The Early Detection of the Protein Toxin using Sanification and Fluorescent Dye in the Field (현장에서 초음파 파쇄와 형광시약을 이용한 단백질 독소의 조기 탐지)

  • This study was carried out to establish the optimum disruption condition of a sonificator for the protein toxin for the purpose of developing automatic biological agent detector equipped a sonificator. One of the best-known collisional quenchers is molecular oxygen, which quenches almost all known fluorophores. The sonification does an excellent job of degassing, which decreased the quenching effect and increased the fluorescence quantity. The fluorescence measurement for the protein using 0.7 X fluorescent dye concentration and above must be done in 1 minute and the fluorescence measurement for the protein using 0.3 X fluorescent dye concentration and below has to be done between 2 and 3 minute. The fluorescence quantity of the sonificatied protein sample was much higher that of the non-sonificatied protein sample. Sonificating the sample turned out to be favorable for the fluorescence measurement when measuring at the low protein concentration.

  • 소니피케이션은 다양한 데이터를 사운드로 변환시키는 과정으로 본 연구에서는 뇌파 신호를 사운드로 생성하는 객관적인 워크플로우를 제시하고자 하였다. 현재까지의 뇌파 소니피케이션은 사운드로의 변환이 인위적이고 임의적으로 진행되어 객관적인 논리를 제시하지 못하고 있는 실정이다. 이에 본 연구에서는 뇌파데이터의 정량적 분석을 통해 파라미터 추출, 사운드 맵핑, 사운드 모델 구축에 대한 논리적 근거를 제시하였으며 이를 통해 뇌파데이터의 객관적인 소니피케이션 과정을 구현하였다. 파라미터 추출을 위해 15Hz High pass filtering이 가장 적절한 방법으로 확인되었으며 뇌파 데이터의 최대값 빈도 분석과 음악코드의 비율 분석을 실제로 맵핑시켜 사운드 모델을 구축하여 사운드 생성을 구현하였다. 결론적으로, 본 연구에서는 뇌파데이터의 소니피케이션 과정에 대한 객관적이고 논리적인 워크플로우를 제시하였으며 이러한 워크플로우가 다양한 분야에서 적용될 수 있을 것으로 예상된다.

  • This study was carried out to establish the optimum condition for cell disruption with a sonificator in the detection of the gram negative bacteria, E. coli for the purpose of developing automatic fluorometer. The efficiency of sonification on the E. coli disruption was greatly dependent on the diameter of sonificator probe tip. The larger sonificator probe diameter showed greater disruption effect. Sonificator probe of 13 mm diameter was the most efficient one for E. coli when sonificated for 20 seconds. The efficiency of the E. coli disruption differed greatly according to the depth of sonificator probe tip sank in the sample solution. The shorter the distance between probe tip end and the bottom of the container, the higher the disruption efficiency. The detection limit of E. coli was $5{\times}10^5CFU/m{\ell}$ when sample was sonificated for 20 seconds with a sonificator probe of 13 mm diameter.

  • This study was carried out to establish the optimum condition for cell disruption with a sonificator in the detection of the gram positive bacteria, Bacillus globigii and Streptococcus epidermidis for the purpose of developing automatic fluorometer. The efficiency of sonificator on the Bacillus globigii and Streptococcus epidermidis disruption differed greatly according to the diameter of sonificator probe tip. The larger sonificator probe diameter showed greater disruption. Bacillus globigii was more disruptive than Streptococcus epidermidis. Sonificator probe of the 13 mm diameter was the most efficient one when sample was sonificated for 20 seconds. The detection limits of Bacillus globigii and Streptococcus epidermidis were $10^5CFU/m{\ell}\;and\;5{\times}10^5CFU/m{\ell}$ respectively when samples were sonificated for 20 seconds with a sonificator probe of 13 mm diameter.

  • The actively soil pollution by the toxic heavy-metals like the arsenic, cadmium, lead due to the industrialization and economic activity. The uses the electrokinetic remediation of contaminated soil has many researches against the fine soil having a small size in the on going. However, it is the actual condition which the research result that is not effective due to the low surface charge of the particle and high permeability shows in the electrokinetic remediation in comparison with the fine soil in the case of the sandy soil in which the particle size is large. In this research, the electrokinetic remediation and ultrasonic wave fetch strategy is compound applied against the sandy soil polluted by the arsenic, cadmium, and lead removal efficiency of the sandy soil through the comparison with the existing electrokinetic remediation tries to be evaluated. First of all, desorption of contaminants in soil by ultrasonic extraction in the Pre-Test conducted to see desorption effective 5~15%. After that, By conducted Batch-Test results frequency output century 200 Khz, reaction time 30 min, contaminated soil used in experiment was 500 g. Removal efficiency of arsenic, cadmium, lead are 25.55%, 8.01%, 34.90%. But, As, Cd, Pb remediation efficiency less than 1% in EK1(control group).

  • There is such a kind of characteristic by using the computer digitization technology's interaction. It not only provides the authoring tool, and also has the user-friendly window. The paper based on this kind of interaction, through the sound and the color exchange of information, as well as each information's digitization, carries on about the scientific dielectric related advance research. This research carries on the analysis to the people to the color and the sound perceptual characteristic first, then makes comparison to the people regarding the color and the music perceptual cognition.. The research is about the research stage of the interactive sound installation creation, which shape is carried on the analysis through the extraction image in color. It is based on the perceptual characteristic,is that the algorithm manufacture reasearch of how to make color sonification. Through capturing the non-stop changing colors by camera,it transforms colors into music. The paper's foundation is the fundamental algorithm manufacture of experimental music composition.

Studies of Morphological Properties and Pyrrolizidine Alkaloids Analysis of Comfrey Cultivating in Korea (국내산 컴프리의 형태학적 특성 및 Pyrrolizidine Alkaloids 분석)

  • This study was carried out to investigate the morphological properties and pyrrolizidine alkaloids of comfrey cultivating in Korea. 12 comfrey samples cultivation in Korea was selected and their appearance (whole plant, leaves, root etc.) were observed by expert and microscopy for morphological analysis. It is confirmed that their species are Symphytum officinale Linnaeus. Samples were extracted by hot MeOH and ultra-sonification. Their extracts contained pyrrolizidine alkaloids, which was identified by TLC analysis. By spraying thin-layer chromatograms of pyrrolizidine alkaloids stable purple spots were developed. But the extracts of chicory, pumpkin and sesame leaves did not show any purple spots. Same HPLC pattern were displayed at about 30 min of retention show peaks an one and the same time.

  • This paper reports the apparatus and method for the safe treatment of pentachlorophenol(PCP)-treated ammunition box by solvent extraction. Experimental variables were chosen as the composition of solvents, types of substance(chips and sawdust), temperatures and sonication to obtain maximum PCP removal from wood samples of the dismantled ammunition box. Up to 99% of PCP in the wood chip was extracted within 2 hours at room temperature when using methanol as the solvent. The extraction volume ratio of methanol per dried sample was about 10. Type of samples, extraction temperature and sonification showed little effects on PCP extraction. Based on this study, a resource recycling system for the treatment of ammunition boxes was recommended.

  • Polyaniline-Camphorsulfonic Acid Emeraldine Salt(PANI-CSA ES) was prepared by doping Polyaniline Ermeralidine Base(PANI EB) with DL-10-Camphorsulfonic Acid(CSA). PANI-CSA ES was solved in an organic solvent by ultrasonification for different periods of time and its surface resistivity was measured. Several PANI-CSA ES solutions solved in different organic solvents were prepared and their surface resistivities were measured. Thermal stability of film casted with PANI-CAS ES solution in m-cresol was estimated by measuring its surface conductivity and the content of this moisture and organic solvents. PANI-CSA ES was blended with different polymeric binders to improve its physical properties and the surface resistivities of several kinds of PANI-CSA ES blends were measured as a function of the content of PANI-CSA ES. PANI-CSA ES polymerized by 1-step oxidative polymerization was prepared and its surface resistivity was measured.
