The Effect of Extrusion Treatment on Aqueous Ammonia Soaking Method in Miscanthus Biomass Pretreatment (억새 바이오매스 전처리에서 압출 처리가 액상 암모니아 침지 처리에 미치는 영향)

  • Pretreatment of cellulosic biomass is necessary before enzymatic saccharification and fermentation. Extrusion is a well established process in food industries and it can be used as a physicochemical treatment method for cellulosic biomass. Aqueous ammonia soaking treatment at mild temperatures ranging from 60 to $80^{\circ}C$ for longer reaction times has been used to preserve most of the cellulose and hemicellulose in the biomass. The objective of this study was to evaluate the effect of extrusion treatment on aqueous ammonia soaking method. Extrusion was performed with miscanthus sample conditioned to 2mm of particle size and 20% of moisture content at $200^{\circ}C$ of barrel temperature and 175rpm of screw speed. And then aqueous ammonia soaking was performed with 15%(w/w) ammonia solution at $60^{\circ}C$ for 1, 2, 4, 8, 12 hours on the extruded and raw miscanthus samples respectively. In the combined extrusion-soaking treatment, most compositions removal occurred within 1~2 hours and on a basis of 1 hour soaking treatment values, cellulose was recovered about 85% and other compositions, including hemicellulose, are removed about 50% from extruded miscanthus sample. The combined extrusion-soaking treated and soaking only treated samples were subjected to enzymatic hydrolysis using cellulase and ${\beta}$-glucosidase. The enzymatic digestibility value of combined extrusion-2 hours soaking treated sample was comparable to 12 hours soaking only treated sample. It means that extrusion treatment can shorten the conventional long reaction time of aqueous ammonia soaking. The findings suggest that the combination of extrusion and soaking is a promising pretreatment method to solve both problems for no lignin removal of extrusion and long reaction time of aqueous ammonia soaking.

Sensory and Mechanical Characteristics of Daechu Injeolmi by Various Soaking time of glutinous rice (수침시간을 달리한 대추인절미의 텍스처 특성)

  • This study was intended to investigate the optimal soaking time of glutinous rice for making jujube Injeolmi, a kind of traditional Korean glutinous rice cake. The jujube Injeolmi was prepared with glutinous rice soaked for various times (2, 4, 6, 8, 10 hr), and the moisture content, chromaticity, sensory qualities and mechanical qualities were measured while storing them up to 72 hr at 20$^{\circ}C$. The moisture content and L-value of jujube Injeolmi were the highest at 8-hr soaking. The a-value was the highest at 6-hr soaking. The hardness was maintained the lowest at the 8-hr soaked sample during storage. The cohesiveness and springiness were not significantly affected by the soaking time. The gumminess was significantly different among the samples during storage. The results of sensory evaluation showed that jujube Injeolmi had the best color at 4-hr soaking and the highest sweetness at 6-hr soaking. The 8-hr soaked glutinous rice gave the highest level of softness, chewiness and moistness in the sample, therefore, 8-hr soaking appeared to be the most desirable method for making jujube Ingeolmi.

  • To examine the difference of castability of non -precious porcelain metal according to 2 type of investing method, experiments were carried out using the method of both existing investing method and the investment with spaces to the upper & lower parts of the ring. The following conclusion were obtained from the result. 1. Seperataly using the existing investment and with spaces to the upper & lower parts of the ring, 44 samples resulted in success with the former method and 52 with the latter out of 84 samples. 2. Under the existing investing method, 44 samples out of 56 total resulted in success when the temperature of the ring was $870^{\circ}C$, and all samples failed when the temperature was $800^{\circ}C$. 3. Under the investment that gives space to the ring, when casted with the temperature of the ring fixed at $870^{\circ}C$, 40 samples out of 56 and 17 samples with the temperature fixed at $800^{\circ}C$ resulted in success. 4. Under the existing investing method with the temperaure of the ring fixed at $870^{\circ}C$ and heat soaking time given an hour, 26 out of 28 samples resulted in success and 18 resulted the same with half an hour heat soaking time. 5. Using the investment that gives space to the ring, at ring temperature $870^{\circ}C$ with heat soaking time 1 hour, all of total 28 rusulted insuccess however only 12 smaples succeeded with 30 minutes heat soaking time. 6. Under the existing investing method, samples with heat soaking time 1 hour equally given at ring temperature $870^{\circ}C$, 26 samples out of 28 succeed however at $800^{\circ}C$ all samples failed. 7. In the case of ring's upper and lower parts opened ring temperature is $870^{\circ}C$ and $800^{\circ}C$ and the heat soaking time was fixed at 1 hour, all 28 samples resulted in success and at $800^{\circ}C$ 17 succeed.

Combinational Effect of Moist Heating and Gamma Irradiation on The Inactivation of Trypsin Inhibitory Activity in Soybean

  • The combinational effect of gamma irradiation and moist heating on the trypsin inhibitor activity (TIA) in soaked and dried soybeans was evaluated by measuring the inhibition using N-benzoyl-DL-arginine-p-nitroanilide (BAPNA) as substrate. Gamma irradiation significantly decreased the TIA level in soybean at doses above 5 kGy, and the $ID_{50}$ (the gamma irradiation dose required to reach 50% inhibition) value for TIA was 13.53 kGy. Soaking prior to gamma irradiation significantly lowered the $ID_{50}$ to 8.44 kGy, and the soaking process enhanced the efficiency to inactivate TIA by as much as 48%. When soaking prior to gamma irradiation was followed by subsequent mild heating ($60^{\circ}C$) process, the $IT_{50}$ (heating time required to reach the 50% inhibition of TIA) value at even 1 kGy (5.28 min) was greatly reduced by over 50% compared to the level for the no-soaking process. In addition, the activation energy of soaking prior to gamma irradiation at 1 kGy was 2.45 kcal/mole, which was also about 50% lower than the 5.10 kcal/mole of dried soybean gamma-irradiated. Based on these results, soaking prior to gamma irradiation is an effective method for TIA inhibition. Furthermore, a combination of two or more processing methods such as soaking, heating and gamma irradiation is much more effective than any single processing method.

  • In this study, the "Multi Well Plate-type cell Apparatus" was designed and setup for performing the producing experiments of methane hydrate by depressurization, heat stimulating methods. In order to characterizing the producing mechanism of hydrate through porous materials, the experiments for various producing methods have been conducted with the aid of the apparatus which has high permeability. In the experimental result of depressurization method, the pressure is temporarily increased unlikely conventional gas reservoir due to the sourcing effect of hydrate dissociation in the pore. Meanwhile, the temperature is decreased because of the endothermic reaction while hydrate is dissociated. In the experimental results of heat stimulating method, the dissociation in depressurization method is more slowly processed than that in thermal method, and hence, its gas production is lower. In the case of production right after heating, hydrate is dissociated only near injecting point and the permeability becomes greater at that area only. It infers that the more gas is produced during relatively earlier producing period. Since then, the hydrate is more slowly dissociated than the case of production after heating and soaking. This time, the performances of pressure and production obtained by thermal method have been analyzed in order to investigate the effect of soaking time on gas recovery. As a result, the gas recoveries in the case of 2 min and 4 min soaking are higher than case in 6 min soaking. This is reason that hydrate is reformed due to the decrease of temperature. It is expected that the experimental results obtained in this work may be more clearly explained by utilizing the lower permeable porous system with the greater hydrate saturation.

  • Effects of soaking time on the physicochemical and structural characteristics of waxy rice flours and starches purified from flours using the alkaline steeping method were investigated. Korean cultivar Sinseonchal waxy rice was washed and soaked in tap water (1:2 w/w) and stored at room temperature for 15 days. On each day of soaking for 0, 1,2 3, 5, 10, and 15 days, pH of soaking water was measured and rice grains were dried, ground, and passed through 100 mesh sieve. The pH was reduced to 3.90 by day 5 and increased to 4.60 by day 15. The protein and ash contents, swelling powers and solubilities of flours and starches decreased with increasing soaking time. The water-binding capacities increased while trends were not similar to soaking time. The flour particle size distribution ranged from two to four peaks with increasing soaking times. Starch granule size decreased with increasing soaking time. The peak, trough, and final viscosities of flours and starches showed similar trends until 10 days and 15 days, respectively. The starches presented higher viscosities than the flours. The branch chain length distributions of amylopectin of starches showed an increaseed DP6~12 portion and decreased DP13~24 portion with increasing soaking time of waxy rice grains.

Effects of Germinating Types of Water-soaking & Air-exposure and Chitosan Treatment on Brown Rice Germination (침수 및 대기 발아방식과 키토산 처리가 현미발아에 미치는 영향)

  • The effects of germinating types and treatments of acetic acid, water-soluble and water-insoluble chitosan on the germination ratio and sprout length of brown rice were investigated in this study. The treatment of 0.001% acetic acid improved the germination ratio and sprout growth of brown rice in the germinating types of a air-exposure method after water-soaking (Type II) and a repetitive method of water-soaking & air-exposure (Type III), not water-soaking method (Type I). The treatment of water-soluble chitosan with higher concentration caused higher germination ratio and faster sprout growth. The treatment of water-insoluble chitosan repressed the germination and the sprout growth of brown rice. The germination ratios of brown rice germinated by the Type III were higher than those by Type I and Type II for all the treatments of acetic acid and water-soluble & water-insoluble chitosan as more than 97% germination ratio. Also, the Type III method accelerated the sprout growth of brown rice compared with Type I and Type II.

  • In this study, gas hydrate production has been followed using swapping method to investigate the effect of injection rate of flue gas and soaking period in unconsolidated artificial sand sample. The results shows that recovery factor of methane gas decreases with increasing the injection rate of flue gas. This indicates that the velocity of flue gas in porous media may act as kinds of inhibitor for production of hydrate. Also recovery factor increases with increasing the soaking time.

  • Cobalt ferrite thin films on Corming glass substrate were fabricated by a sol-gel method. Cobalt ferrite thin films with the grain size of 20-35 nm and thickness of 50nm were obtained. Rapid thermal annealing (RTA) and Annealing processes were adopted for comparison of characteristics of the films. Coercivity values were changed with thermal condition and magnetization values were increased as a function of soaking time. With prolonged soaking time, however, it was decreased because of the diffusion of cations from the glass substrate. The RTA process in preparation of cobalt ferrite thin film was the effective way to prevent and to form a single spinel phase in reduced soaking time. The film heated at 600$^{\circ}C$ for 30 minutes by RTA had coercivity of 2,600 Oe, saturation magnetization 460 emu/㎤, and Mr$.$$\delta$ of 1.43 memu/$\textrm{cm}^2$.

Effect of Natural Fiber Surface Treatments on the Interfacial and Mechanical Properties of Henequen/Polypropylene Biocomposites

  • The surfaces of henequen fibers, which can be obtained from the leaves of agave plants, were treated with two different media, tap water and sodium hydroxide, that underwent both soaking and ultrasonic methods for the fiber surface treatment. Various biocomposites were fabricated with untreated and treated, chopped henequen fibers and polypropylene using a compression molding method. The result is discussed in terms of interfacial shear strength, flexural properties, dynamic mechanical properties, and fracture surface observations of the biocomposites. The soaking (static method) and ultrasonic (dynamic method) treatments with tap water and sodium hydroxide at different concentrations and treatment times significantly influenced the interfacial, flexural and dynamic mechanical properties of henequen/polypropylene biocomposites. The alkali treatment was more effective than the water treatment in improving the interfacial and mechanical properties of randomly oriented, chopped henequen/PP bio-composites. In addition, the application of the ultrasonic method to each treatment was relatively more effective in increasing the properties than the soaking method, depending on the treatment medium and condition. The greatest improvement in the properties studied was achieved by ultrasonic alkalization of natural fibers, which was in agreement with the other results of interfacial shear strength, flexural strength and modulus, storage modulus, and fracture surfaces.
