg/ml, 14.48mg/ml 및 9.93mg/ml이었다. 2. Cassava의 전분을 이용하여 검정곰팡이를 $30^{\circ}C$에서 4일 동안 진탕배양하여 얻어진 균체를 Lowry 방법에 의하여 그 균체단백을 측정한 바 배지 중의 Cassava의 전분의 함량이 4%, 6%, 8% 및 10%일 때 각각의 균체단백은 2.6mg/ml, 2.7mg/ml, 2.5mg/ml 및 1.7mg/ml이었으며 6% 함유구에서 균체단백의 양이 가장 많았다. 3. 배지 중의 Cassava의 전분량을 6%로 맞추고 질소원의 종류를 $NaNO_3,\;(NH_4)_2SO_4$ 및 Urea를 사용하고 질소량을 Czapeck-Dox 배지에 있는 $NaNO_3$의 질소량의 50%, 100%, 200%, 400% 및 $800^{\circ}C$되게 변화시키면서 진탕배양한 바 균체생산량에는 차이가 없었다.

  • 고정화된 Aspenillus niger 세포가 우유공장폐수에서 발효로 구연산을 생산하는데 이용되었다. 구연산 생산균주로서 A. niger ATCC 9142가 3일간 전배양되었으며, 약 2.5∼3.5 mm Ca-alginate beads 로 포획되었다. 최적의 pH와 온도는 각각 3.0과 $30^{\circ}C$였다. 발효에 대한 최적 희석률은 $0.025h^{-1}$ 인 것으로 계산되었다. 구연산의 최대 생산량은 최적화된 발효조건과 함께 4.5 g/$\ell$에서 얻어졌다. 고정화된 Aspergillus niger ATCC 9142에 의해 생산된 구연산의 수율은 70.3%였다. Shake-flask배양실험에 비해 연속식 배양실험 결과가 고정화 세포에 의해 20%증가되었다. 따라서 우유공장폐수는 구연산 발효기질로서의 이용가치가 있다고 볼 수가 있겠다.

Effect of Cultural Conditions on Polysaccharide Production and its Physicochemical Properties in Cordyceps militaris

  • This study was concerned with the optimization of liquid culture conditions for mycelial growth and polysaccharide production and its physicochemical properties in Cordyceps militaris. The one factor at a time method was adopted to investigate the effects of medium composition, environmental factors and C/N ratio. Among the these varialbles, glucose 80g/L, yeast extract 10g/L, $MgSO_4{\cdot}7H_{2}O\;0.5g/L$, $KH_{2}PO_4\;0.5g/L$ were proved to be the most suitable carbon, nitrogen, and mineral sources, respectively. The optimal temperature, initial pH, working volume were identified to be $24^{\circ}C$, 7.0 and 100ml, respectively. Under the optimal conditions, the strategies in shake flask culture and 5L jar fermentor led to mycelial growth of 29.43 g/L, 28.88g/L and polysaccharide production of 2.53g/L, 6.38 g/L, respectively. Among the phisicochemical properties, relative concentrations(w/v) of total sugar, uronic acid, protein and hexoseamine were identified to be 74.07%, 1.13%, 0.91%, and 0.46%, respectively. The fraction of neutral and acidic polysaccharide were identified to be 81.9% and 18.1%, respectively.

Production of Salicylic Acid from Naphthalene by Immobilized Pseudomonas sp. Strain NGK1

  • The Pseudomonas sp. strain NGK1 (NCIM 5120) was immobilized in calcium alginate, agar, and polyacrylamide gel matrices. The salicylic acid-producing capacity of freely suspended cells was compared with immobilized cells in batches with a shake culture and continuous culture system in a packed bed reactor. Freely suspended cells ($4\times10^{10}cfu/ml$) produced 12 mM of salicylic acid, whereas cells immobilized in calcium alginate ($1.8\times10^{11}$cfu/g beads), agar ($1.8\times10^{11}$cfu/g beads), and polyacrylamide ($1.6\times10^{11}$cfu/g beads) produced 15, 11, and 16mM of salicylic acid, respectively, from naphthalene at an initial concentration of 25 mM. The continuous production of salicylic acid from naphthalene was investigated in a continuous packed bed reactor with two different cell populations. The longevity of the salicylic acid-producing activity of the immobilized cells from naphthalene was also studied in semi continuous fermentations. The immobilized cells could be reused 18, 13, and more than 20 times without losing salicylic acid-producing activity in calcium alginate-,agar-, and polyacrylamide-entrapped cells, respectively. The study reveals a more efficient utilization of naphthalene and salicylic acid production by the immobilized Pseudomonas sp. strain NGK1 as compared to the free cells.

Production of Cellulase and Xylanase by Aspergillus niger KKS

  • A fungal strain capable of producing extracellular cellulase was isolated from farmland. It was identified as Aspergillus niger, and named Aspergillus niger KKS. Production of cellulase and xylanase by the A. niger KKS was studied through a shake-flask culture. The effects of culture conditions such as inoculum size, temperature, pH, and medium composition on the cellulase and xylanase production were examined. The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and pH 7.0, respectively. The optimized medium was composed of 2.0% (w/v) rice straw, 0.5% (w/v) proteose peptone, 0.5% (w/v) $KH_2 PO_4$, 0.05% (w/v) yeast extract, 0.01% (w/v) $CoSO_4 \cdot 7H_2O$, and 0.05% (w/v) $CuSO$_4$\cdot 5H_2O$. When the strain was incubated with the optimized medium, it gave the activities of endoglucanase, $\beta$-glucosidase, $\beta$-xylosidase, xylanase were 3.80, 4.20, 4.00, 80.0 (IU/mL), respectively. Filter paper and cotton activities were 0.68 and 0.045 (IU/mL), respectively. The results of this study show that A. niger KKS is a potential organism with a wide spectrum of enzyme activities, such as those of $\beta$-glucosidase, $\beta$-xylosidase, and xylanase.

Effect of pH on the elaboration of pullulan and the production of high molecular weight pullulan by Aureobasidium pullulans.

  • 불완전 곰팡이의 일종인 Aureobasidium pullulans ATCC 42023 균주를 이용한 pullulan의 최적조건 및 pH 조절에 따른 pullulan의 생성 그리고 균체형태와 고분자 pullulan 생산에 미치는 영향을 조사 하였다. 삼각플라스크 내에서 최대의 pullulan 생산은 초기 pH 6.5일때 11.98 g/l의 pullulan을 생산하였고, 회분식 발효에서는 산소 공급속도 0.5 vvm일때 15.16 g/l의 최대의 pullulan생산량을 나타내었다. pH조절에 따른 실험에서는 pH 4.5일때 13.31 g/l의 pullulan 생산량을 보였고, 이때 균체의 형태는 효모형과 균사형태의 성장을 하였다. 그러나 분자량 생성면에서는 pH 6.5일때 100만 이상의 고분자가 가장 많이 생산되었다.
